Abstract:O-Phosphination of α-dicarbonyls via sequential in situ formation of a Kukhtin-Ramirez adduct and a P(NMe2)3-catalyzed process has been exploited for the synthesis of α-phosphoryloxy carbonyls. A range of P(O)-H derivatives, including diarylphosphine oxides, arylphosphinates, and phosphinates, are competent candidates to be introduced into the α-dicarbonyls in this transformation, and various α-phosphoryloxy carbonyls are obtained. This approach possesses advantages of mild conditions, simple operations, atom economy, high efficiency, and gram-scale synthesis, which make it promising in the synthesis toolbox.

Abstract:Here an unprecedented formal deformylative phosphonylation is disclosed. Under the catalysis of 1 mol % 1,2,3,5-tetrakis(diphenylamino)-4,6-dicyanobenzene (4DPAIPN) and the presence of 200 mol % triethylamine as additive, 4-alkyl-1,4-dihydropyridines (DHPs), derived from aldehydes, underwent phosphonylation smoothly with 9-fluorenyl o-phenylene phosphite, allowing facile synthesis of alkylphosphonates. This strategy is applicable to primary, secondary, and even tertiary DHPs, and exhibits a broad substrate scope and excellent functional group tolerance, thereby enabling the late-stage phosphonylation of a series of naturally-ocurring bioactive and biorelevant molecules.

Abstract:Cooperative bimetallic catalysis is a fundamental approach in modern synthetic chemistry. We report bimetallic cooperative catalysis for the direct decarbonylative heteroarylation of ubiquitous carboxylic acids via acyl C-O/C-H coupling. This novel catalytic system exploits the cooperative action of a copper catalyst and a palladium catalyst in decarbonylation, which enables highly chemoselective synthesis of important heterobiaryl motifs through the coupling of carboxylic acids with heteroarenes in the absence of prefunctionalization or directing groups. This cooperative decarbonylative method uses common carboxylic acids and shows a remarkably broad substrate scope (>70 examples), including late-stage modification of pharmaceuticals and streamlined synthesis of bioactive agents. Extensive mechanistic and computational studies were conducted to gain insight into the mechanism of the reaction. The key step involves intersection of the two catalytic cycles via transmetallation of the copper-aryl species with the palladium(II) intermediate generated by oxidative addition/decarbonylation.

Abstract:We describe the direct synthesis of organophosphorus compounds from ubiquitous aryl and vinyl carboxylic acids via decarbonylative palladium catalysis. The catalytic system shows excellent scope and tolerates a wide range of functional groups (>50 examples). The utility of this powerful methodology is highlighted in the late-stage derivatization directly exploiting the presence of the prevalent carboxylic acid functional group. DFT studies provided insight into the origin of high bond activation selectivity and P(O)-H isomerization pathway.

Abstract:The first iron‐catalyzed oxophosphorylation of styrene derivatives with dialkyl H‐phosphonates under open air conditions is developed, which generates a range of previously underexplored α‐substituted β‐ketophosphonates in 55 ~ 70% yields. Furthermore, a convenient one‐pot procedure by direct conversion of styrenes to α, β‐unsaturated ketones is also developed based on the new catalytic reactions, thus offering a protocol to access two classes of valuable products from simple starting materials.

Abstract:Addition of P-H species to carbonyl groups, namely the Pudovik reaction, normally delivers hydroxyl phosphorus compounds, along with phosphate byproducts in some cases. A few controllable systems starting from phosphites were set up to mainly provide the phosphates. Herein, we present a highly selective protocol starting from phosphonate precursors leading to phosphinate derivatives. Enantioenriched phosphinates were successfully achieved from chiral phosphine oxide precursors. Experimental and theoretical investigations were conducted to understand the mechanistic details.

