Abstract:We present a floating reference position (FRP)-based drift correction method for near-infrared (NIR) spectroscopy-based long-term blood glucose concentration (BGC) monitoring. Previously, we reported that it is difficult to quantify the systematic drift caused by the fluctuation of incident light intensity at different source–detector (SD) separations based on the absolute FRP change. We use the relative FRP change as a baseline reference to quantitatively characterize the signal drift at different SD separations. For the wavelengths that were used, a uniform equation was developed to describe the relationship between the drift and the relative FRP change. With the help of this equation, the correction can easily be performed by subtracting the systematic drift estimated by the equation. A theoretical analysis and an experimental phantom study demonstrated that our method could be used for systematic drift correction in NIR spectroscopy for long-term BGC monitoring. Moreover, the analysis method can also be referenced to reduce drifts from multiple sources.
Study on the Method of Combining Double-Beam Measurement with NAS Processing in Quantitative Analysis of Glucose Concentration

Abstract:In the field of noninvasive blood glucose sensing by near-infrared (NIR) spectroscopy, spectra are highly susceptible to the influence of background variations caused by the measuring instruments and physiological variations from the measured object because the concentration range of glucose in blood is usually small. It should be noted that the influence of background variations cannot be entirely removed; reasonable methods should be adopted in order to reduce the consequence of background variations to an acceptable level. One of the most common methods is to select a relatively stable standard material which shows similar optical property to the measured objects as the reference to perform the measurement. In order to maximize the elimination effect on the influence of background variations and realize a relatively accurate extraction of the glucose concentration information, a reference measurement method combined with double-beam spectra collection and net analyte signal (NAS) processing is proposed in this article. Spectra of the measured samples and reference substances are collected simultaneously with a double beam double-detector measuring system. The NASs of glucose are obtained by projecting every spectrum of measured samples on the noise background subspace spanned by the spectrum of the reference substance. Experiments are conducted in pure absorption and strong scattering medium respectively. Two-dimensional correlation spectroscopy (2DCOS) and partial least squares regression (PLSR) are adopted as data processing methods to test the effectiveness of the reference measurement method. Results of 2DCOS show that the specificity of glucose concentration information in samples can be improved to a large extent by NAS processing compared to the reference subtraction method. Meanwhile, the root-mean-square error of prediction (RMSEP) of PLS model predicting glucose concentration decreases by 35. 25% and 37. 95% respectively in the double-beam experiment of glucose aqueous solution and 20%-intralipid solution compared with those in the single-beam experiments. These two RMSEPs further decrease by 26. 11% and 14. 84% after combining NAS processing with the double-beam experiment data. These results prove that the method of combining double-beam measurement with NAS processing is effective in extracting the glucose information and improving the accuracy of the calibration model, which provides more possibilities for noninvasive blood glucose sensing.
Methodology of Effective Glucose-Specific Signal Extraction in Complicated Sample

Abstract:In the area of noninvasive human blood glucose concentration detecting, it has always been a critical task to extract the glucose-specific signal from the highly overlapped and disturbed near-infrared spectrum. In this paper, the methodology of effective glucose-specific signal extraction in complicated non-scattering sample is studied. By analyzing the impact of water displacement upon dissolution of glucose, the relationship between glucose concentration and absorption coefficient of the sample is deduced. Then, the reference wavelength where the absorption coefficient is insensitive to the changes of glucose concentration is put forward theoretically. Accordingly, the validating experiments in aqueous glucose solutions are executed. Both the theoretical and laboratorial results show that the reference wavelength of glucose appears at 1525nm. Based on the reference wavelength, an effective method for extracting the glucose-specific signal in complicated non-scattering samples is proposed and the corresponding validating experiments are constructed with different glucose and albumin concentration. Two different methods, traditional and the novel reference wavelength method are used to extract glucose signal and the corresponding root mean square error of prediction are 19.86mg/dl and 9.87mg/dl respectively. The experiment results indicate that the reference wavelength method can effectively eliminate the influence of various noises on the glucose-specific signal extraction, and thus can remarkably improve the measuring precision in noninvasive near-infrared glucose detecting.

