Abstract:A Cp*Rh-III-catalyzed annulative carbooxygenation of challenging 1,1-disubstituted alkenes triggered by C-H activation of N-aryloxyacetamides has been established, which affords 2,3-dihydrobenzofuran derivatives with a quaternary carbon center in good to excellent yields under mild redox-neutral conditions. An amide group on the alkenes is essential for the process, and may inhibit the beta-H elimination from C(sp3)-Rh species by saturating the rhodium center through coordination. Furthermore, mechanistic insights obtained from control experiments suggest a mechanism involving a Rh-III-Rh-V-Rh-III catalytic cycle.

Abstract:Abstract A Rh-catalyzed electrochemical vinylic C–H annulation of acrylamides with alkynes has been developed in an undivided cell, affording cyclic products in good to excellent yield. Divergent syntheses of α-pyridones and cyclic imidates are accomplished by employing N-phenyl acrylamides and N-tosyl acrylamides as substrates, respectively. Additionally, excellent regioselectivities are achieved when using unsymmetrical alkynes. This electrochemical process is environmentally benign compared to traditional transition metal-catalyzed C–H annulations because it avoids the use of stoichiometric metal oxidants. DFT calculations elucidated the reaction mechanism and origins of substituent-controlled chemoselectivity. The sequential C–H activation and alkyne insertion under rhodium catalysis leads to the seven-membered ring vinyl-rhodium intermediate. This intermediate undergoes either the classic neutral concerted reductive elimination to produce α-pyridones, or the ionic stepwise pathway to produce cyclic imidates.

Abstract:Because of the inherent difficulty in differentiating two olefins, the development of metal-catalyzed asymmetric cyclization of 1,6-dienes remains challenging. Herein, we describe the first rhodium(III)-catalyzed asymmetric borylative cyclization of cyclohexadienone-tethered mono-, 1,1di-, and (E)-1,2-disubstituted alkenes (1,6-dienes), affording optically pure cis-bicyclic skeletons bearing three or four contiguous stereocenters with high yields (25-93%), and excellent diastereoselectivities (>20:1 dr) and enantioselectivities (90-99% ee). This mild catalytic approach is generally compatible with a wide range of functional groups, which allows several facile conversions of the cyclization products. Furthermore, on the basis of our SAESI-MS experiment and computational study, a Rh(I)/(III) catalytic cycle is proposed in this tandem reaction, and the Rh(I) active species catalyzes the overall transformation via sequential oxidative addition of B(2)pin(2), olefin insertion, cyclizing conjugate addition, and reductive elimination. The irreversible conjugate addition determines the overall regioselectivity of borylative cyclization, and the ring strain favors the formation of 5,6-bicyclic structure. This highlights the control of ring strain in diene cyclizations, which provides a useful basis for future reaction designs.
Mechanisms and Origins of Chemo- and Regioselectivities of Ru(II)-Catalyzed Decarboxylative C-H Alkenylation of Aryl Carboxylic Acids with Alkynes: A Computational Study.

Abstract:The mechanisms and chemo- and regioselectivities of Ru(II)-catalyzed decarboxylative C-H alkenylation of aryl carboxylic acids with alkynes were investigated with density functional theory (DFT) calculations. The catalytic cycle involves sequential carboxylate-directed C-H activation, alkyne insertion, decarboxylation and protonation. The facile tether-assisted decarboxylation step directs the intermediate toward the desired decarboxylative alkenylation, instead of typical annulation and double alkenylation pathways. The decarboxylation barrier is very sensitive to the tether length, and only the seven-membered ring intermediate can selectively undergo the designed decarboxylation, suggesting a tether-dependent chemoselectivity. This tether-dependent chemoselectivity also applies to the alkyl tethers. In addition, the polarity of solvent is found to control the chemoselectivity between the decarboxylative alkenylation and [4 + 2] annulation. Solvent with low polarity (toluene) favors the decarboxylation pathway, leading to the decarboxylative alkenylation. Solvent with high polarity (methanol) favors the ionic stepwise C-O reductive elimination pathway, leading to the [4 + 2] annulation. To understand the origins of regioselectivity with asymmetric alkynes, the distortion/interaction analysis was applied to the alkyne insertion transition states, and led to a predictive frontier molecular orbital model. The asymmetric alkynes selectively use the terminal with the larger HOMO orbital coefficient to form the C-C bond in the insertion step.

