Abstract:A novel series of polyhalobenzonitrile quinazolin-4(3H)-one derivatives were synthesized and characterized by NMR, IR, MS, and HRMS spectra. All of the newly prepared compounds were screened for antimicrobial activities against four strains of bacteria (Gram-positive bacterial: Staphylococcus aureus and Bacillus cereus; Gram-negative bacterial: Escherichia coli and Pseudomonas aeruginosa) and one strain of fungi (Candida albicans). Among the synthesized compounds, 5-(dimethylamino)-8-(2,4,5-trichloro-isophthalonitrile) quinazolin-4(3H)-one (7k) exhibited significant activity towards Gram-positive bacterial, Gram-negative bacterial, and the fungi strains. The MIC (0.8-3.3μg/mL) and MBC (2.6-7.8μg/mL) for this compound were close to those of nofloxacin, chlorothalonil, and fluconazole, making it the most potent antimicrobial agents in the series.

Phosphorus Sulfur and Silicon and the Related Elements

Abstract:Three quinoline carbonothioate compounds were synthesized using o-fluoroaniline and ethyl 2-methylacetoacetate as starting materials via multi-steps. The structures of three compounds were confirmed using H-1 NMR and HRMS. The structure of O-ethyl S-(8-fluoro-2,3-dimethylquinolin-4-yl) carbonothioate (3a) was further determined through X-ray single-crystal diffraction. Single-crystal X-ray diffraction analysis indicated that compound 3a crystallizes in the monoclinic space group C2/c, with unit cell parameters: a = 14.3284(6) & Aring;, b = 19.2857(8) & Aring;, c = 10.3990(5) & Aring;, beta = 108.1220(10)degrees, V = 2731.0(2) & Aring;(3). In addition, the physicochemical properties of compound 3a were calculated and discussed using molecular electrostatic potential, frontier molecular orbital and Hirshfeld surface analysis. The antifungal activity of the three compounds was evaluated at 50 mu g/mL, with compound 3b exhibiting good activity (75.8%) against R. solani.

Abstract:Seven new quinoxaline derivatives 3a-3g were synthesized through two steps convenient approach. Their structures were confirmed by Fourier transform infrared, proton nuclear magnetic resonance, and high-resolution mass spectrometry. The synthesized compounds 3a-3g showed good fluorescent intensity in dimethyl sulfoxide. They were also shown to possess good antifungal activities. The structure-activity relationship was studied by density functional theory calculations.

Abstract:A series of 2-substitued-(3-pyridyl)-quinazolinone derivatives were synthesized, characterized, and evaluated for bacteriostatic activity against three species of phytopathogenic bacteria (Xanthomonas oryzae pv. oryzae, Xoo, Ralstonia solanacearum, and Xanthomonas axonopodis pv. citri, Xac). Biological evaluation showed that compounds 4b, 4g, 4h, 4l, and 4m exhibited higher antibacterial activity than bismerthiazol, the positive control, under conditions. In particular, compounds 4l and 4m exhibited significant bacteriostatic activity against Xac.

Abstract:A series of novel quinazolin-4(3H)-one derivatives 3a similar to 3e and 4a possessing 3,4-benzo[c] coumarin and amino side chain were designed and synthesized by the condensation reaction of 2-aminobenzamide and 3,4-benzo[ c] coumarin aldehyde prepared by Heck coupling reaction, followed by S(N)2 substitution reaction with haloalkane. The compounds were evaluated for their anti proliferation activities against four tumor cells and antimicrobial activities. The results showed that 3e showed moderate anti Hela activity with the IC50 value of 22.63 and 23.35 mu mol/L for 4a against MCF-7. Most tested compounds exhibited significant anti Escherichia coli activities, and the inhibition rate was above 89% at the concentration of 50 mu g/mL. The inhibition rates of 2-phenyl-4-(2-(piperidin-1-yl) ethoxy) quinazoline (1b) against Fusarium oxysporum and Rhizoctonia solani and 3d against Verticillium dahliae are 100%.

Abstract:A series of novel 1-methyl-3-substituted quinazoline-2,4-dione derivatives was designed, synthesized, and evaluated for their antimicrobial activities against six strains of bacteria and five fungi in vitro. The synthesized compounds were characterized by spectral methods. The bioactive assays showed that most of the compounds exhibited moderate antimicrobial activities against the tested strains.

Abstract:Fifteen compounds of 6-methyl-l-cyclopropyl-7-(1-piperazinyl-1, 4 - dihydro-4-oxo-3-quinolinecarboxylic acid and its analogues were synthesized. In vitro antibacterial activities of these compounds against three Gram- positive and fifteen Gram -negative organisms were studied by conventional agar dilution procedure. The result of the study showed that antibacterial activities of all of the objective compounds were less than the ciprofloxacin used for comparison.

Abstract:Two series of novel 1,5-benzothiazepine derivatives (23 compounds) were efficiently synthesized and evaluated for antibacterial and antifungal activities. The results indicated that the compounds possessed a broad spectrum of activity against the tested microorganisms and showed higher activity against fungi than bacteria. Compound 2e exhibited the greatest antimicrobial activity. Preliminary study of the structure–activity relationship revealed that substituents in phenyl rings had a great effect on the antimicrobial activity of these compounds.

Abstract:Introduction: Heterocyclic compounds have already provided a platform for the rapid swap of research in the areas of organic, pharmaceutical, analytical, and medicinal chemistry. In the pharmaceutical industry, among the top two hundred branded drugs, more than 75% have heterocyclic fragments in their structures. Methods/Experimental: The compound, 3-(3-methoxyphenyl)-2-thioxo-2,3-dihydro1H-quinazolin-4-one (3) was synthesized by dissolving Methyl anthranilate and N-(3- methoxyphenyl)-methyl dithiocarbamic acid in ethanol and anhydrous potassium carbonate and refluxed for 23 h and re-precipitated by treating with dilute hydrochloric. The 3-(3-methoxyphenyl)-2-thioxo-2,3-dihydro-1Hquinazolin-4-one was dissolved in 40 mL of 2% alcoholic sodium hydroxide solution and dimethyl sulphate was added drop wise with stirring for 1 h, the reaction mixture was then poured into ice water to get 3-(3-methoxyphenyl)-2-methylsulfanyl-3Hquinazolin-4-one (4). The synthesized compounds were screened against various strains of microorganism; Staphylococcus aureus, Bacillus species, Escherichia coli, Klebsiella pneumonia, Enterococcus Feacalis, Pseudomonas aeriginosa, and candida albicans. Compounds 3 and 4 showed significant activity against Staphylococcus aureus, Bacillus species, Escherichia coli, Klebsiella pneumonia, Enterococcus Feacalis, Pseudomonas aeriginosa, and candida albicans, MIC ranging from 6 – 12 mg/mL. Result: The compounds exhibited significant antibacterial activity in comparison to control. Conclusions: From our findings, the compounds synthesized have higher antibacterial activities as compared to the standard antibacterial drug.
