Abstract:A synthesis of 1,2-di-O-acyl-3-O-(alpha-D-galactopyranosyl)-sn-glycerols with different acyl groups has been achieved by an efficient method. Reaction of D-galactose with allyl alcohol provided allyl D-galactopyranside as a mixture of alpha and beta anomers. Subsequent recrystallization furnished the desired a anomer. Protection of the hydroxy with a benzyl group followed by oxidation of the 1-O-allylusing OsO4/NMO (N-methylmorpholine N-oxide) gave a versatile intermediate 3-O-(2', 3', 4', 6'-tetra-O-benzyl-alpha-D-galactopyranosyl)-sn-glycerol as a diastereoisomeric mixture. Acylation of the intermediate with various acyl chlorides afforded the corresponding 1,2-di-O-acyl-3-O-(2', 3',4',6'-tetra-O-benzyl-alpha-D-galactopyranosyl)-sn-glycerols. Finally hydrogenolytic deprotection of the benzyl group provided five title compounds (i. e. 1,2-di-O-lauroyl-3-O-(alpha-D-galactopyranosyl)-sn-glycerol, 1,2-di-O-myristoyl-3-O-( alpha-D-galactopyranosyl)-sn-glycerol, 1,2-di-O-palmitoyl-3-O-(alpha-D-galactopyranosyl)-sn-glycerol, 1,2-di-O-stearoyl-3-O-(alpha-D-galactopyranosyl)-sn-glyceroI and 1,2-di-O-hexanoyl-3-O-(alpha-D-galactopyransyl)-sn-glycerol) in good yields over five steps. The structures, of all compounds were confirmed by H-1 NMR, C-13 NMR, 2D NMR, IR and MS spectroscopy.
