Abstract:Seven new quinoxaline derivatives 3a-3g were synthesized through two steps convenient approach. Their structures were confirmed by Fourier transform infrared, proton nuclear magnetic resonance, and high-resolution mass spectrometry. The synthesized compounds 3a-3g showed good fluorescent intensity in dimethyl sulfoxide. They were also shown to possess good antifungal activities. The structure-activity relationship was studied by density functional theory calculations.

Abstract:Two unprecedented fluorescent nucleosides that feature BN-doped polycyclic aromatic hydrocarbons are presented. One of them, having a BN-modified phenanthrene moiety incorporated, shows blue fluorescence but suffers from poor stability under aqueous conditions. The other nucleoside comprises an internally BN-doped pyrene as the chromophore. It shows green fluorescence in various solvents and is stable under aqueous and alkaline conditions.

Abstract:A series of pyrazole-based 1,3,4-oxadiazole derivatives were rationally designed and synthesized in good yields by following a convenient route. All the newly synthesized molecules were fully characterized by IR, H-1 NMR and elemental analysis. Eight compounds were structurally determined by single crystal Xray diffraction analysis. The fluorescence properties of all the compounds were investigated in dimethyl sulfoxide media. In addition, these newly synthesized compounds were evaluated for in vitro inhibitory activity against commercial enzyme xanthine oxidase (XO) by measuring the formation of uric acid from xanthine. Among the compounds synthesized and tested, 3d and 3e were found to be moderate inhibitory activity against commercial XO with IC50 = 72.4 mu M and 75.6 mu M. The studies gave a new insight in further optimization of pyrazole-based 1,3,4-oxadiazole derivatives with excellent fluorescence properties and XO inhibitory activity. (C) 2015 Elsevier B.V. Abstract:A straightforward and efficient methodology has been employed for the synthesis of a diverse set of base-modified fluorescent nucleoside conjugates via Cu(I)-catalysed cycloaddition reaction of 5-ethynyl-2',3',5'-tri-O-acetyluridine/3',5'-di-O-acetyl-2'-deoxyuridine with 4-(azidomethyl)-N9-(4'-aryl)-9,10-dihydro-2H,8H-chromeno[8,7-e][1,3]oxazin-2-ones in tBuOH to afford the desired 1,2,3-triazoles in 92-95% yields. Treatment with NaOMe/MeOH resulted in the final deprotected nucleoside analogues. The synthesized 1,2,3-triazoles demonstrated a significant emission spectrum, featuring two robust bands in the region from 350-500 nm (with excitation at 300 nm) in fluorescence studies. Photophysical investigations revealed a dual-emissive band with high fluorescence intensity along with excellent Stokes shift (140-164 nm) and superior quantum yields (0.068-0.350). Furthermore, the electronic structures of the synthesized triazoles have been further strengthened by DFT studies. Structural characterization of all synthesized compounds was carried out using various analytical techniques, including IR, 1H-NMR, 13C-NMR, 1H-1H COSY, 1H-13C HETCOR experiments, and HRMS measurements. The dual-emissive nature of these nucleosides would be a significant contribution to nucleoside chemistry as there are limited literature reports for the same.

Abstract:Some novel fluorophores, viz. 6-(6-isobutyrylamino-1,3-dioxo-1H,3H-benzo[de]isoquinolin-2-yl)-hexanoic acid (1), 6-(6-dimethylamino-1,3-dioxo-1H, 3H-benzo[de]isoquinolin-2-yl)-hexanoic acid (2), 6-(6-benzoylamino-1, 3-dioxo-1H, 3H-benzo[de]isoquinolin-2-yl)-hexanoic acid (3), 6-(6-amino-1-oxo-1H, 3H-benzo[de]isoquinolin-2-yl)-hexanoic acid (4) and 6-(6-amino-1H,3H-benzo[de] isoquinolin-2-yl)-hexanoic acid (5) have been designed, synthesized and characterized. Their comparative fluorescence has been studied in different organic solvents and aqueous solutions containing inorganic ions. Out of these, two fluorophores, 1 and 2 have been used for labelling of nucleosides which were finally converted into their phosphoramidites, and used for labelling of oligodeoxyribonucleotides through covalent attachment. These fluorophores after attachment on oligodeoxyribonucleotides showed good fluorescence signals and higher hybridisation affinity than unlabelled oligodeoxyribonucleotides.

