Abstract:Something radical: the first example of alkene-to-allene radical cyclization of allene-enes is reported. The highly chemoselective intermolecular radical addition reaction of the alkene and subsequent, exclusive exo-radical addition to the allene was realized with perfluoroalkyl radicals. A subsequent TBAF-promoted dehydroiodination of the cyclization products forms cyclopentanes and regenerates an allene moiety (TBAF=tetra-n-butylammonium fluoride).

Abstract:Under the catalysis of a rhodium(III) complex, the cyclization of 2,3-allenols was most probably initiated with the C-H bond functionalization of N-methoxybenzamides using the tetramethylcyclopentadiene anion (C5Me4-) as ligand, affording differently substituted 2,5-dihydrofuran derivatives. Abstract:Modern organic synthesis depends on the development of highly selective methods for the efficient construction of potentially useful target molecules. A primary goal in our laboratory is the discovery of new reactions that convert readily available starting materials to complex products with complete control of regio- and stereoselectivity. Allenes are one underused moiety in organic synthesis, because these groups are often thought to be highly reactive. However, many compounds containing the allene group, including natural products and pharmaceuticals, are fairly stable. The chemistry of allenes has been shown to have significant potential in organic synthesis.Electrophilic additions to allenes have often been considered to be synthetically less attractive due to the lack of efficient control of the regio- and stereoselectivity. However, this Account describes electrophilic reactions of allenes with defined regio- and stereoselectivity developed in our laboratory. Many substituted allenes are readily available from propargylic alcohols. Our work has involved an exploration of the reactions of these allenes with many different electrophiles: the E- or Z-halo- or seleno-hydroxylations of allenyl sulfoxides, sulfones, phosphine oxides, carboxylates, sulfides or selenides, butenolides, and arenes, and the halo- or seleno-lactonization reactions of allenoic acids and allenoates.These reactions have produced a host of new compounds such as stereodefined allylic alcohols, ethers, amides, thiiranes, and lactones. In all these reactions, water acts as a reactant and plays an important role in determining the reaction pathway and the stereoselectivity. The differing electronic properties of the two C=C bonds in these allenes determine the regioselectivity of these reactions. Through mechanistic studies of chirality transfer, isolation and reactivity of cyclic intermediates, O-18-labeling, and substituent effects, we discovered that the E-stereoselectivity of some reactions results from the neighboring group participation of functional groups forming cyclic intermediates. We rationalize Z-stereoselectivity under other conditions by soft Lewis acid-base interactions and steric effects. These electrophilic reactions of allenes are efficient and useful methods for the synthesis of stereodefined alkenes and lactones, useful functionalities for synthesis.

Regiochemistry in Radical Cyclization of Allenes

Abstract:The cyclization of allenic radicals was systematically studied for the first time by computational methods. It was found that the theoretical results at the ONIOM(QCISD(T)/6-311+G(2df,2p):UB3LYP/6-311+G(2df,2p)) level were in good agreement with all the available experimental data. For the cyclization of penta-3,4-dien-1-yl radicals the major product was penta-1,2-diene from direct reduction whereas a small amount of vinylcyclopropane may also be produced. For the cyclization of hexa-4,5-dien-1-yl radicals the major product is 1-methyl-cyclopentene. Furthermore, for the cyclization of hepta-5,6-dien-1-yl radicals both vinylcyclopentane and 1-methyl-cyclohexene are produced. Marcus theory analysis indicated that the formation of an olefinic radical product always had a lower intrinsic energy barrier than the formation of an allylic radical product. On the other hand, the formation of an olefinic radical product was always much less favorable than the formation of an allylic radical product in the thermodynamic term. For the cyclization of substituted hexa-4,5-dien-1-yl radicals, substitution at the allene moiety does not affect the regioselectivity where the allylic radical product is always favored. For the cyclization of hepta-5,6-dien-1-yl radicals, substitution at the allene moiety dramatically affects the regioselectivity, where some radical-stabilizing groups such as –CN and –COMe may even completely reserve the regioselectivity.

Abstract:The cadmium iodide‐mediated reaction between an amine and two molecules of terminal alkynes for the synthesis of allenes has been investigated by using DFT calculations. On the basis of these calculations, a plausible mechanism has been proposed. The results also reveal that pyrrolidine serves not only as a reactant but also as a shuttle in a proton relay.

Abstract:Allenes, which contain the functional group of 1,2-propadiene, are highly reactive. In this paper, we review some free-radical reactions of allenes. The application in natural product synthesis is also described. Substituted allenes undergo rather facile radical chain addition reactions. Attack by a radical species can occur at either the central or the terminal C atom. In the case of addition, the preferred route is generally that forming the possibly more stable free-radical intermediates. The steric effect, electronegativity and solvent effect shoulde also be considered. In general, carbon-centered free radicals such as CH3 . and CCl3 . add mostly to the terminal carbon atom while heteroatom-based free radicals such as Br ., RS ., ArSO2 . and R3Sn . add preferentially to the central carbon atom. Substituted allenes can undergo a variety of [2 + 2] cycloaddition reactions with alkenes, allenes, enynes and ketenes and also undergo a cyclopropanation with carbenes via a biradical mechanism.

