Abstract:Asymmetric cyanation of a class of easily available olefins with a favorable cyanide source ethyl cyanoformate (CNCOOEt) was realized by an interesting modular catalyst. High yields and ee values were obtained for a range of substrates under solvent-free and mild reaction conditions. The products obtained could be easily transformed to the enantioenriched useful intermediates 5, 6, and pharmaceutically important gamma-aminobutyric acid 7.

Abstract:Based on different chiral diamine skeletons, a series of bifunctional primary amine-thiophosphoramides were synthesized and screened as the catalysts for the asymmetric Michael addition of acetone to both aromatic and aliphatic nitroolefins. Under the catalysis of a thiophosphoramide derived from 1,2-diphenylethane-1,2-diamine, the corresponding adducts were obtained in high yields (up to >99%) with excellent enantioselectivities (97-99% ee) under mild reaction conditions. Moreover, the catalyst could be recovered via simple phase separation and reused at least five times without any loss of both catalytic activity and stereocontrol.

Abstract:A number of upper rim-functionalized calix[4]thiourea cyclohexanediamine derivatives have been designed, synthesized and used as catalysts for enantioselective Michael addition reactions between nitroolefins and acetylacetone. The optimal catalyst 2 with a mono-thiourea group exhibited good performance in the presence of water/toluene (v/v =1:2). Under the optimal reaction conditions, high yields of up to 99% and moderate to good enantioselectivities up to 94% ee were achieved. Detailed experiments clearly showed that the upper rim-functionalized hydrophobic calixarene scaffold played an important role in cooperation with the catalytic center to the good reactivities and enantioselectivities.

Abstract:A new series of mono-N-sulfonamides of C2-symmetrical 2′-bipyrrolidine have been designed, synthesized, and successfully applied as the organocatalysts to the enantio- and diastereoselective Michael addition of 4H-thiopyran-4-one to a variety of nitroolefins. Catalyst 10 was identified in the examinations to exhibit more superior catalytic ability than the traditional chiral pyrrolidine catalysts, giving high chemical yields, excellent diastereo- and enantioselectivities (up to 99:1 dr and 98% ee). The tert-butyl sulfonamide functionality of catalyst 10 is speculated to play crucial roles in the transition state as both bulky group and hydrogen-bond acceptor. Application of a representative Michael adduct has been also studied and proven to be useful for the diversity-oriented synthesis of enantiopure multiring heterocycles through a unique common bicyclic nitrone intermediate.

Abstract:A chiral dipeptidic proline‐derived thiourea organocatalyst was rationally designed and facilely prepared from dipeptidic proline, a chiral diamine as a linker, and isothiocyanate. This asymmetric catalyst for Michael addition between aldehydes and nitroolefins provided chiral 4‐nitro aldehyde adducts with yields of up to 99 %, 92 : 8 syn‐diastereoselectivity, and 97 % enantiomeric excess at room temperature. The enantiodivergent synthesis of syn‐Michael adducts can be easily achieved by switching the terminal proline in dipeptidic proline‐thiourea catalysts. Abstract:4-Aminothioureaprolinal dithioacetal 4a is a highly efficient catalyst for the asymmetric Michael addition of ketones and aldehydes to nitroolefins requiring only 3 mol-% catalyst loading. The reactions proceeded smoothly and gave syn selective adducts with excellent yields (up to 98% yield), diastereoselectivity (up to >99:1dr), and enantioselectivity (up to 99% ee) under solvent free conditions at room temperature. This extremely simple and practical procedure increases the attractiveness of this reaction.

Abstract:The current work reports an organocatalytic strategy for the asymmetric catalysis of chiral benzofuran-2(3H)-ones bearing 3-position all-carbon quaternary stereocenters. Accordingly, highly enantioselective Michael addition reactions of 3-substituted benzofuran-2(3H)-ones to nitroolefins have been developed by utilizing a bifunctional tertiary-amine thiourea catalyst. The reactions accommodate a number of nitroolefins and 3-substituted benzofuran-2(3H)-ones to give the desired chiral benzofuran-2(3H)-one products with moderate to excellent yields (up to 98%) and moderate to very good selectivities (up to 19 : 1 dr and up to 91% ee). Theoretical calculations using the DFT method on the origin of the stereoselectivity were conducted. The effect of the nitroolefin substituent position on the stereoselectivity of the Michael addition reaction was also theoretically rationalized.

Abstract:Simple pyrrolidine-azole conjugates have been synthesized and found to be efficient catalysts for asymmetric Michael addition to nitrostyrenes. The identified optimal catalysts, pyrrolidine-azoles 2, 8 and 13, could catalyze the asymmetric Michael addition of a range of Michael donors and nitrostyrenes in high yields (up to 99%) and excellent stereoselectivities (up to 99:1 dr and 97% ee).

Abstract:Organo-catalysts for the Michael addition reaction of nitrostyrene with acetone are essential to improve the efficiency and enantioselectivity, and their recycling also faces great challenges. Herein, a sulfonated chiral covalent organic framework (SDA-CCOF) was successfully constructed by post modification of the pre-prepared sulfonated COF with chiral 1, 2-diaminocyclohexane via N-sulfonylation. The catalytic performance was evaluated by the asymmetric Michael addition reaction of β-nitrostyrene with acetone. Under the optimized conditions, 81% conversion of β-nitrostyrene and 90% enantioselectivity were achieved. The generality of the catalyst was examined for diverse substrates with 84–96% enantioselectivity. Moreover, a possible catalytic mechanism of the Michael reaction by SDA-CCOF was speculated based on the characterization results and previous reports. This novel catalyst may provide inspirations for the development of CCOF and the loading of organo-catalysts.

Abstract:We report herein an enantioselective acyl transfer protocol via electrophile activation. The reaction cascade sequence encompasses dinuclear zinc-catalyzed asymmetric Michael addition, intramolecular cyclization, and retro-Claisen reaction, which leads to a step- and atom-economic approach to a variety of protected cyclic tertiary α-hydroxyketones in good yields with excellent enantioselectivities (24 examples, 56%-82% yield, 1.5-13 dr and 79%-96% ee). Besides, the large-scale synthesis and further transformation of the products demonstrate the effectiveness of this method for organic synthesis.

Abstract:Recently the hydrogen-bond activated reactions have attracted much attention.1 Takemoto2 reported a highly enantioselective Michael addition of manolate to nitroolefins catalyzed by a bifunctional organocatalyst with tertiary amine and thiourea moiety. As we known,stereoselective conjugate additions of thiols are interesting due to the standpoint of biological and synthetic importance, however, only very limited good results have been obtained except for the works of Shibasaki3, Kanemasa4 and Deng5 et al.In this letter, we report an efficient catalytic asymmetric Michael reactions of thiols to a,a-unsaturated carbonyl compounds promoted by bifunctional organocatalysts. A series of organocatalysts with chiral amine and thiourea structures were designed and synthesized and have been successfully applied in the conjugated additions of thiols to a,a-unsaturated imides and enones.The reactions got quantitative yields and the ee values were up to 84%. It is noteworthy that the a-asymmetric protonation (up to 43% ee) also could be achieved.The Michael addition between aromatic thiols and a,a-unsaturated carbonyl compounds isdescribed as follows:Works to further increase the enantioselectivity is under investigation in our laboratory.
