Abstract:Individualized treatment is a promising clinical strategy for lung cancer, and drug sensitivity testing is fundamental to this scheme. We aimed to develop an effective drug sensitivity test platform to support individualized treatment. We designed a microfluidic chip-based, three-dimensional (3D) co-culture drug sensitivity test platform. A mono-lung cancer cell line, a mixture of lung cancer and stromal cell lines, and cells from fresh lung cancer tissues were cultured in 3D under continuous media supplementation, mimicking the actual tumor microenvironment in vivo. The cells were treated with anti-cancer drugs according to a gradient concentration generator inside the chips to screen the appropriate chemotherapy schemes. We successfully cultured cell lines or primary cells with this device. We also smoothly assayed the sensitivities of different anti-cancer drugs in parallel and accurately screened appropriate-dose, single and combined-drug chemotherapy schemes for eight patients. Our microfluidic device is a simple, reliable, and high-throughput platform to test drug sensitivity. It would be possible for chemotherapists to screen the appropriate chemotherapy schemes to guide individualized treatment in lung cancer.

Abstract:In recent decades, three-dimensional (3D) cancer cell models have attracted increasing interest in the field of drug screening due to their significant advantages in more accurate simulations of heterogeneous tumor behavior in vivo compared to two-dimensional models. Furthermore, drug sensitivity testing based on 3D cancer cell models can provide more reliable in vivo efficacy prediction. The gold standard fluorescence staining is hard to achieve real-time and label-free viability monitoring in 3D cancer cell models. In this study, a microgroove impedance sensor (MGIS) was specially developed for the dynamic and noninvasive monitoring of 3D cell viability. 3D cancer cells were trapped in microgrooves with gold electrodes on opposite walls for in situ impedance measurement. The change in the number of live cells caused inversely proportional changes to the impedance magnitude of the entire cell/Matrigel construct and reflected the proliferation and apoptosis of the 3D cells. It was confirmed that the 3D cell viability detected by the MGIS was highly consistent with the standard live/dead staining by confocal microscope characterization. Furthermore, the accuracy of the MGIS was validated quantitatively using a 3D lung cancer model and sophisticated drug sensitivity testing. In addition, the parameters of the MGIS in the measurement experiments were optimized in detail using simulations and experimental validation. The results demonstrated that the MGIS coupled with 3D cell culture would be a promising platform to improve the efficiency and accuracy of cell-based anticancer drug screening in vitro.

Abstract:To accelerate the discovery of anti-cancer drugs, there is an urgent need to establish an inexpensive and simple preclinical model that can simulate the tumor microenvironment and screen the drug candidates. Some widely used screening platforms are high-cost and over-simple to verify the cell-drug interactions for predicting human clinical trial outcomes. As a promising technology, microfluidic chip can replace traditional screening methods due to low reagent consumption and the capability of recapitulating the 3D cell culture more representative of the native tumor microenvironment. This review covers the materials for microchip fabrication (especially hydrogel), tumor microenvironment, 2D and 3D tumor culture on chips, drug screening methods (concentration gradient generator-based, microdroplet-based, microarray-based chips, and tumor tissue chip), and their pros and cons for cancer treatment. We also discuss the future development of microfluidic for anti-cancer drug discovery research.
A Novel Micro-Groove Impedance Sensor for 3D Cell Viability Monitoring and High-Throughput Drug Screening

Abstract:Drug screening is traditionally based on the pharmacodynamic models from 2D cell culture or animal experiments. Recently, three-dimensional (3D) tumor cell models have attracted increasing interest due to their great advantages in simulating more accurately the heterogeneous tumor behavior in vivo. Drug screening based on 3D cells can provide more accurate efficacy results. However, it is difficult to realize real-time and label-free monitoring of 3D cell viability by common imaging techniques. To solve this technical difficulty, a novel micro-groove impedance sensor (MGIS) was specially developed for 3D cell viability real-time monitoring. Precultured 3D spheroids cells are trapped in the micro-cavity with opposite gold electrodes for the in-situ impedance measurement. The presence of 3D spheroid cells will lead to the change of electron transfer efficiency on the electrode surface, which will lead to the change of impedance. When antitumor drugs act on the spheroid cells, the spheroids will cleave and the impedance will decrease. In order to verify the accuracy of MGIS chip, we adopted standard live/dead fluorescence staining to validate the activity of 3D cells. Furthermore, anti-tumor drug sensitivity tests were conducted to validate the drug screening ability of MGIS plat-form. All the results demonstrate that the MGIS is able to monitoring 3D cell viability and drug screening.

