Abstract:Two ratiometric probes fluoride ion, Mito-F and Lyso-F, were rationally designed and synthesized with positive charges at physiological conditions. The positive charges functioned as target moieties for subcellular mitochondria and lysosome of living cells, and effective sequesters of fluoride ion for fast and efficient fluorescent detection. In addition, in vivo imaging of fluoride ion in living mice was successfully achieved for the first time using probe Mito-F.

Abstract:We have synthesized four indole derivatives (INS1, IND1, INS2 and IND2) successfully, and experimental results of biochemical RNA fluorescence assays and direct intracellular imaging demonstrated that they are potential highly RNA-selective fluorescent turn-on probes possessing good counterstain compatibility with the classic DNA-staining dye DAPI. Moreover, these sensors exhibit large two-photon excited fluorescence action absorption cross-section in the presence of RNA and are also capable of imaging intracellular RNA in two-photon fluorescence microscopy.

Abstract:Fluoride ions (F-) are one of the essential trace elements for the human body and are widely existed in nature. In this study, we present a novel fluorescent probe (YF-SZ-F) designed and synthesized for the specific detection of F-. The probe exhibits high sensitivity, excellent selectivity, and low cytotoxicity, making it a promising tool for biomedical applications. Imaging experiments conducted on zebrafish and Arabidopsis roots demonstrate the probe's remarkable cellular permeability and biocompatibility, laying a solid foundation for its potential biomedical utility. Abstract:Hydroxyl radicals (•OH) is one of the commonly reactive oxygen species available in biosystem and plays a key role in a number of physiological and pathological processes. Based on our previous study, fluoride could serve as a moderate electron donating group in constructing fluorescence probes. However, there is seldom work on the effects of fluoride-substituted moiety towards the photophysical properties in constructing fluorescence probes. In this work, we developed a series of novel fluorescence probes based on a quinolinium-scaffold containing fluoride substituted donor. From our results, the probe FHyR-3 which has an extended conjugated system could detect hydroxyl radical with high selectivity, sensitivity, and fast response. Furthermore, the probe has the maximum emission at 508 nm with a large stoke shift (114 nm) and exhibits low toxicity, good biocompatibility and mito-targeting. We also demonstrated its capability in tracking hydroxyl radical level in living cells and zebrafish.

Abstract:This work presents the design and synthesis of a new fluorescent probe IF-Br-F for the specific detection of fluoride ions. The IF-Br-F probe has excellent fluorescence properties, and the mechanism of the probe response to fluoride ions was successfully verified via HRMS and DFT calculations. IF-Br-F has high sensitivity and low detection limit (5.82 × 10-7 mol L-1) and successfully detects fluoride ions in actual water samples. The probe can be applied to fluorescence imaging of zebrafish, cells, and Arabidopsis roots and exhibits low cytotoxicity and good biocompatibility.

Abstract:Fluoride ions have the important roles in a lot of physiological activities related with biological and medical system, such as water fluoridation, caries treatment, and bone disease treatment. Great efforts have been made to develop new methods and strategies for F(-) detection in the past decades. Traditional methods for the detection of F(-) including ion chromatography, ion-selective electrodes, and spectroscopic techniques have the limitations in the biomedicine research. The fluorescent probes for F(-) are very promising that overcome some drawbacks of traditional fluoride detection methods. These probes exhibit high selectivity, high sensitivity as well as quick response to the detection of fluoride anions. The review commences with a brief description of photophysical mechanisms for fluorescent probes for fluoride, including photo induced electron transfer (PET), intramolecular charge transfer (ICT), fluorescence resonance energy transfer (FRET), and excited-state intramolecular proton transfer (ESIPT). Followed by a discussion about common dyes for fluorescent fluoride probes, such as anthracene, naphalimide, pyrene, BODIPY, fluorescein, rhodamine, resorufin, coumarin, cyanine, and near-infrared (NIR) dyes. We divide the fluorescent probes for fluoride in cellular application systems into nine groups, for example, type of hydrogen bonds, type of cleavage of Si-O bonds, type of Si-O bond cleavage and cylization reactions, etc. We also review the recent reported carriers in the delivery of fluorescent fluoride probes. Seventy-four typical fluorescent fluoride probes are listed and compared in detail, including quantum yield, reaction medium, excitation and emission wavelengths, linear detection range, selectivity for F(-), mechanism, and analytical applications. Finally, we discuss the future challenges of the application of fluorescent fluoride probes in cellular system and in vivo. We wish that more and more excellent fluorescent fluoride probes will be developed and applied in the biomedicine field in the future.

Abstract:Two polycyclic aromatic hydrocarbons bearing trimethylsilylethynyl groups were designed and synthesized as new colorimetric and ratiometric fluorescent probes for fluoride anion. Both probes exhibit high specific selectivity and sensitivity for F− compared to other anions. When F− is added to the solution of the probe, the trimethylsilyl substituents can be removed immediately, and the color of the solution changes from yellow to colorless under ambient light. Moreover, test papers based on the probes are prepared, which could be applied to detect F− in organic phase.

