Abstract:In recent years, modulating amphiphilic assembly by macrocyclic receptors has been emerging as a fantastic topic. As we know, amphiphiles possess ability of self-assembly in aqueous solution due to their unique structures, resulting in important biological functions and various applications in our daily life and industry. Among methods to modulate their assembly behavior, using macrocyclic compounds as amphiphile adaptors owns advantages of convenience and efficiency. Preorganized structures and cavities of macrocyclic compounds enable them the ability of efficiently interacting with amphiphiles in different manners. These additional interactions alter amphiphilic assembly. In this review, we would like to provide a systemic summary of why and how macrocyclic compounds could behave as amphiphile adaptors. For this objective, we introduce related works from the viewpoint of intermolecular interaction. In general, addition of interactions which decrease electrostatic reputation of amphiphiles could promote amphiphilic assembly, and vice versa. Addition of interactions which prevent hydrophobic interaction could inhibit amphiphilc assembly. On the other hand, complexation of macromolecular compounds and amphiphiles could cause steric hindrance, which is unfavorable for assembly. We also take several classical amphiphiles as examples to clarify how different macrocyclic compounds adapt amphiphile assembly.

Abstract:Macrocycle-based amphiphiles, including macrocyclic amphiphiles and macrocyclic host-based supra-amphiphiles, are a class of amphiphiles which are capable of self-assembling into multidimensional assemblies with defined nanostructures. By involving host-guest recognition, these amphiphiles can be tailored to fabricate new topological structures and fulfill multiple applications. Compared with covalent-bonded traditional amphiphiles and polymeric amphiphiles, the introduction of host-guest interactions facilitates the design, synthesis and controllability of these amphiphilic systems. Moreover, host-guest interactions usually possess stimuli-responsive properties, which further endow self-assemblies of macrocycle-based amphiphiles with fantastic functions. In this review, we summarize recent progress in macrocyclic amphiphiles and macrocyclic host-based supra-amphiphiles, with a focus on novel functions and applications of self-assemblies of these amphiphiles.

Abstract:Macrocyclic compounds play an important role in supramolecular chemistry accounting for their unique recognition and self-assembly properties and potential applications in biomedicine and material science. Azo compounds display promising capability in fields of molecular switches, polymers, smart materials and molecular machines because of their photoactive and electroactive properties. Introducing azo groups into macrocycles gives them signal moieties, additional recognition sites, photo-responsive properties, and so on. Herein, we comprehensively review the structures of azo-containing macrocyclic compounds reported up to now. Then we describe representative works on azo macrocycles with their molecular recognition, self-assembly and application emphasized.

Abstract:A triblock amphiphilic macrocycle consisting of a macrocyclic aromatic segment, a hydrophilic oligo(ethylene oxide) branch, and a hydrophobic alkyl dendron is successfully synthesized and characterized. The resulting cyclic amphiphile is observed to self-assemble into hollow double-layered capsules in aqueous solution, as confirmed by dynamic light scattering and cryogenic transmission electron microscopy investigations. The capsules are able to encapsulate hydrophobic guest molecules through aromatic interactions with high stability.

Abstract:Recently, owing to their special spatial structures, peptide-based macrocycles have shown tremendous promise and aroused great interest in multidisciplinary research ranging from potent antibiotics against resistant strains to functional biomaterials with novel properties. Besides traditional monocyclic peptides, many fascinating polycyclic and remarkable higher-order cyclic, spherical and cylindric peptidic systems have come into the limelight owing to breakthroughs in various chemical (e.g., native chemical ligation and transition metal catalysis), biological (e.g., post-translational enzymatic modification and genetic code reprogramming), and supramolecular (e.g., mechanically interlocked, metal-directed folding and self-assembly via noncovalent interactions) macrocyclization strategies developed in recent decades. In this tutorial review, diverse state-of-the-art macrocyclization methodologies and techniques for peptides and peptidomimetics are surveyed and discussed, with insights into their practical advantages and intrinsic limitations. Finally, the synthetic-technical aspects, current unresolved challenges, and outlook of this field are discussed.

Abstract:Interest in macrocycles as potential therapeutic agents has increased rapidly. Macrocyclization of bioactive acyclic molecules provides a potential avenue to yield novel chemical scaffolds, which can contribute to the improvement of the biological activity and physicochemical properties of these molecules. In this study, we propose a computational macrocyclization method based on Transformer architecture (which we name Macformer). Leveraging deep learning, Macformer explores the vast chemical space of macrocyclic analogues of a given acyclic molecule by adding diverse linkers compatible with the acyclic molecule. Macformer can efficiently learn the implicit relationships between acyclic and macrocyclic structures represented as SMILES strings and generate plenty of macrocycles with chemical diversity and structural novelty. In data augmentation scenarios using both internal ChEMBL and external ZINC test datasets, Macformer display excellent performance and generalisability. We showcase the utility of Macformer when combined with molecular docking simulations and wet lab based experimental validation, by applying it to the prospective design of macrocyclic JAK2 inhibitors.

Abstract:Macrocyclic host molecules play the central role in host-guest chemistry and supramolecular chemistry. The highly structural symmetry of macrocyclic host molecules can meet people's pursuit of aesthetics in molecular design, and generally means a balance of design, synthesis, properties and applications. For macrocyclic host molecules with highly symmetrical structures, building blocks, which could be described as repeat units as well, are the most fundamental elements for molecular design. The structural features and recognition ability of macrocyclic host molecules are determined by the building blocks and their connection patterns. Using different building blocks, different macrocyclic host molecules could be designed and synthesized. With decades of developments of host-guest chemistry and supramolecular chemistry, diverse macrocyclic host molecules with different building blocks have been designed and synthesized. Aromatic building blocks are a big family among the various building blocks used in constructing macrocyclic host molecules. In this feature article, the recent developments of macrocyclic host molecules with aromatic building blocks were summarized and discussed.

