Abstract:Nine new limonoids, meliazedarines A–I (1–9), seven known analogues (10–16), and five known triterpenoids (17–21) were isolated from the fruits of Melia azedarach. Their structures were determined by analysis of 1D and 2D NMR, HRESIMS, X-ray diffraction, and electronic circular dichroism (ECD) data. Abstract:Phytochemical investigations of Melia azedarach fruits have led to the isolation of a novel tirucallane triterpenoid (1), four new limonoids (2–5), and four known limonoids (6–9). Their structures were clarified by comprehensive spectroscopic and spectrometric analyses. The anti-inflammatory activities of isolated compounds were assessed in vitro. Compound 2 exhibited the most potent anti-inflammatory effect, with an IC50 value of 22.04 μM. Additionally, compound 2 attenuated LPS-induced reactive oxygen species (ROS) production and reduced the levels of inflammatory mediators IL-6 and TNF-α. A mechanistic study revealed that limonoid 2 suppresses the expression of iNOS and JAK2 and is implicated in the modulation of the NF-κB signaling cascade, which reveals its anti-inflammatory actions.

Abstract:One new compound, 2H, 3H-cyclopent[b] furo [2,3:4,5] naphtho [2,4-d] heptlactone-[3,7] furan-6aceticacid, 3-(acetyloxy)-8-(3-furyl)-2a, 4a, 4b, 4c,5,5a, 6, 6a, 8, 9,9a, 10a,10b-13 hydrogen-2a,5a,6a,5-tetramethyl-3-[[(2E)-2-methyl-1-oxo-2-butenyl]oxy]-methyl ester, named azadirachta R (1), along with 10 known ones, Azadirachta A, AZ-B, AZ-D, AZ-H, AZ-I, nimbin, deacetylnimbin, salannin, deacetylsalannin and azadiradione were isolated from the crude extracts of neem (Azadirachta indica A. Juss) seeds, which were determined by UV, IR, HR-ESI-MS and NMR data analyses. According to the in vitro antibacterial activity experimental results, this compound showed good antibacterial activity to two bacteria, the minimum inhibitory concentration and the minimum bactericidal concentration of compound 1 to two kinds of bacteria are 50 and 25mg/L, respectively, which were determined by resazurin colour-micro-dilution method. The experimental results provide a theoretical basis for the comprehensive utilisation of azadirachtin compounds in the future.[GRAPHICS].

Abstract:OBJECTIVE:To study the chemical constituents of the peels collected from Citrus changshan-huyou Y. B. Chang, and further screen the bioactive components as the lead structures.METHODS:These compounds were isolated by repeated flash column chromatography on silica gel and Sephadex LH-20. The structures of isolated compounds were elucidated by using IR, EIMS, and NMR analyses.RESULTS:One novel compound along with nine known compounds were obtained and identified as huyoujiasu(I), 3-hydroxy-4-methoxybenzoic acid(II), 3, 4-dihydroxybenzoic acid(III), fatty acid(IV), glyceride(V), 6, 7-dimethoxycoumarin(VI), 6', 7'-dihydroxybergamottin(VII), daucossterol A(VIII), daucossterol B(IX) and huyou-triterpenoid(X).CONCLUSION:One new compound, Huyoujiasu, was obtained from these peels by repeated column chromatography, and the other known compounds were isolated from this fruit for the first time as well.

Abstract:Twelve undescribed limonoids, meliazedarines J–U ( 1 – 12 ), along with a known one, were isolated from the roots of Melia azedarach . Their structures were elucidated by extensive spectroscopic investigations, X-ray diffraction analyses, and ECD calculations. Compounds 1 – 8 were identified as ring intact limonoids, while compounds 9 – 12 were established as ring C- seco ones. The anti-inflammatory potential of compounds 1 – 4 , 6 , 8 , 9 , and 11 – 13 was evaluated on macrophages. Compounds 1 , 3 , 4 , 6 , and 9 significantly suppressed nitric oxide (NO) production in lipopolysaccharide (LPS)-induced RAW 264.7 macrophages, among them compound 3 showed the best inhibitory effect with an IC 50 value of 7.07 ± 0.48 μΜ. Furthermore, compound 3 effectively reduced interleukin-1β (1L-1β) secretion in LPS plus nigericin-induced THP-1 macrophages by inhibiting NLRP3 inflammasome activation. The results strongly suggested that limonoids from the roots of M. azedarach might be candidates for treating inflammation-related diseases.

Abstract:The phytochemical study of the fruits of Melia azedarach (Meliaceae) led to the isolation and characterisation of two novel natural limonoids1-deoxy- 3, 20-dicinnamoyl-11-methoxy-meliacarpinin ( 1 ) and 12 β - O- methyl nimbolinin A ( 2 ), along with twelve known limonoids. Its structure was identified by 1D- and 2D-NMR, HR-ESI-MS and comparison with published data. The anti-inflammatory effect of the compounds was measured in vitro in RAW 264.7 cells by evaluating the production of NO stimulated by LPS. Compounds 1, 8 and 14 indicated significant anti-inflammatory effect with inhibition rate of 11.76, 8.45 and 6.59 μM, respectively. Limonoid 1 significantly inhibited the production of NO, TNF- α and IL-1 β in RAW 264.7 cells. Therefore, limonoid derivative may be a promising source of bioactive metabolite for inflammatory diseases.

