Abstract:Transition metal-catalyzed direct functionalization of C-H bonds is one of the key emerging strategies that is currently attracting tremendous attention with the aim to provide alternative environmentally friendly and efficient ways for the construction of C-C and C-hetero bonds. In particular, the strategy involving regioselective C-H activation assisted by various functional groups shows high potential, and significant achievements have been made in both the development of novel reactions and the mechanistic study. In this review, we attempt to give an overview of the development of utilizing the functionalities as directing groups. The discussion is directed towards the use of different functional groups as directing groups for the construction of C-C and C-hetero bonds via C-H activation using various transition metal catalysts. The synthetic applications and mechanistic features of these transformations will be discussed, and the review is organized on the basis of the type of directing groups and the type of bond being formed or the catalyst.

Abstract:Highly regioselective C(sp3)–H bond functionalization is a very important and attractive research topic as organic compounds usually contain several different kinds of C–H bonds (sp, sp2, and sp3). The introduction of a directing group has become one of the most efficient strategies to achieve this target. In this chapter, we discuss the latest advances in metal-catalyzed arylation, allenylation, amidation, and alkylation of inactivated C(sp3)–H bonds enabled by the directing group strategy.

Abstract:Transition-metal-catalyzed, coordination-assisted C(sp3)-H functionalization has revolutionized synthetic planning over the past few decades as the use of these directing groups has allowed for increased access to many strategic positions in organic molecules. Nonetheless, several challenges remain preeminent, such as the requirement for high temperatures, the difficulty in removing or converting directing groups, and, although many metals provide some reactivity, the difficulty in employing metals outside of palladium. This review aims to give a comprehensive overview of coordination-assisted, transition-metal-catalyzed, direct functionalization of nonactivated C(sp3)-H bonds by covering the literature since 2004 in order to demonstrate the current state-of-the-art methods as well as the current limitations. For clarity, this review has been divided into nine sections by the transition metal catalyst with subdivisions by the type of bond formation. Synthetic applications and reaction mechanism are discussed where appropriate.

During the recent years, significant progress has been made in this fascinating field with various functionalized alkenes, heterocycles and carbocycles being obtained. Abstract:In the past decades, transition metal-catalyzed C-H activation has experienced tremendous growth and revolutionized the field of organic synthesis. Several elegant strategies have been developed to promote reactivity and control precise site-selectivity. Among which, transient directing group strategy has been recognized to be an efficient and powerful approach for selective C-H functionalization. In contrast to traditional directing groups with covalent linkage, transient directing group strategy circumvents the covalent installation and removal of directing groups, which significantly improve the synthetic efficiency and broaden the range of synthetic applications. The recent advances in imine-based transition directing groups are summarized, providing an overview of recent achievements in this cutting-edge research field over the past few years. For clarity, it is classified into two sections according to the type of substrate and the type of activated hydrocarbon bond. Emphasis is placed on the fully discussion of various transient directing groups and their applications. Finally, the limitations of previous works and perspectives on this cutting-edge area are also described.

Abstract:The need to use a static directing group represents a major limitation for the emerging field of ortho-selective C–H activation and functionalization. Chemistry was recently developed that allowed partial removal or minor transformations for certain directing groups. Our recent work demonstrated that triazenes were a class of excellent directing groups for C–H activation/functionalization, and subsequent chemical manipulations generated key compounds with synthetic versatility.

Abstract:Transition-metal-catalyzed C–H bond functionalization has known a rapid evolution in the last years, offering modern strategies for reaching high molecular complexity in a step- and atom-economical way. Despite the indisputable advances, selectivity issues still remain, given the ubiquity of C–H bonds on molecules; thus, several approaches have been developed to tackle this challenge. Among them, the use of a transient directing group has emerged as an effective tool, circumventing the need for extra synthetic steps to install and then cleave a directing group on the molecule. More recently, this strategy has been successfully applied to the even more challenging transition-metal-catalyzed enantioselective C–H bond functionalization. This review will highlight and discuss the main advances made in the use of a chiral transient directing group for the enantioselective functionalization of C(sp2)–H and C(sp3)–H bonds by transition-metal catalysis.This article has not yet been cited by other publications.

Abstract:During the past decades, synthetic organic chemistry discovered that directing group assisted C–H activation is a key tool for the expedient and siteselective construction of C–C bonds. Among the various directing group strategies, bidentate directing groups are now recognized as one of the most efficient devices for the selective functionalization of certain positions due to fact that its metal center permits fine, tunable, and reversible coordination. The family of bidentate directing groups permit various types of assistance to be achieved, such as N,N-dentate, N,O-dentate, and N,S-dentate auxiliaries, which are categorized based on the coordination site. In this review, we broadly discuss various C–H bond functionalization reactions for the formation of C–C bonds with the aid of bidentate directing groups.

