Abstract:Atorvastatin, a lipid-lowering drug that is widely used in the treatment of cardiovascular diseases, has significant clinical significance. This article focuses on the synthetic procedures of atorvastatin, including Paal-Knorr synthesis and several new synthetic strategies. It also outlines chemical and chemo-enzymatic methods for synthesizing optically active side chain of atorvastatin. In addition, a comprehensive overview of the analytical monitoring techniques for atorvastatin and its metabolites and impurities is reported, alongside a discussion of their strengths and limitations.

Abstract:Cinacalcet (I), sold as hydrochloride salt, is a calcimimetic drug which has been approved for the treatment of secondary hyperparathyroidism in patients with chronic renal disease and for the treatment of hypercalcemia in patients with parathyroid carcinoma. Here, an improved method for the synthesis of 3-(3-trifluoromethylphenyl)propanal (II), a key intermediate for the preparation of I, is described. The protocol required a Mizoroki-Heck cross-coupling reaction between 1-bromo-3-(trifluoromethyl)benzene and acroleine diethyl acetal, catalyzed by Pd(OAc)2 in the presence of nBu4NOAc (tetrabutylammonium acetate), followed by the hydrogenation reaction of the crude mixture of products in a cascade process. Palladium species, at the end of the reaction, were efficiently recovered as Pd/Al2O3. The procedure was developed under conventional heating conditions as well as under microwave-assisted conditions. The obtained mixture of 1-(3,3-diethoxypropyl)-3-(trifluoromethyl)benzene (III), impure for ethyl 3-(3-trifluoromethylphenyl) propanoate (IV), was finally treated, under mild conditions, with potassium diisobutyl-tert-butoxyaluminum hydride (PDBBA) to obtain after hydrolysis 3-(3-trifluoromethylphenyl)propanal (II), in an excellent overall yield and very high purity. Microwave conditions permitted a reduction in reaction times without affecting selectivity and yield. The final API was obtained through reductive amination of (II) with (R)-(+)-1-(1-naphthyl)ethylamine (V) using a catalyst prepared by us with a very low content of precious metal.

Abstract:A new and efficient total synthesis has been developed to obtain plagiochin G (22), a macrocyclic bisbibenzyl, and four derivatives. The key 16-membered ring containing biphenyl ether and biaryl units was closed via an intramolecular SNAr reaction. All synthesized macrocyclic bisbibenzyls inhibited Epstein-Barr virus early antigen (EBVEA) activation induced by the tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA) in Raji cells and, thus, are potential cancer chemopreventive agents.

Abstract:An improved synthetic process of captopril intermediate,D-3-acetylthio-2-methylpropanoyl-L-proline,was carried out using potassium hydroxide and potassium dihydrogen phosphate buffer as a condensating agent.Add potassium dihydrogen phosphate into a raw material of the L-proline,then,drop potassium hydroxide to form potassium hydroxide and potassium dihydrogen phosphate buffer as a condensating agent,the title compound was synthesized by dropping another raw material of 3-acetylthio-2-methyl-propanoyl chloride into above prepared solution.The pH of reaction system keeps in from 7.5 to 8.5,the reaction process is simple and efficient,and the average yield of product was improved from 33% to 39.5%.

Abstract:The fluorine-containing drug Cinacalcet is the only oral calcimimetic agent for the management of hyperparathyroidism in China and targets calcium sensitive receptor (CaSR) in organ tissues. The monofluoromethyl (CH 2 F) motif, found in many drug molecules and bioactive molecules, is particularly valuable as the CH 2 F functional group can mimic the methyl (CH 3 ) motif frequently encountered in bioactive molecules. Replacing CH 3 group of Cinacalcet with CH 2 F group will both bring new opportunities for the development of Cinacalcet generic drugs and provide a new option for the F-18 labeling synthesis of Cinacalcet. A Cinacalcet analogue has been successfully synthesized using commercial 2-amino-2-(naphthalen-1-yl)ethan-1-ol and 3-(3-(trifluoromethyl)phenyl)propanal as raw materials in a total yield of 24% in five steps and achieved the F-18 labeling synthesis of this analogue. This strategy is simple and efficient, paving the way for PET imaging of CaSR related diseases and inspiring development of new drugs based on Cinacalcet. Abstract:An improved synthesis of catalpalactone was carried out with a total yield of 18% by using 2-formylbenzoic acid as the starting material followed by a four-steps procedure including Wittig reaction,lactonization,retro-Michael addition and a one-pot manipulation to introduce the double bond.The structures of all of the intermediates as well as the stereo-configurations of double bond in compounds 11,12a and 12b were determined by 1H NMR,13C NMR,MS,1H-1H COSY and NOE techniques.This improved synthetic method provides several advantages such as inexpensive and easily available reagents,mild reaction conditions,simple operations,and high yields.Through being evaluated in vitro against a panel of two human tumor cell lines(MCF-7,BxPC3) which were determined by the SRB assays,catalpalactone displayed good activities against MCF-7 and BxPC3.

