Abstract:Because of strong interaction with diol moieties, boric acid group often plays a critical role in the design of saccharide sensors. We presents an overview about the recent development of saccharide sensors based on various mechanisms, including spectroscopies, pH-indicators and electrochemistry. The chemistry involved in the binding between boric acid and saccharides are described also. Furthermore, potential directions of future research for these boric acid-based saccharide sensors are discussed.

Abstract:Boronic acids can form reversible covalent bonds with 1, 2- or 1, 3-diols to generate five or six-membered cyclic complexes. Thus, boronic acid functionalized compounds and materials have attracted much attention in both chemistry and biology as the recognition motif for chemo/biosensing and enrichment and separation of biomolecules. In this contribution, we addressed the recent progress of boronic acid-based electrochemical sensors for the detection and immobilization of diol-containing biomolecules including dopamine and glycoproteins. This work will be valuable for the development of novel boronic acid-based electrochemical sensors.

Abstract:Fluorescent probes based on boron dipyrromethene functionalized with a phenylboronic acid group (BODIPY-PBAs) were synthesized in high yield for the first time by Suzuki coupling of bis(pinacolato)diboron and 8-(4-bromophenyl)-1,3,5,7-tetramethyl-4,4-difluoro-4-bora-3a,4a-diaza-s-indacene (BODIPY). Wavelength tuning of the fluorophores was achieved by attaching an auxochromic substituent to the 5-position of the BODIPY core structure through Knoevenagel condensation. The emission intensity of fluorophores increases when binding to the analytes with diol groups and forming boronic esters at fixed pH. These compounds can detect monosaccharides in the concentration range of 0.1-100 mM. Whereas glycogen was found to quench the fluorescence of BODIPY-PBAs in an aqueous solution due to the self-quenching of the fluorophores after attaching in the extensively branched and compact glucose polymer, further addition of d-fructose to the solution can release the fluorophores from the polymer and the fluorescence regains. The BODIPY-PBA fluorophore has been applied in polymeric optodes containing anion exchangers to perform repetitive measurement. Such sensors respond to different monosaccharides in the range of 0.1-100 mM and demonstrate an improved selectivity toward d-fructose over other saccharides, compared to the results obtained from homogeneous assay.

Abstract:In this article ferrocene monoamino phenyl boronic acid (FcMAPBA) and ferrocene diamino phenyl boronic acid (FcDAPBA) were synthesized and characterized by 1H NMR and FTIR. Particular emphasis is given to cis-diol and saccharide binding affinities with ferrocene amino phenyl boronic acid (FcAPBAs) which was investigated by 1H NMR and CV (using sorbitol). Free and bound forms of derivatives were depicted from disappearance of boronic acid hydroxyls signal in 1H NMR spectra. The presence of two discrete peaks in the CV curves is in agreement with 1H NMR spectra, as the indication of cis-diol binding with saccharides. It was also proposed that this approach using potential shift difference in CV curves can be applied for surface confined glycated molecules detection.
Boronic Acid Fluorophore/Beta-Cyclodextrin Complex Sensors for Selective Sugar Recognition in Water

Abstract:A novel boronic acid fluorophore 1/beta-cyclodextrin (beta-CyD) complex sensor for sugar recognition in water has been designed. The probe 1 bearing pyrene moiety as a fluorescent signal transducer exhibits no fluorescence emission, due to its aggregation in water containing 2% DMSO; however, the addition of beta-CyD to this solution largely changes UV-vis and fluorescence spectra of 1 by forming an inclusion complex with beta-CyD, and an efficient fluorescence emission response of 1/beta-CyD complex upon sugar binding is found to be obtained at pH 7.5. The pH-fluorescence profile of the 1/beta-CyD complex reveals that the boronate ester formation with fructose induces the apparent pKa shift from 7.95+/-0.03 in the absence of fructose to 6.06+/-0.03 in the presence of 30 mM fructose, resulting in the fluorescence emission response under the neutral condition. The spectral properties of 1 in 95% methanol:5% water (v/v), as well as the fluorescence quenching study of 1-methylpyrene with 4-methoxycarbonylphenyl-boronic acid 2, demonstrate that the response mechanism is based on the photoinduced electron transfer (PET) from the pyrene donor to the acid form of phenylboronic acid acceptor in 1, and thus, the proton dissociation of phenylboronic acid induced by sugar binding inhibits the PET system while increasing the fluorescence intensity of the pyrene moiety. To evaluate the binding ability and selectivity of the 1/beta-CyD complex for monosaccharides in water, the response equilibria have been derived. The 1:1 binding constants of the 1/beta-CyD complex obtained from the equilibrium analysis are in the order: D-fructose (2515+/-134 M(-1)) >> L-arabinose (269 +/- 28 M(-1)) > D-galactose (197+/-28 M(-1)) > D-glucose (79+/-33 M(-1)), which is consistent with the binding selectivity of phenylboronic acid.

