Tissue distribution, elimination of florfenicol and its effect on metabolic enzymes and related genes expression in the white shrimp Litopenaeus vannamei following oral administration

Abstract:Florfenicol (FLR) is a commonly used antibiotic for the treatment of bacterial diseases in white shrimp Litopenaeus vannamei. In this study, we evaluated the tissue distribution and elimination, changes of selected metabolism enzymes and transcriptional activity of metabolism-related genes following oral administration of FLR. Shrimp (9.5 +/- 0.5 cm) were fed coated with FLR 100 mg kg(-1) and 200 mg kg(-1) body weight for 6 days followed by a period of feeding without antibiotic for 10 days to evaluate the elimination pattern. Gill, hepatopancreas and muscle tissues were collected on the 3rd, 6th day of feeding the antibiotics and at 3, 5 and 10 days after FLR withdrawal. The concentration of FLR residue in hepatopancreas was significantly higher than that in other tissues. Ethoxyresorufin-O-deethylase (EROD) and glutathione-S-transferase (GST) activities were significantly increased in gill and hepatopancreas of FLR-fed shrimp until the 6th day post withdrawal. Furthermore, transcriptional profiles of selected genes (CYP1A1, CYP3, CYP4, GST) were dependent on the dosage of antibiotics that was administered and the time of sampling. Results in this study were expected to provide additional contribution to the emerging field of crustaceans in understanding the pharmacokinetic and the metabolic progress of L. vannamei following oral administration.
