Abstract:Machilin D, Virolin, the 8-O-4' neolignan, has been enantioselectively synthesized, with good yield, in eight steps from vanillin using our own method of selective oxidation of benzylic and allylic hydroxyl group of sec-l,2-diols under ultrasound wave promotion and SN2 coupling as the key steps.

Abstract:A novel synthetic route to natural nordihydroguaiaretic acid and Machilin A is described. This method involves two Stobbe condensations to give the skeleton of lignan, then followed by transformation of functional groups to obtain target compounds. The method operation is convenient and practical.

Abstract:ABSTRACT Erythro-1-(4-hydroxy-3-methoxyphenyl)-2-(4-formyl-2-methoxy-phenoxy)-propane-1,3-diol, a 8-O-4′ norneolignan, has been stereoselectively synthesized in eight steps from ferulic acid. In this new route to prepare erythro 8-O-4′ neolignans the cis addition of mCPBA to a double bond with high regioselectivity was a key step, and its mechanism is discussed.

Abstract:A concise, divergent, asymmetric total syntheses of mulinane diterpenoids has been achieved. Specifically, a new strategy was developed featuring a key intramolecular Friedel-Crafts reaction to construct the chiral fused 5-6-6 tricyclic motif, followed by sequential Birch reduction, conjugate methylation, and homologation/ring-expansion reactions to furnish the desired 5-6-7 tricyclic skeleton bearing five contiguous stereocenters. With this efficient strategy, seven mulinane diterpenoids and two analogues were synthesized via late-stage functional modification or functionalization in 8.6-20% overall yields and 11-15 steps.

Abstract:Starting from commercially available lactone 10, a concise and highly diastereoselective synthesis of (−)-(lS,2R,8aS)-2-epi-lentiginosine (2) is described. The synthesis featured an unexpected highly erythro-selective reaction of Grignard reagent 7 with the protected N,O-acetal 8. The stereochemical outcome of this reaction is contrary to the known results involving the reactions with O-benzyl protected aminofuranosides and aminoglycosides. Thus, this method constitutes an extension of the threo-diastereoselective C–C bond formation methodology.

Abstract:AbstractAn asymmetric photoenolization/Diels–Alder (PEDA) reaction between electron‐rich 2‐methylbenzaldehydes and unsaturated γ‐lactones was developed to directly construct the basic tricyclic core of aryltetralin lactone lignans. This methodology enabled the first asymmetric total synthesis of aglacins A, B, and E and revision of the absolute configuration of these natural lignans. The strategy was also used to prepare the naturally occurring aryldihydronaphthalene‐type lignans (−)‐7,8‐dihydroisojusticidin B and (+)‐linoxepin in four and six steps, as well as 27 natural‐product‐like molecules containing a C8′ quaternary center. We believe that the synthetic aglacins and small‐molecule library provide new opportunities to carry out the SAR studies of the podophyllotoxin family of natural products.
