Abstract:Seven new drimane sesquiterpenoids (1-3, 6-9), along with the known compounds deoxyuvidin B (4), strobilactone B (5), and RES-1149-2 (10), were obtained from cultures of the fungus Aspergillus ustus, which was isolated from the marine sponge Suberites domuncula. Their structures were established by means of spectroscopic analyses including one- and two-dimensional NMR spectroscopy and high-resolution MS. Compounds 6, 7, and 10 showed cytotoxic activity against a panel of tumor cell lines, including L5178Y, HeLa, and PC12 cells, with 7 being the most active (EC(50) against L5178Y cell line: 0.6 microg/mL).

Abstract:Four new drimane sesquiterpenoids (1–4) and three known ones (5–7) were isolated from the fermentation broth of the mangrove-derived Aspergillus ustus 094102. Compound 5 was further resolved as four purified compounds 5a–5d. By means of extensive spectroscopic and ECD analysis as well as the chemical transformation, their structures were identified as (2R,3R,5S,9R,10S)-2,3,9,11-tetrahydroxydrim-7-en-6-one (ustusol F, 1), (2R,3R,5R,9S,10R)-2,3,11-trihydroxydrim-7-en-6-one (9-deoxyustusol F, 2), (3S,5R,9R,10R)-3,11,12-trihydroxydrim-7-en-6-one (ustusol G, 3), (5S,6R,9S,10S, 11R,2′E,4′E)-(11-dideoxy-11-hydroxystrobilactone A-6-yl)-5-carboxypenta-2,4-dienoate (ustusolate H, 4), ((5S,6R,9S,10S)-strobilactone A-6-yl) (2E,4E)-6,7-dihydroxyocta-2,4-dienoate (ustusolate I, 5), (2′E,4′E;6′,7′-erythro)-ustusolate I (5a) and (2′E,4′E;ent-6′,7′-erythro)-ustusolate I (5b), (2′E,4′E,6′R,7′R)-ustusolate I (5c) and (2′E,4′E,6′S,7′S)-ustusolate I (5d), (5S,6R,9S,10S,2′E,4′E)-(strobilactone A-6-yl)-5-carboxypenta-2,4-dienoate (ustusolate J, 6), and (2S,5S,9R,10S)-2,9,11-trihydroxydrim-7-en-6-one (ustusol B, 7), respectively. Abstract:A new eremophilane sesquiterpene, capnodiumone (1), along with five known eremophilane sesquiterpenes (2-6) and eight other compounds (7-14), have been isolated from a mangrove-associated fungus Capnodium sp. SZ-F22. The chemical structures were elucidated on the basis of extensive spectroscopic analysis. The broth extract of the fungus exhibited a good inhibitory effect on the mycelium growth against Fusarium graminearum at 100g/mL, however, all the 14 compounds showed no expected antifungal activity. Abstract:Eight drimane sesquiterpenes (1-8), six isochromane derivatives (9-14), and three known compounds, daldinin B (15), 9 alpha-hydroxy-6 beta-[(2 E,4E,6E)-octa-2,4,6-trienoyloxy]-5 alpha-drim-7-en-11,12-olide (16), and pergillin (17), were isolated from the EtOAc extract of the marine-derived fungus Aspergillus ustus 094102. The structures of the new compounds were elucidated on the basis of spectroscopic analysis. The cytotoxic effects on A549 and HL-60 cell lines were evaluated by SRB and MTT methods. Ustusorane E (13) showed significant cytotoxicity against HL-60 cells with an IC50 value of 0.13 mu M. Ustusolates C (6) and E (8) exhibited moderate cytotoxicity against A549 and HL-60 cells with IC50 values of 10.5 and 9.0 mu M, respectively, and ustusolate A (4) showed weak cytotoxicity against HL-60 and A549 cells with IC50 values of 20.6 and 30.0 mu M, respectively.

