Abstract:An odorant-binding protein cDNA (Acer-ASP2) was cloned and characterized from antennae of adult workers of an Asian honey bee, Apis cerana cerana F. (Hymenoptera: Apidae). The full-length open reading frame of Acer-ASP2 cDNA was 429 bp, encoding 142 amino acids. Protein signature analyses revealed that it contained six conserved cysteines with an N-terminal signal sequence of 19 amino acids. The deduced protein sequence shares high homology with Amel-ASP2 from the honey bee, Apis mellifera L., and low similarity with odorant-binding proteins from other species of insects. Immunocytochemical localization showed that Acer-ASP2 was concentrated in the lymph of olfactory sensilla, such as sensilla placodea and sensilla trichodea A. Real-time polymerase chain reaction of Acer-ASP2 transcripts showed that Acer-ASP2 was expressed on antennae but not in other general anatomical regions of the body. Temporally, Acer-ASP2 was expressed at a relatively high level in adults during two periods (9 and 27 vs. 1, 15, and 30 days). This timing is correlated with the production of beeswax and searching behavior for nectar/pollen, respectively. Thus, Acer-ASP2 is a species-specific gene that we propose to be involved in the acquisition of odorant molecules from nectar, pollen, and other general odorant sources.
Study on Specific Apis Cerana Honeybee Queen Pheromone Biosensor Based on Pheromone-Binding Protein ASP1

Abstract:Biosensor is a detection device composed of a biologically active unit and a signal converter. In a eusocial honeybee colony, worker bees have also evolved a sophisticated olfactory system to specifically sense the queen pheromone components. So far, it is not clear whether it is possible to design a biosensor to detect the queen pheromone in vitro by using worker bees' olfactory system. In this study, a specific pheromone biosensor was successfully constructed by immobilizing the recombinant honeybees' pheromone binding protein 1 (PBP1/ASP1) on the forked finger gold electrode using nitrocellulose membrane. The results showed that the maximum resistance charge transfer (Rct) changes of two queen pheromone components [methyl p-hydroxy-benzoate (HOB) and vanillyl alcohol (HVA)] reached 60%, which were significantly higher than those of a bee larval pheromone component (ethyl oleate), and a floral volatile ($\beta $ -ionone). The linear detection of HOB and HVA ranged from 10 -7 to 10 -4 mol/L, and the theoretical detection limit reached 10 -8 mol/L, indicating the wide detection linear range and excellent detection capability of this biosensor. These results indicated that the biosensor was highly sensitive and specific to the queen pheromone components. It is beneficial to protect and maintain the sustainability of honeybee resources in the future.

Abstract:Malate dehydrogenase(MDH) is an important enzyme in glycometabolism. MDH of Apis mellifera showed three enzyme active zones, MDHI,MDHIIand MDHIII. MDHIand MDHIII maintained relative stability in different castes and developmental phases,but MDHIIwas polymorphic,and controlled by three alleles,a,b and c.MDH of Apis cerana was coded by S and F alleles,but some authors reported it is monomorphic.MDH was applied to the studies of A. mellifera, which included several aspects as follows: the number of queen matings,labor division in honeybee societies, the analysis of genetic constitution in honeybee populations and so on. The combination of both MDH and molecular biology will certainly promote honeybee studies.
Evaluation of Cage Designs and Feeding Regimes for Honey Bee (hymenoptera: Apidae) Laboratory Experiments

Abstract:The aim of this study was to improve cage systems for maintaining adult honey bee (Apis mellifera L.) workers under in vitro laboratory conditions. To achieve this goal, we experimentally evaluated the impact of different cages, developed by scientists of the international research network COLOSS (Prevention of honey bee COlony LOSSes), on the physiology and survival of honey bees. We identified three cages that promoted good survival of honey bees. The bees from cages that exhibited greater survival had relatively lower titers of deformed wing virus, suggesting that deformed wing virus is a significant marker reflecting stress level and health status of the host. We also determined that a leak- and drip-proof feeder was an integral part of a cage system and a feeder modified from a 20-ml plastic syringe displayed the best result in providing steady food supply to bees. Finally, we also demonstrated that the addition of protein to the bees' diet could significantly increase the level ofvitellogenin gene expression and improve bees' survival. This international collaborative study represents a critical step toward improvement of cage designs and feeding regimes for honey bee laboratory experiments.
The effects of queen mandibular pheromone on Nosema (Vairimorpha) ceranae infections in caged honey bees

