Abstract:Many questions remain regarding how the brain develops, matures, and ages across the lifespan. Abstract:Brain aging can affect the strength of functional connectivity between brain regions.In recent years,studies have shown that functional connectivity is fluctuant over time,and can reflect more physiological and pathological information.Therefore,in the study resting state functional magnetic resonance imaging (fMRI) data of 32 elderly subjects and 36 younger subjects were selected,and the sliding window technique was used to estimate dynamic functional connectivity network.Then,the dependency of fluctuating energy difference on frequency band was studied using wavelet packet analysis,conducting the linear regression with age at the same time.Results showed that the fluctuating energy in older group was significantly higher than that in the young group in low frequency,and it was significantly lower than that in the young people in high frequency.These results suggested that the dynamic functional connectivity between networks in the elderly exist slow wave phenomenon,which may be related to the decreased reaction rate of the elderly.This article provides new ideas and methods for the research about brain aging,and promotes a theoretical basis for further understanding of the physiological significance of brain dynamic functional connectivity.

Abstract:Healthy aging is usually accompanied by changes in the functional modular organization of the human brain, which may result in the decline of cognition and underlying brain dysfunction. However, the relationship between age-related brain functional modular structure differences and cognition remain debatable. In this study, we investigated the age-associated differences of modules and hubs from young, middle and old age groups, using resting-state fMRI data from a large cross-sectional adulthood sample. We first divided the subjects into three age groups and constructed an individual-level network for each subject. Subsequently, a module-guided group-level network construction method was applied to form a weighted network for each group from which functional modules were detected. The intra- and inter-modular connectivities were observed negatively correlated with age. According to the detected modules, we found the number of connector hubs in the young group was more than middle-age and old group, while the quantity of provincial hubs in middle-age group was discovered more than other two groups. Further ROI-wise analysis shows that different hubs have distinct age-associated trajectories of intra- and inter-modular connections, which suggests the different types of topological role transitions in functional networks across age groups. Our results indicated an inverse association between functional segregation/integration with age, which demonstrated age-associated differences in communication effeciency. This study provides a new perspective and useful information to better understand the normal aging of brain networks.

Abstract:Aging is the fundamental of neurodegeneration and dementia, affecting every organ in the body. With the aggravation of global aging, more and more research is focusing on how brain changes in older adults. This study aimed to uncover the differences in brain functional networks from the perspective of graph theory between young individuals and older individuals. Here, 61 individuals in their 20s and 94 cognitively healthy old individuals in their 70s underwent a resting-state functional magnetic resonance imaging scan. Based on the graph theory method, a functional network was constructed for each participant. Our results revealed that brain functional networks in older adults maintained small-world properties. However other nodal parameters including degree centrality, betweenness centrality, shortest path lengths, local efficiency, nodal efficiency, and cluster coefficients showed significant differences in many nodes (brain regions) between the 2 groups. Moreover, we correlated these nodal parameters with age, exploring 8 brain regions significantly affected with age. 7 out of 8 brain regions including the bilateral superior parietal lobule, bilateral precuneus, right middle cingulate, right inferior parietal lobule and right transverse temporal gyri were distributed in the default mode network. Our findings, based on graph theory, provided evidence for the alteration of the default mode network in older adults from the perspective of the functional network.

Abstract:Abstract Introduction As aging attracted attention globally, revealing changes in brain function across the lifespan was largely concerned. In this study, we aimed to reveal the changes of functional networks of the brain (via local functional connectivity, local FC) in lifespan and explore the mechanism underlying them. Materials and Methods A total of 523 healthy participants (258 males and 265 females) aged 18–88 years from part of the Cambridge Center for Ageing and Neuroscience (CamCAN) were involved in this study. Next, two data‐driven measures of local FC, local functional connectivity density (lFCD) and four‐dimensional spatial‐temporal consistency of local neural activity (FOCA), were calculated, and then, general linear models were used to assess the changes of them in lifespan. Results Local functional connectivity (lFCD and FOCA) within visual networks (VN), sensorimotor network (SMN), and default mode network (DMN) decreased across the lifespan, while within basal ganglia network (BGN), local connectivity was increased across the lifespan. And, the fluid intelligence decreased within BGN while increased within VN, SMN, and DMN. Conclusion These results might suggest that the decline of executive control and intrinsic cognitive ability in the aging population was related to the decline of functional connectivity in VN, SMN, and DMN. Meanwhile, BGN might play a regulatory role in the aging process to compensate for the dysfunction of other functional systems. Our findings may provide important neuroimaging evidence for exploring the brain functional mechanism in lifespan.