Abstract:In order to improve the accuracy of near-infrared (NIR) spectroscopy measurement,the method with medium concentration sample as reference for relative measurement is proposed.Firstly,theoretical analysis of absorbance error sources with background reducted is presented,which shows that errors caused by sample cell and optical fiber loss can be reduced by using medium concentration sample as reference.Furthermore,NIR transmission experiments on glucose aqueous solution and 3％ intralipid solution as phantom are performed with glucose used as analyte.In the double-beam experiment on glucose aqueous solution,root mean square error of calibration (fRMSEC) and root mean square error of cross validation (fRMSECV) of the partial least squares (PLS) regression model using medium concentration sample as reference decrease respectively by 36.6％ and 41.0％ compared to the model with pure water as reference.Meanwhile,in the intralipid phantom experiment,fRMSEC andfRMSECV of the PLS regression model using medium concentration sample as reference decrease respectively by 66.3％ and 39.3％ compared to the model with glucose-free 3％ intralipid solution as reference.The results show that the accuracy of NIR transmission spectroscopy analysis can be effectively improved by using medium concentration sample as reference.
Applying the Reference-Wavelength Method to Improve the Precision of Glucose Measurement by Near Infrared Spectroscopy

Abstract:The reference-wavelength method is proposed to diminish the influence of noises on glucose measurement by differentially processing two signals at the reference and measuring wavelengths. At the reference wavelength, the radiation intensity is insensitive to the changes of glucose concentration. Therefore, it can be used as the internal reference to estimate the noise and then to extract the effective glucose signal at the other wavelengths. The validation experiments are constructed in the non-scattering samples with the reference wavelength of glucose at 1525 nm. The results show that the reference-wavelength-based glucose-specific signal extracting method can largely improve the glucose prediction precision from 17-56 to 8.87 mg/dL in the two-component experiment and from 26.82 to 9.94 mg/dL in the three-component experiment.
Data Correction Method Based on the Floating Reference Point for the Noninvasive Glucose Sensing

Abstract:As an effective noninvasive method for glucose doesn't come into clinical realization due to the weakness of glucose unique signal and complexity of background noise, a method based on a floating reference point and a measuring point, where the diffuse reflectance intensity is insensitive and most sensitive to the variation of glucose concentration, respectively, is applied. In this paper, the data processing method based on the information of reference point was investigated to improve the precision of glucose sensing. The diffuse reflectance of intralipid solution with different glucose concentration in different source-detector distances was obtained by Monte-Carlo simulation. And the radial region selection of reference position and measuring position were discussed. Then in order to simulate the actual measurement condition, the random noise and linear drift were added on the simulated spectra. And the spectra in the proper measuring region corrected by that in the reference point were used to build the multivariate model. Further more, the corresponding optical probe was designed according to the distribution of light intensity in the radial distance and an in vitro experiment about intralipid solution with different glucose concentration was conducted to verify the effect of the data correction based on the information from the reference point. Results showed that, three different measuring regions should be determined in the wavelength of 1100nm-1700nm according to the wavelength characteristic of reference point. And the measuring region should be about 0.2-0.3mm far away from the reference region. For the simulation and in vitro experiment, after the correction by the information from the reference point, the prediction error for glucose was reduced by 46.2% and 23.2%, respectively.
Preliminary Clinical Validation of a Differential Correction Method for Improving Measurement Accuracy in Noninvasive Measurement of Blood Glucose Using Near-Infrared Spectroscopy

Abstract:One of the main challenges in the noninvasive sensing of blood glucose by near-infrared (NIR) spectroscopy is the background variations from light source drift, sweating, and temperature change at the human–machine interface. In this paper, a differential correction method based on the spectra from the floating-reference position and measuring position is proposed to eliminate these spectral variations from background interferences. Its effectiveness was validated by in vitro and in vivo experiments in which the diffuse reflectance of intralipid solutions and human skin was collected at the source distances of 0.6 mm and 2 mm by the custom-built system with six super-luminescent emitting diodes (SLEDs) light source. The results showed that, for the in vitro experiments of intralipid solutions, the coefficients of variations of diffuse reflectance decreased by 20.5% under all the six wavelengths after differential correction. For the in vivo experiments of oral glucose tolerance tests (OGTTs), partial least squares (PLS) regression models between glucose concentrations and the diffuse reflectance from palm skin were built, and the root mean square error of cross validation (RMSECV) decreased by 38.0% on average after the differential correction. Further, the spectra of the oral water tolerance tests (OWTTs) were collected for correlation with glucose concentration in OGTTs, and their correlation coefficients (R) decreased by 35.0% on average after the differential correction. Therefore, this differential correction method based on the spectra from the floating-reference position and measuring position can weaken the influence of background variations on the NIR spectroscopy and has promising potential in in vivo detection, especially for noninvasive blood glucose measurement.