Abstract:Preclinical efficacy and toxicity assessment of drug candidates plays a significant role in drug discovery and development. Traditional planar cell culture is a common way to perform the preclinical drug test, but it is difficult to correctly predict the drug efficacy and toxicity due to the simple two-dimensional (2D) extracellular microenvironment. Compared to the planar cell culture, three-dimensional (3D) cell culture system can better mimic the complex extracellular microenvironment where cells reside in the 3D tissues/organs in vivo. However, the conventional imaging techniques are difficult to achieve the dynamic and label-free monitoring of cellular behavior in thick sample by 3D cell culture. Here, 3D electric cell/matrigel-substrate impedance sensing (3D-ECMIS) is developed for real-time and non-invasive monitoring of 3D cell viability and drug susceptibility. In this study, human hepatoma cells (HepG2) are encapsulated in the matrigel scaffold and cultured in a 3D ECMIS chip which involves a pair of vertical golden electrodes on the opposite sidewalls of the culture chamber for the in-situ impedance measurement. Moreover, a portable multichannel system is developed to monitor the 3D cell/matrigel construct. The number of 3D-cultured cells was inversely proportional to the impedance magnitude of the entire cell/matrigel construct. Furthermore, anti-cancer drug screening will be conducted on the 3D-cultured cells when the cell proliferation reaches to a plateau phase. To validate the performance of 3D-ECMIS for the cell viability and drug susceptibility, the cell live/dead staining are utilized to confirm the results of drug susceptibility by this 3D-cell-based biosensor system. It is demonstrated that the 3D cell-based biosensor and 3D-ECMIS system will be a promising platform to improve the accuracy of cell-based anti-cancer drug screening in vitro.

Abstract:Cell sensor chips for biological impedance detection are fabricated by micromachining technology.The diameter of the interdigital electrode is 50 μm,arranged with space of 50 μm to electrode pairs.The device is equipped with sample giving system and control system.Anti-cancer drugs,such as ginseng saponin Rh2,are applied to cancer cells of human lung adenocarcinoma cell line A549 cultivated directly on the surface of the electrodes.The impedance detecting results suggest that the drug has good inhibitory effect to those cancer cells.It means that the cell microelectronic chip provides a good micro sensor test platform for anti-cancer drugs analysis and selection.
Flow Analysis of a Porous Polymer Based Three Dimensional Cell Culture Device for Drug Screening

Abstract:A porous polymer-based three-dimensional (3D) cell culture device has been developed as an in vitro tissue model system for the cytotoxicity of anticancer drug test. The device had two chambers connected in tandem, each loaded with a 3D scaffold made of highly biocompatible poly (lactic acid) (PLA). Hepatoma cells (HepG2) and glioblastoma multiforme (GBM) cancer cells were cultured in the two separate porous scaffolds. A peristaltic pump was adopted to realize a perfusion cell culture. In this study, we focus on cell viability inside the 3D porous scaffolds under flow-induced shear stress effects. A flow simulation was conducted to predict the shear stress based on a realistic representation of the porous structure. The simulation results were correlated to the cell variability measurements at different flow rates. It is shown that the modeling approach presented in this paper can be useful for shear stress predication inside porous scaffolds and the computational fluid dynamics model can be an effective way to optimize the operation parameters of perfused 3D cell culture devices.

