Synthesis of Stilbenes by Cyanide/Base-Mediated Coupling of Benzylic Chlorides and Alcohols

Abstract:A straightforward procedure has been developed for the direct synthesis of stilbenes from benzylic chlorides and alcohols. The transformation employs a two-step one-pot protocol where the benzylic chloride is first subjected to a substitution with potassium cyanide in o -xylene. Without workup, the resulting arylacetonitrile is then reacted directly with the benzylic alcohol and potassium tert -butoxide to generate the stilbene framework. The condensation has been performed with a variety of commercially available benzylic chlorides and alcohols to afford substituted stilbenes as the pure E -isomers. A kinetic isotope effect of 5.2 has been measured for the overall transformation when comparing benzyl alcohol and α,α- d 2 -benzyl alcohol. The release of cyanide during the final elimination to stilbene has been confirmed by a picrate test. Thus, the potassium tert -butoxide-mediated elimination of cyanide is believed to proceed by an E1cB mechanism where the deprotonation reaction constitutes the rate-determining step. Abstract:(S)-aromatic epoxides are of great importance in the production of pharmaceuticals and biologically active natural products. Consequently, developing efficient, ecofriendly, and economical methods for production of (S)-aromatic epoxides continues to be of tremendous interest in organic synthesis. Herein, we report a novel protocol for the efficient synthesis of (S)-aromatic epoxides by coupling mechanochemical synthesis and biocatalysis from commercially available and cheap materials. The first step of the process is the halogenation of aromatic ketones with N-halosuccinimides (NXS) under ball-milling conditions to form alpha-haloaromatic ketones. This is followed by a one-pot asymmetric bioreduction-cyclization sequence affording the various (S)-aromatic epoxides in good yields, which are pure enough without further purification. Compared to the traditional chemical approaches, this mechano-biocatalytic protocol has prominent advantages, including inexpensive starting materials, short reaction time, simplicity of operation, environmental benignity, broad substrate spectrum, good to excellent enantioselectivities, and easy isolation of products.

Abstract:The development of innovative methods for synthesizing silylcyclopentene compounds is particularly important for enriching and improving the synthetical toolbox of organosilicon compounds. Herein, a facile approach has been developed for the synthesis of silylcyclopentenes promoted by mechanochemically generated organolithium species as silicon nucleophiles under ball milling conditions, avoiding the requirement of large amounts of bulk solvent. This operationally simple method demonstrates good functional group compatibility, which provides a great opportunity for further exploration of the synthetic applications of silylcyclopentenes. Density functional theory calculations indicated that the transient lithiosilole intermediates undergo a stepwise nucleophilic addition process, which governs this mechanic-force-promoted [4+1] cycloaddition reaction.

Abstract:The solvent-free mechanochemical reactions under ball milling have emerged as a promising alternative to traditional solution-based chemistry. This approach not only eliminates the necessity for large quantities of solvents and minimizes waste production, but it also facilitates a unique reaction environment that enables the strategies, reactions, and compound synthesis that are typically unattainable in solution. This solvent-less synthetic strategy under ball-milling has well been employed in synthetic organic chemistry in accessing various potential organic molecules including pharmaceutically important molecules and pharmaceuticals or drug-molecules. This feature article highlights the potential of ball milling in the synthesis of pharmaceutically important classes of molecules without using any solvent (solvent-free conditions).

Abstract:For the first time, ball milling has been employed in the solvent‐free synthesis of sulfur‐functionalized materials from thiols and α,β‐unsaturated ketones and esters, using potassium carbonate as a transition metal‐free catalyst. This environmentally friendly protocol makes use of easily accessible reagents to prepare β‐sulfenylated carbonyl compounds with yields exceeding 91% under ambient air and solvent‐free conditions. Additionally, this innovative synthetic strategy enables the modification of chalcones, compounds with significant medicinal and synthetic potential. The reactions are efficient and easily scalable to gram quantities, offering substantial benefits for practical applications.

Abstract:Herein, we report a ball milling-promoted difunctionalization of alkynyl sulfonium salts with sulfinic acids under catalyst-, additive- and solvent-free conditions. The reaction enables the synthesis of various Z-selective 1,2-disulfonylethenes with wide substrate scope and satisfactory yields. Short reaction time (about 10 minutes), room reaction temperature, as well as simple work up procedure without the need for an extraction process make this method practical and sustainable in organic synthesis.

