Abstract:Two new quassinoids (picraquanines D and E, 1 and 2) and a new alkaloid (picraquanine F, 3), together with twelve known related compounds were obtained from the stems of Picrasma quassioides (D. Don) Benn. Their structures were elucidated on the basis of extensive spectroscopic techniques and chemical methods. The absolute configurations of 1 and 2 were determined by ECD experiments. Compound 10 showed nitric oxide (NO) inhibitory effects against LPS-stimulated RAW 264.7 cells with IC50 value of 96.1 μM.

Abstract:Picrasma quassioides, a member of the Simaroubaceae family, is the subject of research in numerous pharmacological and chemical studies. This plant mainly contains alkaloids, quassinoids and terpenoids. These molecules exhibit various pharmacological benefits, such as anti-inflammatory, anticancer, and anti-viral effects, on the cardiovascular system. Alkaloids make up the majority of these molecules. This review describes 127 alkaloid substances from P. quassioides. These alkaloids can be divided into the following classes: β-carbolines, canthinones and alkaloid dimers. A compilation of their nuclear magnetic resonance spectroscopy data and possible biosynthetic pathways of these compounds and the pharmacological effects of P. quassioides are also included.

Abstract:Abstract Three new phenolic derivatives, picraquanines A–C (1–3), along with 6 known ones 4–9 were obtained from the stems of Picrasma quassioides (D. Don) Benn. The new structures were determined by extensive spectroscopic data analysis, including IR, HRESIMS, 1H-NMR, 13C-NMR, HSQC, HMBC, 1H-1H COSY experiments. The absolute configuration of 1 was determined by comparison of its experimental and calculated ECD spectra. Furthermore, all the compounds were tested for their nitric oxide (NO) inhibitory effects against LPS-stimulated RAW 264.7 cells, however, none of them exhibited inhibitory effects (IC50 >100 μM).

Abstract:Picrasma quassioides (D.Don) Benn is a member of the Simaroubaceae family, which has a long history of medicinal use in China, the composition of compounds is complex, mainly including alkaloids, lignin, triterpenoids, and other compounds. As a traditional Chinese medicine, P. quassioides has pharmacological effects such as anti-inflammatory, antipyretic, antiviral, blood pressure lowering and anticancer. Scholars at home and abroad have been studying P. quassioides for about 50 years. In the present review, the research status of the chemical composition, pharmacological activity and pharmacokinetics of P. quassioides was provided, as a reference for further developing the value of P. quassioides.

Abstract:Two new dimeric β-carboline alkaloids, named kumujansine and kumujantine, were isolated from an alcohol extract of the heartwood of Picrasma quassioides (D. Don.) Benn. (Simaroubaceae) by silica gel and alumina column chromatography.Kumujansine hydrochloride was obtained as light yellow granulous crystal, mp 249～250℃ (dec). Its molecular formula is C_(28)H_(25)N_4O_2Cl. Based on spectral (UV, IR, ~1H-NMR, ~1H-~1H cosy spectrum and ~(13)C-NMR as well as MS) analysis its structure was elucidated as (Ⅰ).Kumujantine hydrochloride is also a light yellow granulous crystal, mp 270℃ (dec). Its molecular formula is C_(30)H_(29)N_4O_4 Cl. Based on analysis of spectral data its structure was identified as (Ⅱ).

Abstract:Phytochemical investigation on the stems of Alstonia scholaris (L.) R. Br. led to the isolation and identification of 30 natural products, including sixteen alkaloids (1–16), nine triterpenoids (17–25), two phenols (26–27), and three lignans (28–30). Their structures were established by comprehensive spectroscopic analyses together with the comparison with reported data. Compound 1 was a new indole alkaloid. Compound 24 was firstly isolated from Apocynaceae family, compounds 22–23 and 26–30 were obtained from the genus Alstonia for the first time, and this is the first report of the presence of 11 and 16 in A. scholaris. The chemotaxonomic significance of these isolated compounds was also discussed.

Abstract:Picrasma quassioides (D. Don) Benn. is a widely used traditional Chinese medicine for anti-inflammation and antibiosis. Canthinone and beta-carboline alkaloids are the main characteristic constituents that possess diverse pharmacological effects, such as anti-inflammatory and anti-infectious properties. In this study, bioautography in thinlayer chromatography indicated that the antiradical activity compound may be alkaloids. Then, a simple, fast, and efficient method was established for the separation and purification of two types of alkaloids from P. quassioides by mass-spectrometry-directed autopurification system. Eight alkaloids were isolated and purified in this one-step methodology. Among them, five compounds (3, 95.1%, 58.8 mg; 4, 98.4%, 71.7 mg; 6, 97.8%, 365.4 mg; 7, 97.7%, 172.7 mg; 8, 98.2%, 180.3 mg) were obtained in large amounts with extremely high purities. Then, the antiradical activities of the isolates showed that 4-methoxy-5-hydroxycanthin-6-one (6) exhibited obvious 1,1-diphenyl-2-picryl-hydrazyl free radical scavenging activity with an IC50 value of 84.037 mu M. This study offers a new method for the preparation of targeted bioactive alkaloids in P. quassioides. This work also provides a reference for the separation of other targeted chemical components with potential activities from traditional Chinese herbal medicines.

