Abstract:Chromatography using polyamide column is one of the most critical purification operations that affect the safety and efficacy of traditional Chinese medicine (TCM) products. To ensure successful operation and reduce time and solvent consumption, UV spectroscopy combined with multivariate data analysis as an online analytical tool was developed for monitoring the polyamide column chromatography of the TCM Danshen. The process trajectories were established by principal component analysis of the UV spectra and used to determine the endpoint of the washing stage and investigate the impacts of the process conditions. The online analysis method developed determined the concentration of salvianolic acid B (an important compound in Danshen) in the effluent rapidly and precisely with a coefficient of determination of 0.9963 and helped to collect salvianolic acid B quantitatively for determining the endpoint of elution. The methodology proposed is an effective approach applicable in guiding successful operations in the chromatographic separation.
RP-HPLC Analysis of the Enantiomers of Esmolol and Its Metabolite in Human Blood with Solid-Phase Extraction

Abstract:To develop a method for simultaneous determination of the enantiomers of esmolol and its metabolite in human whole blood.Methods:The blood samples were pretreated by solid-phase extraction.Esmolol and its metabolite enantiomers were separated on a Aglient Zorbax C_(18) co]utah(4.6 mm×250 mm,5μm)after chiral derivatization with 2,3,4,6-tetra-O-acetyl-β-D-glucopyranosyl isothiocyanate (GITC) employed as a pre-column chiral derivatization reagent.The mobile phase was acetonitrile-0.01 mol·L~(-1) potassium dihydrogen phosphate buffer (pH4.5)(55:45) with the flow rate of 0.80 mL·min~(-1).The detection wavelength was 248 nm. Results:The assay was linear from 0.05 to 10.0μg·mL~(-1) for each enantiomer of esmolol and 0.03 to 10.0μg· mL~(-1)for each enantiomer of metabolite.The absolute recoveries for each enantiomer were greater than 75%.The in- tra-day and inter-day variations were less than 15%.Conclusion:The method is accurate,sensitive,reliable and suitable for simultaneous determination of the enantiomers of esmolol and its metabolite in human blood.
Determination of High Molecular Mass Polycyclic Aromatic in Natural Vitamin E by High-Performance Liquid Chromatography with Fluorescence Detection

Abstract:A method has been developed for the determination of eight high molecular mass polycyclic aromatic hydrocarbons(PAHs)in natural Vitamin E(VE)products by reversed-phase high-performance liquid chromatography(HPLC)with fluorescence detection.PAHs were enriched and purified by an alumina column chromatogram,and separated on XDB-C18 column using acetonitrile-water(20∶80,v/v)as mobile phase.The optimum conditions of alumina column chromatogram were studied.The limits of detection of eight high molecular mass PAHs were between 0.05~0.45ng/g and the limits of quantification were between 0.18~1.50ng/g.The recoveries of spiked standard were between 80.8%~104.0%.Relative standard deviations of heavy PAHs were between 2.0%~9.8%.The method was applied to determination eight high molecular mass PAHs in three different kinds of natural Vitamin E products.good repeatabilities were achieved,and RSD were less than 12.0%.

Abstract:A Vis-NIR (visible-near infrared) spectroscopy based method for the end-point determination of the column chromatographic processes of Ginkgo biloba was established. In-line collected Vis-NIR spectra were used in conjunction with various multivariate data analysis methods, including moving block standard deviation (MBSD), principal component scores distance analysis (PC-SDA), and principal component analysis-moving block standard deviation (PCA-MBSD) for the end-point determination of the elution process. Compared with the results validated by high performance liquid chromatography (HPLC) determination, PCA-MBSD was chosen as the most suitable method. The presented method provided an alternative solution to the end-point of the column chromatographic processes, which in the long term depends upon the workers' operating experience traditionally.

