Abstract:Facile synthesis of the natural oleanane-type triterpenoid saponin 1 isolated from Panax japonicus C. A. Meyer var major (Burk.) C. Y. Wu et K. M. Feng and its derivatives is reported. The glucuronide residue in saponins at a later stage via TEMPO-mediated oxidation of the primary-OH has been achieved. All structures of new compounds were established by means of H-1 NMR, C-13 NMR, HRMS, and optical rotation.

Abstract:Leveraging on Schmidt glycosylation method, a highly efficient approach to obtain oleanane-type triterpene saponins was fixed, whereby oleanyl mono-, disaccharide (guaianin N), trisaccharide (elatoside E), as well as tetrasaccharide (elatoside F) were obtained efficiently. The synthetic investigation has resulted in the discovery of the effect of branch-sugar incorporation sequence on the overall synthetic efficiency. Moreover, through bioactivity investigation, the cytotoxic activity of the obtained triterpenoid saponins was evaluated, and the preliminary structure-activity relationship was deduced.

Abstract:The first synthesis of scabiosaponins E (1), F (2), and G (3), three new oleanolic acid saponins with strong inhibitory activity on pancreatic lipase isolated from the Chinese traditional medicinal herb Scabiosa tschiliensis, was efficiently achieved in an one-pot strategy under the combined use of glycosyl trichloroacetimidates and p-toluene 1-thioglycosides (STol) as donors.

Abstract:Twenty-six naturally occurring oleanolic acid saponins and their derivatives, 16 of which were synthesized in this study, were preliminarily evaluated against human cancer cells. From SAR studies, the presence of alpha-L-rhamnosyl residue at the terminal of both C-3 and C-28 position for oleanolic acid bidesmosides was important to enhance cytotoxicity, and introducing more sugar residues at C-3-OH of compound 12 with C-28 carboxylic acid is a favorable modification to ameliorate the anticancer activity. Furthermore, alpha-L-rhamnosyl moiety linked to C-2-OH of the first monosaccharide (alpha-L-alabinose, beta-D-xylose, beta-n-galactose or beta-D-glucose) in C-3-OH of oleanolic acid was helpful to improve the cytotoxicity. According to the predicted log P values, lipophilicity of the synthesized saponins was not an important factor for cytotoxicity. (C) 2013 Elsevier Masson SAS. All rights reserved.

Abstract:The first synthesis of four natural triterpene saponins, which exhibit significant antitumor activities, was concisely achieved by adopting a stepwise glycosylation. The key intermediate 13 was afforded via Bu(2)SnO-mediated regioseletive benzoylation. During the preparation of the target compounds, it was found that the alpha-L-arabinopyranosyl unit in intermediates 17 and 20 existed in the unusual (1)C(4) conformation, and after removing the benzoyl groups, the alpha-L-arabinopyranosyl unit was normal in a typical (4)C(1) form.

Abstract:3-O-[ss-D-Glucopyranosyl-(1 -> 3)-alpha-L-arabinopyranosyl]-oleanolic acid-28-O-[ss-D-glucopyranosyl] ester 1 was synthesized concisely by a convergent strategy. Using stepwise fashion for the synthesis of saponin 2, 3-O-{[ss-D-glucopyranosyl-(1 -> 2)]-[alpha-L-arabinopyranosyl-(1 -> 3)]-alpha-L-arabinopyranosyl}-oleanolic acid-28-O-(ss-D-glucopyranosyl) ester, an abnormal phenomenon, that the terminal arabinosyl residue took the C-1(4) conformation instead of typical C-4(1) form, was observed. Deprotection or heating could not resume the normal conformation, which resulted in the product of 2 ' not 2.

Abstract:An array of oleanolic acid-type saponins based on beta-hederin has been synthesized in a linear or one-pot manner. The cell viability assays indicate that synthetic saponins show antiproliferation activities in three cancer cell lines with IC50 values of 2.4-15.1 mu M and hederacolchiside A(1) being the most potent. The results demonstrate that the type of terminal monosaccharides and linkage position have apparent effects on cytotoxicities and selectivities of these saponins against cancer cell lines tested. This study is helpful for future development of more potent anticancer leads. (C) 2017 Elsevier Ltd. All rights reserved.

Abstract:From Oxytropis ochrocephala Bunge, four saponins 1, 2, 3, and 4 were isolated. Saponins 1 and 2 are new compounds. The common genuine aglycone of the saponins is soyasapogenol B. The structures of those saponins were established by means of C-13-NMR, MS analyses, and chemical methods. In addition, a new interesting artifact aglycone 6 was obtained from the hydrolysate of these saponins and its structure was elucidated by spectral analyses and chemical transformations.

Abstract:The first synthesis of a natural triterpenoid saponin bearing N-acetylglucosamine, albidoside A, which is isolated from the roots of Acacia albida, is concisely achieved in a convergent strategy. Preliminary pharmacological research shows anticancer activity against HL-60, MCF-7, MDA-MB-231, Hep-2, and Hela cell lines. In particular, it exhibited good selectivity, which is five times more cytotoxic toward Hep-2 cells (IC50 = 8.91 μM) than the Hela cell line.

Abstract:Convenient synthetic strategy toward spinasaponin A methyl ester 1 and calenduloside G methyl ester 2, two natural oleanane-type triterpenoid saponins bearing an unique β-d-glucosyl/galactosyl-(1→3)-β-d-glucuronic acid methyl ester disaccharide moiety, was established. Based on this facile approach, four structurally modified congeners 3–6 with ursolic acid and glycyrrhetinic acid as aglycones were efficiently synthesized. MTT assay revealed the cytotoxicities against cancer cells of the synthesized saponins were varied with the change of aglycones and sugar units. Saponin 2 possessing the most potent cytotoxic effects could induce apoptosis of MCF-7 cells, which was detected by confocal micrographs using DAPI staining and flow cytometry using Annexin V and PI double staining. Furthermore, 2-induced apoptosis in MCF-7 cells was associated with ROS generation and loss of the mitochondria membrane potential (Δψm).

Abstract:Phytochemical analysis of the extract from the seed oil leavings of Xanthoceras sorbifolia Bunge led to the isolation of five new oleanane-type triterpeniod saponins, sorbifoliasides G–K (1–5). Their structures were elucidated on the basis of spectroscopic analysis and chemical derivatization. To our knowledge, compound 5 is the first example of a naturally occurring 3,28-O-bisdesmosic triterpenoid saponin with olefinic linkages at C12 and C15. The cytotoxicity of all compounds against ten selected human cancer cell lines was assayed.
