Abstract:Many solid drugs exist in polycrystalline state, which markedly hinder their pharmacological effectiveness. Given the designability and biocompatibility, the development of peptide-based supramolecular gel that can regulate crystal growth of drug has a strong potential. In this research, the polycrystalline drug carbamazepine (CBZ) was successfully transformed from orthorhombic dihydrate into amorphous form through the introduction of 9-fluorenylmethoxycarbonyl diphenylalanine (FmocFF) gel and co-assembled supramolecular gel was obtained. The intermolecular mechanism of structural transformation was elucidated in detail by various spectral techniques. The dihydrate of CBZ was restrained through the inhibition of hydration of the NH 2 groups and the π-π stacking interaction among the aromatic rings of FmocFF and CBZ. Especially, principal component analysis (PCA), and two-dimensional correlation spectroscopy (2DCOS) techniques were conducted to analyze the time-dependent IR spectra, which indicated that the π-π stacking interaction mainly affected the FmocFF gel fiber elongation after co-assembly with CBZ. In addition, the FmocFF gel matrix significantly influenced the release behavior of CBZ in vitro, which providing theoretical guidance for the development of short-peptide assembly strategies in drug delivery materials.

Abstract:Carbamazepine (CBZ) is an anticonvulsant with very low water solubility, presenting as a white crystalline powder with poor mechanical properties and is hard to bend. To enhance CBZ's physicochemical properties, such as water solubility and mechanical properties, we selected six cocrystal coformers (CCFs): nicotinamide (NIC), benzamide (BZM), salicylic acid (SCA), fumaric acid (FMA), trimesic acid (TMA) and hesperetin (HPE). Six CBZ cocrystals were successfully prepared using the solution method. Fourier transform infrared spectroscopy (FT-IR), powder X-ray diffraction (PXRD), differential scanning calorimetry (DSC) and single crystal X-ray diffraction (SCXRD), were used to characterize the crystal structures and gain comprehensive insights into the six cocrystals. The mechanical, fluorescence and solubility properties of the six cocrystals were tested. The results reveal that most of the prepared cocrystals exhibit improved water solubility and mechanical properties when compared to CBZ. Among them, the dissolution rate of cocrystals excluded CBZ-HPE has increased by an average of 3 or 4 times compared to CBZ, while CBZ-HPE exhibit superior mechanical properties. Moreover, all the six cocrystals possess better fluorescence performance than CBZ. We thoroughly evaluated the mechanical properties of the cocrystals through both experimental and theoretical approaches. This work provides a new direction for studying drug cocrystals to improve the physicochemical properties of drugs.

Abstract:Carbamazepine (CBZ), a Biopharmaceutics Classification System (BCS) class II drug, is widely-used as a first line antiepileptic drug. Numbers of cocrystals have reported to regulate its poor solubility. However, the solubility behaviors are rarely discussed on theoretical calculations and phase solubility diagrams. In this study, two new drug-drug cocrystals (DDC) of CBZ with emodin (EMO, 1: 1) and paeonol (PAE, 1: 1) as conformers, which offer the advantages of synergetic therapy and patient compliance are reported for the first time. The solubilities in water and in pH = 1.2, 6.8 buffer solutions, demonstrating that the rank order is as follows: CBZ-PAE > CBZ > CBZ-EMO. CBZ-PAE presents the biggest solubility at 0.563 ± 0.003 mmoL/L of in water among all the investigated samples. The solubilities of all the starting materials and cocrystals in six pure organic solvents such as methanol, ethanol, 2-propanol, acetone, acetonitrile, ethyl acetate were investigated to discuss the phase solubility diagrams. The results indicate that experimental solubilities are consistent with the theoretical solubilities on consideration of solubility product (Ksp) and solution binding constant (K11) together. The enthalpy differences between cocrystal and its coformer are calculated through the Perdew-Burke-Ernzerhof (PBE) functional with the projector augmented wave (PAW) pseudopotentials. CBZ-EMO presents the lowest enthalpy (−0.38 kcal/mol) with a highly unfavorable solvation energy (2.42 kcal/mol), resulting in unfavorable in an aqueous environment. Overall, the phase solubility diagrams and theoretical calculations support the solubility regulations on DDCs of CBZ.

