Abstract:Novel cADPR mimics, which integrate nucleobase, northern and southern ribose modifications were synthesized. The key steps of the synthesis were a Cu(I)-catalyzed Huisgen [3+2] cycloaddition and a microwave-assisted intramolecular pyrophosphorylation. Preliminary biological investigations showed that these cADPR mimics are membrane-permeating agonists of the calcium signaling pathway. The introduction of chlorine or fluorine at the 2'-position of the southern riboses led to a decrease of activity. The existence of a hydrophobic group on the 3'-OH of the southern riboses does not obviously alter the agonistic activity.

Abstract:New cADPR mimic 5 that integrates the simplified nucleobase and ribose was designed and synthesized by using Cu(Ⅰ)-catalyzed Huisgen [3+2] cycloaddition to build the simplified triazole nucleobase and microwave-assisted reaction to carry out intramolecular pyrophosphorylation.Primary biological investigation showed that the introduction of an ether strand to replace northern and southern riboses could retain calcium inducing activity,but the introduction of a carbon strand to replace the northern ribose could lead to decreased activity.

Abstract:Here, we describe the mechanism of diphenyl α-aminoalkylphosphonate ester derivatives as potent inhibitors of prostate-specific antigen (PSA), a likely protease responsible for the advancement of prostate tumor progression. The AutoDock 4.2 molecular docking suite was utilized to model covalent and noncovalent binding of this class of inhibitors to predict crystallographic poses and compare experimental IC50 dose-response curves and in silico potencies for providing future more specific rational drug design. The new lead compound R/S-diphenyl[N-benzyloxycarbonylamino(4-carbamoylphenyl)methyl]phosphonate is being reported in this study as a potent inhibitor of PSA activity (IC50 = 250 nM; AutoDock Score = -8.29/-9.14 kJ·mol(-1) for R/S). Molecular dynamics (MD) simulations using GROMACS 4.6.5 was used to obtain trajectories of the top ligand and validate key interactions in the binding complex. A hydrogen-bonding map was used to confirm interactions between the lead compound and residues THR190, SER217, and SER227 in the P1 pocket. The modeling study introduces novel aminoalkylphosphonates as a potential drug candidate for targeting PSA by optimizing P1 binding affinities.

Abstract:N-1- [(5"-O-Phosphorylethoxy)methyl] -5'-O-phosphorylinosine 5',5"-cyclicpyrophosphate (cIDPRE 2a) and the 8-substituted derivatives 8-bromo-, 8-azido-, 8-amino-, and 8-Cl-cIDPRE (2b-e) were synthesized from N1- [(5"-acetoxyethoxy)methyl] -2',3'-O-isopropylideneinosine (5) in good yields. The pharmacological activities of cIDPRE and the 8-substituted derivatives (2a-e) were analyzed in intact and permeabilized. human Jurkat T-lymphocytes. The results indicate that cIDPRE permeates the plasma membrane, releases Ca2+ from an intracellular, cADPR-sensitive Ca2+ store, and subsequently initiates Ca2+ release-activated Ca2+ entry. The Ca2+-releasing activity of cIDPRE was confirmed directly in permeabilized. cells. Using time-resolved confocal Ca2+ imaging at the single cell level, the development of global Ca2+ signals starting from local small Ca2+ signals evoked by cIDPRE was observed. 8-N-3-cIDPRE 2c and 8-NH2-cIDPRE 2d were similarly effective in their agonistic activity as compared to cIDPRE 2a, showing almost indistinguishable concentration-response curves for 2a, 2c, and 2d and very similar kinetics of Ca2+ signaling. In contrast, the halogenated derivatives 8-Br- and 8-Cl-cIDPRE (2b and 2e) did not significantly elevate [Ca2+](i). Therefore, cIDPRE 2a, 8-N-3-cIDPRE 2c, and 8-NH2-cIDPRE 2d are novel membrane permeant cADPR mimic and may provide important novel tools to study cADPR-mediated Ca2+ signaling in intact cells.

