Abstract:This study focuses on the aggregation behavior of the biodegradable amphiphilic block copolymers based on methoxy poly(ethylene glycol) (mPEG) as a hydrophilic block and either crystalline poly(caprolactone-b-L-lactide) (P(CL-LLA)) or amorphous poly(caprolactone-b-D, L-lactide) (P(CL-DLLA)) as a hydrophobic block. These block copolymers have a strong tendency to form micelles in aqueous medium, with very low critical micelle concentrations (CMCs). The CMC of P(CL-LLA)-b-mPEG is higher than that of P(CL-DLLA)-b-mPEG when the mPEG block has the same molecular weight. Furthermore, the partition equilibrium coefficient (K-v) of pyrene in the micellar solution of P(CL-LLA)-b-mPEG copolymer was lower than that of P(CL-DLLA)-b-mPEG copolymer when the mPEG block was the same length. These differences were believed to be related to the physical state of the core-forming blocks, i. e., the crystalline P(CL-LLA) block and the amorphous P(CL-DLLA) block. The TEM images showed that micelles formed by P(CL-LLA)-b-mPEG assembled in a cylindrical morphology, whereas those formed by P(CL-DLLA)-b-mPEG took a classical spherical shape. In addition, with differential scanning calorimetry (DSC) and wide-angle X-ray diffraction (WAXD) analyses, it is believed that the crystallization tendency of the core-forming blocks is the main factor governing the morphology of micelles in water. A possible mechanism for the cylindrical assembly morphology was discussed.

Abstract:A series of novel amphiphilic triblock copolymers of poly(ethyl ethylene phosphate) and poly(-caprolactone) (PEEP-PCL-PEEP) with various PEEP and PCL block lengths were synthesized and characterized. These triblock copolymers formed micelles composed of a hydrophobic core of poly(-caprolactone) (PCL) and a hydrophilic shell of poly(ethyl ethylene phosphate) (PEEP) in aqueous solution. The micelle morphology was spherical, determined by transmission electron microscopy. It was found that the size and critical micelle concentration values of the micelles depended on both hydrophobic PCL block length and PEEP hydrophilic block length. The in vitro degradation characteristics of the triblock copolymers were investigated in micellar form, showing that these copolymers were completely biodegradable under enzymatic catalysis of Pseudomonas lipase and phosphodiesterase I. These triblock copolymers were used for paclitaxel (PTX) encapsulation to demonstrate the potential in drug delivery. PTX was successfully loaded into the micelles, and the in vitro release profile was found to be correlative to the polymer composition. These biodegradable triblock copolymer micelles are potential as novel carriers for hydrophobic drug delivery.

Abstract:Biodegradable and amphiphilic triblock copolymers poly(ethyl ethylene phosphate)-poly(3-hydroxy-butyrate)-poly(ethyl ethylene phosphate) (PEEP- b -PHB- b -PEEP) have been successfully synthesized through ring-opening polymerization. The structures are confirmed by gel permeation chromatography and NMR analyses. Crystallization investigated by X-ray diffraction reveals that the block copolymer with higher content of poly(ethyl ethylene phosphate) (PEEP) is more amorphous, showing decreased crystallizability. The obtained copolymers self-assemble into biodegradable nanoparticles with a core-shell micellar structure in aqueous solution, verified by the probe-based fluorescence measurements and transmission electronic microscopy (TEM) observation. The hydrophobic poly(3-hydroxybutyrate) (PHB) block serves as the core of the micelles and the micelles are stabilized by the hydrophilic PEEP block. The size and size distribution are related to the compositions of the copolymers. Paclitaxel (PTX) has been encapsulated into the micelles as a model drug and a sustained drug release from the micelles is observed. MTT assay also demonstrates that the block copolymers are biocompatible, rendering these copolymers attractive for drug delivery.