Abstract:Developments of a drug delivery system (DDS) based on a natural supramolecular hydrogel have been of wide interest due to its biocompatibility, efficacy, and adjustable performance. However, a simple and efficient design of functional hydrogel DDS based on the templated interplay of gelator and model drug is still a challenge. In this work, natural glycyrrhetinic acid (GA) gel was selected as a carrier to encapsulate the model drug pyrazinamide (PZA). It was found that the carboxyl-amide interaction at the interface of gel-drug achieved polymorph control, stabilization, and pH-responsive release. Powder X-ray diffraction confirmed that the metastable γ form of PZA was obtained from the GA gel. Spectral analysis and molecular dynamics simulation showed that the protonation at the amide-O promoted the discretization of PZA molecules in solution, resulting in the polymorphism. Furthermore, the gel-drug interplay increased the stability of the γ form significantly from 2 days to 3 months by in situ encapsulation in the GA gel. In vitro release study indicated that the GA gel achieved targeted control release of PZA due to the pH-responsiveness property of GA. This work provides a promising option for hydrogel-based DDS design combined with polymorph control and stabilization.
Preparation and properties for magnetic/pH double sensitive hydrogel beads carrying carbamazepine

Abstract: BACKGROUND:The faults or defects in pharmaceutical dosage form designed for carbamazepine may lead to irregular drug absorption, great individual differences between the pharmacokinetics, narrow therapeutic concentration range, and the therapeutic drug monitoring for this drug. OBJECTIVE:To prepare a magnetic/pH double sensitive hydrogel beads carrying carbamazepine and to evaluate its properties. METHODS:Using chitosan, alginate and Fe3O4 nanoparticle as carrier materials, the magnetic/pH double sensitive hydrogel beads carrying anti-epileptic carbamazepine were prepared, and the composition and preparation technology were optimized by orthogonal test of L9(34). The surface morphology and structure of the hydrogel beads were characterized by scanning electron microscopy and Fourier transform infrared spectroscopy respectively. Also, the superparamagnetism, swel ing and release in vitro of hydrogel beads were determined. RESULTS AND CONCLUSION:The optimized preparation technology were described as:0.5%(w/v) chitosan, 1.5%(w/v) alginate, 2.0%(w/v) calcium chloride, and 1:2 ratio for magnetic versus carrier materials. The hydrogel beads under the optimal preparation conditions showed a round shape and smooth surface, and average encapsulation efficiency, loading efficiency and hydrogel beads diameter were 94.36%, 25.05%and 1-2 mm respectively. The hydrogel beads appeared to have superparamagnetism, the swel ing degrees were associated with pH value of medium, and the sequential release amount of carbamazepine from the hydrogel beads in simulated gastric fluid was 22.77%for 2 hours. Then, the beads were moved to the simulated intestinal fluid, and this value approached 91.63%for 24 hours. Experimental findings indicate that, the composition and preparation technology of magnetic/pH double sensitive hydrogel beads carrying carbamazepine was rational and feasible, and hydrogel beads show obvious pH sensitivity and magnetic sensitivety. The control ed-release effect of hydrogel beads in vitro is also good.

Abstract:In this study, the existing set of carbamazepine (CBZ) cocrystals was extended through the successful combination of the drug with the positional isomers of acetamidobenzoic acid. The structural and energetic features of the CBZ cocrystals with 3- and 4-acetamidobenzoic acids were elucidated via single-crystal X-ray diffraction followed by QTAIMC analysis. The ability of three fundamentally different virtual screening methods to predict the correct cocrystallization outcome for CBZ was assessed based on the new experimental results obtained in this study and data available in the literature. It was found that the hydrogen bond propensity model performed the worst in distinguishing positive and negative results of CBZ cocrystallization experiments with 87 coformers, attaining an accuracy value lower than random guessing. The method that utilizes molecular electrostatic potential maps and the machine learning approach named CCGNet exhibited comparable results in terms of prediction metrics, albeit the latter resulted in superior specificity and overall accuracy while requiring no time-consuming DFT computations. In addition, formation thermodynamic parameters for the newly obtained CBZ cocrystals with 3- and 4-acetamidobenzoic acids were evaluated using temperature dependences of the cocrystallization Gibbs energy. The cocrystallization reactions between CBZ and the selected coformers were found to be enthalpy-driven, with entropy terms being statistically different from zero. The observed difference in dissolution behavior of the cocrystals in aqueous media was thought to be caused by variations in their thermodynamic stability.