Abstract:A porous polymer-based three-dimensional (3D) cell culture device was developed to test the cytotoxicity of anticancer drugs. The device had two chambers that were connected in tandem, each containing a 3D tissue engineering scaffold made of biodegradable poly(lactic acid) (PLA). The system was driven by a peristaltic pump to realise perfusion. In this paper, we present the design and fabrication of the 3D cell culture device and the selection of the cell culture method and perfusion flow rate. The 3D cell culture device was used to test the cytotoxicity of a cancer treatment drug, Temozolomide (TMZ) to liver cells and glioblastoma multiforme (GBM) cancer cells. As a comparison, the same test was conducted with a two-dimensional (2D) cell culture system. The results show that the cells cultured in 3D were much more resistant to TMZ than those in 2D. Since 3D cell cultures promote the formation of tissue-mimicking structures, the device developed in this study could be used as a more realistic in vitro platform for drug screening.

Abstract:Three-dimensional (3D) cell culture provides an effective way over conventional two-dimensional (2D) monolayer culture to more closely imitate the complex cellular organization, heterogeneity, and interactions as well as tissue microenvironments in vivo. Here we present a novel droplet based 3D cell culture method by using droplet array attached on the sidewall of a PDMS piece. Such an arrangement not only avoids cells from adhering on the chip surface for achieving 3D cell culture in nanoliter-scale droplets, but also facilitates performing multiple operations to cells in droplets, including cell suspension droplet generation, drug treatment, and cell staining with a capillary-based liquid handling system, as well as in situ observation and direct scanning with a confocal laser scanning microscope. We optimized the system by studying the effects of various conditions to cell culture including droplet volume, cell density and fabrication methods of the PDMS pieces. We have applied this system in the 3D culture of HepG2 cells and the stimulation testing of an anticancer drug, doxorubicin, to 3D cell spheroids.

Abstract:Currently, there has been a growing need for developing in vitro models to better reflect organism response to chemotherapy at tissue level. For this reason, a microfluidic platform was developed for mimicking physiological microenvironment of solid tumor with multicellular tumor spheroids (MTS) for anticancer drug screening. Importantly, the power of this system over traditional systems is that it is simple to operate and high integration in a more physiologically relevant context. As a proof of concept, long-term MTS cultures with uniform structure were realized on the microfluidic based platform. The response of doxorubicin and paclitaxel on different types of spheroids were simultaneously performed by in situ Live/Dead fluorescence stain to provide spatial distribution of dead cells as well as cytotoxicity information. In addition, the established platform combined with microplate reader was capable to determine the cytotoxicity of different sized MTS, showing a more powerful tool than cell staining examination at the end-point of assay. The HCT116 spheroids were then lysed on chip followed by signaling transduction pathway analysis. To our knowledge, the on chip drug screening study is the first to address the drug susceptibility testing and the offline detailed drug signaling pathway analysis combination on one system. Thus, this novel microfluidic platform provides a useful tool for drug screening with tumor spheroids, which is crucial for drug discovery and development.

Abstract:Three-dimensional (3D) hydrogel microspheres have aroused increasing attention as an in vitro cell culture model. Yet the preservation of cells' original biological properties has been overlooked during model construction. Here we present an integrated microfluidic device to accomplish the overall process including cell-laden microsphere generation, online extraction, and dynamic-culture. The method extends the noninvasive and nonsuppression capabilities of the droplet preparation system and provides a constant microenvironment, which reduces intracellular oxidative stress damage and the accumulation of mitochondria. Compared to the conventional preparation method, the coculture model of tumor-endothelial construction on an integrated platform displays high-level angiogenic protein expression. We believe that this versatile and biocompatible platform will provide a more reliable analysis tool for tissue engineering and cancer therapy.