Abstract:Amphiphilic copolymers that can assemble into core-shell micelles in aqueous media have potential application as drug carriers for controlled-release. In this article an amphiphilic triblock copolymer poly(ethylene oxide)-block-poly(lactic acid)-block-poly(2-hydroxyethyl methacrylate) (PEG-b-PLA-b-PHEMA) was synthesized by ring-opening reaction and atom transfer radical polymerization. The polymers were characterized by H-1 NMR, FT-IR and gel permeation chromatography. In aqueous solution, PEG-b-PLA-b-PHEMA can self-assemble into micelles. Characterized by DLS and TEM, micelles are found to be spherical with uniform size. Large numbers of hydroxyl groups on the PHEMA chain offer reaction sites for subsequent functional modification, which make it a promising platforms for drug delivery systems.

Abstract:This Letter focuses on the preparation and characterisation of a novel amphiphilic multiblock poly(ether-ester anhydride) copolymer [poly(sebacic acid-b-poly(ethylene glycol)-b-sebacic acid) (PES)], paclitaxel-loaded PES nanoparticles (NPs), in vitro evaluation of the release of paclitaxel and cytotoxicity of NPs. PES was prepared by melt polycondensation and the PES NPs were prepared by nanoprecipitation technique. In vitro release behaviour of the paclitaxel-loaded NPs was investigated by high-performance liquid chromatography (HPLC). In vitro cytotoxicity of NPs was evaluated by MTT assay. The composition and structure of PES are consistent with that of the designed copolymer. The paclitaxel-loaded NPs are of spherical shape and size lower than 200 nm. Paclitaxel can be continuously released from the paclitaxel-loaded NPs. The PES NPs are non-toxic to cells. The results suggest that PES NPs are a potential candidate nanocarrier material for the controlled delivery of paclitaxel and other hydrophobic compounds.
Computational studies on self-assembled paclitaxel structures: Templates for hierarchical block copolymer assemblies and sustained drug release

Abstract:Paclitaxel-loaded poly(ethylene oxide)-b-poly(lactide) (PEO-b-PLA) systems have been observed to assemble into fiber structures with remarkably different properties using different chirality and molecular weight of PLA segments. In this study, dissipative particle dynamics (DPD) simulations were carried out to elaborate the microstructures and properties of pure paclitaxel and paclitaxel-loaded PEO-b-PLA systems. Paclitaxel molecules formed ribbon or fiber like structures in water. With the addition of PEO-b-PDLA, PEO-b-PLLA and their stereocomplex, paclitaxel acted as a template and polymer molecules assembled around the paclitaxel structure to form core/shell structured fibers having a PEO shell. For PEO19-b-PDLA27 and PEO19-b-PLLA27 systems, PLA segments and paclitaxel molecules were distributed homogeneously in the core of fibers based on the hydrophobic interactions. In the stereocomplex formulation, paclitaxel molecules were more concentrated in the inner PLA stereocomplex core, which led to slower release of paclitaxel. By increasing the length of PLA segments (e.g. 8,16,22 and 27), the crystalline structure of paclitaxel was gradually weakened and destroyed, which was further proved by X-ray diffraction studies. All the simulation results agreed well with experimental data, suggesting that the DPD simulations may provide a powerful tool for designing drug delivery systems.

Abstract:A series of biodegradable polydepsipeptides based new triblock copolymers, poly (ethylene glycol)-poly(l-lactide)–poly(3(S)-methyl-morpholine-2,5-dione) (mPEG–PLLA–PMMD) have been synthesized and characterized as self-assembly micelle delivery system for paclitaxel (PTX). Compared to the mPEG2000–PLLA2000 diblock copolymers, the triblock copolymers present more benefits such as lower CMC value, positive-shifted zeta potential, better drug loading efficiency and stability. Among the triblock polymers, mPEG2000–PLLA2000–PMMD1400 micelles present low cytotoxicity and promote the anti-cancer activity of PTX on A-549 and HCT-116cells. In addition, mPEG2000–PLLA2000–PMMD1400 micelles prolongs the circulation time of PTX in rat after i.v. injection (5mg/kg) than that of mPEG2000–PLLA2000 micelles and Taxol®. The half life (t1/2β), mean residence time (MRT), AUC0–∞ and clearance (CL) for PTX-loaded mPEG2000–PLLA2000–PMMD1400 micelles are determined to be 1.941h, 2.683h, 5.220μg/mLh (1.8-fold to mPEG2000–PLLA2000 group), 0.967L/hkg−1, respectively. In conclusion, mPEG2000–PLLA2000–PMMD1400 copolymer could be developed as one of the promising vectors to anti-cancer agents for chemotherapeutics.

