Abstract:The domino reaction of (E)-2-amino-N'-(1-arylethylidene)benzohydrazide and triethyl orthoformate in ionic liquids catalyzed by 10 mol % iodine gave 3a,4-dihydro-2-arylpyrazolo[5,1-b]quinazolin-9(3H)-one derivatives unexpectedly. Abstract:A green method for the synthesis of 2,3-dihydro-2-benzoyl-2-phenylquinazolin-4(1H)-one derivatives, is described in high yields using ionic liquids as green media. The method involves a reaction of 2-aminobenzamide and benzil catalyzed by iodine. The structure of the product 2-benzoyl-3-phenethyl-2-phenyl-2,3-dihydroquinazolin-4(1H)-one (3f) was confirmed by X-ray diffraction analysis.

Abstract:A highly efficient method for the synthesis of 1,2,3,4-tetrahydroquinoline derivatives via a molecular iodine catalyzed domino reaction of anilines with cyclic enol ethers, such as 2,3-dihydrofuran and 3,4-dihydro-2H-pyran, is described. The reaction may proceed through an aza-Diels–Alder process between an in situ generated 2-azadiene and another equivalent of cyclic enol.

Abstract:A highly regioselective approach for constructing 4‐cyclobutene‐isoquinolines through ruthenium‐catalyzed domino C–H activation/cyclization has been demonstrated. This new transformation tolerates various substituted functional groups on imidates and propargylic monofluoroalkynes, resulting in the corresponding products being delivered in moderate yields. Abstract:An efficient and practical one-pot domino synthesis of 1-phenyl-1H-indol-2-amine and 5-amino-indolo[1,2-a]quinazoline derivatives from readily available 2-(2-bromophenyl)acetonitriles was developed. The overall protocol involves a Buchwald-Hartwig type coupling and a base-promoted intramolecular nucleophilic reaction. The reaction scope, advantages and limitations are discussed.

Abstract:An oxidative [3+1+1] convergent domino cyclization is disclosed. This protocol enables to get quinoline, quinoxaline, quinazolin‐4(3H)‐one and benzo[d]thiazole attached 2,4,5‐trisubstituted oxazoles from methyl azaarenes, benzoins and NH 4 OAc in the presence of iodine and molecular sieves without any metal catalyst. The reaction features wide substrate scope, good functional group tolerance, mild reaction conditions, and easily available substrates.

Abstract:A facile one-pot cascade synthesis involving simultaneous in situ pyrazole formation followed by iodine/DMSO-mediated oxidation has been established to afford 1-aryl-2-(3-aryl)-1H-pyrazol-5-yl-ethane-1,2-diones. Primarily, a two-pot approach has been established which includes the reaction of 3-methylthio-1,5-diaryl-2-pentene-1,5-diones with hydrazine in the first step to afford pyrazole, which was eventually oxidized in the next steps in the presence of iodine in DMSO. Further, we performed both steps in the same pot to afford the 1,2-dicarbonyl compounds in good yield. The structure of one of the compounds was confirmed by a single crystal X-ray. DMSO has posed as a solvent as well as an oxidant. Moreover, N-substituted hydrazines provided 1-(1-substituted-3-aryl-1H-pyrazol-5-yl)-2-arylethane-1,2-diones regioselectively. Further, as the synthetic application of this compound, 1-aryl-2-(3-aryl)-1H-pyrazol-5-yl-ethane-1,2-diones was treated with o-phenylenediamine to afford pyrazole functionalized quinoxaline in good yield. The control reaction was carried out to understand the mechanism of product formation.

Abstract:A novel cobalt(II)-catalyzed domino alkene activation/isocyanide insertion/cyclization/directing group migration sequence has been developed for the construction of valuable 2-aminoquinoline scaffolds. By using Co(OAc)2·4H2O as the non-noble catalyst, the reaction of N-(2-vinylphenyl)picolinamides proceeded smoothly to give various 2-aminoquinolines in good yields (up to 91%).

Abstract:A transition-metal-free C═C double bond cleavage reaction employing molecular iodine is described. In the presence of K2CO3 as the base, I2-mediated C═C bond cleavage followed by intramolecular annulation of N-(2-vinylaryl)pyridin-2-amine substrates produces pyrido[2,1-b]quinazolinones and related heterocyclic compounds. This reaction can be completed on a gram scale and has been successfully applied to the synthesis of compounds with important biological properties, including efflux pump inhibitory and antiallergic activities.

Abstract:A variety of 4-(trichloromethyl)pyrido[2',1':3,4]pyrazino[2,1-b]quinazolinones were prepared in moderate to good yields with high regioselectivity through intramolecular 6-endo-dig cyclization and trichloromethylation of N3-alkynyl-2-pyridinyl tethered quinazolinones in chloroform. Mechanistic studies revealed that chloroform might serve as trichloromethyl anion precursor. Furthermore, the reaction could be easily performed at gram scales and an estrone-derived 4-(trichloromethyl)pyrido[2',1':3,4]pyrazino[2,1-b]quinazolinone was prepared over five steps. The present method features broad substrate scope, good functional group tolerance, new dearomatization of pyridine ring, and chloroform as trichloromethylation reagent.
