Abstract:A water culture experiment with 1/2 Hoagland solution was carried out to study the effects of exogenous 24-epibrassinolide (EBR) on the leaf photosynthetic characteristics and polyamines (PAs) content of cucumber seedlings under hypoxia stress. Under the stress, the leaf net photosynthetic rate (Pn), stomatal conductance (g(s)), transpiration rate (Tr), and intercellular CO2 concentration (Ci) decreased significantly while the chlorophyll content was in adverse, and the seedling growth was inhibited. Hypoxia stress increased the leaf Put, Spd, and Spm contents and Put/PAs ratio significantly, but decreased the (Spd+Spm)/Put. Under the stress, exogenous EBR not only increased the Pn, g(s), Tr, and chlorophyll content significantly, but also increased the contents of free Spm, conjugated Spd and Spm, and bound Put, Spd, and Spm significantly. Meanwhile, the PAs content and (Spd + Spm)/Put ratio were further increased, and the Put/PAs ratio decreased. The results suggested that exogenous EBR could regulate the formation and form transformation of PAs in cucumber seedling leaf, made the leaf keep a higher photosynthetic performance, promote the increase of leaf area and dry mass, and alleviate the damage of hypoxia stress.

Abstract:Key messageThe application of exogenous 24-epibrassinolide promotes Brassinosteroids intracellular signalling in cucumber, which leads to differentially expressed proteins that participate in different life process to relieve Ca(NO3)2damage.Abstract NO3− and Ca2+ are the main anion and cation of soil secondary salinization during greenhouse cultivation. Brassinosteroids (BRs), steroidal phytohormones, regulate various important physiological and developmental processes and are used against abiotic stress. A two-dimensional electrophoresis gel coupled with MALDI-TOF/TOF MS was performed to investigate the effects of exogenous 24-epibrassinolide (EBL) on proteomic changes in cucumber seedling roots under Ca(NO3)2 stress. A total of 80 differentially accumulated protein spots in response to stress and/or exogenous EBL were identified and grouped into different categories of biological processes according to Gene Ontology. Under Ca(NO3)2 stress, proteins related to nitrogen metabolism and lignin biosynthesis were induced, while those related to cytoskeleton organization and cell-wall neutral sugar metabolism were inhibited. However, the accumulation of abundant proteins involved in protein modification and degradation, defence mechanisms against antioxidation and detoxification and lignin biosynthesis by exogenous EBL might play important roles in salt tolerance. Real-time quantitative PCR was performed to investigate BR signalling. BR signalling was induced intracellularly under Ca(NO3)2 stress. Exogenous EBL can alleviate the root indices, effectively reduce the Ca2+ content and increase the K+ content in cucumber roots under Ca(NO3)2 stress. This study revealed the differentially expressed proteins and BR signalling-associated mRNAs induced by EBL in cucumber seedling roots under Ca(NO3)2 stress, providing a better understanding of EBL-induced salt resistance in cucumber seedlings. The mechanism for alleviation provides valuable insight into improving Ca(NO3)2 stress tolerance of other horticultural plants.

Abstract:Phytohormone biosynthesis and accumulation are essential for plant growth and development and stress responses. Here, we investigated the effects of 24-epibrassinolide (EBR) on physiological and biochemical mechanisms in cucumber leaves under low-temperature stress. The cucumber seedlings were exposed to treatments as follows: NT (normal temperature, 26 °C/18 °C day/night), and three low-temperature (12 °C/8 °C day/night) treatments: CK (low-temperature stress); EBR (low-temperature and 0.1 μM EBR); and BZR (low-temperature and 4 μM BZR, a specific EBR biosynthesis inhibitor). The results indicated that low-temperature stress proportionately decreased cucumber seedling growth and the strong seedling index, chlorophyll (Chl) content, photosynthetic capacity, and antioxidant enzyme activities, while increasing reactive oxygen species (ROS) and malondialdehyde (MDA) contents, hormone levels, and EBR biosynthesis gene expression level. However, EBR treatments significantly enhanced cucumber seedling growth and the strong seedling index, chlorophyll content, photosynthetic capacity, activities of antioxidant enzymes, the cell membrane stability, and endogenous hormones, and upregulated EBR biosynthesis gene expression level, while decreasing ROS and the MDA content. Based on these results, it can be concluded that exogenous EBR regulates endogenous hormones by activating at the transcript level EBR biosynthetic genes, which increases antioxidant enzyme capacity levels and reduces the overproduction of ROS and MDA, protecting chlorophyll and photosynthetic machinery, thus improving cucumber seedling growth.