Abstract:The diblock copolymer, PEG-b-PMEA, was synthesized by reversible-addition fragmentation chain transfer polymerization (RAFT). The PMEA block contained a polymethacrylamide backbone and twin ortho ester rings in the side-chains. At neutral pH, PEG-b-PMEA self-assembled to form stable micelles. At pH 5, the twin ortho ester rings were quickly hydrolyzed to completion in 12h, and releasing nearly 70% of the encapsulated Nile Red dye. The PEG-b-PMEA micelles were completely nontoxic to cultured cells as determined by the MTT assay. Paclitaxel (PTX)-loaded micelles showed toxicity toward lung cancer cells comparable to that of the free PTX at equivalent doses. These results suggest that the PEG-b-PMEA micelles could be useful nano-carriers for pH-responsive delivery of poorly soluble anticancer drugs.

Abstract:A series of block copolymers of methoxy poly(ethylene glycol) and poly(2-methoxyethyl ethylene phosphate)(MPEG-b-PMOEEP) were synthesized using methoxy poly(ethylene glycol)(MPEG) as the initiator and tin(II) 2-ethylhexanoate as the catalyst.The polymers were characterized by gel permeation chromatograph(GPC) and nuclear magnetic resonance(NMR).After dissolved in water directly,the block polymers self-assembled into polymeric micelles with PMOEEP as the hydrophobic core and MPEG as the hydrophilic shell.They were used for hydrophobic anticancer drug paclitaxel(PTX) solubilization.It was found that the solubility of PTX in aqueous solutions was greatly enhanced after addition of the polymers,and was significantly affected by polymer composition and concentrations.Increasing weight ratio of PMOEEP block of the polymer resulted in enhanced PTX solubility,and PTX solubility was increased with the increase of polymer concentration that was correlative to the micellization process of the block polymer in aqueous solutions.The polymers were proved to be biodegradable and biocompatible.1H-NMR spectra were used to demonstrate the degradability of PMOEEP by phosphodiesterase(I).MTT assay revealed these block copolymers were nontoxic to breast cancer cells MCF-7 up to a concentration of 10 mg mL-1,while the activity of PTX solubilized by MPEG-b-PMOEEP polymer against MCF-7 cells was comparable to free PTX dissolved in dimethyl sulfone,indicating these polymers are potential materials for enhanced PTX solubility.

Abstract:A hydrophilic, thermo-responsive triblock copolymer poly(ethylene oxide) monomethyl ether-block-poly(acrylic acid)-block-poly(N-isopropylacrylamide) (MPEO-b-PAA-b-PNIPAM) was synthesized by sequential atom transfer radical polymerization and hydrolysis. These polymers were characterized in detail by H-1 NMR, FT-IR and gel permeation chromatography. In aqueous solution, MPEO-b-PAA-b-PNIPAM can self-assemble into core-shell-corona micelles above a LCST of the PNIPAM block. Condensation reactions easily cross-link the PAA shells, which enhances micellar stability. Doxorubicin (DOX), a model drug, was loaded into the core of the polymeric micelles and followed a pattern of thermo-sensitive drug release in vitro in cumulative release studies. The average size and zeta potential of PNPP (DL = 8%) in water was about 97.1 +/- 1.2 nm, +13.9 +/- 0.6 mV, respectively. The in vitro drug release profile of PNPP micelles showed a clear slow-release effect. The results suggested that NH2-PEG-PCL copolymer might be an excellent carrier for hydrophobic drugs such as PTX. In particular, the NH2-PEG-PCL polymer has potential value for modifying with ligands to work as active targeting drug delivery carriers, which has great significance for cancer therapeutics.
Amphiphilic block copolymers bearing six-membered ortho ester ring in side chains as potential drug carriers: synthesis, characterization, and in vivo toxicity evaluation