Abstract:Honey bees utilize queen mandibular pheromone (QMP) for maintaining social hierarchy and colony development. In controlled cage studies, synthetic QMP is often introduced to mimic natural conditions. However, questions have arisen about the effects of QMP on nosema disease studies. This short report identifies significant early-stage suppression effects of QMP on Nosema (Vairimorpha) ceranae infections. QMP was found to significantly lower infection rates below the reported infectious dose for 50 % infectivity (ID50) and to slow disease development in a dose-independent manner. These effects diminished at doses exceeding ID100. We recommend that studies investigating treatment effects using caged bees avoid QMP to ensure unambiguous results. Additionally, employing multiple infectious doses with shorter incubation times would be useful for evaluating other treatments that may have subtle effects. Furthermore, our findings support previous field studies suggesting that queen replacement reduces nosema disease at levels similar to treatment with fumagillin.
The Combining Ability and Heterosis Analysis of Royal Jelly Yield and Quality Properties in Western Honeybees

Abstract:The NC II mating design of 2 x 4 was made in order to evaluate The combining ability and heterosis of royal jelly yield and quality properties that included royal jelly yield, royal jelly yield per cup, acceptance ratio and acidity in Apis mellifera. The results were as follows: (1) For all characters, general combining ability in six parents (GCA) achieved highly significant level (P < 0.01), special combining ability (SCA) only significant (P < 0.05), therefore the additive effect is more important for royal jelly yield and quality properties; (2) The GCA of A. m. acervorum in four characters was higher, and the SCA variance of A. m. carpatica, A. m. caucasica was greater, so they were good parents. ZND-1 Apis mellifera lingustica in royal jelly yield, royal jelly yield per cup, acceptance ratio was an excellent parent, but A. m. carnica only could be used in increasing royal jelly quality; (3) The heterosis over high-parent is negative, thus it is difficult to improve royal jelly yield and quality by cross breeding, but by appropriate choice of parents, high yield and quality combination can be obtained.

Abstract:China is the largest royal jelly producer and exporter in the world, and high royal jelly-yielding strains have been bred in the country for approximately three decades. However, information on the molecular mechanism underlying high royal jelly production is scarce. Here, a cDNA microarray was used to screen and identify differentially expressed genes (DEGs) to obtain an overview on the changes in gene expression levels between high and low royal jelly producing bees. We developed a honey bee gene chip that covered 11,689 genes, and this chip was hybridised with cDNA generated from RNA isolated from heads of nursing bees. A total of 369 DEGs were identified between high and low royal jelly producing bees. Amongst these DEGs, 201 (54.47%) genes were up-regulated, whereas 168 (45.53%) were down-regulated in high royal jelly-yielding bees. Gene ontology (GO) analyses showed that they are mainly involved in four key biological processes, and pathway analyses revealed that they belong to a total of 46 biological pathways. These results provide a genetic basis for further studies on the molecular mechanisms involved in high royal jelly production.

Abstract:Propolis is one of the most fascinating honey bee (Apis mellifera L.) products. It is a plant derived product that bees produce from resins that they collect from different plant organs and with which they mix beeswax. Propolis is a building material and a protective agent in the bee hive. It also plays an important role in honey bee social immunity, and is widely used by humans as an ingredient of nutraceuticals, over-the-counter preparations and cosmetics. Its chemical composition varies by geographic location, climatic zone and local flora. The understanding of the chemical diversity of propolis is very important in propolis research. In this manuscript, we give an overview of the available methods for studying propolis in different aspects: propolis in the bee colony; chemical composition and plant sources of propolis; biological activity of propolis with respect to bees and humans; and approaches for standardization and quality control for the purposes of industrial application.
Reproductive traits and mandibular gland pheromone of anarchistic honey bee workers Apis mellifera occurring in China