Abstract:A new sesquiterpenoid derivative, named aspergiketone (1), along with seven known compounds (2-8) were isolated from the coastal saline soil fungus Aspergillus fumigatus. Their structures were elucidated by spectroscopic analysis, and by comparison of experimental and reported data. The absolute configuration of compound 1 was defined by X-ray diffraction analysis. Compound 1 was cytotoxic towards HL-60 and A549 cell lines with IC50 values of 12.4 and 22.1 mu M, respectively.

Abstract:A new diphenyl ether (1) as well as 20 other compounds were identified from the fermentative extracts of marine-derived fungi Phoma sp. CZD-F11 (Compounds 1–8) and Aspergillus sp. CZD-F18(Compounds 9–21). Their structures were elucidated on the basis of extensive spectroscopic analysis. The broth extracts of the fungi exhibited very good anticancer activity against H1975 cells with 5.62 and 25.8% viability at concentration of 10 μg/mL for Phoma sp. CZD-F11 and Aspergillus sp. CZD-F18, respectively. The inhibitory activity of all compounds against PC-3 cell lines, BRD4 and aromatase were evaluated. The results showed compound 7 exhibited moderate anticancer activity with 66.1% inhibition against PC-3 cell lines at the concentration of 10 μg/mL. Compound 7 and 8 exhibited favourable BRD4 inhibitory activity with 78.5 and 76.4% inhibition at the concentration of 10 μg/mL.

Abstract:One new cyclohexenone derivative (1), and two undescribed drimane sesquiterpenes (2 and 3), together with another seven known drimane sesquiterpenes were isolated from a seagrass-derived fungus Aspergillus insuetus SYSU6925. Structures of these metabolites were elucidated by comprehensive spectroscopic analysis, including NMR analysis, mass spectrometry, and ECD calculations. Compounds 1-3, 5 and 7 displayed weak to moderate antifungal activities towards four phytopathogenic fungi, with Minimum inhibition concentration (MIC) values range from 50 to 200 mu g/mL. Compound 1, a rare cyclohexenone derivative with n-propyl group exhibited more potent inhibitory activities (MIC, 50 mu g/mL) against F. oxysporum than positive control (Triadimefon). Compounds 2 and 3 also exhibit potent anti-inflammatory activities by inhibiting the production of nitric oxide (NO) in RAW264.7 cells with IC50 values of 21.5 +/- 1.1 and 32.6 +/- 1.16 mu M, respectively.

Abstract:Fourteen active strains isolated from the leaves or branches of mangrove plants were screened using P388 cell line(mouse leukemia cell line).Five compounds from fungus W-8 were isolated by solvent extraction,bioassay-guided fractionation,silica gel column chromatography,and preparative HPLC.By means of physicochemical properties and spectral analysis,they were identified as aspulvinone H1(1),aspulvinone H(2),butyrolactone I(3),terrein(4),and 3-propylbenzene-1,2-diol(5),respectively.The NMR data of compound 1 were first reported here.Compounds 4 and 5 showed mediate cytotoxicities against A549,K562,and Hela cell lines.

Abstract:Five new diisoprenyl cyclohexene-type meroterpenoids, aspergienynes J–N (1–5), along with three known analogues (6–8), were obtained from the mangrove endophytic fungal strain Aspergillus sp. GXNU-Y85. The chemical structures, including their absolute configurations, were established via spectroscopic data and comparison of experimental and calculated ECD spectra. Cytotoxicity assay results indicated that compound 8 had strong cytotoxicity against HeLa cancer cells, and its IC50 value was 11.8 μM. In addition, flow cytometry analysis revealed that the cytotoxicity of 8 was due to the induction of G1 cell cycle arrest and apoptosis in HeLa cells.