Abstract:The selective rhodium-catalyzed functionalization of arenes is greatly facilitated by oxidizing directing groups that act both as directing groups and internal oxidants. We report density functional theory (B3LYP and M06) investigations on the mechanism of rhodium(III)-catalyzed redox coupling reaction of N-phenoxyacetamides with alkynes. The results elucidated the role of the internal oxidizing directing group, and the role of Rh(III)/Rh(I) and Rh(III)/Rh(V) catalysis of C-H functionalizations. A novel Rh(III)-Rh(V)-Rh(III) cycle successfully rationalizes recent experimental observations by Liu and Lu et al. ( Liu , G. Angew. Chem. Int. Ed. 2013 , 52 , 6033 ) on the reactions of N-phenoxyacetamides with alkynes in different solvents. Natural Bond Orbital (NBO) analysis confirms the identity of Rh(V) intermediate in the catalytic cycle.

Abstract:Cycloaddition and annulation strategies are among the most powerful methods for creating molecular complexity in organic molecules. In this manuscript, we report a highly site-selective palladium-catalyzed decarbonylative [4+2] cyclization of carboxylic acids with terminal alkynes by a sequential C–C/C–H bond activation. Most notably, this method represents the first use of carboxylic acids as the ubiquitous and underdeveloped synthons for intramolecular cycloadditions by decarbonylative C–C bond cleavage. The method provides a solution to the long-standing challenge of the regioselective synthesis of substituted naphthalenes by cycloaddition. Mechanistic studies show that this reaction occurs through a sequential process involving the formation of key palladacycle by a sequential C–C/C–H bond activation and highly regioselective alkyne insertion enabled by cluster catalysis. Wide substrate scope for both carboxylic acids and terminal alkynes is demonstrated with high functional group tolerance. Moreover, this reaction is scalable and applicable to the synthesis of functionalized molecules featuring bioactive fragments. This reaction advances the toolbox of redox-neutral carboxylic acid interconversion to cycloaddition processes. We anticipate that this approach will find broad application in organic synthesis, drug discovery and functionalized material research fields.

Density Functional Theory Study of the Mechanism of the Rhodium(I)-Catalyzed Conjugated Diene Assisted Allylic C–H Bond Activation and Addition to Alkenes Using Ene-2-dienes As Substrates

Abstract:Recently we reported the first conjugated diene. assisted, rhodium-catalyzed allylic C-H bond activation and addition to alkenes to synthesize multifunctional tetrahydropyrroles, tetrahydrofurans, and cyclopentanes from ene-2-diene substrates, with good to excellent diastereoselectivities. Here we report a DFT study of the mechanism of this reaction, aiming to obtain a detailed potential energy surface, to understand factors determining its stereochemistry, and to determine why conjugated diene is very critical for the success of this reaction. DFT calculations unveiled that the catalytic cycle of this reaction involves a sequence of substrate catalyst complex formation, allylic C-H activation, alkene insertion into the Rh-H bond, and di-pi-allyl-assisted C(sp(3))-Rh-C(sp(3)) reductive elimination, among which the C-H activation and alkene insertion steps are reversible. The main reason for the formation of the cis-divinyl product is that the irreversible reductive elimination transition state from the bis-allylic Rh complex favors a cis 5/5 bicyclic conformation to reduce the ring strain. Moreover, formation of the cis-divinyl product is also assisted by the alkene coordination to the Rh center in the reductive elimination transition state. DFT insights revealed that the conjugated diene, which is very critical for the target reaction, disfavors the double-bond isomerization and facilitates the reductive elimination for the bis-allylic Rh complex, causing the C-H activation and alkene insertion to occur. The computational results showed that the bridgehead double-bond distortion, as suggested by the Bredt's rule, is responsible for not generating the type II [4+2] cycloadducts from ene-2-dienes.

Abstract:As a C2 synthon, vinylene carbonate has been used instead of acetylene in transition-metal catalyzed-coupling reactions. In this study, the mechanism of the vinylene carbonate activation mode for the Rh(III)-catalyzed 4 + 2 annulation of benzamide and vinylene carbonate reaction is proposed, and it was investigated by DFT with the M06 density functional. The computational results show that the proposed reaction pathway, which includes the insertion of vinylene carbonate into an Rh-C bond, decarboxylation of the seven-membered rhodacycle intermediate, protonation of the ketonate rhodium intermediate, deprotonation of the benzamide, and catalyst regeneration, is the most favorable pathway. The 1,2-rhodium migration process can assist decarboxylative activation of vinylene carbonate, which is the rate-determining step in the catalytic cycle. The IRC calculation clearly revealed a quasi-synchronized process to directly afford a cyclic ketonate rhodium intermediate by the release of carbon dioxide.