Abstract:In honey bee colonies, workers, in particular of “anarchistic” lineages, can activate their ovaries and lay eggs, even in the presence of the queen. We identified three queenright colonies showing typical signs of worker reproduction. To characterize this new lineage, we extracted the mandibular gland and analyzed it using gas chromatography. The total amounts of the five main components of the mandibular gland, namely methyl p-hydroxyben-zoate (HOB), 9-oxo-2(E)-decenoic acid (9-ODA), (S)-9-hydroxy-(E)-2-decenoic acid (9-HDA), 10-HDA, and 10-hydroxyde-canoic acid (10-HDAA) were significantly higher in the mandibular gland profiles of workers with activated ovaries (AWs, 8.88 ± 1.71 μg) compared to workers with inactivated ovaries (IAWs, 4.00 ± 2.09 μg). Furthermore, the chemical profiles of IAWs were dominated by the “worker substances” 10-HDA (34.64 ± 8.19 %) and its precursor 10-HDAA (22.88 ± 4.95 %), while the chemical profiles in AWs were dominated by the precursor of the queen substance 9-HDA (40.04 ± 7.55 %). The ratios of two precursor substances 10-HDAA/9-HDA of IAWs were more worker like (>1.0) whereas AWs were more queen like (≤1.0). These results suggest that the mandibular pheromones of anarchistic workers resemble a more queen-like reproductive active profile and that these workers may represent a reversion to a more basal reproductive phenotype.

Abstract:The mandibular glands (MGs) of honey bee workers are key exocrine glands responsible for the biosynthesis of multiple royal jelly acids, which have important functions within the colony. However, our knowledge about the nutrition requirement of MGs is quite limited. Here, we artificially manipulated the nutritional status of caged honey bee workers (pollen and sugar candy, sugar candy only, soybean meal and sugar candy, stearic acid supplemented sugar candy or citric acid supplemented sugar candy). Then we measured their impact on the 10-hydroxy-2-decanoic acid (10-HDA) and 10-hydroxydecanoic acid (10-HDAA) content in honey bee heads to evaluate the synthesis ability of workers' MGs, and analyzed the protein content of MGs as well as gene expression in the MGs to reveal the possible physiological changes. Our result showed that nutrition strongly impacted the MGs acid synthesis, the 10-HDA and 10-HDAA contents were significantly higher in workers fed with pollen than with sugar candy only, while an additional supply of protein and organic acid showed no significant contribution. In addition, the 10-HDA/10-HDAA ratio was also significantly affected by the nutrition status. On the contrary, different diets showed no influence on the protein content or gene expressions of MGs, except that organic acid supplements could increase the expression of key genes in fatty acid beta-oxidation pathways. Taken together, our study revealed that honey bee nutritional status is closely related to the function of worker MGs. The nutritional deficiency has a significant negative impact on the synthesis ability of worker MGs, but showed no effect on the physiology of MGs.

Abstract:Analyssis of genotypic effects for colony′s reproductive ability and the weights of work bee′s hypopheryngeal gland of 3 lines of Western Honeybees (Apis melliffera Lingistica)were conducted by using a genotypic model for analyzing the genetic and non-genetic effects.Analysis approaches of conventional and conditional variance and correlation were employed to evaluate the developmental behavior of honeybee′s reproductive ability and the weight of worker bee′s hypopharyngeal gland.The results indicated that significant variance due to genotypic effects were detected for colony′s reproductive ability and the weight of hypopharyngeal gland.Significant variances were found at some stages while no conditional one being detected.Correlation analysis between the same traits at different stages indicated that there existed significant coefficients due to genotypic effects for reproductive ability and the weight of hypopheryngeal gland exception of the first stage of both traits.The analysis of correlation between different pair of traits showed that the coefficients between colony′s RJ yields and reproductive ability,and RJ yields between the weight of hypopharyngeal gland were significant exception of the first stage of both traits.
Pheromonal Predisposition to Social Parasitism in the Honeybee Apis Mellifera Capensis