The Effects of Exogenous 2,4-Epibrassinolide on the Germination of Cucumber Seeds under NaHCO3 Stress

Abstract:This investigation focused on the suppressive impact of varying NaHCO3 concentrations on cucumber seed germination and the ameliorative effects of 2,4-Epibrassinolide (EBR). The findings revealed a negative correlation between NaHCO3 concentration and cucumber seed germination, with increased NaHCO3 concentrations leading to a notable decline in germination. Crucially, the application of exogenous EBR significantly counteracted this inhibition, effectively enhancing germination rates and seed vigor. Exogenous EBR was observed to substantially elevate the activities of superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD), thereby mitigating oxidative damage triggered under NaHCO3 stress conditions. Additionally, EBR improved enzyme activity under alkaline stress conditions and reduced starch content in the seeds. Pertinently, EBR upregulated genes that were associated with gibberellin (GA) synthesis (GA20ox and GA3ox), and downregulated genes that were linked to abscisic acid (ABA) synthesis (NCED1 and NCED2). This led to an elevation in GA3 concentration and a reduction in ABA concentration within the cucumber seeds. Therefore, this study elucidates that alleviating oxidative stress, promoting starch catabolism, and regulating the GA and ABA balance are key mechanisms through which exogenous EBR mitigates the suppression of cucumber seed germination resulting from alkaline stress.

Abstract:To explore the role of exogenous BR on the induced resistance of cucumber seedlings to Ca( NO3)2stress,the effects of different treatments of exogenous BR( 0. 01 mg·L- 1BR soaking seeds,0. 10 mg·L- 1BR spraying leaves and combination of both) on growth,chlorophyll fluorescence indexes,and ultrastructure of cucumber seedlings leaves were determined under 60 mmol·L- 1Ca( NO3)2stress in this paper. The results showed that three treatments of exogenous BR could induced resistance of cucumber seedlings to Ca( NO3)2stress,but the effects were different. The treatment of BR soaking seeds showed the best results,which alleviated the inhibition of Ca( NO3)2stress,kept the number and arrangement uniformity of grana and stroma lamella,and maintain the structural of mitochondria complete. The arrangement of grana lamellae disordered and thylakoid became wrinkled,after BR spraying leaves and BR soaking seeds + spraying leaves of cucumber seedlings under Ca( NO3)2stress. The results indicated that exogenous BR could enhance the resistance of cucumber seedlings to Ca( NO3)2stress by protecting the structural integrity of the organelle and the function of PSⅡ reaction center.