Abstract:In order to directly functionalize C−H bonds of complex molecules and, in particular, to control the regioselectivity of the reaction, a wide range of directing groups has been used. However, these directing groups need to be installed and removed for further applications, which may limit the use of C−H activation in synthesis. Concerning aldehydes and ketones, a transient directing group strategy has recently emerged to overcome this drawback. The addition of an additive, in general an amine, allowed the in situ formation of the real directing group to achieve C−H activation. This review presents the latest developments in the field over the period 2020‐2023.

Abstract:Transition metal-catalyzed direct C-H functionalization is one of the most efficient and powerful tools for the rapid synthesis of organic molecules. The use of functional groups in the molecules or covalently attached coordinating groups as directing groups has been realized as a major strategy to control the selectivity. Noncovalent interactions are of great importance in the field of molecular biology, supramolecular chemistry, material science and drug discovery. More recently, the use of well-designed ligands to enable the site-selective C-H functionalization via noncovalent interactions has emerged as a highly promising yet relatively less explored strategy. In this perspective, recent advances in this cutting-edge area are summarized. The perspective was classified into four sections according to the type of noncovalent interactions, including hydrogen bonding, ion pair, Lewis acid-base interaction and electrostatic interaction. Emphasis is placed on the mode of noncovalent interactions among the transition metals, ligands and substrates. The limitation of current research and the prospect of future work will also be discussed. We anticipate that this strategy might become a promising complementary strategy to control the positional selectivity in C-H functionalization reactions.

Abstract:During the last two decades, impressive advancements have been achieved in transition metal‐catalyzed chelation‐assisted C—H functionalization reaction. While reactions in this field are still dominated by precious 4d or 5d metals (e.g., Pd, Rh, Ir), the 3d base metals (e.g., Ni, Co, Cu, Fe) have made significant headway partially due to their relatively large abundance, low cost, low toxicity as well as their occasionally occurred novel reactivity when compared to their noble cousins. This review will give a comprehensive summary on Ni‐catalyzed functionalization of inert C—H bonds assisted by chelation groups. For clarity, the content is classified by the newly formed chemical bonds, namely C—C, C—N, C—chalcogen and C—halogen bond formation.

Abstract:Direct C–H functionalization has recently emerged as one of the most efficient strategies to access structurally complex molecules from readily accessible feedstocks in an atom- and step-economic manner. In particular, enantioselective C–H activation has garnered increasing attention by enabling chemists to efficiently assemble valuable chiral compounds by asymmetrically manipulating C–H bonds into useful functionalities. Apart from the extensively studied C–C bond formation, very few endeavors have been focused on the C–X formation analogs. Motivated by the utility of the latter approach in constructing academically and industrially important heteroatom-containing chiral compounds, we provide herein an overview on C–X forming asymmetric C–H activation reactions proceeding through C–H metalation. The advancements are organized according to the employed catalytic systems, which include Pd(II) catalysis, group-9 CpxM(III) catalysis, monovalent group-9 metal catalysis, and multi-boryl/silyl Ir(III) catalysis, with emphasis on the design philosophy, mechanism, and mode of enantiocontrol.

Abstract:Over the past few decades, transition-metal catalyzed direct functionalization of unactivated C-H bonds has emerged as an efficient and straightforward strategy for the construction of C-C and C-heteroatom bonds.It provides access to complex molecules via the direct cleavage of C-H bonds without the need of prefunctionalization, rendering this strategy more step-economical and environmentally benign.Recently, tremendous attentions have been devoted to the use of first row-transition metals as most of those metals are naturally abundant, readily available, and inexpensive and have low toxicities in comparison with precious metals.In addition, the exploration of new catalytic systems might lead to the discovery of new mode of reactions.The aim of this review is to give an overview on recent advances in nickel-catalyzed direct functionalization of unactivated C-H bonds, without the discussion of the cleavage of acidic C-H bonds.Firstly, nickel-catalyzed functionalization of C(sp 2 )-H to form new C-C and C-heteroatom bonds is reviewed.Secondly, an overview of Ni-catalyzed C(sp 3 )-H functionalization is provided.In addition, mechanistic hypothesis and general aspects of these reactions have also been summarized.Finally, the limitations and perspectives on nickel-catalyzed C-H functionalization are discussed.