Abstract:For noninvasive blood sensing with attenuated total reflection (ATR)-Fourier Transform infrared spectroscopy(FTIR), the baseline of spectra human skin will drift because of the exitence of pressure changes between ATR prism and human skin. To correct the baseline drifts,In the paper, the fingertips spectra in mid-infrared were recorded under the special conditions. A line equation was calculated using the last 14 spectra coupled with the linear regression analysis between the absorbance spectra at the 1035cm(-1) and the 950cm(-1). And the corrected absorption intensities, after the removal of the baseline drifts, can be applied for the quantification of glucose concentrations in blood easily with the given method in the paper.
Double Beam Near-Infrared Spectrometer for Compensation of Background Water Absorption and Instrumental Drift in Intensity

Abstract:A double beam near-infrared spectrometer is developed to compensate the water absorption and instrumental drift in intensity. The spectrometer may be used for both single and double beam measurements, and the two operation modes are compared. The results show that the double beam technique eliminates instrumental drift in the single beam measurement and therefore the stability of the system increases by more than 20%. The compensation of the double beam system on water absorption is verified by the measurement of fat content in milk. The results show that the spectrum data based on double beam mode get better calibration model and lower prediction error than traditional single beam mode.
An Effective Method Based on Reference Point for Glucose Sensing at 1100-1600Nm

Abstract:Non-invasive blood glucose sensing by near-infrared spectroscopy is easily interrupted by the strong background variations compared to the weak glucose signals. In this work, according to the distribution of diffuse reflectance intensity at different source-detector separations, a method based on a reference point and a measuring point, where the diffuse reflectance intensity is insensitive and most sensitive to the variation of glucose concentration, respectively, is applied. And the data processing method based on the information of two points is investigated to improve the precision of glucose sensing. Based on the Monte Carlo simulation in 5% intralipid solution model, the corresponding optical probe is designed which includes two detecting points: a reference point located at 1.3-1.7mm and a measuring point located at 1.7-2.1mm. Using the probe, the in vitro experiment with different glucose concentrations in the intralipid solution is conducted at 1100-1600nm. As a result, compared to the PLS model built by the signal of the measuring point, the root mean square error of prediction (RMSEP) and root mean square error of cross calibration (RMSEC) of the corrected model built by reference point and measuring point reduces by 45.10%, and 32.15% respectively.

Abstract:For the non-invasive blood glucose concentration sensing by the near-infrared spectroscopy, the signal to noise ratio of the optical measurement system is very low. Both the content of glucose in body and the absorption coefficient of glucose in the near-infrared region are quite weak. More over, the structure of spectral noise is complicated and the variation of noise intensity is very large. The background correction is one of the most effective pre-processing methods to improve the signal to noise ratio of the near-infrared optical detection system. In this paper, the theory expression formula of traditional background correction method was induced firstly. Then in order to avoid the influence from the variation of optical characteristics in the sample and the drift in the optical system, the similar background correction method was proposed, that is, the background which has the similar optical characteristics with the sample was chosen as the reference. The in vitro experiments of pure absorption media and scattering media were conducted to validate the effect. The results showed that, for the glucose in the blood plasma solution and Intralipid-2% solution, after the correction of the background which has the similar optical characteristics with the sample, the prediction precision of multivariate model for glucose concentration has been improved by 25.9% and 40.1%, respectively.
The Validation of the Effect of Correcting Spectral Background Changes Based on Floating Reference Method by Simulation

Abstract:There are some challenges in near-infrared non-invasive blood glucose measurement,such as the low signal to noise ra-tio of instrument,the unstable measurement conditions,the unpredictable and irregular changes of the measured object,and etc. Therefore,it is difficult to extract the information of blood glucose concentrations from the complicated signals accurately.Ref-erence measurement method is usually considered to be used to eliminate the effect of background changes.But there is no refer-ence substance which changes synchronously with the anylate.After many years of research,our research group has proposed the floating reference method,which is succeeded in eliminating the spectral effects induced by the instrument drifts and the measured object’s background variations.But our studies indicate that the reference-point will changes following the changing of measurement location and wavelength.Therefore,the effects of floating reference method should be verified comprehensively.In this paper,keeping things simple,the Monte Carlo simulation employing Intralipid solution with the concentrations of 5% and 10% is performed to verify the effect of floating reference method used into eliminating the consequences of the light source drift. And the light source drift is introduced through varying the incident photon number.The effectiveness of the floating reference method with corresponding reference-points at different wavelengths in eliminating the variations of the light source drift is esti-mated.The comparison of the prediction abilities of the calibration models with and without using this method shows that the RMSEPs of the method are decreased by about 98. 57%(5%Intralipid)and 99. 36%(10%Intralipid)for different Intralipid.The results indicate that the floating reference method has obvious effect in eliminating the background changes.
A Reference-Wavelength-Based Method for Improved Analysis of Near-Infrared Spectroscopy

