Abstract:We successfully extended our Rh/bisphosphine-thiourea (ZhaoPhos) catalytic system to asymmetric hydrogenation of alpha,beta-unsaturated N-acylpyrazoles affording products with high yields and excellent enantioselectivities (up to 97% yield, 99% ee). The pyrazole moiety played an important role in providing H-bond acceptor sites, which is critical for achieving high reactivities and enantioselectivities.

Abstract:Herein,an organocatalytic enantioselective desymmetrizing hydroalkoxylationof 1,4- and 1,8-diynes is disclosed, which represents a unique chiralBronsted acid-catalyzed desymmetrization of diynes. This protocolprovides a facile and practical access to enantioenriched 1,3-diaminopropanolderivatives and & gamma;-butyrolactones with a wide substrate scopeand generally high enantioselectivities. Besides, the backbones ofthese products constitute the structural core of numerous bioactivemolecules, and they can serve as valuable precursors for the expeditiousassembly of versatile N- and O-heterocycles. Moreover, control experimentsand theoretical calculations are employed to confirm the mechanisticrationale and elucidate the origin of enantioinduction.

Abstract:Highly enantioselective cobalt-catalyzed hydrogenation of 1,1-diarylethenes was developed by using bench stable chiral oxazoline iminopyridine-cobalt complexes as precatalysts. A unique o-chloride effect was observed to achieve high enantioselectivity. Easy removal as well as further transformations of the chloro group make this protocol a potentially useful alternative to synthesize various chiral 1,1-diarylethanes. This process can be successfully performed under 1 atm of hydrogen at room temperature on gram scale.

Abstract:A highly enantioselective Ir-catalyzed asymmetric hydrogenation of 2,5-disubstituted pyrrolo[1,2-a][1,2,4]triazolo[5,1-c]pyrazines containing four nitrogen atoms has been first realized. Under additive-free conditions, a variety of chiral 2,5-disubstituted 5,6-dihydropyrrolo[1,2-a][1,2,4]triazolo[5,1-c]pyrazines can be afforded in high yields (86-98%) with excellent enantioselectivities of up to 99% ee. This method provides a straightforward strategy for the efficient synthesis of chiral multinitrogen polyheterocyclic compounds.

Abstract:Direct enzymatic hydroxylation of prochiral alkylnitriles represents a green and efficient synthesis of chiral β-hydroxy nitriles, but reported examples are very inefficient and limited. Here we describe the development of an evolved P450pyr monooxygenase for enantioselective benzylic hydroxylation of alkylnitriles. By the use of an engineered P450pyr-D5M3 mutant, a variety of pharmaceutically relevant chiral aryl-substituted β-hydroxy nitriles were prepared from prochiral 3-arylpropanenitriles in moderate yields with excellent enantioselectivities.

Abstract:A highly enantioselective rhodium-catalyzed hydrogenation of both (Z)- and (E)-β-arylenamides was developed by using monodentate chiral spiro phosphite and phosphine ligands, respectively. Abstract:The (+/-)-cis-5-arylcarbamoyl-2-ethoxycarbonylpyrrolidines 6a-g were firstly synthesized in 53-64% yields by using meso-diethyl-2,5-dibromoadipate 3 and (S)-(-)-1-phenylethylamine in three steps. The diastereomeric mixture (S;2S,5R)-(-)-7 and (S;2R,5S)-(+)-8 were prepared by the Grignard reaction and separated by a flash column chromatography in 29 and 52% yields. The absolute configurations of (+)-8 was confirmed by X-ray crystallographic analysis and the enantiopure pyrrolidines (2S,5R)-(-)-9/(2R,5S)-(+)-9 and (2S,5R)-(-)-10/(2R,5S)-(+)-10 were obtained in good yields.

Herein, we report a Brønsted acid-catalysed intramolecular hydroamination, which provides such pyrrolidines from simple starting materials in high yield and enantioselectivity. Key to an efficient reaction was the use of an electron-deficient protective group on nitrogen, the common nosyl protecting group, to avoid deactivation of the Brønsted acid by deprotonation. The reaction proceeds as a stereospecific anti-addition suggesting a concerted reaction. Furthermore, kinetic studies show Michaelis-Menten behaviour suggesting the formation of a precomplex similar to those observed in enzymatic catalysis.

Abstract:The palladium-catalyzed asymmetric carboamination reaction is one of the most significant transformations in organic chemistry. Herein, we report the first palladium-catalyzed asymmetric alleneamination of β,γ-unsaturated hydrazones with propargylic acetates. This protocol enables the efficient installation of various multisubstituted allene groups onto dihydropyrazoles in good yields with excellent enantioselectivities. The chiral sulfinamide phosphine ligand Xu-5 exhibits highly efficient stereoselective control in this protocol. The salient features of this reaction include the readily available starting materials, a broad substrate scope, an easy scale-up, mild reaction conditions and versatile transformations.

Abstract:Comprehensive Summary Chiral ruthenium‐catalyzed enantioselective hydrogenation of tetrapyridine‐type N‐heteroarenes was developed. The partially reduction of adjacent tetraheteroaromatic substrates proceeded smoothly in the presence of phosphine‐free chiral cationic ruthenium diamine complexes, affording unprecedented high reactivity, enantioselecitivity and diastereoselectivity (up to 93% yield, >99% ee and 92:8 dr). The potential application of chiral tetradentate pyridine‐amine products as chiral ligands has been demonstrated in the Cu‐catalyzed asymmetric Friedel–Crafts alkylation reaction between indoles and nitroalkenes.