Abstract:A series of macrocycles, including crown ethers, cyclodextrins, calixarenes, pillararenes and cucurbiturils, are well known to be able to associate various organic/inorganic/biological guest molecules and ions in their well-defined cyclic cavities to form stable host-guest complexes and supramolecular systems through the cooperative contributions of various non-covalent interactions. When one or more functional groups are attached to the cavity of macrocycles or guest molecules, enhanced and/or controlled host-guest associations may take place, leading to not only improved host-guest binding abilities but also fascinating properties. In this review, some representative contributions in the construction of controllable macrocyclic supramolecular assemblies in aqueous solution are presented with an emphasis on the stimuli-responsive control manner and wide applications of this property.

Abstract:Gene therapy, with an important role in biomedicine, often requires vectors for gene condensation in order to avoid degradation, improve membrane permeation, and achieve targeted delivery. Macrocyclic molecules are a family of artificial receptors that can selectively bind a variety of guest species. Amphiphilic macrocycles, particularly those bearing cationic charges and their various assemblies represent a new class of promising non-viral vectors with intrinsic advantages in gene condensation and delivery. The most prominent examples include amphiphilic cyclodextrins, calixarenes and pillararenes. Herein, we systemically reviewed reported assemblies of amphiphilic macrocycles for gene delivery and therapy. The advantages and disadvantages of each type of macrocyclic amphiphiles for gene delivery, as well as the perspectives on the future development of this area are discussed.

Abstract:For decades, macrocyclic compounds have been widely applied in various fields owing to essential physicochemical properties such as their rigid cyclic structures, geometric dimensions (diameter and height), hydrophobic cavity, and hydrophilic interface. This review is an attempt to summarize various research accomplishments involving macrocyclic compounds for drug and gene delivery in immune-modulating therapies: the structures and benefits of main host molecules, their mechanisms regulating the immune system from cell uptake to activation of dendritic cells and T helper lymphocytes, as well as their potential immunotherapy for different diseases. Macrocyclic compounds including cucurbiturils (CBs), calixarenes, pillararenes, cyclodextrins (CyDs), macrocyclic peptides and metallo-supramolecular compounds, have their own unique physicochemical properties and functional derivatizations that enable to improve the biocompatibility, responsiveness to stimuli, and effectiveness of immune-modulating therapy. Based on abundant clarifications of the biological immunity mechanisms, representative constructions of macrocyclic compounds for immune therapies have been conducted for the investigation of treatment of different diseases including cancer, atherosclerosis, Niemann-Pick type C1 disease (NPC1), diabetes, and inflammations. Although there are critical challenges that remain to be conquered, we believe the future of macrocyclic compounds in the immune-modulating therapy must be bright.

Abstract:In the past half-century, macrocycles with different structures and functions, have played a critical role in supramolecular chemistry. Two macrocyclic moieties can be linked to form bismacrocycle molecules. Compared with monomacrocycle, the unique structures of bismacrocycles led to their specific recognition and assembly properties, also a wide range of applications, including molecular recognition, supramolecular self-assembly, advanced optical material construction, etc. In this review, we focus on the structure of bismacrocycle and their applications. Our goal is to summarize and outline the possible future development directions of bismacrocycle research.

Abstract:pi-Conjugated macrocycles have been highly attractive due to their challenging synthesis, fascinating aesthetic structure and unique physical and chemical properties. Although some progress has been made in synthesis, the study of pi-macrocycles with different structural characteristics and supramolecular interactions still faces major challenges. In this paper, two new single-bond linked macrocycles (MS-4T/MS-6T) were reported, and the corresponding vinyl-bridged heterocycloarenes (MF-4T/MF-6T) were synthesized by the periphery fusion strategy. Further studies have indicated that the structure of these four macrocycles is determined by both size and curvature, showing unique variations from nearly planar to bowl and then to saddle. Interestingly, the nearly planar MS-4T with a small size and the rigid saddle-shaped MF-6T show no obvious response to fullerenes C-60 or C-70, while the bowl-shaped MS-6T and MF-4T demonstrate a strong binding affinity towards fullerenes C-60 and C-70. What's more, two kinds of co-crystals with capsule-like configurations, MS-6T@C-60 and MS-6T@C-70, have been successfully obtained, among which the former shows a loose columnar arrangement while the latter displays a unique three-dimensional honeycomb arrangement that is extremely rare in supramolecular complexes. This work systematically studies the pi-conjugated macrocycles and provides a new idea for the development of novel host-guest systems and further multifunctional applications.

Abstract:The development of supramolecular chemistry has always been accompanied by the innovation of macrocyclic hosts. The synthesis of novel macrocycles with unique structures and functions will bring new development opportunities for supramolecular chemistry. As a new generation of macrocyclic hosts, biphenarenes have customizable cavity sizes and diverse backbones, overcoming the limitation that the cavities of traditionally popular macrocyclic hosts are generally smaller than 10 Å. These features undoubtedly endow biphenarenes with distinguished host–guest properties, which have attracted more and more attention. In this review, the structural characteristics and molecular recognition properties of biphenarenes are summarized. In addition, the applications of biphenarenes in adsorption and separation, drug delivery, fluorescence sensing and other fields are introduced. Hopefully, this review will provide a reference for the study of macrocyclic arenes, especially biphenarenes.
