Abstract:Ferric chloride hexahydrate was shown to be an efficient catalyst for the conjugate addition of 2,3-allenoates with alkyl-, aryl-, or vinyl-Grignard reagents to synthesize polysubstituted beta,gamma-unsaturated alkenoates with high regio- and stereoselectivity. The in situ formed magnesium dienolate may readily react with different electrophilic reagents under different reaction conditions with or without a catalyst to construct an allylic quaternary carbon at the alpha-position of the ester group and a stereocontrol able retention of the carbon-carbon double bond.

Abstract:The direct functionalization of allylic C-H bonds is distinguished by enabling rapid assembly of structural complexity from simple molecules. Although Pd-catalyzed allylic C-H functionalization has been extensively studied, the discovery of new catalytic systems remains fairly underdeveloped. Here, we disclose a Pt-catalyzed allylic C-H alkylation of a wide range of alpha-alkenes by using phosphoramidites as ligands and malononitriles as alkylating reagents. Notably, the combination of chiral ureacatalyzed Michael addition and Pt-catalyzed allylic C-H alkylation can serve as an efficient protocol to access chiral tetrahydropyran with high levels of diastereo- and enantioselectivity. Mechanistic studies suggest that the Pt-catalyzed allylic C-H activation proceeds through a concerted proton and two-electron transfer process, which is analogous to transition state geometries of Pd catalysis.

Abstract:Metal-catalyzed enantioselective allylation, which involves the substitution of allylic metal intermediates with a diverse range of different nucleophiles or S(N)2'-type allylic substitution, leads to the formation of C-H, -C, -O, -N, -S, and other bonds with very high levels of asymmetric induction. The reaction may tolerate a broad range of functional groups and has been applied successfully to the synthesis of many natural products and new chiral compounds.

Abstract:The Michael addition of malonates to α,β-unsaturated aldehydes catalyzed by O-TMS protected diphenylprolinols and acetic acid in water occurs at 0°C to rt. In most cases, the reaction runs to completion in less than 24h. A wide range of aldehydes including β-aryl, β-alkyl and β-alkenyl acroleins are found to be compatible with these conditions, providing the corresponding adducts in good yields and with good to excellent enantioselectivities.

Abstract:A highly enantioselective Pd-catalyzed amination of allenyl phosphates generating 2,3-allenyl amines with central chirality has been developed. Under the optimized conditions, chiral 2,3-allenyl amines with or without (an) additional C-C double or triple bond(s) have been prepared at 0 °C with up to 90% yield and 94% ee by identifying (R)-3,4,5-(MeO)3-MeOBIPHEP as the ligand.

Abstract:CuBr and ZnI2 have been developed as catalysts or subcatalysts for the efficient asymmetric synthesis of axially chiral allenols with up to 97% ee from readily available propargylic alcohols, aliphatic or aromatic aldehyde, pyrrolidine, and commerically available ligands. The alcohol unit in the terminal alkynes plays a very important role for ensuring high enantioselectivity via coordination.

Abstract:A range of primary allylic amines were resolved with selectivity factors of up to 491 through [Pd(allyl)Cl](2)/(S)-BINAP-catalyzed and mesitylsulfonyl hydrazide-accelerated asymmetric allylic alkylation of malononitriles involving enantioselective C-N bond cleavage under aerobic conditions. Moreover, the reaction proved useful for the asymmetric synthesis of a-branched allyl-substituted malononitriles with high enantiopurity.

Abstract:An efficient synthetic approach for chiral malonates was established via enantioselective phase transfer catalysis. The α-alkylation of 2,2-diphenylethyl tert-butyl α-methylmalonates with (S,S)-3,4,5-trifluorophenyl-NAS bromide as a phase-transfer catalyst under phase-transfer catalytic conditions successfully produced corresponding α-methyl-α-alkylmalonates; these compounds are versatile chiral building blocks containing a quaternary carbon center in high chemical yields (up to 99%) with excellent enantioselectivities (up to 98% ee). α,α-Dialkylmalonates were selectively hydrolyzed to the corresponding chiral malonic monoacids under basic (KOH/MeOH) and acidic conditions (TFA/CH 2 Cl 2 ), showing the practicality of the method.

Abstract:Highly enantioselective Michael addition of malonates to enones catalyzed by dipeptide-derived multifunctional phosphonium salts has been developed. The newly established catalytic system was characterized with its wide substrate scope featured with aliphatic aldehyde-derived enones and substituted malonates. The gram scale-up synthesis of adducts can also be successfully achieved under optimal conditions with both excellent yield and enantioselectivity.

Abstract:A series of Schiff-based ligands consisting of both tertiary amines and lipophilic groups were designed and synthesized. Using these ligands, a new chiral surfactant-type metallomicellar catalyst was developed in water, and this was identified by SEM/TEM analyses. These metallomicelles can be empolyed in asymmetric Michael addition reactions in water, delivering the corresponding adducts with excellent yields and enantioselectivities.

Abstract:Herein we report the first example of Pd-catalyzed highly selective three-component reaction of alkyn-1,4-diol dicarbonates, organoboronic acids, and malonate anions for the efficient synthesis of trisubstituted 2,3-allenyl malonates not readily available by the known protocols. The reaction demonstrates an excellent regio- and chemoselectivity for both the oxidative addition referring to the two C-O bonds and the subsequent coupling with the nucleophile with a remarkable functional group compatibility. A series of control experiments confirm a unique mechanism involving β-O elimination forming alka-1,2,3-triene and the subsequent insertion of its terminal C=C bond into Ar-Pd bond.

Abstract:Abstract Chiral aldehyde catalysis is a burgeoning strategy for the catalytic asymmetric α-functionalization of aminomethyl compounds. However, the reaction types are limited and to date include no examples of stereodivergent catalysis. In this work, we disclose two chiral aldehyde-catalysed diastereodivergent reactions: a 1,6-conjugate addition of amino acids to para -quinone methides and a bio-inspired Mannich reaction of pyridinylmethanamines and imines. Both the syn - and anti -products of these two reactions can be obtained in moderate to high yields, diastereo- and enantioselectivities. Four potential reaction models produced by DFT calculations are proposed to explain the observed stereoselective control. Our work shows that chiral aldehyde catalysis based on a reversible imine formation principle is applicable for the α-functionalization of both amino acids and aryl methylamines, and holds potential to promote a range of asymmetric transformations diastereoselectively.