Abstract:Developing a bio-functional model in vitro to study cancer resistance, which is a big challenge for clinical cancer therapy, is of great interest. Such reliable model requires appropriate drug diffusion kinetics simulation and a microenvironment that allows cell-cell and cell-matrix interactions. In this work, a special hydrogel-based three-dimensional (3D) microfluidic chip was constructed to simulate tumour-vascular microenvironment. The self-healing hydrogel supports long-time cell survival and proliferation, effective cellular metabolism of cancer drugs and cell-cell interaction between different types of cells. In the effective near-physiological tumour-vascular microenvironment, the endothelial and fibroblast cells are spread on different sides of a porous membrane, while sensitive and resistant breast tumour cells are separately cultured in the dynamic hydrogel consisting of glycol chitosan and telechelic difunctional poly (ethylene glycol) in the upper chambers. Nutrients and drugs are introduced through the bottom channel and diffuse into the cancer cells. Doxorubicin molecules pass first through blood vessel endothelial cells and act on the tumour cells surrounded by fibroblasts. Tumour cells respond differently to drug when they are cultured in the microenvironment. Sensitive breast tumour cells have a 47% increase in viability than those cultured without fibroblasts and endothelial cells. Both sensitive and resistant tumour cells can be analysed under the same chemical environment. This work represents a multi-functional in vitro platform that allows near-physiological simulation, effective drug metabolism and cellular response to extracellular stimuli and has great potential to make drug discovery speedy and precise.

Abstract:Cell-based assays can be used prior to in vivo tests to enable quantitative examinations of the responses of cells and, on the tissue scale, tumor-stromal mimics to drugs. However, the challenge is always to reproduce as closely as possible the native-state tumor microenvironment so as to accurately predict in vivo drug responses. In this chapter, we will list the advantages of 3D models over conventional 2D culture, review the current progresses in developing anti-cancer drug tests based on 3D in vitro culture systems, and discuss the importance of mechanics in designing these 3D models. We will highlight various techniques that have been used to construct 3D models that reflect the native physiology/pathology of a solid tumor as closely as possible in order to evaluate these drug effects, and detail specific research questions addressed using these 3D models. We will conclude this review with an outlook of the future of 3D models for improved in vitro drug assays.

Abstract:Traditional monolayer cell cultures often fail to accurately predict the anticancer activity of drug candidates, as they do not recapitulate the natural microenvironment. Recently, three-dimensional (3D) culture systems have been increasingly applied to cancer research and drug screening. Materials with good biocompatibility are crucial to create a 3D tumor microenvironment involved in such systems. In this study, natural silk fibroin (SF) and chitosan (CS) were selected as the raw materials to fabricate 3D microscaffolds; Besides, sodium tripolyphosphate (TPP), and 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC) were used as cross-linking agents. The physicochemical properties of obtained scaffolds were characterized with kinds of testing methods, including emission scanning electron microscopy, x-ray photoelectron spectroscopy, fourier transform infrared spectroscopy, water absorption, and swelling ratio analysis. Cancer cell lines (LoVo and MDA-MB-231) were then seeded on scaffolds for biocompatibility examination and drug sensitivity tests. SEM results showed that EDC cross-linked scaffolds had smaller and more uniform pores with great interconnection than the TPP cross-linked scaffolds, and the EDC cross-linked scaffold exhibited a water absorption ratio around 1000% and a swelling ratio of about 72%. These spatial structures and physical properties could provide more adhesion sites and sufficient nutrients for cell growth. Moreover, both LoVo and MDA-MB-231 cells cultured on the EDC cross-linked scaffold exhibited good adhesion and spreading. CCK8 results showed that increased chemotherapeutic drug sensitivity was observed in 3D culture compared with 2D culture, particularly in the condition of low drug dose (<1 μ M). The proposed SF/CS microscaffold can provide a promising in vitro platform for the efficacy prediction and sensitivity screening of anticancer drugs.

Abstract:Multi-drug combinations therapy is a promising method in the fight against cancer cells. Multi-drug combinations cannot be practically elucidated using conventional devices owing to structural limitations or operational complexity. 3D printing has recently attracted attention as a way to fabricate complicated and interconnected microfluidic channels, which is of design flexibility. Here, an easy-to-use, high-throughput, multi-drug combinations 3D printed microfluidic chips were brought forward. 36 discrete concentration combinations have been generated by repeated splitting-and-mixing of four inlet drugs in interconnected network channels, which are used to determine the optimal concentration of inhibition of cancer cells survival. We evaluate the potential to advance multi-drug combinations efficiency by optimizing the geometric parameters of tree-shaped branch unit, as well as the chip performance by dye visualization. The microfluidic chip was further applied to study multidrug combinations responses, and the 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt (MTS) assay was employed to test the cytotoxic effect of four drugs combinations on A549. The experiment results demonstrated clinically relevant antagonistic, synergistic interactions between the four different antitumor drugs. Moreover, this chip has outstanding features, e.g., more compact structure, more accurate combination of concentrations, greater space for scalability. As such, it will be an effective multi-drug combinations screening platform for applications in biomedical research and clinical medicine.

