Abstract:Described herein is the development of a palladium(II)-catalyzed two- or three-component reaction of 2-(1-alkynyl)-2-alken-1-ones with nucleophiles and allylic chlorides. Various types of nucleophiles such as O-, N-, C-based nucleophiles and olefin-tethered O-, N-, C-based nucleophiles were investigated. The scope, mechanism and application of this Pd(II) -catalyzed domino reaction were studied. In these transformations, the palladium catalyst exhibits a dual role, serving simultaneously as a Lewis acid and a transition metal. Two possible reaction pathwavs (cross-coupling reaction vs. Heck reaction) from the same intermediate furanylpalladium species were observed. The reaction pathway is dependent on the property of the nucleophile and the length of the tethered chain as well. When olefin-tethered O- based nucleophiles were used., only the cross-coupling reaction pathway was observed, in contrast, both reaction pathways were observed when olefintethered C-based nucleophiles were employed. The product ratio is dependent on the length of the tethered chain. Furthermore, ring-closing metathesis (RCM) of corresponding furans with C=C bonds provides an easy method for the preparation of functionalized oxygen-heterocycles - 3,4-fused bicyclic furans. It is also noteworthy that allylic chloride can be as an oxidant besides its well known function as an alkylating reagent.

Abstract:Transition metal-catalyzed domino reactions have been used as powerful tools for the preparation of polysubstituted furans in a one-pot manner. In this paper, an efficient synthetic method was developed for the construction of tri- or tetrasubstituted furans from electron-deficient alkynes and 2-yn-1-ols by a silver-catalyzed domino reaction. It is especially noteworthy that a 2,3,5-trisubstituted 4-ynyl-furan was formally obtained in an extremely direct manner without tedious stepwise synthesis. In addition, regio-isomeric furans were observed when substituted aryl alkynyl ketones were employed. This methodology represents a highly efficient synthetic route to electron-deficient furans for which catalytic approaches are scarce. The reaction proceeds efficiently under mild conditions with commercially available catalysts and materials.

Abstract:Recently, we have developed highly regio- and stereoselective carbometalation of 2-alkynols and 2,3-allenols. The organometallacyclic intermediates may be trapped with I(2) to afford 3-iodoalk-2-en-1-ols. These 3-iodoalk-2-en-1-ols may readily undergo the Sonogashira coupling with terminal propargyl alcohols to form 4-alkyn-2-ene-1,6-diols. Subsequent cycloisomerization in DMA or CH(2)Cl(2) with Au(PPh(3))Cl and AgOTf as the catalyst would afford polysubstituted 2-(1-alkenyl)furans; with PdCl(2) as the catalyst and the reaction in DMA in the presence of allylic bromides, the same substrates afforded polysubstituted 2-(1,4-alkadienyl)furans. In both types of catalyzed cyclization reactions, the elimination of H(2)O promoted the aromatization to form the furan ring. Different alkyl or aryl groups could be introduced into different positions of furans due to the substituent-loading capability of 3-iodoalkenols and diversity of the terminal propargyl alcohols and allylic bromides.

Abstract:A variety of 2-aroyl(acyl, or carboxyl)-3-vinyl benzo[b]furans have been prepared via C–C bond formationin good to excellent yields by Pd/C-catalyzed cyclization/isomerization of propargylic compounds. Abstract:Herein, we devised a synthetic approach for skeletally diversified furfuryl sulfides and 4‐alkylthiopyridines involving the heterocyclization of flexible α‐acyl ketene dithioacetals, enabled by cooperative Pd/Cu catalysis. The overall transformation was initiated by acyl‐directed desulfurative Sonogashira coupling, which was proposed as the key step in surmounting the competitive carbothiolation pathway. This step was followed by 1,6‐addition of either a S‐ or N‐nucleophile and intramolecular cyclization. Notably, the two different annulation reactions featured readily accessible starting materials, selective cleavage and reassembly of multiple chemical bonds, broad functional group tolerance, pharmacologically significant heterocyclic products with high regio‐ and chemoselectivities, and high atom economy.

Abstract:A highly efficient palladium-catalyzed cascade reaction of propargyl carboxylates bearing a furanyl group with organoborons was developed. This methodology offers rapid access to polycyclic Diels-Alder cycloadducts in good to high yields. The thus-formed oxygen-bridged products were further converted into anthracene derivatives in a chemoselective manner under mild conditions.