Abstract:A novel cross-linked pyridine anion exchange membrane was synthesized via thermal cross-linking methods. 2,5-bis(2,3,4,5,6-pentafluorophenyl)-1,3,4-oxadiazole (FPOx) and diallyl bisphenol A (DABPA) were reacted to obtain the main polymer chain (MP). MP was substituted with 4-bromopyridine and 4-hydroxypyridine to generate two pyridine polymers conjugated pyridine polymer (CPP) and non-conjugated pyridine polymer (NPP). The different conjugation statues of CPP and NPP led to distinct mechanical and electrochemical properties. The diallyl groups in MP acted as the cross-linking structures which strengthened the mechanical properties of CPP and NPP and provided more flexible side chains. The ultimate membrane CPP-membrane and NPP-membrane were prepared by solution casting methods. Compared with NPP-membrane, the ionic conductivity of CPP can achieve to 20.1 mS/cm at 20 °C with excellent mechanical and thermal characteristics. Quantum theory computation and AFM morphology were carried out to figure out the reason of the difference in ionic conductivities and physical properties.

Abstract:We report the design, synthesis and evaluation of a novel macromolecular membrane permeabilizer displaying geminally substituted crown-ethers (18-crown-6) on every third carbon alongside the backbone. The polymer has a rather high affinity with potassium as well as permeabilization properties towards K+, Na+ and Ca2+, including single-channel behavior.

Abstract:A new family of aryl-extended calix[4]pyrroles with two phoshonate groups can act as ion-pair receptors for guest alkylammonium/phosphonium chloride salts, and the ion-pair binding mode and binding affinity of these hosts are different. In the present contribution, the origin of opposite ion-pair binding behavior for two typical hosts ii and oo towards primary ammonium chloride salt and quaternary phosphonium chloride salt was investigated using quantum mechanics (QM) calculations and a new noncovalent weak interaction analysis method. Two types of arrangements - separated and contact - were taken into account for each complex. The binding energy suggests that contact and separated arrangements are the favorable binding modes for receptor ii and oo respectively. Furthermore, the primary alkylammonium chloride salt binds stronger to ii than to oo, while the quaternary phosphonium chloride salt binds stronger to oo than to ii, in agreement with the experiment. Moreover, geometry, charge transfer based on natural bond orbital (NBO) and noncovalent weak interactions analysis suggest that the hydrogen bonding and cation-pi interactions play a critical role in ion-pair recognition of ii and oo respectively due to the different orientation of the P=O groups. This work further unveils the mechanism of ion-pair recognition by new calix[4]pyrrole bis-phosphonate receptors, while opening exciting perspectives for the design of stronger and more efficient calix[4]pyrrole-based ion-pair receptors.

Abstract:A novel trifluoromethylated analogue of cADPR, 8-CF3-cIDPDE (5) was designed and synthesized via construction of N-1, N-9-disubstituted hypoxanthine, trifluoromethylation and intramolecular condensation. A series of acyclic analogues of cADPR were also designed and synthesized. These compounds could be useful molecules for studying the structure-activity relationship of cADPR analogues and exploring the cADPR/RyR Ca2+ signalling system.

Abstract:For the investigation of the structure-activity relationship of the analogues of cyclic ADP-ribose (cADPR), a novel cyclic IDP-ribose analogue, cIDPRN, in which the northern ribose was replaced by N-carbobenzyloxy-alkylimine bridge was designed and synthesized. The enzymatic stability of cIDPRN was evaluated by the incubation with Jurkat T-lymphocytes for 0, 2 and 18 h. The result analyzed by capillary electrophoresis indicated that cIDPRN antagonized the hydrolysis, whereas cADPR was easily hydrolyzed. The Ca2+ signal release behavior was investigated in intact Jurkat T-lymphocytes by spectrofluorometer. It showed that cIDPRN induced calcium release either in extracellular Ca2+-containing or Ca2+-free condition. The result indicated that cIDPRN was a mild membrane-permeant agonist of cADPR/RyR signaling system. This study provided further information for understanding the effect of structure of northern ribose moiety of cIDPR on the calcium motivation activity.