Abstract:The objectively-defined subtle cognitive decline individuals had higher progression rates of cognitive decline and pathological deposition than healthy elderly, indicating a higher risk of progressing to Alzheimer's disease. However, little is known about the brain functional alterations during this stage. Thus, we aimed to investigate the functional network patterns in objectively-defined subtle cognitive decline cohort. Forty-two cognitive normal, 29 objectively-defined subtle cognitive decline and 55 mild cognitive impairment subjects were included based on neuropsychological measures from the Alzheimer's disease Neuroimaging Initiative dataset. Thirty cognitive normal, 22 objectively-defined subtle cognitive declines and 48 mild cognitive impairment had longitudinal MRI data. The degree centrality and eigenvector centrality for each participant were calculated by using resting-state functional MRI. For cross-sectional data, analysis of covariance was performed to detect between-group differences in degree centrality and eigenvector centrality after controlling age, sex and education. For longitudinal data, repeated measurement analysis of covariance was used for comparing the alterations during follow-up period among three groups. In order to classify the clinical significance, we correlated degree centrality and eigenvector centrality values to Alzheimer's disease biomarkers and cognitive function. The results of analysis of covariance showed significant between-group differences in eigenvector centrality and degree centrality in left superior temporal gyrus and left precuneus, respectively. Across groups, the eigenvector centrality value of left superior temporal gyrus was positively related to recognition scores in auditory verbal learning test, whereas the degree centrality value of left precuneus was positively associated with mini-mental state examination total score. For longitudinal data, the results of repeated measurement analysis of covariance indicated objectively-defined subtle cognitive decline group had the highest declined rate of both eigenvector centrality and degree centrality values than other groups. Our study showed an increased brain functional connectivity in objectively-defined subtle cognitive decline individuals at both local and global level, which were associated with Alzheimer's disease pathology and neuropsychological assessment. Moreover, we also observed a faster declined rate of functional network matrix in objectively-defined subtle cognitive decline individuals during the follow-ups.

Our findings highlighted the notable dynamic characteristics in large-scale brain networks across the adult lifespan, which enhanced our understanding of the neural substrate in various cognition during ageing. These findings also provided further evidence that dedifferentiation and compensation are the outcomes of functional brain interactions.

Abstract:Introduction: There are still uncertainties about the true nature of age related changes in topological properties of the brain functional network and its structural connectivity during various developmental stages. In this cross- sectional study, we investigated the effects of age and its relationship with regional nodal properties of the functional brain network and white matter integrity. Method: DTI and fMRI data were acquired from 458 healthy Chinese participants ranging from age 8 to 81 years. Tractography was conducted on the DTI data using FSL. Graph Theory analyses were conducted on the functional data yielding topological properties of the functional network using SPM and GRETNA toolbox. Two multiple regressions were performed to investigate the effects of age on nodal topological properties of the functional brain network and white matter integrity. Result: For the functional studies, we observed that regional nodal characteristics such as node betweenness were decreased while node degree and node efficiency was increased in relation to increasing age. Perversely, we observed that the relationship between nodal topological properties and fasciculus structures were primarily positive for nodal betweenness but negative for nodal degree and nodal efficiency. Decrease in functional nodal betweenness was primarily located in superior frontal lobe, right occipital lobe and the global hubs. These brain regions also had both direct and indirect anatomical relationships with the 14 fiber bundles. A linear age related decreases in the Fractional anisotropy (FA) value was found in the callosum forceps minor. Conclusion: These results suggests that age related differences were more pronounced in the functional than in structural measure indicating these measures do not have direct one-to-one mapping. Our study also indicates that the fiber bundles with longer fibers exhibited a more pronounced effect on the properties of functional network.
Changes in Topological Organization of Functional PET Brain Network with Normal Aging.