Abstract:Nigakialcohol A (1), as unusual cyclization ionone derivative, together with eight known ones (2-9), were isolated from the leaves of Picrasma quassioides (D. Don) Benn (Simaroubaceae). Their structures were elucidated by extensive spectroscopic analyses and comparison with literature data. Compound 2 showed a weak inhibitory effect on NO production at non-cytotoxic concentration (100 mu M) with inhibitory rate of 59%, and thus it should be regarded as potential anti-inflammatory agents. Abstract:Electro-thermal transport in a new class of nanotube bundle transistors and their application to electronic displays is considered. The transistors are made of nanocomposites composed of isotropic 2D ensembles of nanotubes or nanowires in a plastic or glass substrate. The random nanotube network is generated numerically and simulated using a finite volume scheme. The voltage and spatial power distribution in the transistor is computed in the linearly-biased regime. The computed power distribution is used as Joule heat source in a Fourier heat transport model for the nanotube network and substrate and the temperature rise in tube network and substrate are predicted. The effect of tube-tube contact conductance, tube-substrate contact conductance and substrate-tube conductivity ratio on the temperature rise is analyzed. The effect of convective cooling on the temperature rise is investigated for a range of heat transfer coefficients between the display surface and the ambient air. The tube-substrate contact resistance emerges as a dominant resistive component for the tube temperature rise, but is found to be insignificant for the substrate temperature rise. Tube conductivity and tube-tube contact conductance are found to have only a minor effect on lateral heat spreading because the dominant resistance to heat removal lies on the substrate and the air side for the range of parameters investigated

Abstract:This study aimed to investigate the molecular mechanism of Picrasma quassioides Benn against inflammation by means of network pharmacology. The paper will provide a reference for multi-target and multi-channel treatment of inflammation with traditional Chinese medicine. Through screening and analysis, 11 active ingredients and 109 anti-inflammation prediction targets were obtained and constructed a compound-target network. The targets such as VEGFA, TLR4 and STAT3 may play a crucial role. Network enrichment analysis showed that the 109 potential targets constitute a number of pathways or inflammatory reactions closely related to inflammation, including NF-κB signaling pathway and MAPK signaling pathway. The docking results indicated that the binding energy of Picrasidine Y and the inflammatory factors VEGFA is the highest. This study predicted the role of multiple active compounds in the alkaloids of Picrasma in the inflammatory response, and provided a theoretical basis for the anti-inflammatory mechanism of Picrasma.

Abstract:A bioactivity-guided study on the leaves of Picrasma javanica led to the isolation of 19 quassinoids, including 13 new compounds. The structures of the new compounds were elucidated by a combination of spectroscopic data analysis, X-ray crystallography studies, and electronic circular dichroism (ECD) data interpretation. Compounds 1–7 are rare examples of quassinoids with a keto carbonyl group at C-12. The biological activities of 11 of the more abundant isolates were evaluated against five phytopathogenic fungi in vitro, and several of them including 6 and 15 showed moderate inhibitory effects that were comparative to those of the positive control, carbendazim. Abstract:Three unusual sesterterpenoids featuring unprecedented rearranged colquhounane (C25) and tetranorcolquhounane (C21) frameworks, colquhounoids E (1) and F (3) and norcolquhounoid F (2), were isolated from a Lamiaceae medicinal plant Colquhounia coccinea var. mollis. Their structures were elucidated by spectroscopic analysis and quantum chemical calculations. A biomimetic inspired regioselective cyclopropane cleavage was achieved under acidic conditions. The immunosuppressive activities of these new sesterterpenoids were also evaluated.

Abstract:Three new C-20 quassinoids, namely, nigakilactones R (1), S (2), and T (3), along with 13 known ones, were isolated on Picrasma quassioides leaves. Their structures were elucidated by an extensive spectroscopic analysis. All the compounds were tested for their inhibitory activities against the six major CYP450 enzymes CYP1A2, 2C8, 2C9, 2C19, 2D6, and 3A4. Compounds 1, 10, 11, and 12 showed potential as inhibitor of CYP2C19 with IC50 values of 62.6, 160.0, 61.5, and 35.8 mu M, respectively. Compounds 1, 3, and 6 exhibited considerable inhibition against CYP3A4 with IC50 values of 51.4, 97.2, and 62.8 mu M, respectively. CYP2C19 and 3A4 are important metabolic enzymes involved in many drugs. Our study may provide a cautionary measure to avoid the combined use of CYP2C19- and 3A4-catalyzed drugs with the quassinoid-containing herbs and related formulae.

Abstract:A novel alkaloid with a hexa-tetra-hexa-cyclic skeleton, Bi-4-methoxycarbonyl-2-quinolone ( 1 ), was discovered during the investigation of Brucea javanica . Additionally, six known alkaloids ( 2 – 7 ) were also found. The chemical structures of these compounds were identified using HRESIMS and 1D and 2D NMR spectroscopic analysis. Additionally, the absolute configuration of the new compound 1 was determined through X-ray single crystal diffraction. Compound 1 exhibited significant anti-inflammatory activity in RAW264.7 cells and demonstrated promising anti-cancer effects in Lewis cells.

Abstract:A new alkaloid, identified as 1-benzyl-2-nitroso-1,2,3,4-tetrahydroisoquinoline-6,7-diol, named oleraisoquinoline ( 1 ), and five organic acids and two esters, identified as 5-(hydroxymethyl)furan-2-carboxylic acid ( 2 ), 1 H -pyrrole-2,5-dicarboxylic acid ( 3 ), (7 E ,10 E )-octadeca-7,10-dienoic acid ( 4 ), (10 E ,13 E )-octadeca-10,13-dienoic acid ( 5 ), (7 E ,10 E )-hexadeca-7,10-dienoic acid ( 6 ), methyl tridecanoate ( 7 ) and methyl (9 E ,12 E )-octadeca-9,12-dienoate ( 8 ), were isolated from Portulaca oleracea L., among which compounds 2 and 4 ‒ 7 were isolated for the first time.