Abstract:In this study, two novel fluorescent probes MD1 and MD2 were designed and synthesized to detect BF3 based on natural and renewable isolongifolanone. Upon reaction with BF3, the probe MD1 exhibited a significant turn-on fluorescence enhancement. Different from MD1, the probe MD2 displayed an observable ratiometric fluorescence response to the presence of BF3. The sensing mechanisms were carefully verified by H-1 NMR, F-19 NMR, HRMS and density functional theory (DFT) studies. Meanwhile, two different molecular logic gates were rationally constructed based on the fluorescent sensing properties of the probes. Furthermore, these probes were successfully employed to prepare two test paper-based portable detecting devices for monitoring BF3 in the gas phase. All the studies and facts demonstrated that these smart probes have great potential in industrial applications.

Abstract:Fluorescent probes with a microsecond lifetime have attracted much attention in biological detection. The luminescence properties and responsive mechanisms of a probe [DCF-MPYM-lev-H]- for detecting sulfite and its corresponding product [DCF-MPYM-2H]2- are studied based on density functional theory (DFT) and time-dependent density functional theory (TD-DFT) calculations as well as the thermal vibration correlation function method. It is found that the luminescence efficiency of the probe increases obviously after reacting with sulfite, which is induced by increased radiative decay rates and decreased nonradiative rates. In addition, the thermally activated delayed fluorescence (TADF) properties of products are confirmed by analyzing the spin-orbital constants and energy gaps between the singlet excited states and the triplet excited states. The calculation results favor the understanding of the luminescence properties and responsive mechanism of a turn-on TADF probe for sulfite, which may provide a theoretical reference for the development of new TADF probes.

Abstract:A novel water soluble fluorescein-based fluorescent probe for superoxide detection was developed. The probe is fairly stable under neutral and acidic conditions. It can be used to detect superoxide both in solution with the detection limit of 2.2 μM and in living cells. Cell imaging experiments indicated that such a probe displayed good cell penetration and O2•- could be detected with PMA-stimulated HepG2 cells in both mitochondria and lysosome. Such a probe is the first dual mitochondria- and lysosome- targetable fluorescent chemodosimeter. Additionally, O2•- in intact live zebrafish embryos was successfully visualized under PMA-stimulated conditions and the possible detection mechanism was studied as well.

Abstract:In daily life, sulfite and bisulfite have been extensively used as antimicrobial agent and antioxidant for food products, beverages and pharmaceutical products, but, SO3 2−/HSO3 − are fantastically harmful to human health, so it is meaningful to develop an effective probe for these two anions. In this work, a new fluorescent material probe 1 was designed and constructed, which was able to sense sulfite and bisulfite through fluorescence “turn-on” manner. Compared with correlative fourteen other anions used, probe 1 showed great selectivity toward sulfite or bisulfite. Furthermore, the probe can detect SO3 2−/HSO3 − with fast response (within 4min) as well as low detection limit (8.8nM) compared with the previous work. We characterized the adduct through NMR and X-ray diffraction analysis and further confirmed the detection mechanism of probe 1. Finally, fluorescence imaging of MCF-7 cells indicated that 1 could be used for monitoring SO2 derivatives in living cells with low cytotoxicity.

Abstract:Currently, there is a great need to develop methods for the selective detection of fluoride anions (F(-) ) owing to their toxicity in the environment and biological function in living systems. In this study, we developed a new fluorescent probe (probe 1) employing a Si-O bond as a highly selective recognition receptor for detecting F(-) via intramolecular charge transfer. Probe 1 could detect F(-) quantitatively using the turn-on fluorescence spectroscopy method with excellent sensitivity in the range of 4-38 μM and a detection limit of 0.26 μM; the detection time was < 17 min. We anticipate that probe 1 would be used widely to monitor F(-) in the environment. Copyright © 2015 John Wiley & Sons, Ltd.

Abstract:A novel fluorescent probe was developed by reacting with fluorene core with azacyclobutane. The homemade portable device was exploited and applied for improving the practicability of the probe on site detection. This method provides a new idea for designing efficient fluorescent probes based on dipstick and mobile phone rapid detection. Fluorene nucleus derivatives show great potential for building outstanding fluorescence probes. In this paper, a novel fluorescent probe was developed by reacting with fluorene core with azacyclobutane, which exhibits typical solvation chromogenic effect in solvent. The fluorescence of the probe quenched in highly polar solvent. Based on this phenomenon, a novel fluorescence system for trace water was constructed. The response of this probe was fast (30 s) and sensitive for the detection of trace water in organic solvents, and the detection limit of water content in DMSO reached 0.13%. In addition, the probe can also be made as a test strip combined with homemade portable device and a smartphone for rapid detection of trace water. The luminescence mechanism of the probe is theoretically calculated based on time‐contained density functional theory (TDDFT). To showcase its practicality, it has been applied for the detection of trace water in honey and alcohol by dipstick. This method provides a new idea for designing efficient fluorescent probes based on dipstick and mobile phone rapid detection.

Abstract:A dual-channel fluoride anions probe has been synthesized from 4-hydroxy-1,8-Naphthalimide and t-butyldiphenylsilyl derivative. The probe presents selective coloration for fluoride anion that can be recognized by naked-eye. And it can quantitatively determine fluoride anion in aqueous and organic media. In MeCN/H2O (v/v, 50:50), the detection limit of the probe is found to be 0.35 mg L−1 which satisfied EPA's requirements. While density function theory and time-dependent density function theory calculations were conducted to rationalize the optical response of the probe. Furthermore, the biological application shows that it could be successfully employed as a selective fluoride probe in the fluorescence imaging of living cells.