Abstract:Current fundamental investigations of human biology and the development of therapeutic drugs commonly rely on 2D monolayer cell culture systems. However, 2D cell culture systems do not accurately recapitulate the structure, function or physiology of living tissues, nor the highly complex and dynamic 3D environments in vivo. Microfluidic technology can provide microscale complex structures and well-controlled parameters to mimic the in vivo environment of cells. The combination of microfluidic technology with 3D cell culture offers great potential for in vivo-like tissue-based applications, such as the emerging organ-on-a-chip system. This article will review recent advances in the microfluidic technology for 3D cell culture and their biological applications.

A Perfused Two-Chamber System For Anticancer Drug Screening

Abstract:A cell culture microfluidic device has been developed to test the cytotoxicity of anticancer drugs while reproducing multi-organ interactions in vitro. Cells were cultured in separate chambers representing the liver and tumor. The two chambers were connected through a channel to mimick the blood flow. Glioblastoma (GBM) cancer cells (M059K) and hepatoma cells (HepG2) were cultured in the tumor and the liver chambers, respectively. The cytotoxic effect of cancer treatment drug Temolozomide (TMZ) was tested using this two chamber system. The experimental results showed that with the liver cells, the cancer cells showed much higher viability than those without the liver cells. This indicates that the liver metabolism has strong effect on the toxicity of the anticancer drug. The results demonstrated that the perfused two chamber cell culture system has the potential to be used as a platform for drug screening in a more physiologically realistic environment.

Abstract:Tumor metastasis presents a formidable challenge in cancer treatment, necessitating effective tools for anti-cancer drug development. Conventional 2D cell culture methods, while considered the "gold standard" for invasive studies, exhibit limitations in representing cancer hallmarks and phenotypes. This study proposes an innovative approach that combines the advantages of 3D tumor spheroid culture with impedance-based biosensing technologies to establish a high-throughput 3D cell invasion assay for anti-metastasis drug screening through multicellular tumor spheroids. In addition, the xCELLigence device is employed to monitor the time-dependent kinetics of cell behavior, including attachment and invasion out of the 3D matrix. Moreover, an iron chelator (deferoxamine) is employed to monitor the inhibition of epithelial–mesenchymal transition in 3D spheroids across different tumor cell types. The above results indicate that our integrated 3D cell invasion assay with impedance-based sensing could be a promising tool for enhancing the quality of the drug development pipeline by providing a robust platform for predicting the efficacy and safety of anti-metastatic drugs before advancing into preclinical or clinical trials.

Abstract:Three-dimensional (3D) tumor spheroids offer unprecedented capability for drug screening due to their unique features such as spatial 3D structure, relevant physiological responses, more complex intercellular network and stroma-cancer cell interactions. Microfluidic technology provides a facile strategy to make uniform tumor spheroids with potential of high-throughput production. In this article, we develop a microfluidic approach to produce core-shell alginate particles, which allows the separate confinement of different cells in the core and shell structure. To reconstitute the complex tumor structure, we encapsulated tumor cells in the core and stromal fibroblast cells in the shell. These co-culture tumor spheroids were applied for drug evaluation showing similar drug resistance as those prepared using conventional methods in well plates. Abstract:Compared to the conventional two-dimensional (2D) culture,in vitro three-dimensional cell culture (3DCC) can mimic the microenvironment more exactly.Compared to in vivo animal models,3DCC is less time-consuming and can be real-time recorded.Thus,3DCC fills the gap between 2D culture and animal models,which is used for oncologic research quickly.In 3DCC,the characteristics of cancer cells,such as morphology,the capability of invasion and migration and the resistance to chemo-radiotherapy,are quite different from those in 2D culture.The development of 3DCC technology provides a platform for oncology,organization engineering and gene or drug screening.
