Abstract:The 1-N-benzyl-5-iodo(or bromo)uracil undergoes Pd-catalyzed [Pd-2(dba)(3)] direct arylation with benzene and other simple arenes in the presence of TBAF in DMF without the necessity of adding any ligands or additives to give 5-arylated uracil analogues. The TBAF-promoted coupling also occurs efficiently with electron rich heteroarenes at 100 degrees C (1 h) even with only small excess of heteroarenes. The protocol avoids usage of the arylboronic acid or stannane precursors for the synthesis of 5-(2-furyl, or 2-thienyl, or 2-pyrroly)uracil nucleosides, which are used as important RNA and DNA fluorescent probes. The fact that 1-N-benzyl-3-N-methyl-5-iodouracil did not undergo the TBAF-promoted couplings with arenes or heteroarenes suggests that the C4-alkoxide (enol form of uracil) facilitates coupling by participation in the intramolecular processes of hydrogen abstraction from arenes. TBAF-promoted arylation was extended into the other enolizable heterocyclic systems such as 3-bromo-2-pyridone. The it-excessive heteroarenes also coupled with 5-halouracils in the presence of Pd(OAc)(2)/Cs2CO3/PivOH combination in DMF (100 degrees C, 2 h) to yield 5-arylated uracils.

Abstract:An efficient synthesis of novel fused heterocycles has been established via microwave-assisted palladium-catalyzed intramolecular direct arylation. The acyclic aryl bromides are readily available from microwave-assisted one-pot annulation of N-(2-bromobenzyl)-2-aminophenois and ethyl 2-bromoalkanoates. The intramolecular direct arylation is then performed in the presence of palladium(II) acetate and dppf (10 mol% each) in toluene using potassium carbonate (2 equiv) as the base under microwave heating (150 degrees C, 1 h) to afford the products in 43-99% yields. Steric effect is observed for Ar-Ar bond formation, giving a substantial amount of the debromination byproduct.

Abstract:A Lewis-acid-promoted approach to the synthesis of highly functionalized dihydrobenzofuran derivatives was developed. A diverse range of functional groups are tolerated in this type of reaction. The reaction mechanism investigation indicates that the highly reactive phenyl cation intermediate is probably involved in this process. The chirality of substrate is retained under the reaction conditions.

Abstract:A method for the direct arylation of 5-iodouracil and 5-iodouridine was found to proceed in moderate yields. By irradiating mixtures of 5-iodouracil or 5-iodouridine and a series of five-membered heterocycles such as 1H-pyrrole, furan, 2-methylfuran, 1-methyl-1H-pyrrole, thiophene, as well as benzene in MeCN/H2O with a Hg lamp, 5-aryluracils and 5-aryluridines were synthesized. The reaction proceeded smoothly without the requirement of adding any transition metals or ligands.

Abstract:Herein we report the first versatile and expeditious method for the site-selective C-H fluoromethylation of aryl iodides via Pd/norbornene cooperative catalysis, which could work as a robust toolbox for the diversity-oriented synthesis (DOS) of fluoromethylated arenes. This methodology features the use of the low-cost industrial raw material CH<sub>2</sub>IF as the fluoromethyl source, an excellent functional group tolerance, and a broad <i>ipso</i> termination scope and can be expanded to the late-stage modification of biorelevant molecules.

Abstract:The reactivity of di-, tri- and tetra-fluoroalkoxy-substituted bromobenzenes in the direct arylation of 5-membered ring heteroarenes using palladium catalysis was explored. High yields in arylated heteroarenes were obtained using only 1 mol-% of Pd(OAc)(2)catalyst with KOAc as an inexpensive base. Similar yields were obtained witho/m/ptrifluoromethoxy-,o/pdifluoromethoxy-, and tetrafluoroethoxy-substituents on the aryl bromide. A bromo-substituted difluorobenzo[d][1,3]dioxole was successfully coupled. The major side-products of the reaction are HBr/KOAc. Therefore, this synthetic scheme is very attractive for the access to such polyfluoroalkoxy-containing arylated heteroaromatics in terms of cost, simplicity, and low environmental impact, compared to reactions involving arylation of heteroarenes with bromophenols followed by polyfluoroalkylation.

Abstract:Reported herein is a Pd-catalyzed regioselective C-H activation method that is used for C-H deuteration, carbonylation, halogenation, and oxidation of arene substrates substituted by two N-heterocycles. When conducted in acetic acid (AcOH), these reactions occur at the five-membered palladacycle sites, whereas they switch to the six-membered palladacycle sites in trifluoroacetic acid (TFA). This controllable regioselective C-H activation is applied for late-stage functionalization of bioactive molecules. A mechanism study indicated that the regioselectivity is achieved by Brönsted acid-Lewis base interactions and electronic effects (in TFA) and the different kinetic stabilities of palladacycle intermediates (in AcOH).