Abstract:Near-infrared (NIR) spectroscopy has been widely used in many industrial applications. It also has tremendous potential for trace element detection and noninvasive human physiological measurements. In NIR spectroscopy, however, the measurement precision is often dependent on temperature, measurement position, and sample status. In order to improve measurement precision, a method using spectral information at a reference wavelength is developed in this paper. Based on the displacement effect between solvent and solute molecules in a solution, the signal at the reference wavelength is used as an internal reference to correct the spectrum of the sample under test. As an example, the spectra of glucose aqueous solutions under different temperatures are measured, and our method for eliminating the temperature disturbance is evaluated. The experimental results obtained show that the relative error of glucose concentration prediction is 330% per degree before the spectrum correction. After the correction, the relative error is reduced to 5.12%, and the error is no longer dependent on temperature. As the displacement effect can be found commonly in various solutions, the method described in this work may be used to improve the accuracy of spectral analysis of many other solutions.
Correlation Analysis Combined with a Floating Reference Measurement to Improve the Prediction Accuracy of Glucose in Scattering Media.

Abstract:Noninvasive sensing of blood glucose based on near-infrared (NIR) spectroscopy is a research hotspot in the biomedical field. However, its accuracy is severely limited by the weak specific signal of glucose and the strong background variations caused by other constituents in the blood, the measuring instrument, and the environment. In this paper, special source–detector distances, defined as the floating reference position, are used to conduct relative measurements and correct for background variations. These floating reference positions are chosen so that the diffuse reflectance is not sensitive to the change in glucose concentration due to the combined effects of absorption and scattering. Nine 10% intralipid samples with glucose concentrations in the range of 1000–5000 mg dL–1 at an interval of 500 mg dL–1 were prepared. Using a custom-built, continuously moving, spatially resolving, double-fiber measurement system with a superluminescent diode (SLD) as the light source, the diffuse reflectance of intralipid samples containing glucose under different source–detector distances (0.2–5 mm, with intervals of 0.2 mm) were collected. Then, a correlation analysis between the spectra and the glucose concentration was carried out to determine the floating reference position and the optimal measuring position. The signal in the floating reference position was used to correct the background variation because it contains the same systematic drift and interference as the signal in the optimal measuring position. The results showed that the correlation between the diffuse reflectance and the glucose concentration was increased significantly compared with traditional correction by subtracting the nearest spectrum of pure 10% intralipid solution. The correlation between the diffuse reflectance and the concentration of glucose is significantly increased, which indicated that the combination of the correlation analysis and a floating reference is able to eliminate the influence of background variations.
Research on Temperature Disturbance of Glucose Solution With Two-Trace Two-Dimensional Correlation Spectrum Method

Abstract:The near-infrared absorption features are easily affected by temperature changes, which reduce the accuracy of quantitative analysis. This paper presents the methods for analysing the source and amplitude of temperature disturbance based on the theory of two-trace two-dimensional correlation spectroscopy (2T2D-COS). The absorption spectra of glucose solution, with a concentration of 0 similar to 200 mg.dL(-1), are measured in the temperature range of 34 similar to 38.5 degrees C with a stepwise of 0.5 degrees C. After baseline correction and noise filtering, the concentration-dependent and temperature-dependent spectra are analyzed with the 2T2D-COS algorithm. In the asynchronous spectrum of temperature-dependent spectra, the cross-peaks are observed at 1 474 and 1 410 nm, corresponding to strong and weak hydrogen-bonded water, respectively. While in the asynchronous spectrum of concentration-dependent spectra, the cross peaks at 1 450 and 1 595 nm are related to the characteristic absorption of water and glucose. From the slice spectrum at 1 410 nm, the intensity of the peaks in the range of 1 410n similar to 1 600 nm is increased with temperature, demonstrating good correlation between the cross peaks and temperature variance. The calibration model is established for the quantitative analysis of sample temperature with the cross peak intensity range of (1 475 +/- 4) nm. The root mean square error achieved 0.125 9 degrees C for sample temperature prediction with linear regression. The experimental results demonstrate that the temperature disturbances could be identified and qualified with distinctly separate cross peaks in the asynchronous spectrum, obtained using only a pair of spectra. The proposed approach also provides a reasonable way for researchers to simplify the in vivo measurement system and establish a more stable calibration model.
Extraction of Glucose Information in Blood Glucose Measurement by Noninvasive Near-Infrared Spectra