Abstract:A new type of amphiphilic block copolymers, poly(ethylene glycol)-block-poly(2-methyl-acrylicacid 2-methoxy-5-methyl-[1,3]dioxin-5-ylmethyl ester) (PEG-b-PMME), bearing acid-labile six-membered ortho ester rings in side chains was synthesized by reversible addition-fragmentation chain-transfer polymerization, and the influence of chain length of the hydrophobic PMME block on micelle properties was investigated. The PEG-b-PMME micelles were stable in aqueous buffer at physiological pH with a low critical micelle concentration. Nile Red as a model drug was encapsulated into the micelles to explore the release profiles. The Nile Red-loaded polymeric micelles showed rapid release of Nile Red in weakly acidic environments (pH 5) but slow release under physiological condition (pH 7.4), due to different hydrolysis rate of ortho ester side chains of PEG-b-PMME. The Paclitaxel (PTX)-loaded micelles retained potency in killing lung cancer cells (A549), compared with the free PTX. No obvious toxicity was found in vitro and in vivo after intraperitoneal injection of the micelles, which confirms that the PEG-b-PMME micelles with unique acid-labile characteristic have great potential as nano-scaled carriers for drug delivery.

Abstract:Aliphatic polyesters and polyphosphoesters (PPEs) have received much interest in medical applications due to their favorable biocompatibility and biodegradability. In this work, novel amphiphilic triblock copolymers of PPE and poly(L-lactic acid) (PLLA) with various compositions were synthesized and characterized. The blocky structure was confirmed by GPC analyses. These triblock copolymers formed micelles composed of hydrophobic PLLA core and hydrophilic PPE shell in aqueous solution. Critical micellization concentrations of these triblock copolymers were related to the polymer compositions. Incubation of micelles at neutral pH followed by GPC analyses revealed that these polymer micelles were hydrolysized and resulted in decreased molecular weights and small oligomers, whereas its degradation in basic and acid mediums was accelerated. MTT assay also demonstrated the biocompatibility against HEK293 cells. These biodegradable polymers are potential as drug carriers for biomedical application. (C) 2008 Wiley Periodicals, Inc.

Abstract:The hydrophobic block of polymeric micelles formed by amphiphilic copolymers has no direct therapeutical effect, and the metabolites of these hydrophobic segments might lead to some unexpected side effects. Here the hydrophobic core of polymeric micelles is replaced by highly water-insoluble drugs themselves, forming a new micellar drug delivery system. By grafting hydrophobic drugs of paclitaxel (PTX) onto the surface of hydrophilic hyperbranched poly(ether-ester) (HPEE), we constructed an amphiphilic copolymer (HPEE-PTX). HPEE-PTX could self-assemble into micellar nanoparticles in aqueous solution with tunable drug contents from 4.1 to 10.7%. Moreover, the hydrolysis of HPEE-PTX in serum resulted in the cumulative release of PTX. In vivo evaluation indicated that the dosage toleration of PTX in mice had been improved greatly and HPEE-PTX micellar nanoparticles could be used as an efficient prodrug with satisfactory therapeutical effect. We believe that most of the lipophilic drugs could improve their characters through this strategy.