Abstract:Overall, the review paper discusses the scope of future work in the field of CBZ cocrystallization. It highlights the need for further investigations on the optimization of cocrystal synthesis, characterization techniques, and formulation strategies. Moreover, the development of innovative coformers, exploration of new cocrystallization techniques, and investigation of the in vivo performance of CBZ cocrystals are suggested as potential areas for future research. Carbamazepine (CBZ), a widely used antiepileptic drug, is known for its therapeutic efficacy but exhibits suboptimal physicochemical characteristics, including limited bioavailability, low solubility, and poor dissolution rates. In recent years, cocrystallization has emerged as a promising approach to improve these properties and enhance the overall performance of CBZ. This review paper presents a comprehensive exploration of the advances in CBZ cocrystals, focusing on their role in enhancing bioavailability, solubility/dissolution rates, morphology, and stability compared to raw CBZ. It delves into the in‐depth examination of computational techniques, such as molecular modelling and crystal structure prediction, that play a pivotal role in the rational design and prediction of CBZ cocrystals, thereby expediting the development process. Additionally, the morphological attributes of CBZ cocrystals are discussed, shedding light on how the unique crystal structures affect their physical properties. Stability is another critical aspect addressed in this review, encompassing thermal, physical, and chemical stability. The thermal behavior of CBZ cocrystals is analyzed through differential scanning calorimetry (DSC), and their crystal structures are characterized by techniques such as X‐ray diffraction (XRD) and Fourier‐transform infrared spectroscopy (FTIR). These analyses reveal the stability and structural changes of CBZ cocrystals under various conditions, providing valuable insights for formulation and storage. In summary, this comprehensive review paper amalgamates the latest advancements in CBZ cocrystals, demonstrating their capacity to significantly improve bioavailability, solubility, dissolution rates, and stability.

Abstract:Introduction of compartments with defined spaces inside a hydrogel network brings unique features, such as cargo quantification, stabilization and diminishment of burst release, which are all desired for biomedical applications. As a proof of concept, guest-modified cowpea chlorotic mottle virus (CCMV) particles and complementary guest-modified hydroxylpropyl cellulose (HPC) were non-covalently cross-linked through the formation of ternary host-guest complexes with cucurbit[8]uril (CB[8]). Furthermore, CCMV based virus-like particles (VLPs) loaded with tetrasulfonated zinc phthalocyanine (ZnPc) were prepared, with a loading efficiency up to 99%, which are subsequently successfully integrated inside the supramolecular hydrogel network. It was shown that compartments provided by protein cages not only help to quantify the loaded ZnPc cargo, but also improve the water solubility of ZnPc to avoid undesired aggregation. Moreover, the VLPs together with ZnPc cargo can be released in a controlled way without an initial burst release. The photodynamic effect of ZnPc molecules was retained after encapsulation of capsid protein and release from the hydrogel. This line of research suggests a new approach for sophisticated drug administration in supramolecular hydrogels.
A pH-responsive hydrogel system based on cellulose and dopamine with controlled hydrophobic drug delivery ability and long-term bacteriostatic property

Abstract:An interpenetrating double network hydrogel based on cellulose backbone and poly-dopamine dendritic structure was successfully prepared with the advantages of controlled hydrophobic drug loading, pH-responsive ability, and long-term bacteriostatic property. 13C-NMR was used to process the structural characterization and explain the mechanism of hydrogel system’s construction. Hydrophobic drug of ciprofloxacin with carbonyl group can evenly carry in the hydrogel system, and the highest loaded ratio of ciprofloxacin was reach to 80.56%. Besides, the swelling property and the controlled drug loading ability of our prepared hydrogel were carefully studied, which showed a close relation with the hydrogel design. Moreover, the in vitro drug release experiments demonstrated that the hydrogel had an obvious pH-responsive release property of ciprofloxacin, and the result of bacteriostatic experiment also showed a long-term bacteriostatic effect on Escherichia coli, which showed potential application on biological and medical fields, including wound treatment.Graphical abstract.

Abstract:A chiral supramolecular hydrogel (CSH) is designed for selective loading of chiral drugs and pH-responsive drug delivery. A derivative of phenylalanine is first synthesized as the gelator, which can be self-assembled to the CSH via intermolecular hydrogen bonding. The resultant CSH is composed of spiral nanofibers and exhibits reversible phase transition behavior (sol–gel) depending on heating/cooling process and pH values. More importantly, the CSH can be used for the selective loading of chiral drug naproxen (NPX) due to the stronger attractive interactions between CSH and S-NPX resulted from their same optical activity. Interestingly, the delivery of S-NPX from the CSH is pH-sensitive and the cumulative release of S-NPX is significantly enhanced in alkaline solutions (pH = 10.0), which might be ascribed to the broken hydrogen bonding and the consequently ruined nanostructure of the CSH under alkaline conditions.