Abstract:The effects of 0.1 μM 24-epibrassinolide (EBL) on plant growth (plant height, leaf area, fresh weight, and dry weight), chlorophyll content, photosynthetic characteristics, antioxidant enzymes, and chloroplast ultrastructure were investigated using cucumber seedlings (Cucumis sativus L. cv. Jinyou No. 4) with 80 mM Ca(NO3)2 to induce stress. The presence of Ca(NO3)2 caused significant reductions in net photosynthetic rate (PN), stomatal conductance (Gs), intercellular CO2 concentration (Ci), and transpiration rate (Tr) of leaves. In addition, Ca(NO3)2 markedly reduced the chlorophyll content and inhibited photochemical activity, including the actual photochemical efficiency (ΦPSII). In contrast, EBL increased the chlorophyll content, especially chlorophyll b, and minimized the harmful effects on photosynthesis caused by the Ca(NO3)2. The application of EBL to the plants subjected to Ca(NO3)2-enhanced photochemical activity. EBL protected the photosynthetic membrane system from oxidative damage due to up-regulating the capacity of the antioxidant systems. Microscopic analyses revealed that Ca(NO3)2 affected the structure of the photosynthetic apparatus and membrane system and induced damage of granal thylakoid layers, while EBL recovered the typical shape of chloroplasts and promoted the formation of grana. Taken together, EBL compensated for damage/losses by Ca(NO3)2 due to the regulation of photosynthetic characteristics and the antioxidant system.

Abstract:We studied the effects of exogenous spermidine(Spd) on free polyamines content and polyamine biosynthesis gene expression in cucumber(Cucumis sativus L.cv.Jinchun No.2) seedlings under salt stress with solution culture.Plant height,stem diameter,and fresh and dry weight were significantly reduced by treatment with 75 mmol/L NaCl,while the inhibition of seedling growth was alleviated by spraying 1 mmol/L Spd.Free polyamine content was significantly increased in leaves when exposed to salt stress,and exogenous Spd application to leaves further promoted free Spd and spermine(Spm),but reduced free putrescine(Put).Expression analysis of polyamine synthase genes showed that salt stress up-regulated adc,odc,samdc and spds gene expression,and exogenous Spd application further up-regulated samdc gene expression and down-regulated adc,odc,spds gene expression.The results revealed that exogenous Spd regulated polyamine metabolism in cucumber seedlings under salt stress,inhibited free Put accumulation by reducing adc,odc gene expression,increased free Spd and Spm accumulation by up-regulated samdc gene expression,and alleviated the inhibition of plant growth under salt stress.
Exogenous brassinosteroid induced the salt resistance of cucumber （Cucumis sativus L.） seedlings

Abstract:【Objective】To ascertain the induced role of exogenous brassinosteroid (BR) on the salt resistance of cucumber (Cucumis sativus L.) seedling. 【Method】Different BR concentrations (0, 0.005, 0.01, 0.05, 0.1 and 0.2 mg·L-1) were applied to the seedling by root injection and foliar spray under salt stress. Physiological parameters were then determined. 【Result】The result of exogenous BR application indicated that the BR distinctly lowered the salt injured index of seedlings by 40.2 percent point, significantly increased the contents of free-proline and soluble sugar and the activity of antioxidative enzymes ( such as superoxide dismutase,peroxidase and catalase ) in seedling leaves (P0.01), and thereby effectively protected membrane from salt stress. 【Conclusion】 The exogenous BR has an important role to induce salt resistance of cucumber seedlings and the 0.01–0.05 mg·L-1 concentration range is considered the optimum density scope.

A Role for Brassinosteroids in the Regulation of Photosynthesis in Cucumis Sativus

Abstract:The effects of 24-epibrassinolide (EBR) spray application on gas-exchange, chlorophyll fluorescence characteristics, Rubisco activity, and carbohydrate metabolism were investigated in cucumber (Cucumis sativus L. cv. Jinchun No. 3) plants grown in a greenhouse. EBR significantly increased the light-saturated net CO(2) assimilation rate (A(sat)) from 3 h to 7d after spraying, with 0.1 mg l(-1) EBR proving most effective. Increased A(sat) in EBR-treated leaves was accompanied by increases in the maximum carboxylation rate of Rubisco (V(c,max)) and in the maximum rate of RuBP regeneration (J(max)). EBR-treated leaves also had a higher quantum yield of PSII electron transport (phi(PSII)) than the controls, which was mainly due to a significant increase in the photochemical quenching (q(P)), with no change in the efficiency of energy capture by open PSII reaction centres (F'(v)/F'(m)). EBR did not influence photorespiration. In addition, significant increases in the initial activity of Rubisco and in the sucrose, soluble sugars, and starch contents were observed followed by substantial increases in sucrose phosphate synthase (SPS), sucrose synthase (SS), and acid invertase (AI) activities after EBR treatment. It was concluded that EBR increases the capacity of CO(2) assimilation in the Calvin cycle, which was mainly attributed to an increase in the initial activity of Rubisco.
24-Epibrassinolide promotes NO3− and NH4+ ion flux rate and NRT1 gene expression in cucumber under suboptimal root zone temperature

