Abstract:In this work, compound 1 has been rationally designed and synthesized as a new fluorescent probe for biological thiols. Notably, probe 1 has almost no background fluorescence (Phi(f) < 0.0001) in aqueous solutions; however, it exhibited fluorescence turn-on response to thiols with high sensitivity (a 246-fold fluorescence enhancement and a low detection limit of 0.22 mu M for Cys). Moreover, probe 1 showed excellent thiol specificity over other biologically relevant species. The kinetic studies indicated that the probe responded to thiols rapidly, and the pseudo-first-order rate constants of probe 1 reaction with Cys, Hcy, and GSH were determined to be 1.85842, 0.67656, and 0.51519 min(-1), respectively. A possible detection mechanism was proposed to involve the Michael addition of the thiol to the alpha,beta-unsaturated ketone, followed by a cleavage of the hemiketal group, thereby leading to the formation of a fluorescent 7-hydroxyl coumarin derivative. Furthermore, the optical responses of probe 1 to thiols were studied by TD-DFT calculations. Finally, probe 1 has been successfully applied to the detection of biological thiols in human blood serum. And the intracellular imaging applications established that probe 1 can be used to detect different concentrations of intracellular thiols in living cells.

Abstract:Two probes, structurally symmetric CBFB and asymmetric CBFM, constructed by a D–A–D (donor–acceptor–donor) type curcuminoid as the fluorophore and the DNBS (2,4-dinitrobenzenesulfonyl) group as the biothiol recognition site were designed and synthesized here. The DNBS group can quench the emission of the fluorophore by the PET (photoinduced electron transfer) process, and in the presence of biothiols, the emission of the probe was switched on as a result of the cleavage of the quencher by a nucleophilic aromatic substitution reaction. Experimental analyses and theoretical calculations revealed that two recognition moieties in the molecule can quench the fluorescence more efficiently, therefore, CBFB showed a much higher SNR (signal to noise ratio) than CBFM in biothiol detection with an emission maximum at 610 nm. This “low background” and “turn-on” fluorescent probe, CBFB, was successfully utilized to map endogenous biothiols in living cells.

Abstract:A new turn-on fluorescent probe for the selective detection of thiol over other amino acids was synthesized. Probe possesses the widely-used thiol-selective 2,4-dinitrobenzenesulfonyl (DNBS) group which can react with thiol and release the fluorescein which has strong fluorescence. Fluorescein, a well known xanthene fluorescent dye, has two states at different environment. Fluorescein is in the state of spirocyclization when connected with 2,4-dinitrobenzenesulfonyl (DNBS) group which has no fluorescence. However, it is in the state of open form when it reacts with thiol which has a strong fluorescence. The transition of the two states can be used to selectively detect thiol and the color can change from colorless to yellow which can be differentiated by naked eyes. Upon the titration of thiol, the absorption band at 454nm rises gradually and the fluorescence emerges at 521nm and the detection limit can be as low as 0.16μM. All of such good properties prove it to be a good sensor for the selective detection of thiol and it shows a potential use in bioimaging applications.

Abstract:A new ratiometric fluorescent probe is designed and prepared based on the concept of supramolecular encapsulation and dye competition. This supramolecular probe is based on two commercially-available dyes, one common guest and a simple-to-synthesize host. Fluorescence spectroscopy confirms that the supramolecular probe is capable of detecting thiols quantitatively with a broad linear region in phosphate buffered saline or fetal bovine serum. Mechanistic study shows a reaction between thiol specie and the guest to alter the distribution of encapsulated dyes. The supramolecular probes are demonstrated to quantitatively detect extracellular biological thiols by plate reader, which shows it keeps its effectiveness in complex buffered systems.

Abstract:In recent years, targeting drugs made by physical loading or chemical bonding of drugs on small molecular carriers have shown a very wide application prospect in the field of tumor and cancer treatment. How to achieve the release of drugs in cancer cells has become the core of this research. One of the most important bases for drug localization is to use the difference of small molecular biothiol concentration between cancer cells and normal cells. Details of the changes of biothiol levels in the growth and reproduction of cancer cells are still poorly understood, and the main reason is the lack of sensitive real-time imaging tools for biothiols in cancer cells. In this work, we reasonably designed and synthesized the combination of 4-hydroxy-1,8-naphthalimide and NBD-Cl as a concise fluorescent probe HN-NBD for imaging biothiols in live cells and zebrafish. In addition, due to the advantages of HN-NBD design, it is sufficiently sensitive to biothiols, and further imaging can distinguish cancer cells from normal cells. Probe HN-NBD would be of great significance to biomedical researchers for the study of biothiol-related diseases, the screening of new anticancer drugs, and the early diagnosis and treatment of cancers.