Peg-B-Ptba-B-Phema Well-Defined Amphiphilic Triblock Copolymer: Synthesis, Self-Assembly, And Application In Drug Delivery

Abstract:A series of well-defined amphiphilic triblock copolymers, poly(ethylene glycol)-b-poly(tert-butyl acrylate)-b-poly(2-hydroxyethyl methacrylate) (PEG-b-PtBA-b-PHEMA), were synthesized via successive atom transfer radical polymerization (ATRP). ATRP of tBA was first initiated by PEG-Br macroinitiator using CuBr/N,N,N',N",N"-pentamethyldiethylenetriamine as catalytic system to give PEG-b-PtBA diblock copolymer. This copolymer was then used as macroinitiator to initiate ATRP of HEMA, which afforded the target triblock copolymer, PEG-b-PtBA-b-PHEMA. The critical micelle concentrations of obtained amphiphilic triblock copolymers were determined by fluorescence spectroscopy using N-phenyl-1-naphthylamine as probe. The morphology and size of formed aggregates were investigated by transmission electron microscopy and dynamic light scattering, respectively. Abstract:A set of amphiphilic poly(ethylene glycol)-b-poly(ethylene brassylate) (PEG-b-PEB) copolymers based on the PEB hydrophobic block was first synthesized by ring-opening polymerization of ethylene brassylate with an organic catalyst. The EB/PEG molar ratios and reaction times were adjusted to achieve different chain lengths of PEB. Block copolymers that were characterized by 1H NMR and GPC could self-assemble into multimorphological aggregates in aqueous solution, which were characterized by DLS and TEM. The hydrophobic doxorubicin (DOX) was chosen as a drug model and successfully encapsulated into the nanoparticles. The release kinetics of DOX were investigated.

Abstract:A novel biodegradable amphiphilic brush-coil block copolymer consisting of poly(epsilon-caprolactone) and PEGylated polyphosphoester was synthesized by ring opening polymerization. The composition and structure of the copolymer were characterized by 1H NMR, 13C NMR, and FT-IR, and the molecular weight and molecular weight distribution were analyzed by gel permeation chromatograph (GPC) measurements to confirm the diblock structure. These amphiphilic copolymers formed micellar structures in water, and the critical micelle concentrations (CMCs) were around 10(-3) mg/mL, which was determined using pyrene as a fluorescence probe. Transmission electron microscopy (TEM) images showed that the micelles took an approximately spherical shape with core-shell structure, which was further demonstrated by laser light scattering (LLS) technique. The degradation behavior of the polymeric micelle was also investigated in the presence of Pseudomonas lipase and characterized by GPC measurement. Such polymer micelles from brush-coil block copolymers are expected to have wide utility in the field of drug delivery.

Abstract:A series of amphiphilic poly(L-leucine)-block-poly(ethylene glycol)-block-poly(L-leucine) (PLL-PEG-PLL) hybrid triblock copolymers have been synthesized. All the blocks in this system have good biocompatibility and low toxicity. The PLL-PEG-PLL copolymers could self-assemble into micelles with PLL blocks as the hydrophobic core and PEG blocks as the hydrophilic shell, which were characterized by FT-IR, (1) H NMR, and transmission electron microscopy analysis. The critical micellar concentration of the copolymer was 95.0 mg · L(-1) . The circular dichroism spectrum shows that the PLL segments adopt a unique α-helical conformation, which is found to play an important role in controlling the drug release rate. The drug release could be effectively sustained by encapsulation in the micelles. The copolymers may have potential applications in drug delivery.

Abstract:This paper describes the synthesis, characterization, and self-assembly behavior of amphiphilic polyethylene-block-poly(l-glutamate) (PE-b-PGA) diblock copolymers. PE-b-PGA diblock copolymers were obtained by ring-opening polymerization (ROP) of γ-benzyl-l-glutamate-N-carboxyanhydride (BLG-NCA) using PE–COOCH(iPr)NH2 as a macroinitiator and subsequent deprotection of the benzylester groups. The self-assembly behaviors of the PE-b-PGA copolymers in water were studied as a function of pH and ionic strength by means of fluorescence spectroscopy, laser light scattering, UV-circular dichroism, and transmission electron microscopy. The size of the polymeric micelles decreases with a decreasing pH value even at high salt concentrations because the solvating PGA units can perform a coil-to-helix transition.