Abstract:Abstract Soil-derived fungi represent an insufficiently tapped reservoir for discovering new and bioactive natural products (NPs), and despite an ever-increasing number of unknown NPs have been discovered over the past few decades, much of the hidden biosynthetic potential is still in an urgent need to be disclosed. In this research, a chemical investigation was performed on a wetland soil-derived fungus Aspergillus calidoustus TJ403-EL05, leading to the isolation of a total of fourteen drimane sesquiterpenoids (1–14), incorporating three new ones, namely ustusols F–H (1–3). Their structures, comprising absolute configurations, were completely authenticated by widespread spectroscopic data, quantum chemical 13C NMR and ECD calculations, and X-ray crystallography experiments. Compound 14 exhibited moderate anti-inflammatory activity by inhibiting the LPS-induced NO release (IC50 = 25.6 μM). Graphical Abstract

Abstract:Fifteen terpenoid derivatives, including six new sesquiterpenoids (1 – 5 and 10), one new ophiobolin sesterterpenoid (11), and one new 3,5-dimethylorsellinic acid (DMOA)-based meroterpenoid (15), as well as seven known compounds (6 – 9 and 12 – 14) were isolated and characterized from fungus Aspergillus calidoustus, which was separated from the wetland soil collected at Dianchi Lake, Yunnan Province. Compound 15 was a rare austin analogue with the opened A ring, and also featured an undescribed oxygen bridge between C-3 and C-16 to construct an unexpected furan ring. Their structures were established by means of spectroscopic methods and single-crystal X-ray diffraction. All the isolated drimane sesquiterpenoids were evaluated for the in vitro cytotoxicity against five tumor cell lines, including SW480 (colon cancer), IOMM-Lee (meningioma), HeLa (cervical cancer), FARAGE (diffuse large B-cell lymphoma), and SU-DHL-4 (diffuse large B-cell lymphoma). Compound 9 exhibited significant cytotoxicity against FARAGE and SU-DHL-4 tumor cell lines with IC50 values of 5.54 and 9.78μM, respectively. Further mechanism study showed that 9 could significantly promote apoptosis in FARAGE and SU-DHL-4 cell lines by interfering with mitochondrial function.

Abstract:The mangrove ecosystem is a rich resource for the discovery of actinomycetes with potential applications in pharmaceutical science. Besides the genus Streptomyces, Micromonospora is also a source of new bioactive agents. We screened Micromonospora from the rhizosphere soil of mangrove plants in Fujian province, China, and 51 strains were obtained. Among them, the extracts of 12 isolates inhibited the growth of human lung carcinoma A549 cells. Strain 110B exhibited better cytotoxic activity, and its bioactive constituents were investigated. Consequently, three new isoflavonoid glycosides, daidzein-4-(2-deoxy--l-fucopyranoside) (1), daidzein-7-(2-deoxy--l-fucopyranoside) (2), and daidzein-4,7-di-(2-deoxy--l-fucopyranoside) (3) were isolated from the fermentation broth of strain 110B. The structures of the new compounds were determined by spectroscopic methods, including 1D and 2D nuclear magnetic resonance (NMR) and high-resolution electrospray ionization mass spectrometry (HR-ESIMS). The result of medium-changing experiments implicated that these new compounds were microbial biotransformation products of strain M. aurantiaca 110B. The three compounds displayed moderate cytotoxic activity to the human lung carcinoma cell line A549, hepatocellular liver carcinoma cell line HepG2, and the human colon tumor cell line HCT116, whereas none of them showed antifungal or antibacterial activities.

Abstract:Two new coumarin derivatives, 4,4'-dimethoxy-5,5'-dimethyl-7,7'-oxydicoumarin (1), 7-(gamma,gamma-dimethylallyloxy)-5-methoxy-4-methylcoumarin (2), a new chromone derivative, (S)-5-hydroxy-2,6-dimethyl-4H-furo[3,4-g] benzopyran-4,8(6H)-dione (5), and a new sterone derivative, 24-hydroxylergosta-4,6,8(14), 22-tetraen-3-one (6), along with two known bicoumarins, kotanin (3) and orlandin (4), were isolated from an endophytic fungus Aspergillus clavatus (collection No. R7), isolated from the root of Myoporum bontioides collected from Leizhou Peninsula, China. Their structures were elucidated using 1D-and 2D-NMR spectroscopy, and HRESIMS. The absolute configuration of compound 5 was determined by comparison of the experimental and calculated electronic circular dichroism (ECD) spectra. Compound 6 significantly inhibited the plant pathogenic fungi Fusarium oxysporum, Colletotrichum musae and Penicillium italicum, compound 5 significantly inhibited Colletotrichum musae, and compounds 1, 3 and 4 greatly inhibited Fusarium oxysporum, showing the antifungal activities higher than those of the positive control, triadimefon.