Abstract:Saccharides - a versatile class of biologically important molecules - are involved in a variety of physiological and pathological processes, but their detection and quantification is challenging. Herein, surface plasmon resonance and self-assembled monolayers on gold generated from bis-boronic acid bearing a thioctic acid moiety, whose intramolecular distance between the boronic acid moieties is well defined, are shown to detect d-glucose with high selectivity, demonstrating a higher affinity than other saccharides probed, namely d-galactose, d-fructose and d-mannose.

Abstract:It is of great importance to have sensitive and accurate detection of cis-diol-containing biologically related substances because of their important functions in the research fields of metabolomics, glycomics, and proteomics. Boronic acids can specifically and reversibly interact with 1,2- or 1,3-diols to form five or six cyclic esters. Based on this unique property, boronic acid-based materials have been used as synthetic receptors for the specific recognition and detection of cis-diol-containing species. This review critically summarizes the recent advances with boronic acid-based materials as recognition elements and signal labels for the detection of cis-diol-containing biological species, including ribonucleic acids, glycans, glycoproteins, bacteria, exosomes, and tumor cells. We also address the challenges and future perspectives for developing versatile boronic acid-based materials with various promising applications.

Abstract:A novel boron-containing pi-conjugated compound BCCB has been synthesized by the introduction of electron-acceptors (dimesitylboron groups) at the 3,3'-positions of a carbazole dimer (electron-donor). The compound BCCB possesses excellent electrochemical properties and high fluorescence quantum yields. In addition, BCCB is a sensitive fluorescence sensor with remarkable colour changes and the results could be confirmed through theoretical calculations of the compounds BCCB and [(Bu4N)-Bu-n](2)(+)[BCCB center dot(F)(2)](2-). Our studies indicate that BCCB could be used as an excellent optoelectronic material in OLED devices and a ratiometric fluorescent chemosensor.

Abstract:Diabetes is a kind of metabolic disease caused by the inability of the pancreas to control the blood glucose concentration. An effective management of diabetes relies on the frequent monitoring of blood glucose concentrations and the glucose responsive insulin release. Glucose-sensitive systems, resembling the pancreas with the abilities of feedback and equilibrium, can be responsive to the changes of environmental glucose concentration and exhibit extensive application prospect in controlling and treating diabetes. Carbohydrate, which is treated as one of the three substantial units as protein and nucleic acid, plays a great role in life body as an indispensable energy resource to biological processes. Therefore, the recognition of saccharides is very important in the fields of medical and life sciences. Phenylboronic acid is frequently used for recognizing and perceiving carbohydrate due to the high affinity between phenylboronic group and glycol. A fluorescent receptor sensing for saccharide can be constructed while a phenylboronic acid group is linked to a fluorophore. In this paper, the current research situations of phenylhoronic acid-containing glucose-sensitive materials in the field of biomedicine are reviewed, and the latest research progress of such materials in the fields of drug sustained-controlled release systems and fluorescent probes used as the sugar molecule recognition are introduced emphatically. Furthermore, the future research direction of these glucose-sensitive materials is predicted.

Abstract:Small-molecule sensors that are selective for particular sugars are rare. The synthesis of BODIPYs appended with two boronic acid units is reported, alongside cellular staining/labelling and turn-on fluorescence binding data for carbohydrates. The structural frameworks were designed using computational methods, leaning on the chelation characteristics of bis(boronic acids) and the photophysical properties of BODIPYs. Selective binding to glucose is demonstrated via emission and absorption methods, and the challenges of using NMR data for studying carbohydrate binding are discussed. Furthermore, crystal structures, cell permeability and imaging properties of the BODIPYs appended with two boronic acid units are described. This work presents boronic-acid-appended BODIPYs as a potential framework for tunable carbohydrate sensing and chemical biology staining.

Abstract:Boronate affinity materials, as unique sorbents, have emerged as important media for the selective separation and molecular recognition of cis-diol-containing compounds. With the introduction of boronic acid functionality, boronate affinity materials exhibit several significant advantages, including broad-spectrum selectivity, reversible covalent binding, pH-controlled capture/release, fast association/desorption kinetics, and good compatibility with mass spectrometry. Because cis-diol-containing biomolecules, including nucleosides, saccharides, glycans, glycoproteins and so on, are the important targets in current research frontiers such as metabolomics, glycomics and proteomics, boronate affinity materials have gained rapid development and found increasing applications in the last decade. In this review, we critically survey recent advances in boronate affinity materials. We focus on fundamental considerations as well as important progress and new boronate affinity materials reported in the last decade. We particularly discuss on the effects of the structure of boronate ligands and supporting materials on the properties of boronate affinity materials, such as binding pH, affinity, selectivity, binding capacity, tolerance for interference and so on. A variety of promising applications, including affinity separation, proteomics, metabolomics, disease diagnostics and aptamer selection, are introduced with main emphasis on how boronate affinity materials can solve the issues in the applications and what merits boronate affinity materials can provide.

