Abstract:We report a facile method for forming amide bonds between acylsilanes and a wide range of amines in the presence of a mild chlorinating agent under aqueous acidic conditions. The reaction is highly chemoselective, as exemplified by the late-stage modification of a panel of approved drugs and natural products containing reactive functionalities.

Abstract:It is shown here for the first time that diethyl azodicarboxylate promotes dehydrogenation of tertiaryamines to afford enamines, which subsequently take place in tandem reactions with sulfonyl azides to give the N-sulfonyl amidine derivatives. A number of different substituted tertiaryamines and sulfonyl azides can successfully be coupled, and several functionalized groups are tolerated in this system. The reaction described here is mild, general, and efficient, thus providing an extremely preferable method for synthesis of a variety of N-sulfonyl amidine derivatives.

Abstract:A copper‐catalysed (diacetoxyiodo)benzene‐promoted aerobic esterification reaction of acetoacetamides was developed for the synthesis of oxamates, which are useful precursors in synthetic organic chemistry. This practical and mild synthetic approach proceeded at 25 °C under open‐air conditions and afforded methyl 2‐oxo‐2‐(phenylamino)acetates in good to excellent yields combined with C−C σ‐bond cleavage and formal oxidative C−H bond functionalization. A mechanism is proposed.

Abstract:In this communication, we report a photocatalyzed amidation strategy from carboxylic acids and tertiary amines through C-N bond cleavage. A wide scope of structurally diverse carboxylic acids participate smoothly in the reaction, providing the desired tertiary amides with moderate-to-good yields (34 examples, up to 93% yield). This amidation strategy provides an alternative way to address the regioselectivity between nucleophilic functional groups, thus complementing the functional group compatibility of classical amidation protocols. Its synthetic robustness is also proved by the late-stage modification of several complex molecules and gram-scale applications.

Abstract:The development of catalysts for the amidation of tertiary amines with acyl chlorides through oxidative C–N bond cleavage is rather challenging. By employing iodine as the catalyst, a broad range of aromatic acyl chlorides and tertiary amines are efficiently converted into amides in good yields under mild conditions. A plausible mechanistic pathway is proposed for this transformation and is supported by appropriate control experiments. Abstract:Herein, the first electrochemical strategy for direct acyloxylation of benzylic C(sp3)–H with amides is described. The method features transition metal- and oxidant-free conditions with H2 evolution, the excellent functional group (including fluoro, chloro, bromo, iodo, cyano, trifluoromethyl, aldehyde, ketone, carboxyl, ester, and ether) tolerance, up to 82% yield in an operation-friendly undivided cell under the open air, and ease of scale up. A number of well-designed mechanistic experiments [e.g., radical trapping, electron paramagnetic resonance (EPR) and isotope labeling] strongly support the radical-involved process of benzylic C(sp3)–H functionalization and the process of amide C–N bond breaking.

Abstract:Amides are important intermediates in organic chemistry and the pharmaceutical industry, but their low reactivity requires catalysts and/or severe reaction conditions for esterification. Here, a novel approach was devised to convert amides into esters without the use of transition metals. The method effectively overcomes the inherent low reactivity of amides by employing dimethylsulfate-mediated reaction to activate the C-N bonds. To confirm the proposed reaction mechanism, control experiments and density functional theory (DFT) calculations were conducted. The method demonstrates a wide array of substrates, including amides with typical H/alkyl/aryl substitutions, N,N -disubstituted amides, amides derived from alkyl, aryl, or vinyl carboxylic acids, and even amino acid substrates with stereocentres. Furthermore, we have shown the effectiveness of dimethylsulfate in removing acyl protective groups in amino derivatives. This study presents a method that offers efficiency and cost-effectiveness in broadening the esterification capabilities of amides, thereby facilitating their increased utilization as synthetic compounds in diverse transformations.

Abstract:A new synthetic method for ((4R,6S)-6-hydroxymethyl-2,2-dimethyl-1,3-dioxinan-4-yl)-acetic acid tert-butyl ester was accomplished by a chemoenzymatic approach. Racemic 4-(4-methylbenzyloxy)-3-hydroxybutyronitrile (3) was firstly synthesized via condensation and epoxy opening reactions. (S)-4-(4-Methylbenzyloxy)-3-hydroxybutyronitrile (4) was obtained from resolution of racemic 3 by the lipase from Artgribacter sp. with a high enantiomeric excess (98% ee) in the mixture solvent of acetonitrile/hexane (V : V= 1 : 4). Dihydroxy ester 7 was synthesized by a syn-selective triethylborane mediated reduction with a de value of 97%. After acid-catalyzed protection of 7 and hydrogenolysis of the benzyl protecting group, the objective 9 was obtained with a high optical purity.

Abstract:By DFT calculation, we found that acid-catalyzed carboxylic acid esterification and ester hydrolysis are brief two-step reactions. First, the carboxylic acid hydroxyl-oxygen or ester alkyl-oxygen is protonated, which generates a highly active acylium ion. The protonation requires an activation energy (Ea) of 4-10 kcal mol(-1), and is the rate-controlling step of the esterification or hydrolysis. Sequentially, the acylium ion spontaneously reacts with two alcohol or two water molecules to form a neutral product molecule; this is a trimolecular reaction. The acylium ion is the highly active intermediate shared by esterification and hydrolysis. ESI-MS data for several typical carboxylic acids confirmed that their acylium ions are easily generated. For 2,4,6-trialkylbenzoic acid and its ester, the two unsubstituted carbons in the benzene ring are very easily protonated, and we have thus revealed the root of the success of Newman's method. Based on these results, the popular esterification and hydrolysis mechanism in organic chemistry textbooks is incorrect.
Computational Study On The Aminolysis Of Beta-Hydroxy-Alpha,Beta-Unsaturated Ester Via The Favorable Path Including The Formation Of Alpha-Oxo Ketene Intermediate

Abstract:The possible mechanisms of the aminolysis of N-methyl-3-(methoxycarbonyl)-4-hydroxy-2-pyridone (beta-hydroxy-alpha,beta-unsaturated ester) with dimethylamine are investigated at the hybrid density functional theory B3LYP/6-31G(d,p) level in the gas phase. Single-point computations at the B3LYP/6-311++G(d,p) and the Becke88-Becke95 1-parameter model BB1K/6-311++G(d,p) levels are performed for more precise energy predictions. Solvent effects are also assessed by single-point calculations at the integral equation formalism polarized continuum model IEFPCM-B3LYP/6-311++G(d,p) and IEFPCM-BB1K/6-311++G(d,p) levels on the gas-phase optimized geometries. Three possible pathways, the concerted pathway (path A), the stepwise pathway involving tetrahedral intermediates (path B), and the stepwise pathway via alpha-oxo ketene intermediate due to the participation of beta-hydroxy (path C), are taken into account for the title reaction. Moreover, path C includes two sequential processes. The first process is to generate a-oxo ketene intermediate via the decomposition of N-methyl-3-(methoxycarbonyl)-4-hydroxy-2-pyridone; the second process is the addition of dimethylamine to a-oxo ketene intermediate. Our results indicate that path C is more favorable than paths A and B both in the gas phase and in solvent (heptane). In path C, the first process is the rate-determining step, and the second process is revealed to be a [4+2] pseudopericyclic reaction without the energy barrier. Being independent of the concentration of amine, the first process obeys the first-order rate law.