Abstract:Objective To further study on the chemical constituents in the peels of Citrus changshan-huyou.Methods The chemical constituents were separated by repeated silica gel column chromatography,the structures were determined by spectral analyses.Results Ten compounds were obtained and identified as huyoujiasu(2'-hydroxy-malonyl-3,4-dihydroxybenzoic acid,Ⅰ),3'-hydroxy-4',5,6,7,8-pentamethoxyflavone(Ⅱ),eriodictyol(Ⅲ),tetracosanoic acid(Ⅳ),glyceryl dodecanoate(Ⅴ),neoeriocitrin(Ⅵ),naringin(Ⅶ),neohesperidin(Ⅷ),4-methoxy-3-hydroxybenzoic acid(Ⅸ),and 3,4-dihydroxybenzoic acid(Ⅹ).Conclusion 2'-hydroxy-malonyl-3,4-dihydroxybenzoic acid is a new compound and the above ten compounds are obtained from this plant for the first time.

Abstract:Two new triterpenoids (1, 2) and two new steroids (3, 4) along with twelve related known compounds (5-16) were isolated from the bark of Melia azedarach. The new structures were elucidated by means of spectroscopic methods and molecular modeling studies and found to be 21,24-cycloeupha- 7-ene-3 beta,16 beta,21 alpha,25-tetrol (1), 3 beta-acetoxy-12 beta-hydroxy-eupha-7,24-dien-21,16 beta-olide (2), 29-hydroperoxy-stigmasta-7,24(28)E-dien-3 beta-ol (3), and 24 xi-hydroperoxy-24-vinyl-lathosterol (4). All isolated compounds were tested for their cytotoxic activity against three human cancer cell lines (A549, H460, HGC27) using the Cell-Titer Glo (TM) luminescent cell viability assay. Among them, compounds 2-4, 24 xi-hydroperoxy-24-vinyl-cholesterol (6), kulinone (7), meliastatin 3 (8), 3-oxo-olean-12-en-28-oic acid (10), and (22E,24S)-5 alpha,8 alpha-epidioxy-24-methyl-cholesta-6,22-dien-3 beta-ol (12) were found to have cytotoxic effects, with IC50 values of 5.6-21.2 mu g/mL.

Abstract:Limonoids, as the vital bioactive chemical compounds in genus Melia plants, have attracted significant attention owing to their exclusive structural characteristics and remarkable biological activity. These compounds can be usually classified into two categories, including the ring-intact group and the ring-C-seco group. Benefiting from the development of separation and analysis technology, more than 200 limonoids have been isolated and identified from this genus. There is growing evidence that limonoids from genus Melia possess diverse pharmacological activities, especially anti-cancer effects, insecticidal activities, and anti-botulism effects. Toosendanin, one of the paramount limonoids, was considered as the pivotal bioactive marker in two medicinal herbs, including Melia toosendan Sieb. et Zucc and Melia azedarach L. In particular, limonoids are found to exhibit non-negligible toxic effects, a finding which needs further research. Besides this, the lack of clinical research data seriously hinders its further development and utilization, and necessary clinical trials should be taken into consideration. In this review, we systematically summarized the phytochemical compounds and their synthesis methods, pharmacological activities, and the structure-activity relationship, pharmacokinetics, and toxicology of genus Melia-derived limonoids. We believe that this up-to-date review could provide scientific evidence for the application of limonoids as agents beneficial to health in future clinical practice.

Abstract:In this study five new limonoids, toosendone [24,25,26,27-tetra-nor-6alpha-acetoxy-21,22-epoxy-7alpha-tigloyl-1alpha,3alpha,28-trihydroxyapotirucalla-(apoeupha)-14,20,22-trien-12-one, 1] and 12-ethoxynimbolinins A-D (2-5), together with five known limonoids, 1-acetyltrichilinin (6), 1-cinnamoyltrichilinin (7), trichilinin B (8), 1,7-di-O-acetyl-14,15-deoxyhavanensin (9) and 12-O-methylnimbolinin B (10),were isolated from the fruits of Melia toosendan. Their structures and relative configurations were established based on spectroscopic analysis. Compound 4 exhibited significant antibacterial activity against the oral pathogen, Porphyromonas gingivalis ATCC 33 277, with an MIC value of 15.6 microg/mL. Compounds 7 and 8 were also active against P. gingivalis ATCC 33 277, with MIC values of 31.3 and 31.5 microg/mL respectively.
Mexicanolide limonoids from the seeds of Khaya ivorensis with antimicrobial activity

Abstract:The methanol extract of the seeds of Khaya ivorensis afforded two new mexicanolide limonoids, ivorensines A and B (1 and 2), together with one known compound, ruageanin D (3). The structures of the isolated compounds were established based on 1 D and 2 D (1H-1H COSY, HMQC, and HMBC) NMR spectroscopy, in addition to high resolution mass spectrometry. The isolated limonoids were tested in vitro for antimicrobial potentials against 5 pathogenic microorganisms. As a result, compounds 1-3 exhibited antimicrobial activity against the tested Gram negative bacteria at the minimum inhibitory concentration values less than 50 μg/ml.

