Down-regulation of Apelin in obesity-related hypertensive rats induced by high-fat diet: Possible role of angiotensin II–AT1R system

Abstract:Objective: Apelin, a new adipocytekine exerting potent hypotensive effect, is expected to be involved in the pathogenesis of obesity-related hypertension. We investigated the expression of Apelin-APJ system in obesity-related hypertensive rats induced by high-fat diet, and also studied the effects of Angiotensin II (Ang II) on Apelin-APJ expression in 3T3-L1 adipocytes. Methods: Obesity related hypertensive (OH) rats were established in Sprague–Dawley rats fed a high-fat diet for 15 weeks. OH rats and normal-fat diet (NF) rats were then randomly assigned to 1 of 2 groups and administered vehicle or perindopril for 4 weeks (OH, OH-P, NC and NC-P). Results: Plasma Apelin levels and the expression of peri-renal adipose tissue Apelin-APJ mRNAs were reduced in OH group than that of NC rats. Perindopril increased this expression in OH-P group rats. The plasma Apelin levels and the levels of peri-renal adipose tissue Apelin-APJ mRNAs are negatively related to systolic blood pressure and plasma Ang II level. Ang II reduced the levels of mRNAs and proteins of Apelin and APJ in both dose- and time-dependent manner in 3T3-L1 adipocytes. Losartan abrogated the negative effects of Ang II. Adipocytes incubated with only losartan could upregulate the expression of Apelin and APJ mRNAs. Finally, the phosphorylation levels of Akt protein were decreased by Ang II. Conclusions: The decreased expression of adipose tissue Apelin-APJ system may play a role in the pathogenesis of hypertension in obesity-related hypertensive rats induced by high-fat diet. The expression of Apelin-APJ system appears to be suppressed by Ang II–AT1R system through AT1R in adipocytes, by interfering Akt phosphorylation.