Abstract:Five isocoumarin derivatives including three new compounds, aspergisocoumrins A–C (1–3), together with two known analogues, 8-dihydroxyisocoumarin-3-carboxylic acid (4) and dichlorodiaportin (5) were obtained from the culture of the endophytic fungus Aspergillus sp. HN15-5D derived from the fresh leaves of the mangrove plant Acanthus ilicifolius. Their structures were elucidated using comprehensive spectroscopic methods. The double bond geometry of compounds 1 and 2 were assigned as E and Z on the basis of the distinct coupling constants, respectively. Compounds 1 and 2 showed cytotoxicity against MDA-MB-435 with IC50 values of 5.08 ± 0.88 and 4.98 ± 0.74 μM, respectively.

Abstract:Four new isochromane derivatives (1-4) along with the known peniciphenol (5) and (R)-2-(hydroxymethyl)-3-(2-hydroxypropyl)phenol (6) were isolated from the EtOAc extract of the fermentation broth of the mangrove fungus, Aspergillus ustus 094102. The structures of the new compounds including the absolute configuration were elucidated on the basis of spectroscopic analysis, CD and ECD calculation. Compounds 1 and 2 exhibited alpha-glucosidase inhibition and antioxidation against DPPH radical with IC50 values of 1.4 mM and 25.7 mu M, respectively.

Abstract:Four new compounds, including three drimane sesquiterpenoids purpurides E-G(1-3) and one isocoumarin peniisocoumarin H(4), together with 15 known compounds, were isolated from the marine-sourced fungus Penicillium minioluteum ZZ1657. Their structures were elucidated by a combination of the HRESIMS data, extensive NMR spectroscopic analyses, and single crystal X-ray diffractions. Purpurides E(1) and F(2) are two N-acetyl-L-valine-conjugated drimane sesquiterpenoids and showed antimicrobial activities against methicillin-resistant Staphylococcus aureus, Escherichia coli, and Candida albicans with MIC values of 6-12 and 3-6 mu g/mL, respectively. Abstract:Objective To study the metabolites of mangrove fungus Penicillium oxalicum from the south China sea and search for new anti-tumor compounds.Methods Compounds were isolated by silica gel,ODS column chromatography,Sephadex LH-20 column chromatography and reversed-phase HPLC purification.Structural elucidation was achieved by physico-chemical constants and spectroscopic analysis.MTT assay was used to evaluate the bioactivities in vitro.Results Six cyclic dipeptides were isolated from the acetone extracts of mycelium of fungus and elucidated as cyclo-(Phe-Ile)(1),cyclo-(Phe-Val)(3),cyclo-(Ile-Leu)(3),cyclo-(Val-Val)(4),cyclo-(Pro-Val)(5),cyclo-(Pro-Gly)(6).Coumpouds 2,3 and 5 could evidently inhibit the growth of cancer cell lines HepG Ⅱ and LNCaP at the concentration of 50 μg·mL-1.Conclusions All of Compounds are obtained from the marine fungus for the fist time.Compounds 2,3 and 5 show cytotoxic activity in vitro.

The structure of the new compound was established by analysis of spectroscopic data. The hypothetical biogenic relationship of four sesquiterpene analogues was also described in this paper. Furthermore, in the cytotoxicity assays, compound 1 showed potent inhibiting activity towards MCF-7 and HL-60 cancer cell lines with 50% inhibition of cell growth (IC50) values of 4.49 and 3.43M, respectively, and compound 4 exhibited promising activity against HL-60 cell line with an IC50 value of 0.6M.