Abstract:The choice of drug content is a critical factor as far as the solid dispersion is concerned. This investigation aims to build the relationship between the drug content, intermolecular hydrogen bonding and the crystalline of the carbamazepine-polyvinyl pyrrolidone solid dispersion. In this work, the microstructural changes of solid dispersions were investigated using experimental characterization combined with molecular simulation. Experimental investigations demonstrated that increasing the drug content enhances the intermolecular hydrogen bonding between drugs, resulting in the crystalline phase of the drug emerged in the solid dispersion. This negatively affects the solubility and stability of solid dispersions. Molecular simulations were then used to analyze the changes of intermolecular hydrogen bonding at different drug content in the system. It revealed a tenfold increase in drug-drug hydrogen bonding concentration as drug content elevated from 10% to 50%, while the drug-excipient hydrogen bonding concentration decreased by 45%. The correlation analysis proves the significant relationships among the drug content, intermolecular hydrogen bonding, and crystallinity of solid dispersion. Using polynomial fitting analysis, the quantitative relationships between the drug content and crystalline properties were investigated. This study will offer valuable insights into the impact of drug content on the performance of solid dispersion.

Abstract:With excellent biocompatibility and biodegradability,natural polysaccharides and their derivative s have exhibited great potential in constructing drug delivery ve hicles for tissue engineering and therapeutics.Cucurbit[n]uril（CB [n]）-mediated reversible crosslinking of polysaccharides possess intrinsic stimuliresponsiveness towards competitive guests and have been extensively investigated to fabricate various particles and hydrogels for multiple stimuli-re sponsive drug release by incorpo ration with other stimuli including photo,redox,and enzyme.Through host-guest interactions between CB[6] and aliphatic diamines,functional tags covalently connected with CB[6] can be readily anchored into polysaccharidebased hydrogels,realizing multiple functionalization.The rheological prope rty and drug release profile of polysaccharide-based supramolecular hydrogels can be facilely tuned through CB [8]-mediated dyna mic homo or hetero crosslinking of polysaccharides and/or other polymers.In this review,we introduce and summarize recent progress regarding polysaccharide-based supramolecular drug delivery systems mediated via host-guest interactions of CB[6] and CB[8],covering both bulk hydrogels and particular systems.At the end,possible utilization of CB[7]-based host-guest interactions in constructing polysaccharide-based drug delivery systems and future perspectives of this research direction are also discussed.

Abstract:Self-assembled supermolecular hydrogels of therapeutic agents without structural modification are of great significance in biomedical applications. Nevertheless, the complex conformations and elusive interactions of therapeutic molecules limit the controlled assembly of hydrogels. Molecules at the interface might have different arrangements and assemblies compared to those in bulk aqueous solution, which could potentially alter the selectivity of supramolecular polymorphs. However, this effect is still not well understood. Here, we demonstrate the interface-induced self-assembly of fibers for hydrogels, which is distinct from the spherical aggregates in the bulk aqueous solution, using cephradine (CEP) as a model compound. This phenomenon is caused by the packing of anisotropic molecules at the interface, and it can be applied to control the supramolecular polymorphism for the direct self-assembly of hydrogels of therapeutic agents. The interface-induced hydrogel exhibits a high degree of adjustable release and a long-acting bactericidal effect.

Abstract:A fibrillar hydrogel was obtained by covalent crosslinking via Diels–Alder reaction of two types of cellulose nanocrystals (CNCs) with furan and maleimide groups. Gelation has been studied at various ratios of components and temperatures in the range from 20 to 60 °C. It was shown that the rheological properties of the hydrogel can be optimized by varying the concentration and ratio of components. Due to the rigid structure of the CNCs, the hydrogel could be formed at a concentration of at least 5 wt%; however, it almost does not swell either in water with pH 5 or 7 or in the HBSS buffer. The introduction of aldehyde groups into the CNCs allows for the conjugation of physiologically active molecules containing primary amino groups due to the formation of imine bonds. Here, we used benzocaine as a model drug for conjugation with CNC hydrogel. The resulting drug-conjugated hydrogel demonstrated the stability of formulation at pH 7 and a pH-sensitive release of benzocaine due to the accelerated hydrolytic cleavage of the imine bond at pH < 7. The developed drug-conjugated hydrogel is promising as wound dressings for local anesthesia.