Abstract:Recent studies about brain network have suggested that normal aging is associated with alterations in coordinated patterns of the large-scale brain functional and structural systems. However, age-related changes in functional networks constructed via positron emission tomography (PET) data are still barely understood. Here, we constructed functional brain networks composed of 90 regions in younger (mean age 36.5 years) and older (mean age 56.3 years) age groups with PET data. 113 younger and 110 older healthy individuals were separately selected for two age groups, from a physical examination database. Corresponding brain functional networks of the two groups were constructed by thresholding average cerebral glucose metabolism correlation matrices of 90 regions and analysed using graph theoretical approaches. Although both groups showed normal small-world architecture in the PET networks, increased clustering and decreased efficiency were found in older subjects, implying a degeneration process that brain system shifts from a small-world network to regular one along with normal aging. Moreover, normal senescence was related to changed nodal centralities predominantly in association and paralimbic cortex regions, e.g. increasing in orbitofrontal cortex (middle) and decreasing in left hippocampus. Additionally, the older networks were about equally as robust to random failures as younger counterpart, but more vulnerable against targeted attacks. Finally, methods in the construction of the PET networks revealed reasonable robustness. Our findings enhanced the understanding about the topological principles of PET networks and changes related to normal aging.

Abstract:Intrinsic brain activity in a resting state incorporates components of the task negative network called default mode network (DMN) and task-positive networks called attentional networks. In the present study, the reciprocal neuronal networks in the elder group were compared with the young group to investigate the differences of the intrinsic brain activity using a method of temporal correlation analysis based on seed regions of posterior cingulate cortex (PCC) and ventromedial prefrontal cortex (vmPFC). We found significant decreased positive correlations and negative correlations with the seeds of PCC and vmPFC in the old group. The decreased coactivations in the DMN network components and their negative networks in the old group may reflect age-related alterations in various brain functions such as attention, motor control and inhibition modulation in cognitive processing. These alterations in the resting state anti-correlative networks could provide neuronal substrates for the aging brain.

Abstract:Normal aging is typically characterized by abnormal resting-state functional connectivity (FC), including decreasing connectivity within networks and increasing connectivity between networks, under the assumption that the FC over the scan time was stationary. In fact, the resting-state FC has been shown in recent years to vary over time even within minutes, thus showing the great potential of intrinsic interactions and organization of the brain. In this article, we assumed that the dynamic FC consisted of an intrinsic dynamic balance in the resting brain and was altered with increasing age. Two groups of individuals (N = 36, ages 20–25 for the young group; N = 32, ages 60–85 for the senior group) were recruited from the public data of the Nathan Kline Institute. Phase randomization was first used to examine the reliability of the dynamic FC. Next, the variation in the dynamic FC and the energy ratio of the dynamic FC fluctuations within a higher frequency band were calculated and further checked for differences between groups by non-parametric permutation tests. The results robustly showed modularization of the dynamic FC variation, which declined with aging; moreover, the FC variation of the inter-network connections, which mainly consisted of the frontal-parietal network-associated and occipital-associated connections, decreased. In addition, a higher energy ratio in the higher FC fluctuation frequency band was observed in the senior group, which indicated the frequency interactions in the FC fluctuations. These results highly supported the basis of abnormality and compensation in the aging brain and might provide new insights into both aging and relevant compensatory mechanisms.
Alterations of Functional Connectivities from Early to Middle Adulthood: Clues from Multivariate Pattern Analysis of Resting-State Fmri Data.