Abstract:Allenes are valuable organic molecules that feature unique physical and chemical properties. They are not only often found in natural products, but also act as versatile building blocks for the access of complex molecular targets, such as natural products, pharmaceuticals, and functional materials. Therefore, many remarkable and elegant methodologies have been established for the synthesis of allenes. Recently, more and more methods for radical synthesis of allenes have been developed, clearly emphasizing the associated great synthetic values. In this Perspective, we will discuss recent important advances in the synthesis of allenes via radical intermediates by categorizing them into different types of substrates as well as distinct catalytic systems. The mechanistic studies and synthetic challenges will be highlighted.

Abstract:An efficient Rh(III)-catalyzed stepwise ortho allylation of N-methoxybenzamides 1 with polysubstituted allenes is reported. This C-H functionalization involving allenes is conducted under very mild conditions (-20 °C or room temperature) and compatible with ambient air and moisture, and it can be applied to terminal or internal allenes with different synthetically attractive functional groups. Highly efficient axial chirality transfer has been realized, yielding optically active lactones.

Different radical partners including carbon radicals and heteroatom radicals are discussed in the reactions of allenes. Generally, the radical formed in situ would attack the allene at the central carbon leading to allyl radical intermediate. Abstract:We report the first example of intermolecular allenyl functionaliza-tion of alkenes by a radical-polar crossover process. This reaction leads to homoallenic alcohols and amides with high synthetic value. It is initiated by the addition of an allenyl radical to an alkene, a chal-lenging step because of the short lifetime of the allenyl radical and its strong tendency to produce propargyl radicals. The electrophi-licity of transient allenyl radicals can be efficiently improved by a strategically located sulfone, an outcome that is supported by density functional theory calculations. Solvents are engaged as nucleophiles in this radical-polar crossover, and the products can be converted to diversely complex molecules. This reaction proceeds under mild photoredox neutral conditions and shows exclusive regioselectivity, broad functionality tolerance, and high product diversity.

This organocatalytic strategy was suitable for a broad spectrum of substrates to efficiently synthesize allenic ketones bearing diverse substituents. Abstract:Allenes with two carbon–carbon double bonds belong to a unique class of unsaturated hydrocarbons. The central carbon atom of allene is sp hybridized and forms two σ-bonds and two π-bonds with two terminal sp2 hybridized carbon atoms. The chemistry of allenes has been well documented over the last decades. They are more reactive than alkenes due to higher strain and exhibit significant axial chirality, thus playing a vital role in asymmetric synthesis. Over a variety of organic transformations, allenes specifically undergo classical metal catalyzed cycloaddition reactions to obtain chemo-, regio- and stereoselective cycloadducts. This review briefly describes different types of annulations including [2+2], [2+2+1], [3+2], [2+2+2], [4+2], [5+2], [6+2] cycloadditions using titanium, cobalt, rhodium, nickel, palladium, platinum, gold and phosphine catalyzed reactions along with a mechanistic study of some highlighted protocols. The synthetic applications of these reactions towards the synthesis of natural products such as aristeromycin, ent-[3]-ladderanol, waihoensene(−)-vindoline and (+)-4-epi-vindoline have also been described.

Abstract:The first enantioselective copper-catalyzed cyanation of propargylic C-H bonds via radical relay was established using novel BoxOTMS ligands, providing an efficient and straightforward tool for the construction of structurally diverse chiral allenyl nitriles in good yields with excellent enantioselectivities. This reaction features high functional group tolerance and mild conditions. In addition, the chiral allene products can be readily converted to other chiral compounds via axis-to-center chirality transfer.

Abstract:Allenenitriles bearing different synthetically versatile functional groups have been prepared smoothly from 5-alkynyl fluorosulfonamides in decent yields with an excellent chemo- and regio-selectivity under redox neutral conditions. The resulting allenenitriles can be readily converted to useful functionalized heterocycles. Based on mechanistic study, it is confirmed that this is the first example of radical-based non-activated propargylic C-H functionalization for allene syntheses.

Abstract:The asymmetric 1,4‐difunctionalization of 1,3‐enynes represents a concise and highly efficient strategy for the preparation of chiral allenes. Herein, we developed a copper‐catalyzed three‐component asymmetric radical 1,4‐aryl/alkynylation of 1,3‐enynes by employing inexpensive, readily available, and highly reactive diaryliodonium salts as precursors to generate aryl radicals. The success of this reaction relied on the use of iminophenyl oxazolinylphenylamines (IPOPA) as chiral ligands. The reaction exhibited good compatibility with various alkynes, 1,3‐enynes, and diaryliodonium salts, providing an effective method for constructing highly enantioselective tetrasubstituted axially chiral allene compounds.