Abstract:Near infrared spectroscopy has been proposed as an effective way for measuring blood glucose non-invasively. However the change of spectrum due to an increase in glucose level is very small compared to the changes due to other variations such as absorption of major blood components, skin surface reflectance, temperature and pressure and so on. So the complexity of spectrum makes it difficult to identify unique glucose information. In this paper, the effect of background correction is discussed firstly. Then a simple substitution is proposed to compute the net analyte signal of glucose using the subspace spanned by the background spectra. For the in vitro experiment, the net analyte signals of glucose using the traditional methods and the subspace spanned by background have the same peaks in the absorption peaks of glucose for the glucose aqueous solution. For in vivo experiment, there is significant spectral difference between the subject who took OGTT test and the subject who took no glucose or water. And the net analyte signal of glucose is computed for OGTT test based on the subspace spanned by the spectra of subject who didn’t take glucose. Results show that, the spectral information induced by glucose taking is quite significant but it does not have the same peak at the absorption peak of glucose in near-infrared region.

Abstract:We present an approach for accurate glucose sensing in turbid media using a spectrally resolved reflectance setup. Our proposed reflectance setup uses specialized source-detector separations (SDSs) to enable an effective separation of diffusion and absorption signals. Additionally, we adjust the selected SDSs to their optimal values to acquire maximum sensitivity to glucose in the two signals. The separation can help to enhance the sensitivity to glucose both for the diffusion and absorption signals, as they always suppress each other by causing opposite effects on the reflected diffuse light intensity. Monte Carlo simulations and experiments for glucose sensing are used to test the method. The acquired optimal SDSs could provide a reference for noninvasive blood glucose sensing.
Conditions And Methods Of High Precision Measurement Of Blood Glucose Concentration Using Mid-Ir Atr Spectroscopy

Abstract:This paper involves conditions and methods of high precision measurement of blood glucose concentration using Mid-IR ATR spectroscopy: (1) Comparing the spectra of saline solution, plasma, and whole blood to which adding glucose respectively, we find that in these three backgrounds with different complexity, the absorption peaks of glucose are at the same wavelengths in the band of 1200 to 900 cm(-1). (2) Analyzing experimental results of concentration gradient built by adding glucose to a series of plasma, we obtained a high correlation (greater than 0.98) between glucose concentration and absorbance in the band of 1200 to 900 cm(-1), it provides us the feasibility of building calibration model of glucose concentration in this specific band. (3) In the process of building calibration model for glucose concentration of 40 whole blood samples (blood glucose concentration varied from 73 to 167 mg/dL, mean was 94.46 mg/dL, standard error was 17.68 mg/dL) by utilizing PLS method optimized with full cross validation, we applied genetic algorithms to the selection of optimal subsets of wavelengths, the number of wavelengths for calibration was decreased to 14 and the precision of the model was improved considerably, RMSEP was 10.56 mg/dL. This proves that high precision measurement of blood glucose concentration can be achieved in narrow-band width (1200-900 cm(-1)) when few wavelengths are selected.

Abstract:Relative measurement of near-infrared (NIR) spectroscopy is of great significance for the high precision detection of human blood glucose concentration in vivo. Although the similar background subtraction and double-beam design are frequently used in the in vitro experiments, they are not suitable for the human body detection because of the complex background variations. Reference measurement based on position is one of the most promising methods to realize in vivo reference measurement. The author proposes differential floating reference measurement method for in vivo reference measurement. The differential floating reference measurement is a universal method. However, it is difficult to determine the radial detection positions and extract the effective information from differential signals in practical use. To solve these problems, differential floating reference based on NAS (Net Analyte Signal)-VIP (Variable Importance in Projection)-SPXY (Sample set Partitioning based on joint X-Y distances)-PLS (Partial Least Square) method is proposed and the feasibility is investigated byin vitro and in vivo experiments, the results shows that the prediction accuracy and precision are improved through this method.

Abstract:In the near-infrared region, we can test the blood glucose by non-invasive method. Based on this theory, combined photoacoustic principle, we had research about the simulated samples of blood glucose, measured the concentration of glucose solution. We have discussed the optimal wavelength in the near-infrared region (780-2400nm), and did the experiment repeatedly, and then analyzed data through the linear, polynomial and exponential fitting of the Generalized Least Squares, the measuring accuracy of the glucose solution had improved. These research results provide the guidance on the practice and experimental evidence for the further research of measuring.