Abstract:Mechanochemical reactions achieved by processes such as milling and grinding are promising alternatives to traditional solution-based chemistry. This approach not only eliminates the need for large amounts of solvents, thereby reducing waste generation, but also finds applications in chemical and materials synthesis. The focus of this study is on the synthesis of quinazolinone derivatives by ball milling, in particular evodiamine and rutaecarpine analogues. These compounds are of interest due to their diverse bioactivities, including potential anticancer properties. The study examines the reactions carried out under ball milling conditions, emphasizing their efficiency in terms of shorter reaction times and reduced environmental impact compared to conventional methods. The ball milling reaction of evodiamine and rutaecarpine analogues resulted in yields of 63–78% and 22–61%, respectively. In addition, these compounds were tested for their cytotoxic activity, and evodiamine exhibited an IC 50 of 0.75 ± 0.04 μg mL −1 against the Ca9-22 cell line. At its core, this research represents a new means to synthesise these compounds, providing a more environmentally friendly and sustainable alternative to traditional approaches.

Abstract:The milling ball is the catalyst. We introduce a palladium-catalyzed reaction inside a ball mill, which makes catalyst powders, ligands, and solvents obsolete. We present a facile and highly sustainable synthesis concept for palladium-catalyzed C-C coupling reactions, exemplarily showcased for the Suzuki polymerization of 4-bromo or 4-iodophenylboronic acid giving poly(para-phenylene). Surprisingly, we observe one of the highest degrees of polymerization (199) reported so far.

Abstract:A mechanochemical‐enabled Ru‐catalyzed regioselective synthesis of 3,4‐isoxazoles and 3,4,5‐isoxazoles from terminal and internal alkynes and hydroxyimidoyl chlorides is reported. This solid‐state and solvent‐free approach carefully examines the impact of the milling conditions on regiocontrol in 1,3‐dipolar cycloadditions using mechanochemical means. The study reveals that milling frequency, jar material, and the choice of liquid additive for liquid‐assisted grinding (LAG) significantly influence the catalytic activity of the Ru catalyst. Transmission electron microscope (TEM) analysis confirms the crucial role of coordinating liquid additives such as acetone and cyclopentyl methyl ether (CPME) in stabilizing and reducing the size of the in‐situ formed Ru nanoparticles, which is essential for catalytic activity. The applicability of this protocol is further demonstrated through the synthesis of a library of 3,4‐ and 3,4,5‐isoxazoles from a wide range of terminal and internal alkynes with varying physical states and electronic properties that highlights the potential of this method for the synthesis of more complex target molecules.

Abstract:We hereby report a simple and efficient method for the preparation of 4-methylcoumarins series, including Coumarin 120 (7-amino-4-methylcoumarin) from phenols (or naphthols) and ethyl acetoacetate in the presence of 3 mol% InCl3. Coumarins were obtained in good yields (52–92%) through Pechmann condensation, under a rapid and environmentally friendly protocol using a high-speed ball mill mixer at room temperature, with short reaction times, under solvent-free conditions.

Abstract:This study describes an accessible approach to various solvent-free transition metal-catalyzed C–H and C–X functionalizations. Conditions from eight different Rh-, Ru-, Ir- and Pd-based mechanochemical reactions are adapted to proceed with generally high efficiency by substituting automated ball milling with pestle and mortar grinding and subsequent heating. This is a low-cost, operationally simple method that requires no specialized equipment and offers similar sustainability benefits. The reaction scope encompasses 10 heterocycle types, many pendant functional groups and is demonstrated on the late-stage modification of a variety of bioactive compounds.

Abstract:Mechanochemistry, more specifically high-speed ball milling, has garnered significant attention in several areas of chemistry, particularly for the synthesis of inorganic materials, cocrystals, organic compounds, discrete metal complexes and metal organic frameworks. This methodology is creating exciting research opportunities because, unlike traditional synthesis, a reaction carried out in a high-speed ball mill does not necessarily need a solvent, thus representing an environmentally friendly solution to the issue of solvent waste. This Chemistry Frontiers article delves into a unique area of ball milling that capitalizes on the solventless nature of the synthesis by using reaction vials, balls, foils, and pellets as both reaction medium and the catalyst. Several examples are highlighted, from nanoparticle synthesis and nanocatalysis to using transition metals in their metallic forms as catalysts. This article is aimed to show both the advantages and challenges present in the field, and to spark interest in further development of this research area.