Abstract:A novel DCC reaction between aromatic aldehydes or ketones and H-phosphonates has been developed for the synthesis of p-formyl or p-acylphenylphosphonates. The synthetic method has excellent para regioselectivities, good yields, and broad substrate scopes and is more benign to the environment. The DCC reaction also tolerates many functional groups, and results in a series of new p-formyl and p-acylphenylphosphonates, which should be important building blocks for the synthesis of versatile arylphosphonate derivatives.

Abstract:Organophosphines are versatile compounds with widespread applications in both organic synthesis and the development of biologically active molecules. Herein, we describe a manganese-catalyzed three-component deoxygenative 1,3-carbophosphination of allylic alcohols with alcohols and phosphines using a borrowing hydrogen strategy. This novel multi-component process offers a direct approach to a diverse library of valuable ε-hydroxy phosphines with moderate to good yields. The method demonstrates a broad substrate scope and good functional group tolerance. The synthetic versatility of distal hydroxyphosphine compounds is showcased through downstream transformations, leading to a wide range of ε-functionalized phosphines. Mechanistic studies suggest a rate-determining step in the manganese-catalyzed dehydrogenation of secondary alcohols and a rapid Michael addition of α, β-unsaturated carbonyl by phosphines.

Abstract:A base-catalyzed O-phosphorylation of aldehyde/ketone with R P -menthyl phenylphosphinate afforded phosphonates. The mechanism was proposed as an intermolecular SN@P reaction of α-hydroxy phosphinate. In an intermediate, the C-P bond was transferred from the equatorial to axial position via Berry pseudorotation, then was cleaved with the leaving of a carbon anion. The P-retention and C-racemization could be satisfactorily explained by the mechanism.

Abstract:A first simple palladium catalyzed allylation of vinylethylene carbonates with β ‐ketophosphonates has been developed. This method provides access to ( Z )‐tri‐ and tetrasubstituted homoallylic phosphonates with exclusive regioselectivity, chemoselectivity and ( Z )‐stereoselectivity. The reaction tolerates a wide substrate scope of vinylethylene carbonates and β ‐ketophosphonates with electron‐donating and electron‐withdrawing groups and works well on the gram scale.

Abstract:A general and practical cross-dehydrogenative coupling protocol between readily available trisubstituted α,β-dehydro α-amino carboxylic esters and H-phosphites is described. This C(sp2)–H phosphorylation reaction proceeds with absolute Z-selectivity promoted by silver salt in a radical relay manner. Abstract:Herein we report a facile method for the synthesis of arylphosphonates via copper‐mediated the cross‐coupling reaction in the presence of 1, 10‐Phenanthroline and Ag2O. This method features a broad scope with respect to both the arylsilanes and H‐phosphonate diesters. Mechanistically, the reaction is proposed to involve oxidation by phosphinyl radical and reductive elimination process as the two key steps. With the optimized conditions established, various arylphosphonates were obtained up to 74% yield. This protocol is the first example of copper mediated the Ar−P bond formation of various arylsilanes with H‐phosphonates, and this novel methodology expands the applications of low cost and toxicity copper reagent in Ar−P bond construction.

Abstract:Different from the reported work focusing on the construction of single P- or C-stereocenter via hydrophosphinylation of unsaturated carbon bonds, the highly diastereo- and enantioselective hydrophosphinylation reaction of allenes, conjugated enynes and 1,3-dienes is achieved via a designed Pd/Co dual catalysis and newly modified masked phosphinylating reagent. A series of allyl motifs bearing both a tertiary C- and P-stereocenter are prepared in generally good yields, >20:1 dr, >20:1 rr and 99% ee. The unprecedented diastereo- and enantioselective hydrophosphinylation of 1,3-enynes is established to generate skeletons containing both a P-stereocenter and a nonadjacent chiral axis. The first stereodivergent hydrophosphinylation reaction is also developed to achieve all four P-containing stereoisomers. The present protocol features the use of only 3-minutes reaction time and 0.1% catalyst, and with the observation of up to 730 TON. A set of mechanistic studies reveal the necessity and roles of two metal catalysts and corroborate the designed synergistic process.
