Abstract:A new sesquiterpenoid, diaporol A (1), possessing a unique tricyclic lactone framework, eight new drimane sesquiterpenoids, diaporols B-I (2-9), and the known compounds 10 and 11 were isolated from a culture of the mangrove-derived endophyte Diaporthe sp. The absolute configurations of 1-5 were determined by low-temperature (100 K) single-crystal X-ray diffraction with Cu Kα radiation. The compounds were evaluated for cytotoxic activity; however, no compound showed significant cytotoxicity against the tested cell lines at a concentration of 20 μM.

Abstract:A new eremophilane sesquiterpene, capnodiumone (1), along with five known eremophilane sesquiterpenes (2-6) and eight other compounds (7-14), have been isolated from a mangrove-associated fungus Capnodium sp. SZ-F22. The chemical structures were elucidated on the basis of extensive spectroscopic analysis. The broth extract of the fungus exhibited a good inhibitory effect on the mycelium growth against Fusarium graminearum at 100g/mL, however, all the 14 compounds showed no expected antifungal activity. Abstract:Phytopathogenic species of Diaporthe are associated with melanose, stem-end rot and gummosis diseases of Citrus. However, little is known about the occurrence of species of Diaporthe as endophytes and saprobes. In this study, we obtained 58 strains of Diaporthe, including 44 endophytic isolates from cultivated Citrus in China. The nuclear ribosomal internal transcribed spacer (ITS), partial sequences of translation elongation factor 1-α (EF1-α), beta-tubulin (TUB), and histone-3 (HIS) gene regions were analysed to determine the species of the isolates collected. In combined analysis of four gene regions, these strains grouped in 16 distinct clades in Diaporthe. The isolates were identified in Diaporthe arecae species complex, Diaporthe citri, Diaporthe citriasiana, Diaporthe citrichinensis, Diaporthe endophytica, Diaporthe eres, Diaporthe hongkongensis, and Diaporthe sojae based on molecular phylogeny and morphology. Seven new species are described from Citrus namely, Diaporthe biconispora, Diaporthe biguttulata, Diaporthe discoidispora, Diaporthe multigutullata, Diaporthe ovalispora, Diaporthe subclavata, and Diaporthe unshiuensis with descriptions and illustrations.

Abstract:Four new eudesmane-type sesquiterpenoids, penicieudesmol A-D (1-4), were isolated from the fermentation broth of the mangrove-derived endophytic fungus Penicillium sp. J-54. Their structures were determined by spectroscopic methods, the in situ dimolybdenum CD method, and modified Mosher's method. The bioassays results showed that 2 exhibited weak cytotoxicity against K-562 cells.

Abstract:Six new xanthone dimers, diaporxanthones A-F (1-6), and an unusual xanthone monomer diaporxanthone G (7), in addition to seven known analogues (8-14), were isolated and identified from endophytic Diaporthe goulteri L17 harbored in the fruits of the salt-tolerant plant Vitex trifolia. The chemical structures of these metabolites were elucidated on the basis of nuclear magnetic resonance, high-resolution electrospray ionization mass spectrometry, and reported data in the literature. Their absolute configurations were established by single-crystal X-ray diffraction analysis together with time-dependent density functional theory electronic circular dichroism calculations. Among these compounds, compounds 1 and 6 exhibited moderate antifungal activities against Nectria sp. and Colletotrichum musae and compound 4 showed significant cytotoxicity against all selected five cancer cell lines.

Abstract:A new sesquiterpene, named (1S, 4aS, 7 R, 8 R, 8aR) Decahydro-1,4a-dimethyl-7-(1-methylethyl)-1,8-naphthalenediol (1), and six known compounds (2-7) were obtained from Colletotrichum sp. B-89, an endophytic fungus isolated from Dracaena cochinchinensis collected at Xishuangbanna, Yunnan. Their structures were elucidated by detailed analysis of comprehensive spectroscopic data, and the structure of 1 was further determined by X-ray diffraction analysis. The antibacterial activities of above compounds were assayed and compound 6 exhibited certain activities against S. aureus ATCC 25923, B. subtilis ATCC 6633, E. coli ATCC 25922, and K. pneumoniae ATCC 13883.

Abstract:Two novel 3,5-dimethylorsellinic acid derived meroterpenoids, peniciterpenolides A (1) and B (2) individually containing a 2-oxaspiro[4,5]deca-6,8-diene-1,4-dione and a 6-oxabicyclic[3,2,1]oct-3-en-7-one motif, one new spongian-type diterpene, peniciterpenolide C (3), and one new drimane-type sesquiterpene, peniciterpenolide D (4), together with the known meroterpenoid atlantinone B (5) were isolated from Penicillium chrysogenum MT-40, an endophytic fungus identified from Huperzia serrata. All the five compounds additionally contain a unique 2-oxabicyclo[2.2.2]octan-3-one moiety formed by the annulation of the C-3 hydroxyl and the oxidized methyl on C-10. The structures were established by the analysis of their spectroscopic data, single-crystal X-ray diffraction, and quantum chemical calculations of electronic circular dichroism spectra. Compounds 1-5 showed inhibition of nitric oxide production in lipopolysaccharide-activated RAW 264.7 macrophage cells with IC50 values in the range of 20.6-46.9 mu M.