Abstract:In contrast to extended research interests in the maturation and aging of human brain, alterations of brain structure and function from early to middle adulthood have been much less studied. The aim of the present study was to investigate the extent and pattern of the alterations of functional interactions between brain regions from early to middle adulthood. We carried out the study by multivariate pattern analysis of resting-state fMRI (RS-fMRI) data of 63 adults aged 18 to 45years. Specifically, using elastic net, we performed brain age estimation and age-group classification (young adults aged 18–28years vs. middle-aged adults aged 35–45years) based on the resting-state functional connectivities (RSFCs) between 160 regions of interest (ROIs) evaluated on the RS-fMRI data of each subject. The results indicate that the estimated brain ages were significantly correlated with the chronological age (R=0.78, MAE=4.81), and a classification rate of 94.44% and area under the receiver operating characteristic curve (AUC) of 0.99 were obtained when classifying the young and middle-aged adults. These results provide strong evidence that functional interactions between brain regions undergo notable alterations from early to middle adulthood. By analyzing the RSFCs that contribute to brain age estimation/age-group classification, we found that a majority of the RSFCs were inter-network, and we speculate that inter-network RSFCs might mature late but age early as compared to intra-network ones. In addition, the strengthening/weakening of the RSFCs associated with the left/right hemispheric ROIs, the weakening of cortico-cerebellar RSFCs and the strengthening of the RSFCs between the default mode network and other networks contributed much to both brain age estimation and age-group classification. All these alterations might reflect that aging of brain function is already in progress in middle adulthood. Overall, the present study indicated that the RSFCs undergo notable alterations from early to middle adulthood and highlighted the necessity of careful considerations of possible influences of these alterations in related studies.
Brain-wide functional connectome analysis of 40,000 individuals reveals brain networks that show aging effects in older adults

Abstract:The functional connectome changes with aging. We systematically evaluated aging related alterations in the functional connectome using a whole-brain connectome network analysis in 39,675 participants in UK Biobank project. We used adaptive dense network discovery tools to identify networks directly associated with aging from resting-state fMRI data. We replicated our findings in 499 participants from the Lifespan Human Connectome Project in Aging study. The results consistently revealed two motor-related subnetworks (both permutation test p-values <0.001) that showed a decline in resting-state functional connectivity (rsFC) with increasing age. The first network primarily comprises sensorimotor and dorsal/ventral attention regions from precentral gyrus, postcentral gyrus, superior temporal gyrus, and insular gyrus, while the second network is exclusively composed of basal ganglia regions, namely the caudate, putamen, and globus pallidus. Path analysis indicates that white matter fractional anisotropy mediates 19.6% (p<0.001, 95% CI [7.6% 36.0%]) and 11.5% (p<0.001, 95% CI [6.3% 17.0%]) of the age-related decrease in both networks, respectively. The total volume of white matter hyperintensity mediates 32.1% (p<0.001, 95% CI [16.8% 53.0%]) of the aging-related effect on rsFC in the first subnetwork.

Abstract:At rest, the brain's sensorimotor and higher cognitive systems engage in organized patterns of correlated activity forming resting-state networks. An important empirical question is how functional connectivity and structural connectivity within and between resting-state networks change with age. In this study we use network modeling techniques to identify significant changes in network organization across the human lifespan. The results of this study demonstrate that whole-brain functional and structural connectivity both exhibit reorganization with age. On average, functional connections within resting-state networks weaken in magnitude while connections between resting-state networks tend to increase. These changes can be localized to a small subset of functional connections that exhibit systematic changes across the lifespan. Collectively, changes in functional connectivity are also manifest at a system-wide level, as components of the control, default mode, saliency/ventral attention, dorsal attention, and visual networks become less functionally cohesive, as evidenced by decreased component modularity. Paralleling this functional reorganization is a decrease in the density and weight of anatomical white-matter connections. Hub regions are particularly affected by these changes, and the capacity of those regions to communicate with other regions exhibits a lifelong pattern of decline. Finally, the relationship between functional connectivity and structural connectivity also appears to change with age; functional connectivity along multi-step structural paths tends to be stronger in older subjects than in younger subjects. Overall, our analysis points to age-related changes in inter-regional communication unfolding within and between resting-state networks.