Abstract:Objective:In a chromatographic cycle,the adsorption process is a critical unit operation that has a significant impact on downstream processes and,ultimately,the quality of the final products.The development of a rapid method to determine the endpoints of adsorption processes in a large-scale manufacturing is of substantial importance for herbal medicine(HM)manufacturers.Methods:In this study,the adsorption of saponins on a macroporous resin column chromatograph,a critical unit operation in Panax notoginseng(Burkill)F.H.Chen injection manufacturing,was considered as an example.The evaluation results of in-line ultraviolet and visible spectra combined with various multivariate analysis methods,including the moving block standard deviation(MBSD),difference between the moving block average and the target spectrum(DMBA-TS),soft-independent modeling of class analogy(SIMCA),and partial least-squares discriminant analysis(PLS-DA),were compared.Results:MBSD was unsuitable for adsorption processes.The relative standard errors of prediction between the predicted and experimental endpoints were 13.2％,4.67％,and 5.71％using DMBA-TS,SIMCA,and PLS-DA,respectively.Conclusions:Among the considered analysis methods,SIMCA and PLS-DA were more effective for endpoint determination.The results of this study provide a more comprehensive overview of the effectiveness of various multivariate analysis methods to facilitate the selection of the most suitable method.This study was also conducive to address the issues of the in-line detection of adsorption endpoints to guide practical HM manufacturing.

Abstract:Objective:To establish a new method of assaying rapidly of active components in steviol glycoside solution. Method: The near infrared transmittance spectroscopy was used to quantify the different batches of steviol glycoside. According to selected different preprocessions and wave bands for information extraction and analysis from spectrums, the best factors and wave bands for quantitative analysis were determined. The calibration models were established using the PLS (partial least squares). Result: The correlation coefficients(R2) for steviolbioside(SX), dulcoside A(DA), stevioside (STV), rebaudioside C(RC), Rebaudioside A(A3) and steviosides(STS) were 0.9271, 0.9355, 0.9759, 0.9706, 0.9500 and 0.9870, and the RMSEP were 0.0004, 0.0015, 0.0150, 0.0043, 0.0170 and 0.0199, respectively. Conclusion: The proposed method was simple and fast, and it provides a novel efficient and environmental friendly approach for the fast determination of the active components in steviol glycoside.

Abstract:An in-line monitoring method for the elution process of Ginkgo biloba L. leaves using visible and near-infrared spectroscopy in conjunction with multivariate statistical process control (MSPC) was established. Experiments, including normal operating batches and abnormal ones, were designed and carried out. The MSPC model for the elution process was developed and validated. The abnormalities were detected successfully by the control charts of principal component scores, Hotelling T2, or DModX (distance to the model). The results suggested that the established method can be used for the in-line monitoring and batch-to-batch consistency evaluation of the elution process.

Abstract:Objective:To establish a method for simultaneous determination of Gastrodin and p-2-hydroxybenzyl methanol in biotransformation system.Method:RP-HPLC was used in the study,the mobile phase consists of 0.02% phosphoric acid solution (pH 2.6) and methanol (5:95,V/V),the flow rate was 0.5 mL/min,all chromatographic operations were carried out at the temperature of 35 ℃ with UV detector at 220 nm.Results:Under the above condition,good linearity was obtained for both Gastrodin and p-2-hydroxybenzyl alcohol with a correlation coefficient 0.9999,recoveries ranged between 95% and 102%,while the precision of the method was <5% (RSD).Conclusion:The content of gastrodin and p-2-hydroxybenzyl alcohol in biotransformation system can be accurately determine simultaneously by this method.

Abstract:A speedy and selective ultra-HPLC-MS/MS method for simultaneous determination of deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-ADON), 15-ADON. nivalenol and fusarenon X in traditional Chinese medicines (TCMs) was developed. The method was based on one-step sample cleanup using reliable homemade cleanup cartridges. A linear gradient mobile-phase system, consisting of water containing 0.2% aqueous ammonia and acetonitrile/methanol (90:10, v/v) at a flow rate of 0.4 mL/min, and an Acquity UPLC HSS T3 column (100 mm x 2.1 mm, 1.8 mu m) were employed to obtain the best resolution of the target analytes. [C-13(15)]-DON was used as the internal standard to accomplish as accurate as possible quantitation. The established method was further validated by determining the linearity (R-2 >= 0.9990), sensitivity (LOQ, 0.29-0.99 mu g/kg), recovery (88.5-119.5%) and precision (RSD <= 15.8%). It was shown to be a suitable method for simultaneous determination of DON, 3-ADON, 15-ADON, nivalenol and fusarenon X in various TCM matrices. The utility and practical impact of the method was demonstrated using different TCM samples.