Melanogenesis-Inhibitory and Cytotoxic Activities of Limonoids, Alkaloids, and Phenolic Compounds from Phellodendron amurense Bark

Abstract:Four limonoids, 1 - 4, five alkaloids, 5 - 9, and four phenolic compounds, 10 - 13, were isolated from a MeOH extract of the bark of Phellodendron amurense (Rutaceae). Among these, compound 13 was new, and its structure was established as rel-(1R,2R,3R)-5-hydroxy-3-(4-hydroxy-3-methoxyphenyl)-6-methoxy-1-(methoxycarbonylmethyl)indane-2-carboxylic acid methyl ester (γ-di(methyl ferulate)) based on the spectrometric analysis. Upon evaluation of compounds 1 - 13 against the melanogenesis in the B16 melanoma cells induced with α-melanocyte-stimulating hormone (α-MSH), four compounds, limonin (1), noroxyhydrastinine (6), haplopine (7), and 4-methoxy-1-methylquinolin-2(1H)-one (8), exhibited potent melanogenesis-inhibitory activities with almost no toxicity to the cells. Western blot analysis revealed that compound 6 inhibited melanogenesis, at least in part, by inhibiting the expression of protein levels of tyrosinase, TRP-1, and TRP-2 in α-MSH-stimulated B16 melanoma cells. In addition, when compounds 1 - 13 were evaluated for their cytotoxic activities against leukemia (HL60), lung (A549), duodenum (AZ521), and breast (SK-BR-3) cancer cell lines, five compounds, berberine (5), 8, canthin-6-one (9), α-di-(methyl ferulate) (12), and 13, exhibited cytotoxicities against one or more cancer cell lines with IC50 values in the range of 2.6 - 90.0 μm. In particular, compound 5 exhibited strong cytotoxicity against AZ521 (IC50 2.6 μm) which was superior to that of the reference cisplatin (IC50 9.5 μm).

Abstract:A structurally novel 8,9-seco-eudesma-9,4:8,12-dilactone sesquiterpenoid, chlorajapodiolide (1), a new eudesmane sesquiterpenoid, chlorajapotriol (2), a new lindenane-type sesquiterpenoid dimer, chlorajaponilide I (3), and four known lindenane-type sesquiterpenoid dimers were isolated from the whole plant of Chloranthus japonicus. Their structures were established through extensive analysis of NMR spectroscopic data in combination with mass spectrometry. All compounds were evaluated for cytotoxicities against four human cancer cell lines A549, HCT116, MGC803, and HepG2.

Abstract:Four previously unreported tirucallane-type triterpenoids ( 1–4 ), together with four known analogues ( 5–8 ), were isolated from the fruits of Melia toosendan Sieb. et Zucc. Their planar structures were comprehensively elucidated by detailed analyses of HRESIMS, 1D and 2D NMR spectra data. The relative configurations of 1–4 were determined by NOESY experiments. The comparison of experimental and calculated electronic circular dichroism (ECD) spectra led to the establishment of the absolute configurations of new compounds. All isolated triterpenoids were evaluated for their α -glucosidase inhibitory activities in vitro . Compounds 4 and 5 showed moderate α -glucosidase inhibitory activities with IC 50 values of 120.3 ± 5.8 and 104.9 ± 7.1 μM, respectively.

Abstract:From the extracts of Dimocarpus longan Lour leaves, 2 unusual flavonol glycosides, quercetin 3-O-(3″-O-2‴-methyl-2‴-hydroxylethyl)-β-D-xyloside (1) and quercetin 3-O-(3″-O-2‴-methyl-2‴-hydroxylethyl)-α-L-rhamnopyranoside (2), as well as 10 known compounds including 2 flavonol glycosides, afzelin (3) and kaempferol-3-O-α-L-rhamnopyranoside (4), 2 flavans, (-)-epicatechin (5) and proanthocyanidin A-2 (6), 3 triterpenoids, friedelin (7), epifriedelanol (8) and β-amyrin (9), a peptide, N-benzoylphenylalanyl-N-benzoylphenylalaninate (10), and 2 sterols, β-sitosterol (11) and daucosterol (12) were isolated and identified by using combination of mass spectrometry and various 1D and 2D NMR techniques. This is the first report of flavonoid glycosides possessing a 2-methyl-2-hydroxylethoxyl group in sugar moiety from D. longan.