Abstract:<h3>Background</h3>Suboptimal root zone temperature (RZT) causes a remarkable reduction in growth of horticultural crops during winter cultivation under greenhouse production. However, limited information is available on the effects of suboptimal RZT on nitrogen (N) metabolism in cucumber seedlings. The aim of this study is to investigate the effects of 24-Epibrassinolide (EBR) on nitrate and ammonium flux rate, N metabolism, and transcript levels of NRT1 family genes under suboptimal RZT in cucumber seedlings.<h3>Results</h3>Suboptimal RZT (LT) negatively affected on cucumber growth and proportionately decreased EBR contents, bleeding rate, root activity, enzyme activities of nitrate reductase (NR), nitrite reductase (NiR), glutamine synthetase (GS), and glutamate synthase (GOGAT), nitrate (NO3−) influx rate, ammonium (NH4+) efflux rate, and transcript levels of nitrate transporter (NRT1) encoding genes. However, exogenous EBR reduced the harmful effects of suboptimal RZT and increased endogenous EBR contents, bleeding rate, root activity, enzyme activities of NR, NiR, GS, and GOGAT, NH4+ and NO3− flux rates and contents, and N accumulation. EBR-treated seedlings also upregulated the transcript levels of nitrate transporters CsNRT1.1, CsNRT1.2A, CsNRT1.2B, CsNRT1.2C, CsNRT1.3, CsNRT1.4A, CsNRT1.5B, CsNRT1.5C, CsNRT1.9, and CsNRT1.10, and downregulated CsNRT1.5A and CsNRT1.8. LT treatment upregulated the expression level of CsNRT1.5A, while exogenous BZR application downregulated the expression level of NRT1 genes.<h3>Conclusion</h3>These results indicate that exogenous application of EBR alleviated the harmful effects of suboptimal RZT through changes in N metabolism, NH4+ and NO3− flux rates, and NRT1 gene expression, leading to improved cucumber seedlings growth. Our study provides the first evidence of the role of EBR in the response to suboptimal RZT in cucumber, and can be used to improve vegetable production.

Exogenous Salicylic Acid Alleviates Salt Stress by Improving Leaf Photosynthesis and Root System Architecture in Cucumber Seedlings

Abstract:Exogenous salicylic acid (SA) can alleviate salt stress in plants. Previous researches have been focused on SA's role in leaf morphological and physiological characteristics, however, little information is available on its role in both leaf photosynthesis and root system architecture (RSA). A hydroponic study was conducted to analyze biomass, leaf photosynthesis, RSA parameters and expression of RSA genes in cucumber seedlings grown under four treatments, including control (CK), 50 mM NaCl (NaCl), 0.3 mM SA (SA) and 50 mM NaCl plus 0.3 mM SA (NaCl + SA). The results showed that compared with CK, both NaCl and SA treatments severely reduced relative growth rate (RGR) and leaf photosynthetic parameters (F-v/F-m, Phi(II) and Pn). However, compared with NaCl treatment, NaCl + SA treatment significantly increased RGR, leaf photosynthetic (F-v/F-m, Phi(II) and Pn) and RSA (total root length, root surface area, number and total length of primary and secondary LR) parameters. In addition, compared with CK, although both NaCl and SA treatments down-regulated expression of GL2 gene and up-regulated expression of RHD2 gene, NaCl + SA treatment notably up-regulated expression of NAC1, NAC2, GL2, EXP and down-regulated expression of RHD2. It is concluded that exogenous SA alleviated salt stress via effectively promoting leaf photosynthesis and RSA in cucumber seedlings. SA-mediated increment in expression of RSA genes, total length and number of primary and secondary LR could contribute to the improved RSA.