Abstract:In this article, we present the synthesis and the optical properties of three original molecules as potential fluorescent ribonucleoside analogues incorporating a 1,6-naphthyridin-7(6H)-one scaffold as a fluorescent nucleobase and a 1,2,3-triazole as a linkage. The nucleosides were prepared via a Cu alkyne-azide cycloaddition (CuAAC) reaction between a ribofuranosyl azide and a 4-ethynylpyridine partner. Construction of substituted 1,6-naphthyridin-7(6H)-ones was achieved through two additional steps. Optical property studies were investigated on nucleoside analogues. Powerful fluorescence properties have been evidenced with a remarkable change of emissivity depending on the polarity of the solvent, making these molecules suitable as a new class of artificial fluorescent nucleosides for investigating enzyme binding sites as well as probing nucleic acids. In addition, we are convinced that such analogues could be of great interest in the search for new antiviral or antitumoral drugs based on nucleosides.

Abstract:A series of novel 5-(3-aryl-1H-pyrazol-5-yl)-2-(3-butyl-1-chloroimidazo[1,5-a]- pyridin-7-yl)-1,3,4-oxadiazole derivatives has been synthesized from 3-butyl-1-chloroimidazo[1,5-a]pyridine-7-carboxylic acid and ethyl 3-aryl-1H-pyrazole-5-carboxylate. The compounds were characterized using IR, (1)H NMR, HRMS and UV-vis absorption. The fluorescence spectral characteristics of the compounds in dichloromethane were investigated. The results showed that absorption λmax and emission λmax was less correlated with substituent groups on N-1 position of pyrazole moiety and para position of benzene moiety. The calculated molecular orbital correlates well with their absorption.

Abstract:Post-synthetic conversion of the trifluoromethyl group to a heteroaryl group at the C5 position of the pyrimidine base in the oligonucleotide was achieved. Specifically, oligonucleotides containing 5-trifluoromethylpyrimidine bases were treated with o -phenylenediamines and o -aminothiophenols as nucleophiles to afford the corresponding 5-(benzimidazol-2-yl)- and 5-(benzothiazol-2-yl)-pyrimidine-modified bases. Furthermore, evaluation of the fluorescence properties of the obtained oligonucleotides revealed that among them the oligonucleotide containing 5-(5-methylbenzimidazol-2-yl)cytosine exhibited the highest fluorescence intensity. These results indicated that post-synthetic trifluoromethyl conversion, which is practical and operationally simple, is a powerful tool for exploring functional oligonucleotides.

Abstract:A series of indolizines were synthesized. The fluorescence spectra and fluorescence quantum yields of the compounds 3a - 3k, 4a - 4b and 5 - 6 were examined. It was found that the quantum yields increased if the cyano group was introduced into the indolizines, while the presence of carbonyl groups as substituents of indolizines resulted in fluorescence quenching. The indolizines 3a - 3b and 3g - 3i with high fluorescence quantum yields are shown to be useful fluorophores to the construction of probe compounds.

Abstract:A series of small-molecular fluorescent probes based on nitrogen-fused pyridazinone scaffold were developed in this report.The design strategy involved two steps:1) enhancing the electron-withdrawing ability of the acceptor by incorporating an N-heterocyclic aromatic ring (pyridine or pyrazine) at the C4 and C5 positions of the pyridazinone skeleton and 2) anchoring a triphenylphosphine or morpholine tail as the subceilular targeting group.These fluorescent probes displayed excellent properties in live cell and brain tissue imaging.

Abstract:The creation of new functional molecules is a central task in chemical synthesis. Herein, we report the synthesis of a new type of fluorophore, bisbenzo[f]isoindolylidenes, from easily accessible dipropargyl benzenesulfonamides. Wavelength-tunable fluorophores emitting strong fluorescence of green to red light were obtained in this reaction. Late-stage modifications and incorporation of bioactive molecules into these fluorophores exhibit potential applications in biological studies. Detailed computational and experimental studies were conducted to elucidate the mechanism, and suggest a reaction sequence involving Garratt-Braverman type cyclization, isomerization, fragmentation, dimerization and oxidation.

Abstract:Imidazo[1,5‐a]quinolines emit intense blue luminescence, suggesting their possible use as emitter molecules in organic light‐emitting diodes. We synthesized several new imidazo[5,1‐a]isoquinolines and found they possessed a characteristic hypsochromic effect and even increased quantum efficiency compared to the homolog imidazo[1,5‐a]quinolines, as measured by UV‐vis‐ and fluorescence spectroscopy in solution. The energies of the highest occupied and lowest unoccupied molecular orbitals were calculated using density functional theory and experimentally determined by cyclic voltammetry in solution. We obtained a maximum fluorescence quantum yield of 48% at 446 nm for the newly synthesized 3‐(4‐cyanophenyl)‐1‐(pyridyl)imidazo[5,1‐a]isoquinoline with an optimized substitution pattern. For 1‐phenyl‐3‐(2‐pyridinyl)imidazo[5,1‐a]isoquinoline, a quantum yield of 33% was obtained at an even shorter emission wavelength of 431 nm.