Abstract:A computational study has been performed to investigate the mechanism of Rh-III-catalyzed C-H bond activation using sulfoxonium ylides as a carbene precursor. The stepwise and concerted activation modes for sulfoxonium ylides were investigated. Detailed theoretical results showed that the favored stepwise pathway involves C-H bond activation, carbonization, carbene insertion, and protonation. The free energy profiles for dialkylation of 2-phenylpyridine were also calculated to account for the low yield of this reaction. Furthermore, the substituent effect was elucidated by comparing the energy barriers for the protonation of meta- and para-substituted sulfoxonium ylides calculated by density functional theory.

Abstract:The asymmetric total synthesis of clovan-2,9-dione with a [6.3.1.01,5]dodecane skeleton has been achieved. Abstract:Rh(III)-catalyzed C-H activation assisted by an oxidizing directing group has evolved to a mild and redox-economic strategy for the construction of heterocycles. Despite the success, these coupling systems are currently limited to cleavage of an oxidizing N-O or N-N bond. Cleavage of an oxidizing C-N bond, which allows for complementary carbocycle synthesis, is unprecedented. In this article, α-ammonium acetophenones with an oxidizing C-N bond have been designed as substrates for Rh(III)-catalyzed C-H activation under redox-neutral conditions. The coupling with α-diazo esters afforded benzocyclopentanones, and the coupling with unactivated alkenes such as styrenes and aliphatic olefins gave ortho-olefinated acetophenoes. In both systems the reactions proceeded with a broad scope, high efficiency, and functional group tolerance. Moreover, efficient one-pot coupling of diazo esters has been realized starting from α-bromoacetophenones and triethylamine. The reaction mechanism for the coupling with diazo esters has been studied by a combination of experimental and theoretical methods. In particular, three distinct mechanistic pathways have been scrutinized by DFT studies, which revealed that the C-H activation occurs via a C-bound enolate-assisted concerted metalation-deprotonation mechanism and is rate-limiting. In subsequent C-C formation steps, the lowest energy pathway involves two rhodium carbene species as key intermediates.

Abstract:We report the first Rh2(II)-catalyzed asymmetric cycloisomerization of activated enynes to provide cyclopropane-fused tetra-hydropyridines in good yields and excellent enantioselectivities under mild conditions. The activated group, CHZ (Z is electron-withdrawing group (EWG)), in the enyne substrates exerts two synergetic roles, one is to activate alkyne for the cyclopropanation reaction; the other is to introduce the C-H···O interaction between substrate and catalyst (reducing the energy barrier of the reaction). This double-mode activation was supported by both density functional theory (DFT) calculations and experimental tests. This strategy was also extended to other CH2Z (Z can be OH, OMe, F) as activating groups that made the CH2 more acidic so that the substrates could also form increased C-H···O interaction with the catalyst.

Abstract:Developing new reaction to synthesize challenging eight-membered carbocycles is a research frontier of organic synthesis. Reported here is the development of the first Rh-catalyzed [5 + 1 + 2] cycloaddition of yne-3-acyloxy-1,4-enyne (Yne-ACEs, short as YACEs) and CO, in which sequentially 5-carbon (generated from 3-acyloxy-1,4-enynes), 1-carbon (CO) and 2-carbon (alkynes) are embedded into the final 5/8 scaffold containing cyclooctatrienone structure. This reaction has broad scope and can be carried out under mild conditions. Keys to the success of the present [5 + 1 + 2] reaction, discovered and supported by experiments and ab initio calculations, include: using terminal alkyne in the 3-acyloxy-1,4-enyne moiety of the substrates so that 1,2-acyloxy migration (instead of 1,3-acyloxy migration, a step required for competing [4 + 2 + 1] reaction) can be realized; applying electron-rich aryl group (here is p-dimethylamino aryl) in the acyloxy group to make [5 + 1] pathway disfavored. Quantum chemical calculations have also been used to answer why this reaction is [5 + 1 + 2] but not [5 + 2 + 1] (where alkyne insertion is ahead of CO insertion), and the factors disfavoring the competitive [5 + 2], [5 + 1] and [4 + 2 + 1] reactions.

Abstract:The C–O and C–H bond cleavages of simple allylic alcohols have witnessed tremendous progress in recent years. However, an efficient method for cleaving the C–C bond via β-carbon elimination is still comparatively underexploited. In this study, we demonstrate rhodium(I)-catalyzed deketonative transfer vinylation and transfer dienylation of various tertiary allylic alcohols with ketones via β-vinyl and β-dienyl elimination. Under mild reaction conditions, allylic and dienyl alcohols with quaternary centers were thus selectively and efficiently constructed in moderate to good yields. The methodologies of transfer vinylation and transfer dienylation, which would provide access to a range of useful organic compounds, pave a significant way for transition-metal-catalyzed C–C activation in organic synthesis.