Abstract:Three new azaphilone compounds, isochromophilones X–XII ( 1–3 ), together with two known ones sclerotioramine ( 4 ) and isochromophilone VI ( 5 ) were isolated from the cultures of an endophytic fungus Diaporthe sp. The structures were elucidated by extensive HRESIMS and NMR spectroscopic analyses. All compounds were tested for their cytotoxicities against five human cancer cell lines by MTT method, among which compound 1 showed moderate inhibitory effects on these cell lines. This was the first report of azaphilones isolated from Diaporthe sp.

Abstract:Chemical investigation of the culture extract of an endophyte Xylaria curta YSJ-5 from Alpinia zerumbet (Pers.) Burtt. et Smith resulted in the isolation of eight previously undescribed compounds including five eremophilane sesquiterpenes xylarcurenes A-E, one norsesquiterpene xylarcurene F, and two α-pyrone derivatives xylarpyrones A-B together with eight known related derivatives. Their chemical structures were extensively established based on the 1D- and 2D-NMR spectroscopic analysis, modified Mosher's method, electronic circular dichroism calculations, single-crystal X-ray diffraction experiments, and the comparison with previous literature data. All these compounds were tested for in vitro cytotoxic, anti-inflammatory, α-glucosidase inhibitory, and antibacterial activities. As a result, 6-pentyl-4-methoxy-pyran-2-one was disclosed to display significant antibacterial activity against Staphylococcus aureus and methicillin-resistant S. aureus with minimal inhibitory concentration value of 6.3 μg/mL.

Abstract:A new polyketide named (1R,2E,4S,5R)-1-[(2R)-5-oxotetrahydrofuran-2-yl]-4,5-dihydroxy-hex-2-en-1-yl(2E)-2-methylbut-2-enoate (1), along with eight known polyketide including one monoterpene (2), three linear furanopolyketides (3-5) and four lovastatin analogues (6-9), was isolated from the endophytic fungal strain Diaporthe sp. SXZ-19 of Camptotheca acuminate. The chemical structures of compounds 1-9 were elucidated on the basis of extensive spectroscopic analyses, including FT-ICR-MS, IR and 1D and 2D NMR experiments. The in vitro cytotoxicity of 1 against the human colon cancer cell line HCT 116 was evaluated but showed no evident activity.

Abstract:Four new highly oxidized caryophyllene-type sesquiterpenes (1-4), one new caryophyllene-type natural product (5), and two new drimane-type sesquiterpenes (6, 7), along with eight known compounds have been isolated from an endophytic fungus Pestalotiopsis adusta. Their structures were elucidated on the basis of HRESIMS, 1D and 2D NMR spectroscopic data analyses. The absolute configurations for 1 and 5 were determined by single-crystal X-ray crystallographic analysis using Cu K alpha radiation. The absolute configuration of 6 was determined by CD spectrum associated with TD-DFT calculation of its benzoylated derivative 6a. The in vitro bioassay revealed that 8 and 9 presented cytotoxicity against cancer cell lines A549, HeLa, and SMMC-7721, in which 8 showed comparable activity (IC50 = 28.3 mu M) to the positive control etoposide (IC50 = 23.2 mu M) against SMMC-7721.

Abstract:A chemical investigation of the endophytic fungus Diaporthe destruens from the Hernandiaceae plant Illigera orbiculata C. Y. Wu collected from southern Yunnan Province, China, led to the isolation of six undescribed compounds, including two azaphilone analogs, which are a pair of epimers (13R-hydroxy-chermesinone A and 13S-hydroxy-chermesinone A); a pyrrole derivative (1-(4-(methoxymethyl)-1H-pyrrol-3-yl)ethan-1-one); an isoindolone derivative (4-hydroxy-6-methoxyisoindolin-1-one); a benzylbenzene derivative (destruensine A) and a conjectural fragment of polyketide ((2R,4R)-2-(methoxymethyl)pentane-1,4-diol) along with nine known compounds. Their structures were elucidated by spectroscopic methods and HRESIMS, and the absolute configurations were further confirmed by electronic circular dichroism (ECD) and chemical derivatization. The antimicrobial activities, anti-acetylcholinesterase activities, antiproliferation, and NO production inhibitory effects of compounds 1–15 were evaluated.

Abstract:Bioinformatics analysis of the synthetic gene clusters in association with the qRT-PCR detection led to speculate a marine derived fungus, Penicillium brasilianum WZXY-m122-9, to be potential to produce meroterpenoids. Chromatographic separation of the EtOAc extract of this fungal strain on a large-scale fermentation resulted in the isolation of six new DMOA-related meroterpenoids, namely brasilianoids G‒L (1–6), together with nine known meroterpenes. Their structures were determined by extensive analyses of spectroscopic data, including the X-ray diffraction and the ECD data for configurational assignment. Brasilianoid G is characterized by the presence of an unprecedented scaffold bearing a 6/6/5/5/5 pentacyclic nucleus, while brasilianoid K possessing a 7/6/6/6/5 pentacyclic system is uncommonly found from nature. The biogenetic relationship among the isolated compounds was postulated based on the comparative analyses of gene clusters. Brasilianoid L (6), austinol (12), and dehydroaustin (14) exhibited significant inhibition against bacteria invasion into host cells, that was suggested to be mediated by preventing the polymerization of actin in the host cells.