Reactive oxygen species are involved in brassinosteroid-induced stress tolerance in cucumber

Abstract:Brassinosteroids (BRs) induce plant tolerance to a wide spectrum of stresses. To study how BR induces stress tolerance, we manipulated the BR levels in cucumber (Cucumis sativus) through a chemical genetics approach and found that BR levels were positively correlated with the tolerance to photo-oxidative and cold stresses and resistance to Cucumber mosaic virus. We also showed that BR treatment enhanced NADPH oxidase activity and elevated H(2)O(2) levels in apoplast. H(2)O(2) levels were elevated as early as 3 h and returned to basal levels 3 d after BR treatment. BR-induced H(2)O(2) accumulation was accompanied by increased tolerance to oxidative stress. Inhibition of NADPH oxidase and chemical scavenging of H(2)O(2) reduced BR-induced oxidative and cold tolerance and defense gene expression. BR treatment induced expression of both regulatory genes, such as RBOH, MAPK1, and MAPK3, and genes involved in defense and antioxidant responses. These results strongly suggest that elevated H(2)O(2) levels resulting from enhanced NADPH oxidase activity are involved in the BR-induced stress tolerance.
24-Epibrassinolide Alleviates Organic Pollutants-Retarded Root Elongation by Promoting Redox Homeostasis and Secondary Metabolism in Cucumis Sativus L.

Abstract:Environmental pollution by organic pollutants (OPs) has become a global concern due to its detrimental effects on the environment and human health. As plants are used to remediate contaminated sites, understanding the responses of plants to various OPs and fortification of plant tolerance are of great significance. In this work, we studied the biochemical and molecular responses of cucumber plants to three well-known OPs, 2,4,6-trichlorophenol, chlorpyrifos and oxytetracycline in the absence or presence of 24-epibrassinolide (EBR), a potent regulator of plant growth and stress tolerance. The results showed that the selected three OPs retarded root elongation; however, the phytotoxic effects of OPs were attenuated by exogenous EBR. OPs induced accumulations of both hydrogen peroxide (H2O2) and nitric oxide (NO) in root tips and resulted in an increased malondialdehyde (MDA) content, an indicator of membrane lipid peroxidation. Exogenous EBR reduced accumulations of H2O2, NO and MDA in the roots by increasing the expression of antioxidant and detoxification genes and the activities of the corresponding enzymes. Intriguingly, EBR not only promoted the activities of glutathione S-transferase and glutathione reductase, but also increased the content of reduced glutathione without altering the content of oxidized glutathione, which resulted in a reduced redox state under OPs stress. Furthermore, EBR increased the free radical scavenging capacity, flavonoid content and the activity and transcription of secondary metabolism related enzymes. Our results suggest that EBR treatment may fortify secondary metabolism to enhance antioxidant capacity in response to OPs treatment, which might have potential implication in phytoremediation of OPs. Abstract:The present experiment was designed to assess the effects of seed soaking with 24-epibrassinolide (EBR) on the physiology of Brassica juncea L. seedlings grown under imidacloprid (IMI) toxicity. Application of EBR increased the length of seedlings, dry weight, and pigment contents, polyphenols, total phenols, and organic acids under IMI toxicity. The expression of genes coding key enzymes of pigment, phenols, polyphenols, and organic acid biosynthetic pathways was also studied including CHLASE (chlorophyllase), PSY (phytoene synthase), CHS (chalcone synthase) and PAL (phenylalanine ammonialyase), CS (citrate synthase), SUCLG1 (succinyl Co-A ligase,), SDH (succinate dehydrogenase), FH (fumarate hydratase), MS (malate synthase). Multiple linear regression (MLR) analysis revealed that IMI application regressed negatively on seedling length, dry weight and total chlorophyll content. However, EBR seed treatment regressed positively on all the parameters studied. Moreover, interaction between IMI and EBR showed positive regression for growth parameters, content of pigments, total polyphenol, total phenol and malate, and expression of PSY and PAL. Negative interactions were noticed for the contents of fumarate, succinate and citrate, and expression of CHS and all genes studied related to organic acid metabolism. In conclusion, EBR enhanced the growth and contents of all studied metabolites by regulating the gene expression of B. juncea seedlings under IMI stress.