Abstract:Fluorescent base analogues (FBAs) have become useful tools for applications in biophysical chemistry, chemical biology, live‐cell imaging, and RNA therapeutics. Herein, two synthetic routes towards a novel FBA of uracil named qU (quadracyclic uracil/uridine) are described. The qU nucleobase bears a tetracyclic fused ring system and is designed to allow for specific Watson‐Crick base pairing with adenine. We find that qU absorbs light in the visible region of the spectrum and emits brightly with a quantum yield of 27% and a dual‐band character in a wide pH range. With evidence, among other things, from fluorescence lifetime measurements we suggest that this dual emission feature results from an excited‐state proton transfer (ESPT) process. Furthermore, we find that both absorption and emission of qU are highly sensitive to pH. The high brightness in combination with excitation in the visible and pH responsiveness makes qU an interesting native‐like nucleic acid label in spectroscopy and microscopy applications in, for example, the field of mRNA and antisense oligonucleotide (ASO) therapeutics.

Abstract:The donor-acceptor design is a classic method of synthesizing new fluorescent molecules. In this study, a series of new fluorescent compounds (1-10) were synthesized based on 2-(3,5-bis(trifluoromethyl)phenyl)-quinazoline acceptor and various amino donors. The fluorescent emissions of1-10cover the spectrum from 414 nm to 597 nm in cyclohexane solutions with various amino donors on 4- or 7-positions of quinazoline. Ultimately, compounds1and2presented the highest photoluminescence quantum yield (QY) over 80%, while compound10provided the largest Stokes shift (161 nm) in cyclohexane. Most of them have strong emissions in aggregated states such as in nanoparticles, in powders, in crystals and in films. Mechanochromic properties were observed for compounds1,2,4and7. Furthermore, blue OLEDs were fabricated by using compound2or7as the active layer.

Abstract:It is now possible to prepare 2-oxo-2 H -pyrano[2,3- b ]indolizine-3-carboxylates with an ordered arrangement of various substituents directly from pyridinium salts and diethyl 2-(ethoxymethylene)malonate, allowing for refined control of their photophysical properties. Facile entry into some previously unknown derivatives is disclosed to demonstrate the potential of this method. The use of substituted picolinium salts, as well as further functionalization of the pyrrole ring, permitted easy introduction of new moieties upon the dye, which enabled fine-tuning of the photophysical properties. The obtained dyes possess absorption and emission spectrum in the blue–green region and fluorescence quantum yields reaching 92%. The parent 2-oxo-pyrano[2,3- b ]indolizine-3-carboxylate turned out to be an electron-deficient system with a low-lying LUMO, an electronic transition energy of 2.7 eV and possessing a large oscillator strength. Almost complete overlap of the HOMO and LUMO in the 2-oxo-pyrano[2,3- b ]indolizine core is responsible for the large fluorescence quantum yields for almost all prepared derivatives. The reason for maintaining the large emission intensity in polar solvents is that the increase in the dipole moment is accompanied with a significant change in its orientation in space. Fluorescence imaging studies have proven that 2-oxo-pyrano[2,3- b ]indolizines penetrate the membrane of living cells. A positively charged analog was synthesized and used to stain intracellular organelles in the H9c2 cell line. This compound did not penetrate the cell membrane, however after permeabilization, it specifically stained the nucleus.

Abstract:A series of pi-conjugated compounds ending with dibenzosuberenylidene were synthesized efficiently. Their luminescence efficiencies were well-tuned by structural modification. Moreover, relative emission intensities were strongly affected by their existing appearances and exhibited aggregation-induced emission (AIE) behavior. Thus, emissions from nanoparticles, films, or powders were found to be more efficient than those from solutions. It demonstrated that these synthesized compounds might be practically used as fluorescent materials for potential optoelectronic applications.

Abstract:A series of bicyclic furo[2,3-d]pyrimidine nucleosides conjugated with N-diisopropylphosphoryl amino acids were synthesized from 2′-deoxy-5-iodouridine. The key step is its cross coupling with the 5-phthalimido-1-pentyne under Sonogashira reaction condition. 5-Endo-dig electrophilic cyclization catalyzed by AgNO3 is used to afford the furo[2,3-d]pyrimidines.