Abstract:Carbamazepine (CBZ) is a poorly water soluble drug owing to the Biopharmaceutic Classification System (BCS) class II. It is characterized by a variable bioavailability and by the presence of different polymorphs. In this paper the effects of CaCO3 on the physicochemical properties of CBZ and its solubility and release were evaluated. CaCO3 is a naturally non-toxic biomineral and was chosen because it is a safe, cheap and eco-friendly excipient able to dissolve in an acidic environment. Composites with different CBZ loadings were prepared by ball milling and antisolvent method. The composites were characterized by X-ray powder diffraction, differential scanning calorimetry analysis and attenuated total reflectance FT-IR which revealed that both the presence of CaCO3 and the preparation procedure affect the polymorphic form crystallinity and intermolecular interactions among the drug molecules. Scanning electron microscopy showed that small drug crystals with different crystalline forms were deposited on the surface of the CaCO3 particles. Solubility and dissolution tests showed an increase in the apparent solubility of CBZ and improved drug release. These results demonstrated that CaCO3 affected the drug release properties likely due to its pH-sensitive characteristics.

Three supramolecular hydrogels were obtained via cooling the gelator aqueous solutions at 25 degrees C, 0 degrees C and at 25 degrees C under ultrasonic treatment, respectively. The results indicated that the gelator molecules assembled into fibrillar networks during forming the gels with the network density and gel stability increased by the order of the above formation methods. The electron spectroscopic spectrum of the xerogel surface reveals that the nitrogen atom locates mainly within the fiber, indicating the molecular assembling in water is affected by the amphiphilic gelator molecule. The assembling fashion of the gelator was further investigated by analyzing the single crystal structures of the complex formed from 4-hydroxypyridine and o-phthalic acid, and 1,2,4-benzene tricarboxylic acid, which shows that the molecules assembled into branched fiber via different hydrogen bonding. Moreover, the powder X-ray diffraction diagram of the xerogel indicates that the gelator molecules assembled in a preferred direction during forming the gel.

Abstract:A simple drug compound, 4-oxo-4-(2-pyridinylamino) butanoic acid (defined as AP), was able to gel water at 4wt% concentration under various conditions. In the superstructure, AP molecules assembled into fibrous aggregates driving by hydrogen bonds and π–π stacking interaction. The gels with different backbone structures released drug molecules in different speeds.

Abstract:Supramolecular hydrogels made from guanine derivatives have attracted great interest for various applications. The gelation of guanine analogues usually involves the formation of guanine/K+ quartets. Functional cargoes such as fluorophores and drugs are usually decorated onto the guanine quartet gels via non-covalent pi-pi interactions or dynamic covalent linkages to render the gel with new functionalities. Here, a class of antiviral guanine quartet hydrogels have been developed via direct gelation of clinically available antiviral drugs such as entecavir, penciclovir, and ganciclovir in the presence of K+ ions. The prepared gels were stable at ambient temperature, non-toxic to normal cell lines, and maintained their inherent antiviral activities against hepatitis B virus and Herpes simplex virus, respectively. This study expands the function of guanine-based supramolecular gels and permits the development of a new class of antiviral gels.

Abstract:As a novel class of three-dimensional (3D) hydrophilic cross-linked polymers, supramolecular hydrogels not only display unique physicochemical properties (e.g., water-retention ability, drug loading capacity, biodegradability and biocompatibility, biostability) as well as specific functionalities (e.g., optoelectronic properties, bioactivity, self-healing ability, shape memory ability), but also have the capability to undergo reversible gel-sol transition in response to various environmental stimuli inherent to the noncovalent cross-linkages, thereby showing great potential as promising biomaterial scaffolds for diagnosis and therapy. In this Review, we summarized the recent progress in the design and synthesis of supramolecular hydrogels through specific, directional noncovalent interactions, with particular emphasis on the structure-property relationship, as well as their wide-ranging applications in disease diagnosis and therapy including bioimaging, biodetection, therapeutic delivery, and tissue engineering. We believe that these current achievements in supramolecular hydrogels will greatly stimulate new ideas and inspire persistent efforts in this hot topic area in future.

Abstract:A new dendritic gelator with carbazole as the building block (HBCD) was synthesized. It was found that H-bonding between the amide groups and pi-pi interaction between the aromatic rings played predominant roles in the gel formation. Meanwhile, significant aggregation-induced emission enhancement was observed in the gel state due to the formation of J-aggregates and the restricted molecular motion. Notably, the gel state of HBCD can be destroyed upon addition of F-, accompanied by fluorescence enhancement on account of the formation of N-H center dot center dot center dot F-, which could further lead to the increased coplanarity of HBCD. The sensory capability of HBCD exhibited a high selectivity towards F- instead of the Cl-, Br-, I- and AcO- anions, which could be explained by the fact that the steric hindrance of the dendrimer would go against the interactions between the larger anions and HBCD.