Abstract:Herbal medicines have played a vital role in maintaining the health of the world population in the past thousands of years, and have proved to be an effective therapy. It is important to improve our understanding of the effects of the multi-step processing in herbal medicines on the chemical changes to ensure product quality. A proton nuclear paramagnetic resonance (1H NMR)-based evaluation strategy was developed for an efficient process variation exploration and diversified metabolite identification. In this study, 48 process intermediates from 6 commercial batches of the multi-step manufacturing chain of Danshen processing were obtained. Hierarchical classification analysis (HCA) tree based on 1H NMR spectra clustered the samples according to the processing steps, which indicates that 1H NMR has the potential capability for critical control point identification based on its adequate information of the organic compounds. Then, principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA) were applied to distinguish the major metabolite differences between the intermediates before and after the critical control point. In this case, the alkali-isolation and acid-dissolution method was recognized as the most critical process in the multi-step chain of Danshen extract manufacturing. Potential metabolites with the larger amplitude of variation and contributing the most to the discrimination were found to be potential quality markers by S-plot, including several previously undetected amino acids. The results in this study are consistent with previous research studies and reference experiments conducted with other analytical tools. Taken together, they prove that 1H NMR with chemometrics is a very effective process quality control tool to provide comprehensive information on the chemical changes during the processing of herbal medicines, and help with the identification of critical control points and potential critical quality markers.

Abstract:OBJECTIVES To investigate the feasibility of using near-infrared spectroscopy for rapid determination of main organic acids in Ginkgo biloba leaf extract (EGBL). METHODS Main organic acids in EGBL were assayed using the HPLC method. Critical factors of the chromatographic separation were optimized by a novel analytical quality by design approach. Partial least squares-discriminant analysis (PLS-DA) was performed to screen the marker components, and principal component analysis (PCA) was utilized to distinguish the different samples. Then, spectral quantification potential was investigated using PLS and support vector machine (SVM) approaches. For modelling, different spectral preprocessing and wavelength selection methods were systematically compared. KEY FINDINGS It was found that quinic acid, protocatechuic acid and 6-hydroxykynurenic acid were identified as possible index components. PLS-DA based on contents and PCA based on near-infrared spectra can both effectively distinguish the different EGBL samples. The calibration models with wonderful prediction performance can be both developed by the PLS and SVM algorithms. CONCLUSIONS NIR spectroscopy combined with chemometrics can realize the rapid and non-destructive qualitative and quantitative analysis of EGBL. The proposed method may be applied to quality control of EGBL and other natural products in commercial use.

Abstract:Background Quality control of the herbal medicines (HMs) still hits technical bottlenecks due to their complicated multi-step pharmaceutical processes. Nuclear magnetic resonance (NMR) spectroscopy could serve as a powerful technique able to obtain comprehensive information of chemical compounds in the manufacturing process of HMs by a single analytical test. In this study, ethanol precipitation of Salvia miltiorrhiza Radix et Rhizoma (Danshen in China), a critical purification process of a particularly commonly used herbal medicinal material, was employed as the research subject. Methods Processed with different variables, including ethanol-material ratio, apparent ethanol concentration and moisture concentration of the raw material, 40 lab-scale experiments of ethanol precipitation were orchestrated by a full factor design with additional central composite circumscribed experiments. After 1H NMR tests, chemometrics methods, including orthogonal partial least squares-discriminant analysis (OPLS-DA), principle component analysis (PCA) and multiple linear regression (MLR), were applied to analyze the spectra of samples. Results 16 metabolites were recognized as the most critical potential chemical markers in the process of Danshen ethanol precipitation by 1H NMR spectra and OPLS-DA, including γ-aminobutyrate, pyroglutamate, succinate, acetate, malonate, fructose, sucrose, raffinose, stachyose, mannotriose, danshensu, lithospermic acid, protocatechuic aldehyde, rosmarinic acid, salvianolic acid A and B. PCA reasonably distinguished all the process intermediates as a function of different process variables adopted during the ethanol precipitation. MLR made the statistical interpretation by quadratic equations to characterize how do significant variables affect the chemical markers. Conclusions Our study indicates that NMR can track detailed potential chemical markers of ethanol precipitation process, which could promote the further understanding of ethanol precipitation process and open up possibilities to develop a more effective process quality control and consistency evaluation methodology of an HM, through providing a more in-depth and improved theoretical process understanding and characterization.
Simultaneous Determination of Xanthohumol,Isoxanthohumol and 8-Prenylnaringenin in Hop using RP-HPLC