Abstract:The title compound, C34H33N4, is a new 3,5-dimethylpyrazole-substituted paracyclonaphthane. Each of the two symmetry-independent molecules in the asymmetric unit occupies a special position on a twofold axis and exhibits an intramolecular pi,pi stacking interaction between the naphthalene moieties. The geometry of both molecules is essentially the same, the bridged rings of the naphthalene nuclei being deformed into a boat shape.

Abstract:The design, synthesis and structure-activity relationship studies of some novel trisubstituted pyrimidine amide derivatives prepared as CCR4 antagonists are described. The activities of these compounds were evaluated by the CCR4-MDC chemotaxis inhibition assay. Compound 1, which we have previously reported as a potent antagonist of CCR4, was employed as the positive control. The results indicated that most of the synthesized compounds exhibited some chemotaxis inhibition activity against CCR4. Of these new compounds, compounds 6c, 12a and 12b, with IC50 values of 0.064, 0.077 and 0.069 μM, respectively, showed higher or similar activity compared with compound 1 (IC50 of 0.078 μM). These compounds provide a basis for further structural modifications. The systematic structure-activity relationship of these trisubstituted pyrimidine amide derivatives was discussed based on the obtained experimental data. The results from the SAR study may be useful for identifying more potent CCR4 antagonists.

Abstract:C-2 dimethylated/unmethylated thiazolidine-4-carboxylic acid and C-2 dimethylated oxazolidine-4-carboxylic acid were introduced into the insect kinin core pentapeptide in place of Pro(3), yielding three new analogues. NMR analysis revealed that the peptide bond of Phe(2)-pseudoproline (Psi Pro)(3) is practically 100% in cis conformation in the case of dimethylated pseudoproline-containing analogues, about 50% cis for the thiazolidine-4-carboxylic acid analogue and about 33% cis for the parent Pro(3) peptide. The diuretic activities are consistent with the population of cis conformation of the Phe(2)-Psi Pro(3)/Pro(3) peptide bonds, and the results confirm a cis Phe-Pro bond as bioactive conformation. Abstract:A novel series of cyanoguanidine-piperazine P2X7 antagonists was designed based upon the structure of A-740003. Structure–activity relationship (SAR) studies focused on the piperazine moiety and the right hand side substitution. Compounds were assayed for activity at human and rat P2X7 receptors and compound 29 was found to possess potent activity (IC50 = 30–60 nM) at both species.

Abstract:PDZ domains are protein-protein interaction modules that coordinate multiple signaling and trafficking pathways in the cell and that include active therapeutic targets for diseases such as cancer, cystic fibrosis, and addiction. Our previous work characterized a PDZ interaction that restricts the apical membrane half-life of the cystic fibrosis transmembrane conductance regulator (CFTR). Using iterative cycles of peptide-array and solution-binding analysis, we targeted the PDZ domain of the CFTR-Associated Ligand (CAL), and showed that an engineered peptide inhibitor rescues cell-surface expression of the most common CFTR disease mutation ΔF508. Here, we present a series of scaffolds containing chemically modifiable side chains at all non-motif positions along the CAL PDZ domain binding cleft. Concordant equilibrium dissociation constants were determined in parallel by fluorescence polarization, isothermal titration calorimetry, and surface plasmon resonance techniques, confirming robust affinity for each scaffold and revealing an enthalpically driven mode of inhibitor binding. Structural studies demonstrate a conserved binding mode for each peptide, opening the possibility of combinatorial modification. Finally, we diversified one of our peptide scaffolds with halogenated substituents that yielded modest increases in binding affinity. Overall, this work validates our approach and provides a stereochemical foundation for further CAL inhibitor design and screening.