Abstract:Deregulation of ceramide metabolism is a hallmark of human cancer. Ceramide analogues thereby represent a new class of anti-cancer agents. We aimed at developing effective and low toxic ceramide analogues and synthesized a new class of ceramide analogues starting from l-threonine. Several analogues exhibit potent cytotoxicity against human cancer cells in vitro with IC50 as low as 4.8 μM. These ceramide analogues decreased xIAP and Bcl-xL level and exhibited significant sensitization activity to overcome human cancer cell resistance to TRAIL, a cancer-selective agent that are being tested in human clinical trials. Furthermore, we determined that these ceramide analogues effectively suppress human cancer xenograft growth in vivo with no significant toxicity at the efficacious dose. Therefore, we have developed a simple and effective method to synthesize functional ceramide analogues using l-threonine as starting material and these analogues have the great potential to be further developed as anti-cancer agents in human cancer therapy.

Abstract:Caffeic acid (CA) is one of the most abundant natural compounds present in plants and has a broad spectrum of beneficial pharmacological activities. However, in some cases, synthetic derivation of original molecules can expand their scope. This study focuses on the synthesis of caffeic acid phosphanium derivatives with the ambition of increasing their biological activities. Four caffeic acid phosphanium salts (CAPs) were synthesized and tested for their cytotoxic, antibacterial, antifungal, and amoebicidal activity in vitro, with the aim of identifying the best area for their medicinal use. CAPs exhibited significantly stronger cytotoxic activity against tested cell lines (HeLa, HCT116, MDA-MB-231 MCF-7, A2058, PANC-1, Jurkat) in comparison to caffeic acid. Focusing on Jurkat cells (human leukemic T cell lymphoma), the IC50 value of CAPs ranged from 0.9 to 8.5 μM while IC50 of CA was >300 μM. Antimicrobial testing also confirmed significantly higher activity of CAPs against selected microbes in comparison to CA, especially for Gram-positive bacteria (MIC 13–57 μM) and the yeast Candida albicans (MIC 13–57 μM). The anti-Acanthamoeba activity was studied against two pathogenic Acanthamoeba strains. In the case of A. lugdunensis, all CAPs revealed a stronger inhibitory effect (EC50 74–3125 μM) than CA (>105 μM), while in A. quina strain, the higher inhibition was observed for three derivatives (EC50 44–291 μM). The newly synthesized quaternary phosphanium salts of caffeic acid exhibited selective antitumor action and appeared to be promising antimicrobial agents for topical application, as well as potential molecules for further research.

Abstract:A novel, practical, and scalable approach for the synthesisofeldecalcitol (2), via the Horner-Wadsworth-Emmons(HWE) olefination of A-ring and C/D-ring fragments, has been described.The improved route was carried out to produce the key intermediatechiral epoxide 17 in good yield (13 steps, 9.0% overallyield) and high optical purity based on the utilization of d-mannose as a readily accessible, economically attractive chiralraw material. This process employed d-mannose as the chiralpool to directly assemble all the stereocenters of the A-ring fragment,thereby reducing the complexity of the chiral separation process,eliminating the need for asymmetric oxidation required for the constructionof the C-3 stereocenter, and ultimately increasing the efficiency.

Abstract:OBJECTIVE:To establish a new approach to synthesis and purification of calcium 5-formytetrahydrofolate.METHODS:Target compound was synthesized by the use of folic acid as starting material via reduction, formylation, hydrolysis and salt formationRESULTS:The structure of calcium 5-formytetrahydrofolate was confirmed by UV, 1H-NMR and elemental analysis and the overall yield was 54%-59%, 14%-17% higher than the reported yield.CONCLUSION:A convenient synthetic route was developed and it would be suitable for industrial production.

Abstract:OCT, a non-calcemic analogue of 1,25(OH)2D3 has been found to have a more potent activity than that of 1,25(OH)2D3 regarding cell differentiation and immunopotentiation activity, and to prolong the average life span of MRL/l mice. Recently, we found that OCT effectively suppressed the secretion and synthesis of PTH without inducing hypercalcemia. In primary cultures of bovine parathyroid cells, OCT was capable of suppressing PTH release in a dose-dependent manner. OCT was also active in vivo, and, like 1,25(OH)2D3, decreased the pre-pro(PTH) mRNA levels. In a group of rats with CRF, daily administration of OCT, 8 ng i.p. for 2 weeks returned PTH levels to normal without changes in serum calcium. Preliminary results in dogs with CRF indicated that after the administration of OCT 5 micrograms i.v., N-terminal PTH decreased by 76% without changes in Ca. In conclusion, OCT may provide a unique contribution to the treatment of secondary hyperparathyroidism.
