Abstract:Treatment of polyfluorophenyl perfluoroalkanesulfonates RfSO3C6F4X-p (1, X = F, Cl) with alkynes in the presence of Pd(PPh3)2Cl2 or Pd(PPh3)4 and Et3N in DMF gave polyfluorophenylalkynes in good to high yields. The same products were also prepared in a one-pot reaction from the corresponding perfluoroalkanesulfonyl fluoride and polyfluorophenol in comparable yields.

Abstract:<p>1,3-Butadiene plays a key role in modern synthetic chemistry and biochemistry because it is a key intermediate in the synthesis of many drugs. A new and effective method for the synthesis of 4-trifluoromethylated 1,3-butadiene through the fluorinated Heck reaction catalyzed by Pd(0) is described. Without additives, 1-chloro-3,3,3-trifluoropropene (an inexpensive CF3 structural unit that is harmless to ozone) reacts with enamide to synthesize 4-trifluoromethylated 1,3-butadienes with good yield, high regioselectivity and chemical selectivity, and strong tolerance of substrate functional groups such as alkynyl, aldehyde, and ester groups.</p><p>This article is protected by copyright. Abstract:Various new transformations of gem-difluoroalkenes leading to trifluoromethyl substituted compounds have been well established in the past years. However, the development of new transformations of gem-difluoroenynes lags much behind. Herein is reported the fluoroarylation of 1,1-difluoro-1,3-enynes with aryl halides in the presence of silver fluoride affording trisubstituted trifluoromethyl allenes under the catalysis of palladium. The reaction features mild conditions, high functional-group tolerance, and high regioselectivity.

Abstract:A palladium-catalyzed cross-coupling of gem-difluorinated cyclopropanes with boronic acids, providing the corresponding arylated/alkenylated/alkylated 2-fluoroallylic scaffolds, is generated. This new approach has good functional group compatibility for both gem-difluorinated cyclopropanes and boronic acids; thus, an array of synthetic building blocks of monofluoroalkene scaffolds including conjugated fluorodiene and skipped fluorodiene gave good yields with high Z-selectivity. Moreover, proficient application was described for monofluoroalkene, whereas the corresponding alkyl fluoride was constructed through hydrogenation.

Abstract:Palladium-catalyzed enantioselective domino Heck/intramolecular C-H functionalization reaction, as a valuable strategy for creating molecular diversity, has remained a prominent challenge. Here, we describe a Pd/XuPhos catalyst for asymmetric domino Heck/intermolecular C-H alkylation of unactivated alkenes with diverse polyfluoro- and heteroarenes in a highly chemo- and enantioselective manner. This process enables efficient synthesis of various dihydrobenzofurans, indolines and indanes, which are of interest in pharmaceutical research and other areas. Late-stage modifications of the core structures of natural products are also well showcased. Moreover, synthetic transformations create a valuable platform for preparing a series of functionalized molecules. Several control experiments for mechanistic study are conducted to pursue a further understanding of the reaction.

Abstract:A highly enantioselective palladium‐catalyzed intramolecular Heck reaction of ortho‐iodophenol‐derived allyl ether, which provides an efficient method to synthesize various chiral 3,3‐disubstituted 2,3‐dihydrobenzofuran compounds bearing an all‐carbon quaternary stereocenter has been developed. Comprehensive Summary A highly enantioselective palladium‐catalyzed intramolecular Heck reaction of unactivated alkenes is developed, which provides a powerful route to access a broad range of chiral 3,3‐disubstituted‐2,3‐dihydrobenzofurans bearing all‐carbon quaternary stereocenter of interest in pharmaceutical research. Its salient features include high yield, excellent chemo‐ and enantioselectivity, mild conditions, a broad substrate scope as well as versatile transformations of the product.

Abstract:Palladium-catalyzed CF bond formation via directed CH activation has recently emerged as a significant method to introduce fluorine atoms into organic molecules in an atom- and step-economical path. At the interface between fluorine chemistry and synthetic organic chemistry, the development of direct fluorinations of the CH bond remains sluggish, due to the major challenge of reductive elimination to afford the CF bond. One strategy is to oxidize palladium(II) species to more electron-deficient palladium(IV or III) fluorides to promote the CF bond forming reductive elimination, which opens up palladium-catalyzed CH fluorination as a powerful transformation. This focused review gives a brief summary of the mere handful of examples, including C(sp(2))F and C(sp(3))F bond formations, in this field so far.
