Abstract:Herein, a general strategy for the regioselective dearomative 1,2-hydrocyanoalkylation of chromium-bound (hetero)arenes with simple alkylnitriles as pronucleophiles was disclosed, providing rapid access to 1,3-cyclohexadienes possessing useful alkylnitrile groups. The versatility of this methodology further enabled a selective dearomative deuteration reaction. Finally, synthetic applications of the method in the formal synthesis of natural products, including erysotramidine, demethoxyerythratidinone, and morphine, were demonstrated.

Mechanistic studies demonstrated that the dearomatized alkylation product is the thermodynamically more stable compound. Abstract:Abstract Dearomative functionalization reactions represent an important strategy for the synthesis of valuable three-dimensional molecules from simple planar aromatics. Naphthalene is a challenging arene towards transition-metal-catalyzed dearomative difunctionalization reactions. Reported herein is an application of naphthalene as a masked conjugated diene in a palladium-catalyzed dearomative 1,4-diarylation or 1,4-vinylarylation reaction via tandem Heck/Suzuki sequence. Three types of 1,4-dihydronaphthalene-based spirocyclic compounds are achieved in excellent regio- and diastereoselectivities. Key to this transformation is the inhibition of a few competitive side reactions, including intramolecular naphthalenyl C-H arylation, intermolecular Suzuki cross-coupling, dearomative 1,2-difunctionalization, and dearomative reductive-Heck reaction. Density functional theory (DFT) calculations imply that the facile exergonic dearomative insertion of a naphthalene double bond disrupts the sequence of direct Suzuki coupling, leading to the tandem Heck/Suzuki coupling reaction. The observed regioselectivity towards 1,4-difunctionalization is due to the steric repulsions between the introduced aryl group and the spiro-scaffold in 1,2-difunctionalization.

Abstract:Density functional theory calculations have been performed to investigate the mechanisms of the Pd-catalyzed dearomatization reaction between chloromethylnaphthalene and the cyclic amine morpholine. The calculation results indicate that the reductive elimination leading to the formation of the dearomatic product takes place via an intramolecular C-N bond coupling between the para carbon of an eta(3)-exo-(naphthyl)methyl ligand and the nitrogen atom of the amide ligand. The free energy barrier is calculated to be only 13.1 kcal/mol, significantly lower than that (37.8 kcal/mol) through the eta(3)-endo-(naphthyl)methyl intermediate originally thought For comparison, various C-N coupling reaction pathways leading to the formation of dearomatic and aromatic products have also been examined.

Abstract:Organocatalytic dearomatization of β‐naphthols has been achieved via an 1,6‐conjugated addition of 2‐naphthols to alkynyl 7‐methylene‐7H‐indoles in situ generated from α‐(7‐indolyl)methanols. In the presence of racemic phosphoric acid, a series of tetrasubstituted allenes was obtained in high yields. Particularly, with the aid of chiral phosphoric acid, asymmetric dearomatization of β‐naphthols was investigated, affording axially chiral tetrasubstituted allenes with moderate enantioselectivity.

Abstract:Herein, we report a novel palladium (Pd)-catalyzed asymmetric domino Heck/carbocyclization/Suzuki reaction of N-(2-bromoaryl)-2-naphthylacrylamides. The reaction involves a novel and enantioselective dearomative 1,2-insertion of the naphthalene group, thus providing a unique dearomatization strategy of nonactivated naphthalenes. A new phosphoramidite ligand L12, which displayed excellent reactivity and enantioselectivity in the reaction, has been devel-oped. The reaction employs readily available starting materials, and provides straightforward access to a diverse array of spirooxindoles bearing three stereogenic centers in high yields under mild conditions.

Abstract:The catalytic asymmetric dearomatization reaction of non-functionalized arenes poses a long-standing synthetic challenge to organic chemists because of the relative ease of rearomatization via the departure of a proton. In this work, the direct dearomatization of non-functionalized 1-naphthols was achieved through a phosphoric acid-catalyzed asymmetric (3+2) annulation reaction with azoalkenes. The reported convergent method allows the use of readily available simple 1-naphthols without pre-functionalization under mild conditions, furnishing a variety of tricyclic hexahydrobenz[e]indole derivatives in good yields and excellent enantioselectivities (75%-96% yields and 92%-99% ee). Density functional theory (DFT) calculations were performed to gain insights into the origins of the observed chemo-, regio-, and enantioselectivities. It is noteworthy that using azoalkenes as 1,3-dipole surrogates, in combination with powerful chiral phosphoric acid (CPA) catalysis, was key to the success of current reaction.

Abstract:Abstract Substituted aromatic compounds play important roles in materials, biological agents, dyes, etc. Thus, the synthesis of substituted aromatic compounds has been a hot topic throughout the history of organic chemistry. Traditionally, the Friedel–Crafts reaction was a powerful tool for synthesizing substituted aromatic compounds. Abstract:A catalytic asymmetric dearomatization reaction of benzyl 1-naphthyl ethers accelerated by a chiral N,Nʹ-dioxide/Co(II) complex is disclosed. The reaction proceeds via an enantioselective [1,3] O-to-C rearrangement through a tight ion pair pathway, providing a wide array of α-naphthalenone derivatives bearing an all-carbon quaternary center in high yields with excellent ee values.

Abstract:A protocol for palladium-catalyzed dearomative functionalization of simple, nonactivated arenes with Grignard reagents has been established. This one-pot method features a visible-light-mediated [4+2] cycloaddition between an arene and an arenophile, and subsequent palladium-catalyzed allylic substitution of the resulting cycloadduct with a Grignard reagent. A variety of arenes and Grignard reagents can participate in this process, forming carboaminated products with exclusive syn-1,4-selectivity. Moreover, the dearomatized products are amenable to further elaborations, providing functionalized alicyclic motifs and pharmacophores. For example, naphthalene was converted into sertraline, one of the most prescribed antidepressants, in only four operations. Finally, this process could also be conducted in an enantioselective fashion, as demonstrated with the desymmetrization of naphthalene.