Abstract:Glycoproteins influence a broad spectrum of biological processes including cell-cell interaction, host-pathogen interaction, or protection of proteins against proteolytic degradation. Abstract:We design a new saccharides sensor based on the ensemble of compound 2 with a boronic group and compound 3 with two phenolic –OH groups, taking advantage of the fluorescence quenching ability of nitronyl nitroxides and reversible boronate formation between boronic acid and diol. The results show that the fluorescence of compound 2 was largely quenched upon addition of compound 3 due to the formation of the intermediate nitronyl nitroxide–anthracene dyad 1. Sequential addition of saccharides such as fructose to the ensemble of compounds 2 and 3 together with dyad 1 induced the fluorescence enhancement. These results clearly demonstrate the possibility to employ the ensemble of compounds 2 and 3 (with dyad 1) to sense saccharides.

Abstract:The affinity of boronic acids to cis-diol-containing biomolecules has found wide applications in many fields, such as sensing, separation, drug delivery, and functional materials. A sound understanding of the binding interactions will greatly facilitate exquisite applications of this chemistry. Although a few analytical tools have been available for the characterization of the interactions, these techniques cc are associated with some apparent drawbacks, so they are only applicable to a limited range of boronic acids and cis-diol-containing biomolecules. Therefore, a widely applicable method is still greatly needed. In this work, an affinity capillary electrophoresis (ACE) method was established and validated to probe the interactions between boronic acids and cis-diol-containing biomolecules. The method was proven to be applicable to almost all types of cis-diol-containing biomolecules and boronic acids. Based on this method, a quantitative, comparative study on the interactions between 14 boronic acids that have important potentials for application with 5 typical monosaccharides of biological importance was carried out. The findings provided new insights into boronate affinity interactions, particularly the relationship between the binding strength with the molecular structures of the binding species. Besides, effects of pH and temperature on the binding strength were also investigated. This method exhibited several significant advantages, including (1) possibility of simultaneous study of multiple interactions, (2) low requirement on the purity of the binding species, (3) wide applicability, and (4) high accuracy and precision.

Abstract:Due to the formation of dynamic boronate ester bonds between phenylboronic acid (PBA) and 1,2- and 1,3-diols, PBA-containing biomaterials show increasing applications in biomedical areas, including responsive nanobiomaterials for targeting delivery of chemical drugs, protein drugs, gene drugs and imaging agents in vitro and in vivo for detection of various bioactive molecules such as sialic acid, saccharides, adenosine triphosphate (ATP) and dopamine. With the specific reaction between PBA and sialic acid, which is overexpressed in many kinds of cancer cells, PBA has been used as a targeting moiety for tumor-targeting drug delivery. Additionally, PBA as an electron acceptor affords nanomaterials Lewis acid-base coordination with electron donor atoms such as nitrogen and oxygen, which can be exploited for developing drug delivery systems with high drug loading. Furthermore, PBA-containing materials can stoichiometric consume reactive oxygen species (ROS) and show a nucleus targeting ability. This current review outlined PBA-containing biomaterials with various responsive abilities including sialic acid-targeting, sugars-binding, ROS-response, ATP-response, dopamine-binding, and nuclear targeting. On this basis, their biomedical applications were summarized.
Chiral donor photoinduced-electron-transfer (d-PET) boronic acid chemosensors for the selective recognition of tartaric acids, disaccharides, and ginsenosides

Abstract:A modular approach was proposed for the preparation of chiral fluorescent molecular sensors, in which the fluorophore, scaffold, and chirogenic center can be connected by ethynyl groups, and these modules can easily be changed to other structures to optimize the molecular sensing performance of the sensors. This modular strategy to assembly chiral sensors alleviated the previous restrictions of chiral boronic acid sensors, for which the chirogenic center, fluorophore, and scaffold were integrated, thus it was difficult to optimize the molecular structures by chemical modifications. We demonstrated the potential of our new strategy by the preparation of a sensor with a larger scaffold. The photoinduced electron-transfer (PET) effect is efficient even with a large distance between the N atom and the fluorophore core. Furthermore, the rarely reported donor-PET (d-PET) effect, which was previously limited to carbazole, was extended to phenothiazine fluorophore. The contrast ratio, that is, PET efficiency of the new d-PET sensor, is increased to 8.0, compared to 2.0 with the previous carbazole d-PET sensors. Furthermore, the ethynylated phenothiazine shows longer excitation wavelength (centered at 380 nm) and emission wavelength (492 nm), a large Stokes shift (142 nm), and high fluorescence quantum yield in aqueous solution (Φ=0.48 in MeOH/water, 3:1 v/v). Enantioselective recognition of tartaric acid was achieved with the new d-PET boronic acid sensors. The enantioselectivity is up to 10 (ratio of the binding constants toward D- and L-tartaric acid, k(D)/k(L)). A consecutive fluorescence enhancement/decrease was observed, thus we propose a transition of the binding stoichiometry from 1:1 to 1:2 as the analyte concentration increases, which is supported by mass spectra analysis. The boronic acid sensors were used for selective and sensitive recognition of disaccharides and glycosylated steroids (ginsenosides).