Abstract:Exploring brain changes across the human lifespan is becoming an important topic in neuroscience. Though there are multiple studies which investigated the relationship between age and brain imaging data, the results are heterogeneous due to small sample sizes and relatively narrow age ranges. Here, based on year-wise estimation of 5967 subjects from 13 to 72 years old, we aimed to provide a more precise description of adult lifespan variation trajectories of gray matter volume (GMV), structural network correlation (SNC) and functional network connectivity (FNC) using independent component analysis and multivariate linear regression model. Our results revealed the following relationships: 1)GMV linearly declined with age in most regions, while parahippocampus showed an inverted U-shape quadratic relationship with age; SNC presented a U-shape quadratic relationship with age within cerebellum, and inverted U-shape relationship primarily in the default mode network (DMN) and frontoparietal (FP) related correlation. 2) FNC tended to linearly decrease within resting-state networks (RSNs), especially in visual network and DMN. Early increase was revealed between RSNs, primarily in FP and DMN, which experienced decrease at older ages. U-shape relationship was also revealed to compensate for the cognition deficit in attention and subcortical related connectivity at late years. 3) The link between middle occipital gyrus and insula, as well as precuneus and cerebellum, exhibited similar changing trends between SNC and FNC across the adult lifespan. Collectively, these results highlight the benefit of lifespan study and provide a precise description of age-related regional variation and SNC/FNC changes based on a large dataset.

Abstract:At rest, the brain's sensorimotor and higher cognitive systems engage in organized patterns of correlated activity forming resting-state networks. An important empirical question is how functional connectivity and structural connectivity within and between resting-state networks change with age. In this study we use network modeling techniques to identify significant changes in network organization across the human lifespan. The results of this study demonstrate that whole-brain functional and structural connectivity both exhibit reorganization with age. On average, functional connections within resting-state networks weaken in magnitude while connections between resting-state networks tend to increase. These changes can be localized to a small subset of functional connections that exhibit systematic changes across the lifespan. Collectively, changes in functional connectivity are also manifest at a system-wide level, as components of the control, default mode, saliency/ventral attention, dorsal attention, and visual networks become less functionally cohesive, as evidenced by decreased component modularity. Paralleling this functional reorganization is a decrease in the density and weight of anatomical white-matter connections. Hub regions are particularly affected by these changes, and the capacity of those regions to communicate with other regions exhibits a lifelong pattern of decline. Finally, the relationship between functional connectivity and structural connectivity also appears to change with age; functional connectivity along multi-step structural paths tends to be stronger in older subjects than in younger subjects. Overall, our analysis points to age-related changes in inter-regional communication unfolding within and between resting-state networks.

Abstract:Aging is associated with changes in brain functional patterns as well as cognition. The present research sought to investigate longitudinal changes in whole brain functional connectivity strength (FCS) and cognitive performance scores in very old cognitively unimpaired individuals. We studied 34 cognitively normal elderly individuals at both baseline and 4-year follow-up (baseline age = 78 +/- 3.14 years) with resting-state functional magnetic resonance imaging (r-fMRI), structural MRI scans, and neuropsychological assessments conducted. Voxel-based whole brain FCS was calculated and we found that bilateral superior parietal and medial frontal regions showed decreased FCS, while the supplementary motor area (SMA) and insula showed increased FCS with age, along with a decrease in bilateral prefrontal cortical thickness. The changes of FCS in left precuneus were associated with an aging-related decline in global cognition. Taken together, our results suggest changes in FCS with aging with the precuneus as a hub and this may underlie changes in global cognition that accompany aging. These findings help better understand the normal aging mechanism.

Abstract:Across the adult lifespan, there are changes in how emotions are perceived and regulated. As individuals age, there is an observed improvement in emotion regulation and overall quicker recovery from negative emotions. While previous studies have shown differences in emotion processing in late adulthood the corresponding differences in large scale brain networks remain largely underexplored. By utilizing large scale datasets such as the Human Connectome Project ((HCP-Aging, N=621, 349 females) and Cambridge Centre for Ageing and Neuroscience (Cam-CAN, N=333, 155 females), we were able to investigate how emotion regulation networks' functional topography differs across the entire adult lifespan Based on previous meta analytic work that identified four large scale functional brain networks involved in emotion generation and regulation, we found an increase in the functional integration of the emotional control network among older adults. Additionally, confirming through the nonlinear model, individuals around the age of 70 showed a steadier decline in integration of a network mediating emotion generation and regulation via interoception. Furthermore, the analyses revealed a negative association between age and perceived stress and loneliness that could be attributed to differences in large scale emotion regulation networks. Our study highlights the importance of identifying topological changes in the functional emotion network architecture across the lifespan as it allows for a better understanding of emotional aging and psychological well being in late adulthood.