Abstract:Biological thiols, such as glutathione, cysteine, and homocysteine, are closely associated with human health and disease. In this work, we demonstrated a general strategy for the design of fluorescent probes for the detection of various biological related thiols. The preferred strategy we employed was to introduce a 3,5-dinitropyridyl moiety to a selected fluorophore via ether linkage to afford fluorescent probe. Once the ether bond was thiolyzed by biothiol, the hydroxy-functionalized fluorescent dye would be released and fluorescence emission restored. Herein, some novel fluorescent probes based on this strategy have been developed for sensing biothiols in living cells. All these probes presented here showed desirable properties, such as high sensitivity, high selectivity and preferred reaction kinetics. Moreover, the probes have been used for imaging of intracellular biothiols with emission colors ranging from green to near-infrared (NIR).

Abstract:Developing fluorescent probes for specific detection of extremely toxic thiophenols is pretty significant in the field of environment, chemistry and biology. We report herein a turn-on red fluorescent xanthene-based probe (RD-Probe) for detecting thiophenol with high selectivity over other analytes including aliphatic thiols. The probe could play the part of a “naked-eye”colorimetric indicator toward thiophenol. Moreover, the relative fluorescence intensity at 653 nm displayed good linearity with the concentration of thiophenol ranging from 0 to 6 μM, and the limit of detection for thiophenol could be as low as 15 nM. Furthermore, the practicability of RD-Probe has been successfully proved through the quantitative thiophenol detection in real water samples and fluorescence bioimaging of thiophenol in HeLa cells.

Abstract:A new green-emitting fluorescent probe 1 was developed for biothiol detection. The sensing mechanism was considered to be biothiol-induced cleavage of the 2,4-dinitrobenzene- sulfonate group in probe 1 and resulting inhibition of the probe's photoinduced electron transfer (PET) process. Probe 1 exhibited favorable properties such as excellent selectivity, highly sensitive (0.12 mu M), large Stokes shift (117 nm) and a remarkable turn-on fluorescence signal (148-fold). Furthermore, confocal fluorescence imaging indicated that probe 1 was membrane-permeable and suitable for visualization of biothiols in living A549 cells.

Abstract:Bio-thiols such as cysteine (Cys), homocysteine (Hcy), and glutathione (GSH) play a crucial role in physiological processes. Distinguishing between Cys, Hcy, and GSH remains a challenge because of their similar structure and reactivity. Herein, we conceptualized and synthesized a near-infrared (NIR) fluorescent probe TAI based on acridine-conjugated indole salts. This innovative probe demonstrated effectiveness in sensitively distinguishing Cys, Hcy and GSH by NIR dual-channel and time-dependence fluorescence. Probe displays a significant turn-on NIR fluorescent response toward Cys at 654nm under 488nm excitation with Large Strokes shift (about 166nm) and high selectivity, but also shows turn-on NIR fluorescent response for Hcy/GSH at 695nm under 640nm excitation. More interestingly, the probe also exhibited an NIR fluorescence response at 654nm to Hcy and no fluorescence response to GSH when the response time is over 3h. The sensing mechanism was fully confirmed by NMR, HRMS spectra and DFT theoretical calculations. Moreover, successful intracellular imaging experiments in living cells suggest that the probe is suitable for discrimination imaging endogenous and exogenous bio-thiols. This work offers a promising near-infrared fluorescent imaging tool for the differential detection of bio-thiols.

It shows turn-on properties with red emission towards biothiols in acetonitrile-PBS buffer (1 : 9, v/v) solution, and its properties of rapid response, high selectivity and high sensitivity make it a potential probe with real applications. Moreover, the in vitro assays show that the probe can be used in fluorescence imaging for the detection of intracellular biothiol levels.

Abstract:In this study, a new near-infrared (NIR) fluorescent turn-on probe featuring a large Stokes shift (198 nm) was developed for the detection of biothiols. The probe was based on a dicyanoisophorone derivative serving as the fluorophore and a 2,4-dinitrobenzenesulfonyl (DNBS) group functioning as both a recognition site and a fluorescence quencher. In the absence of biothiols, the fluorescence of the probe was low due to the photoinduced electron transfer (PET) effect between the fluorophore and DNBS. Upon the presence of biothiols, the DNBS group underwent a nucleophilic aromatic substitution reaction with the sulfhydryl group of biothiols, leading to the release of the fluorophore and a notable emission peak at 668 nm. This developed probe exhibited exceptional selectivity and sensitivity to biothiols in solution, with an impressive detection limit of 28 nM for cysteine (Cys), 22 nM for homocysteine (Hcy), and 24 nM for glutathione (GSH). Furthermore, the probe demonstrated its applicability by successfully visualizing both endogenous and exogenous biothiols in living systems.