Abstract:In order to achieve simultaneous determination of xanthohumol,isoxanthohumol and 8-prenylnaringenin in hop,a method based on RP-HPLC was developed in this study.An ODS C18 column(250 mm×4.6 mm,4 μm) with temperature 30 ℃ was used.Acetonitrile-1% formic acid solution were used as mobile phase at a flow rate of 1.0 mL/min with a nonlinear gradient elution program.The detection wavelengths were controlled at 370 nm and 288 nm.Results showed that under the optimized chromatographic condition,good linearity was obtained for all of these three components with correlation coefficients exceeding 0.998,recoveries ranged between 93% and 106%,while the precision was <1%.The evaluation parameters indicate that this method can accurately determine the content of xanthohumol,isoxanthohumol and 8-prenylnaringenin in hop.In addition,the effect of different extraction solvents was investigated using the developed method and some middle-polar solvents had been found having good extraction performance with high content of these three components.
In situ derivatization-ultrasound-assisted dispersive liquid-liquid microextraction for the determination of neurotransmitters in Parkinson's rat brain microdialysates by ultra high performance liquid chromatography-tandem mass spectrometry

Abstract:Simultaneous monitoring of several neurotransmitters (NTs) linked to Parkinson's disease (PD) has important scientific significance for PD related pathology, pharmacology and drug screening. A new simple, fast and sensitive analytical method, based on in situ derivatization-ultrasound-assisted dispersive liquid-liquid microextraction (in situ DUADLLME) in a single step, has been proposed for the quantitative determination of catecholamines and their biosynthesis precursors and metabolites in rat brain microdialysates. The method involved the rapid injection of the mixture of low toxic bromobenzene (extractant) and acetonitrile (dispersant), which containing commercial Lissamine rhodamine B sulfonyl chloride (LRSC) as derivatization reagent, into the aqueous phase of sample and buffer, and the following in situ DUADLLME procedure. After centrifugation, 50μL of the sedimented phase (bromobenzene) was directly injected for ultra high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) detection in multiple reaction monitoring (MRM) mode. This interesting combination brought the advantages of speediness, simpleness, low matrix effects and high sensitivity in an effective way. Parameters of in situ DUADLLME and UHPLC-MS/MS conditions were all optimized in detail. The optimum conditions of in situ DUADLLME were found to be 30μL of microdialysates, 150μL of acetonitrile containing LRSC, 50μL of bromobenzene and 800μL of NaHCO3-Na2CO3 buffer (pH 10.5) for 3.0min at 37°C. Under the optimized conditions, good linearity was observed with LODs (S/N>3) and LOQs (S/N>10) of LRSC derivatized-NTs in the range of 0.002-0.004 and 0.007-0.015 nmol/L, respectively. It also brought good precision (3.2-12.8%, peak area CVs%), accuracy (94.2-108.6%), recovery (94.5-105.5%) and stability (3.8-8.1%, peak area CVs%) results. Moreover, LRSC derivatization significantly improved chromatographic resolution and MS detection sensitivity of NTs when compared with the reported studies through the introduction of a permanent charged moiety from LRSC into NTs. Taken together, this in situ DUADLLME method was successfully applied for the simultaneous determination of six NTs in biological samples.
Determination of polyphenolic compounds in raw plant materials by NMR spectroscopy

Abstract:Environmental monitoring and assessment of the prospects for extracting biologically active substances (BAS) from various types of plant biomass requires the development of simple and fast methods for measuring their content in raw materials. For this purposes massspectrometry and NMR spectroscopy are widely used. Both methods have their own advantages and disadvantages. But NMR spectroscopy is a nondestructive method and requires simple sample preparation. In addition, quantitative studies using NMR spectroscopy do not require authentic standard samples. We have developed various techniques for targeted metabolomic analysis of plant origin samples using NMR spectroscopy, including: determination of anthraquinone derivatives in extracts from roots and rhizomes of Rubia tinctorum L., determination of diosmin in Hyssopus officinalis L., determination glucofrangulin A in extracts from leaves of Rhamnus frangula L.
Simultaneous Determination of Four Kinds of Alkanolamines in Desulfurization Solution by Ion Chromatography

Abstract:A new ion chromatographic method for simultaneous separation and determination of isopropanolamine (IPA), triisopropanolamine (TIPA), methyldiethanolamine (MDEA), Dimethylethanolamine(DMAE) and six kinds of cations was developed.The optimal chromatographic conditions were as following: a TSK gelSuper IC-CR cation exchange column, the mixture of 2.0 mmol/L 18-Crown-6, 2.2 mmol/L Methanesulfonic acid and 0.5 mmol/L L-Histidine as eluent, a flow rate of 0.5 mL/min, a suppressed conductivity detection, and the injection volume of 30 μL.The limit of detection for DMAE, MDEA, TIPA, IPA was 0.48, 36.9, 47.0 and 1.39 μg/L, respectively.The average recovery for DMAE, MDEA, TIPA, IPA was 103.15%,95.48%,97.64%,92.41% , respectively.The method has the advantages of simplicity, fast speed, high sensitivity and good anti-interference capability.It has been applied for the determination of DMAE, MDEA, TIPA, and IPA in the desulfurization solution with satisfactory results.