Abstract:In honeybees, worker reproduction is mainly regulated by pheromones produced by the brood and the queen. The source of one of the queen pheromones influencing worker reproduction has been located in the mandibular glands. In nonlaying workers, this gland's profile is dominated by fatty acids that are incorporated into the food given to the brood and to nest mates. After queen loss and onset of reproductive activity, workers are able to synthesize different fatty acids, which are normally only produced by queens and that contribute to their reproductive success. Apis mellifera capensis workers have the ability to rapidly produce queen-like mandibular profiles that could represent an important factor in their ability to behave as facultative intraspecific social parasites. Indeed, A. m. capensis workers can take over reproduction from the host queens in colonies of other subspecies. Here, we show that in the presence of their own queen, the mandibular gland profile of A. m. capensis workers is dominated by the precursor of the major compound of the queen pheromone. This is a unique trait among honeybee workers and suggests that A. m. capensis workers are primed for reproduction and that this phenomenon represents a pheromonal predisposition to social parasitism. We identified geographical variation in the ratio of queen-to worker-specific compounds in themandibular gland profile of A. m. capensis workers, which corresponds with the introgression with the neighboring subspecies A. m. scutellata.
Quantum Chemical Study on the Stability of Honeybee Queen Pheromone Against Atmospheric Factors

Abstract:The managed honeybee, Apis mellifera, has been experienced a puzzling event, termed as colony collapse disorder (CCD), in which worker bees abruptly disappear from their hives. Potential factors include parasites, pesticides, malnutrition, and environmental stresses. However, so far, no definitive relationship has been established between specific causal factors and CCD events. Here we theoretically test whether atmospheric environment could disturb the chemical communication between the queen and their workers in a colony. A quantum chemistry method has been used to investigate for the stability of the component of A. mellifera queen mandibular pheromone (QMP), (E)-9-keto-2-decenoic acid (9-ODA), against atmospheric water and free radicals. The results show that 9-ODA is less likely to react with water due to the high barrier heights (~36.5 kcal · mol−1) and very low reaction rates. However, it can easily react with triplet oxygen and hydroxyl radicals because of low or negative energy barriers. Thus, the atmospheric free radicals may disturb the chemical communication between the queen and their daughters in a colony. Our pilot study provides new insight for the cause of CCD, which has been reported throughout the world.
Comparative transcriptome analysis on the synthesis pathway of honey bee (Apis mellifera) mandibular gland secretions.

Abstract:Secretions from mandibular glands (MGs) have important caste-specific functions that are associated with the social evolution of honey bees. To gain insights into the molecular architecture underlying these caste differences, we compared the gene expression patterns of MGs from queens, queenright workers (WQRs) and queenless workers (WQLs) using high-throughput RNA-sequencing technology. In total, we identified 46 candidate genes associated with caste-specific biosynthesis of fatty acid pheromones in the MG, including members of cytochrome P450 (CYP450) family and genes involved in fatty acid β-oxidation and ω-oxidation. For further identification of the CYP450s genes involved in the biosynthesis of MG secretions, we analyzed by means of qPCR, the expression levels of six of the CYP450 genes most abundantly expressed in the transcriptome analysis across different castes, ages, tasks and tissues. Our analysis revealed that CYP6AS8 and CYP6AS11, the most abundantly expressed CYP450 genes in worker and queen MGs, respectively, are selectively expressed in the MGs of workers and queens compared to other tissues. These results suggest that these genes might be responsible for the critical bifurcated hydroxylation process in the biosynthesis pathway. Our study contributes to the description of the molecular basis for the biosynthesis of fatty acid-derived pheromones in the MGs.
The effect of essential oils of sweet fennel and pignut on mortality and learning in africanized honeybees (Apis mellifera L.) (Hymenoptera: Apidae)

Abstract:It was recently discovered that exposure to small concentrations of the essential oils of sweet fennel (Foeniculum vulgare Mill) or pignut [Hyptis suaveolens (L.) Poit] can be used to control aphids. What is not known is whether these oils also influence honeybee behavior. Experiments using both harnessed and free-flying foragers at concentrations used to control aphids showed that bees readily associated the odors with a reward, discriminated between them, and were not repelled. Honeybees, however, would not consume the oils when mixed with sucrose to create an unconditioned stimulus. An experiment in which harnessed bees consumed various concentrations showed that concentrations greater than 50% were detrimental. The experiments reported here provide further evidence supporting the use of conditioning techniques to evaluate the use of essential oils on honey bee behavior.
The Effects of Sub-lethal dose of Insecticides on Honeybee Behavior and Transcriptome