Abstract:To investigate the effects of exogenous Spd on proteomic changes under normal condition and NaCl stress of 3 days in cucumber seedling leaves, a 2-DE gel electrophoresis and MALDI-TOF/TOF MS was performed. A total of 63 differentially expressed proteins responded to salt stress or exogenous Spd treatments, and they were all successfully identified by MALDI-TOF/TOF MS. Many changes were observed in the levels of proteins involved in energy and metabolic pathways, protein metabolic, stress defense, and other functional proteins. Increased salt tolerance by exogenous Spd would contribute to higher expressions of proteins involved in the SAMs metabolism, protein biosynthesis, and defense mechanisms on antioxidant and detoxification. Meanwhile, the regulation of Calvin cycle, protein folding assembly and the inhibition of protein proteolysis by Spd might play important roles in salt tolerance. This study provides insight that may facilitate a better understanding of the salt resistance by Spd in cucumber seedlings.
Brassinosteroids-Induced Systemic Stress Tolerance Was Associated With Increased Transcripts Of Several Defence-Related Genes In The Phloem In Cucumis Sativus

Abstract:Brassinosteroids (BRs), a group of naturally occurring plant steroidal compounds, are essential for plant growth, development and stress tolerance. Recent studies showed that BRs could induce systemic tolerance to biotic and abiotic stresses; however, the molecular mechanisms by which BRs signals lead to responses in the whole plant are largely unknown. In this study, 24-epibrassinosteroid (EBR)-induced systemic tolerance in Cucumis sativus L. cv. Jinyan No. 4 was analyzed through the assessment of symptoms of photooxidative stress by chlorophyll fluorescence imaging pulse amplitude modulation. Expression of defense/stress related genes were induced in both treated local leaves and untreated systemic leaves by local EBR application. With the suppressive subtractive hybridization (SSH) library using cDNA from the phloem sap of EBR-treated plants as the tester and distilled water (DW)-treated plants as the driver, 14 transcripts out of 260 clones were identified. Quantitative Real Time-Polymerase Chain Reaction (RT-qPCR) validated the specific up-regulation of these transcripts. Of the differentially expressed transcripts with known functions, transcripts for the selected four cDNAs, which encode an auxin-responsive protein (IAA14), a putative ankyrin-repeat protein, an F-box protein (PP2), and a major latex, pathogenesis-related (MLP)-like protein, were induced in local leaves, systemic leaves and roots after foliar application of EBR onto mature leaves. Our results demonstrated that EBR-induced systemic tolerance is accompanied with increased transcript of genes in the defense response in other organs. The potential role of phloem mRNAs as signaling components in mediating BR-regulated systemic resistance is discussed.

