Abstract:A simple, highly efficient, one-pot synthesis of 3,5-diaryl-4-bromopyrazoles via 1,3-dipolar cycloaddition of diazo compounds and alkynyl bromides has been developed. The diazo compounds and alkynyl bromides were generated in situ from tosylhydrazones and gem-dibromoalkenes, respectively. When ketone-derived hydrazones were used, 3,5-diaryl-4-bromo-3H-pyrazoles were obtained and the isomerization products 3,5-diaryl-4-bromo-1H-pyrazoles were formed when using aldehyde-derived hydrazones. The reaction system exhibited high regioselectivity and good functional group tolerance. Both electron-rich and electron-deficient substituents on the aromatic ring of the hydrazones or the gem-dibromoalkenes gave desired products in moderate to good yields (67-86%).

Abstract:An efficient synthesis of 1,3-diaryl-4-halo-1H-pyrazoles was achieved. The synthesis involves the [3 + 2] dipolar cycloaddition of 3-arylsydnones and 2-aryl-1,1-dihalo-1-alkenes. The process proceeds smoothly in moderate to excellent yields. 1,3-Diaryl-4-halo-1H-pyrazoles are found to be important intermediates that can easily be converted into 1,2,5-triaryl-substituted pyrazoles via Pd-catalyzed C-H bond activation.

Abstract:The first conjugate addition of alpha-isocyanoacetates with in situ generated azoalkenes from alpha-halogeno hydrazones in the presence of Cs2CO3 has been developed. The reaction condition was suitable to various alpha-aryl substituted isocyanoacetates as well as aryl and alkyl-substituted alpha-halogeno hydrazones, affording the corresponding products in good to high yields. The adducts can be easily transformed into biologically attractive 1,4-dihydropyrimidine derivatives via an intramolecular cyclization by treating with AgOAc. The one-pot conjugate addition/intramolecular cyclization has also been investigated to furnish the formal [3 + 3] cycloadduct, 1,4-dihydropyrimidine derivatives.(C) 2021 Elsevier Ltd. Abstract:A formal [3 + 3] cycloaddition reaction between alkyl 2-aroyl-1-chlorocyclopropanecarboxylates 1 and common hydrazones 2 was developed. This process readily proceeded in diastereo- and regioselective fashion and gave alkyl 1,4,5,6-tetrahydropyridazine-3-carboxylates in high yields under mild basic conditions. Treatment of the annulation products with DDQ leads to the formation of functionalized pyridazine-3-carboxylates, analogues of nicotinic acid ester.