Abstract:Abstract Exposure to insecticides is the main cause of honeybee (Apis mellifera) population decline. At sub-lethal doses, these chemicals have been shown to negatively affect honeybee physiological development and behavior. Previously, we found the insecticide imidacloprid and deltamethrin significant reduced honeybee dancing and foraging efficiency. As a follow up, we performed a deep RNA-seq analysis to reveal the gene regulatory mechanisms underlying the altered foraging behavior. Genes involved in detoxification were up-regulated in both imidacloprid and deltamethrin treatment groups. Gene members in immune pathways, odorant receptors and major royal jelly protein families showed significant up or down regulation in treatment groups compared with controls. This fluctuating gene expression profile reflects that multifaceted aspects of honeybee physiology were affected by the two insecticides, leading to inaccurate communication and impaired learning and memory. Our findings reveal candidate molecular mechanisms under impaired dance performance in honeybees exposed to insecticides.

Abstract:This study investigates the impact of bee pollen nutrition on the royal jelly production of honey bees ( Apis mellifera ). Results demonstrate that pollen diet significantly impacts hypopharyngeal gland (HPG) development and the expression of genes associated with royal jelly biosynthesis. Bees fed Brassica napus pollen exhibited superior HPG development, and increased mrjp1 expression (encoding a key royal jelly protein). While the cyp450 6AS8 gene expression (encoding a key enzyme in 10-HDA biosynthesis) was increased by pollen consumption, no distinct expression patterns were observed among the different pollen types tested. An in vitro bee cage platform for royal jelly production has been established to further understand the mechanisms behind royal jelly production in bees. The experiment demonstrated a positive correlation between the number of worker bees and the total yield of royal jelly per cage. However, when the number of worker bees is low, the amount of royal jelly each individual worker bee needs to produce increases. In conclusion, these findings enhance our understanding of the role of bee pollen nutrition in royal jelly production. Furthermore, the results from this in vitro bee cage platform suggest that the number of worker bees is a critical factor in royal jelly production, and that bees may possess a controllable mechanism for regulating royal jelly secretion.

Abstract:Honey bee populations have been declining precipitously over the past decade, and multiple causative factors have been identified. Recent research indicates that these frequently co-occurring stressors interact, often in unpredictable ways, therefore it has become important to develop robust methods to assess their effects both in isolation and in combination. Most such efforts focus on honey bee workers, but the state of a colony also depends on the health and productivity of its queen. However, it is much more difficult to quantify the performance of queens relative to workers in the field, and there are no laboratory assays for queen performance. Here, we present a new system to monitor honey bee queen egg laying under laboratory conditions and report the results of experiments showing the effects of pollen nutrition on egg laying. These findings suggest that queen egg laying and worker physiology can be manipulated in this system through pollen nutrition, which is consistent with findings from field colonies. The results generated using this controlled, laboratory-based system suggest that worker physiology controls queen egg laying behavior. Additionally, the quantitative data generated in these experiments highlight the utility of the system for further use as a risk assessment tool.
Molecular recognition of floral volatile with two olfactory related proteins in the Eastern honeybee (Apis cerana)

Abstract:The honeybee relies on its sensitive olfaction to perform the foraging activities in the field. In the antennal chemoreception system of honeybee, odorant-binding proteins (OBPs) and chemosensory protein (CSPs) are major two protein families capable of binding with some plant volatiles and chemical ligands. However, the chemical binding interaction of plant odors with OBPs and CSPs in the honeybee olfactory system is still not clear yet. Hence, complex fluorescent spectra, ultraviolet absorption spectra, circular dichroism spectra and molecular docking were used to investigate the binding property of AcerASP2 (an OBP of Apis cerana) and AcerASP3 (a CSP of Apis cerana) with β-ionone, one of ordinary floral volatiles in botanical flower. As a result, β-ionone had a strong capability to quench the fluorescence that the two proteins produced, and their interaction was a dynamic process that was mainly driven by a hydrophobic force. AcerASP2 had a larger hydrophobic cavity than that of AcerASP3 and the conformation of AcerASP2 was changed less than AcerASP3 when binding with β-ionone. Our data suggests that OBPs like AcerASP2 might make a large contribution toward assisting the honeybee in sensing and foraging flowers, and A. cerana has evolved a good circadian rhythm to perceive a flower's odor following the fluctuation of temperature in the olfactory system. This significantly extends our knowledge on how to strengthen the honeybees' pollination service via manipulation of target proteins in the olfactory system.