Abstract:Salt stress is a major abiotic factor that negatively impacts the growth, performance, and secondary metabolite production in pepper (Capsicum annuum L.) plants. Brassinosteroids (BRs) play a crucial role in enhancing plant tolerance to abiotic stress, yet their potential in mitigating salt stress in pepper plants, particularly by promoting sugar and glycine betaine accumulation, remains underexplored. In this study, we investigated the effects of the foliar application of 2,4-epibrassinolide (EBR) on salt-stressed pepper seedlings. Our findings revealed that EBR treatment significantly increased the levels of proline, sugar, and glycine betaine under salt stress compared to untreated controls. Moreover, EBR enhanced the antioxidant defense mechanisms in pepper seedlings by increasing sugar and glycine betaine levels, which contributed to the reduction of reactive oxygen species (ROS) and malondialdehyde (MDA) accumulation.

Abstract:Strigolactones (SLs) play crucial roles in plant growth and development and during the response to multiple biotic and abiotic stresses. In this study, we investigated the role of exogenous SLs analog GR24 in amelioration of salt damage and analyzed the interaction between GR24 and other signaling molecules in cucumber seedlings under salt stress by using physiological, biochemical and RNA-seq analysis approaches. Higher chlorophyll contents, stomatal conductance, greater efficiency of photosynthetic were observed in GR24-pretreated plants than in non- GR24-pretreated plants under salt stress. In addition, exogenous GR24 pretreatment can also alleviate salt-induced photodamage, enhance the efficiency of ascorbate-glutathione (AsA-GSH) cycle, and scavenge excessive reactive oxygen species (ROS), thus alleviating oxidative stress under salt stress in cucumber. RNA-seq analysis revealed that there were 1678 differentially expressed genes (DEGs) in D (NaCl)_vs_LD (GR24+NaCl), including 1219 up-regulated genes and 459 down-regulated genes. There were 66 DEGs related to photosynthesis, MAPK cascade pathways, oxidation and antioxidant system screened by GO classification and KEGG enrichment analysis, and the expression levels of these genes were consistent with the changes of physiological parameters. However, the effects of GR24 on the photosynthetic of cucumber seedlings under salt stress, as well as the expression levels of MAPK3/4/6 genes were significantly inhibited by diphenyleneiodonium chloride (DPI, NADPH oxidase inhibitor) or dimethylthiourea (DMTU, H2O2 scavenger). These findings suggested that H2O2 and MAPK cascade pathways might play a positive role in exogenous GR24 alleviating salt stress in cucumber seedlings. In conclusion this study provides insights for further investigating the mechanism of SLs in alleviating adversity stress.

Abstract:Zinc (Zn) is an important element in plants, but over-accumulation of Zn is harmful. It is well-known that brassinolide (BR) plays a key role in the regulation of abiotic stress responses in plants. However, the effects of brassinolide on alleviating Zn phytotoxicity in watermelon ( Citrullus lanatus L.) seedlings are not clear. The purpose of this study was to study the effect of 24-epibrassinolide (EBR, one of the bioactive BRs) on Zn tolerance of watermelon seedlings and its potential resistance mechanism. Exposure to excessive Zn significantly inhibited shoot and root fresh weight of watermelon, but this could be significantly alleviated by the optimum 0.05 μM EBR. Exogenous spraying EBR increased the pigments and alleviated oxidative damage caused by Zn through reducing Zn accumulation and the levels of reactive oxygen species (ROS) and malonaldehyde (MDA) and increasing the activities of antioxidant enzymes and contents of ascorbic acid (AsA) and glutathione (GSH). Importantly, the relative mRNA levels of antioxidant genesincluding Cu/Zn-superoxidedismutase ( Cu-Zn SOD ), catalase ( CAT ), ascorbic acid peroxidase ( APX ), and glutathione reductase ( GR ) were significantly induced after EBR treatment. In addition, EBR pre-treatment induced lignin accumulation under Zn stress, and the activity of phenylalanine ammonia-lyase (PAL) and 4-coumaric ligase (4CL), two key enzymes regulating lignin synthesis, also tended to be consistent. Collectively, the present research proves the beneficial effects of EBR in response to Zn stress through enhancing antioxidant defense and lignin accumulation and provides a new insight into the mechanism of BR-enhancing heavy metal tolerance.