Abstract:A novel photoconvertible fluorescent probe, which can be activated by intracellular thiols, has been synthesized. Such a molecular probe comprises three parts: a 7-aminocoumarin phototrigger, a thiol-removable energy acceptor, and a caged fluorescein scaffold with intracellular thiols reactivity as the fluorescent reporter. Extracellularly, the energy acceptor blocks the emission of the coumarin that regulates the photocleavage and photoactivation of the fluorescein. Intracelluarly, the high concentration of thiols releases the energy acceptor, thus activating the S1 state of the phototrigger, which emits coumarin blue fluorescence for pre-visualization and liberates the caged green-fluorescent fluorescein to highlight the specific cell upon illumination. Compared to traditional photoactivated organic dyes, the intracellular thiols activated probe requires double activations: one by intracellular thiols and the other by light activation. The dual activations restrict fluorescence precisely inside live cells and at the particular spatial region of light activation, thus a probe with precise spatial accuracy in live cells.

Abstract:In this work, a dicyanoisophorone-based fluorophore, which showed an attractive near-infrared fluorescence as well as a favorable large stokes shift, was used to develop a novel probe (NIR-Probe 1) for effective detection of biothiols. It displayed distinct color changes and a turn-on NIR fluorescence response with a high off/on ratio toward biothiols. We demonstrated that NIR-Probe 1 exhibited fast sensing speed, high selectivity for biothiols and possessed the function of detecting low concentration of biothiols with impressive limit of detection: 12.1 nM for Cys, 14.5 nM for Hcy and 11.9 nM for GSH. Abstract:Development of multi-input fluorescent probes would facilitate the monitoring of multiple biomolecular events leading to a common disease pathology. Here, we reported a two input fluorescent probe(QME-N-3) based on quinoline scaffold with two distinct reactive sites, the alpha,beta-unsaturated carbonyl at 2 site and 6-azido group, which were specific acceptors of RSH and H2S respectively. The turn-on fluorescent probe could react with the two molecules via the Michael addition at the double bond for RSH and the reduction of the azide by H2S without mutual interference, to generate the intensively fluorescent product, which could act as a two-input AND logic gate. Furthermore, QME-N-3 has very low cytotoxicity and excellent stability, and the resulting sensing product has a high two-photon absorption cross section. Consequently, detection of intracellular thiols and H2S can be achieved by cell fluorescence imaging under both one- and two-photon excitations.

Abstract:Bio-thiols play an important role in living organisms. Both elevated levels and deficiency of bio-thiols will induce some diseases, such as cardiovascular disease and Alzheimer's diseases, skin lesions, and weakness. Therefore, fast and accurate detection of bio-thiols is of great significance. We have synthesized a ratiometric probe that can respond to bio-thiols with the fluorescence intensity at 568 nm (F-5(68)) increasing and taht at 420 nm (F-420) decreasing. The responding process reaches equilibration in 1 min in the present of Cys or Hcy while in about 50 min with GSH. This probe can further detect bio-thiols in human breast cancer cells (MDA-MB-231) by confocal laser scanning microscope. Fluorescent ratio (R-Y/B) of yellow channel with blue channel increase as reacting with bio-thiols in cells. Ratiometric probe SH-1 can realize the fast detecting of bio-thiols with R-Y/B as detection signal both in vitro and in living cells.

Abstract:Photochromic molecules can achieve reversible isomerization upon alternate light irradiations, which offers a great opportunity to improve the precision of analytes detection and imaging in complicated biological environments. Previous reported photochromic probe exhibited only mono-color switching and an initially fluorescence-ON state that may cause high background signal and impose an adverse impact on the desired sensing precision. To overcome this set-back, we developed a novel photochromic probe with an analyte-activation mode for ratiometric sensing of toxic thiols in both real water samples and live cells. The dynamic dual-fluorescence signal is released only after the fast and selective cleavage of the 2,4-dinitrophenyl sulfonate by the targeted thiophenol derivatives. Consequently, a “double-check” with synchronized dual-fluorescence blinking for analyte detection is successfully employed upon alternate light triggers with rapid response ( k =7.2×10 −2 s −1 ), high sensitivity (LOD=6.1 nM) as well as selectivity of thiophenol derivatives over other common thiol species (e.g., GSH, Cys and Hcy). The photochromic probe was successfully introduced to the fast and on-site detection of highly toxic thiophenols in real waste water samples. Moreover, by using confocal laser-scanning microscopy (CLSM) and flow cytometric analysis, the potential applications of this ratiometric photochromic probe for trace toxic thiol sensing in live cells are examined.