Abstract:A novel and highly sensitive metastable state nanoparticle-enhanced Raman spectroscopy (MSNERS) was reported in this work, which employed an amphiphilic polymer polyurethane-Ag nanoparticle (AgNPs) as the MSNERS substrate. Polyurethane could form micelle to incorporate nanoparticles and analytes, where targets could be in close contact with the metal surface, which was effective for further enhancing the detection sensitivity. Time-dependent visible absorption spectra and time-dependent Raman spectra indicated that polyurethane could greatly enhance the stability of AgNPs aggregates during the volatilization process, which was a great improvement of MSNERS substrate. Combining this MSNERS substrate with the rapid separation method of thin layer chromatography (TLC), this TLC-MSNERS was successfully applied to analyze mixed pesticides on fruit skin and the detection limits of thiabendazole, triazophos and phosmet were 0.02 mu g/mL, 0.8 mu g/mL, and 0.6 mu g/mL, respectively. It enhanced 1 order of magnitude the signals of analytes in comparison to that of traditional TLC-SERS method.

Abstract:Ion mobility spectrometry(IMS) is difficult to identify some organic compounds with closed reduced mobility values,such as the isomeric compounds of xylene.A tandem analytical technique,UV photoionization ion mobility spectrometry(UV-IMS) coupled with a packed column gas chromatography (GC),GC-UV-IMS,was developed for the efficient separation and detection of aromatics compounds.The gas chromatograms and ion mobility spectra were measured at different column temperatures and the flows of nitrogen carrier gas in order to obtain optimized experimental parameters.The gaseous mixture of benzene,toluene,o-xylene,m-xylene and p-xylene was detected with a homemade GC-UV-IMS apparatus. The results show that three isomeric compounds o-xylene,m-xylene and p-xylene can not be distinguished when only using the UV photoionization IMS.However,two dimensional gas chromatogram-ion mobility spectrum can greatly enhance resolution of these organic compounds on the basis of their retention time and drift time.This demonstrates that the established GC-UV-IMS has a good ability to discriminate and detect trace volatile organic compounds.

Abstract:Use of liquid chromatography/tandem mass spectrometric (LC/MS(n)) molecular fingerprinting is systematically demonstrated as a very effective tool for rapid structural elucidation of pharmaceutical impurities through a case study in which three isomers of betamethasone sodium phosphate (BSP) were rapidly identified as degradants formed due to the D-homoannular ring expansion of the steroid core structure of BSP in the solid state. The rapid structural elucidation of these degradants was achieved by matching or closely matching the UV profiles, molecular weights, and more importantly the fragmentation patterns obtained from the LC/MS(n) (n = 1 to 3) analysis of their enzyme-catalyzed hydrolytic products, respectively, with those of a D-homoannular isomer of betamethasone available in our LC/MS(n) molecular fingerprint database. This strategy of using LC/MS(n) molecular fingerprinting to obtain high-confidence structures of unknown species is then validated by structure verification through one- (1D) and two-dimensional (2D) nuclear magnetic resonance (NMR) experiments.

Abstract:Qualitative and quantitative analysis of photosynthetic glycolipids mixture was Studied by means of the ultra performance liquid chromatography-quadrupole-time of flight mass spectrometry(UPLC-Q-TOF-MS) using electron spraying ionization. The results showed that the ionization mode was depending on the structural types, namely, monoglycosyldiglycerides (MGDG) and diglycosyldiglycerides(DGDG) in positive ion mode, whereas sulfoquinovosyl diacylglycerol (SQDG) in negative ion mode. Each glycolipid component in the mixture can be separated effectively using gradient elution on BEH C-18 column. The characteristic fragment ions were shown in the. mass spectra indicating the polar group and acyl chains, the relative intensity of the two acyl chains were clearly correlated with their regiostereochemistry. The quantitation of glycolipid has been carried Out by accounting their composition in isocratic elution.
