Abstract:Self-organized TiO2-nanotube layers can be used for immunoassay-type sensing in combination with amplifying CdTe labels in a direct and very sensitive electrochemiluminescent (ECL) configuration. Key properties for this method are the conductivity of the TiO2 nanotubes, and their transparency for light emitted from the CdTe labels at approximately 2.4 eV. To demonstrate the potential of this platform, we constructed a sandwich-type immunoassay onto the TiO2-nanotube wall with a layer of (3-aminopropyl)triethoxysilane as the cross-linker for antibody immobilization. For the counter part of the sandwich, we created an amplification system consisting of TiO2 nanobeads carrying the secondary antibody and multiple CdTe quantum dots (multiQD). For antigen (IgG) detection, we find that this combination of 3D transparent electrode with multiQD labels allows for an ECL detection limit of 0.05 pg mL(-1) and a linearity of the signal in the range of 0.1-10(8) pg mL(-1).

Abstract:CdS nanocrystals (NCs) are decorated homogeneously on the TiO2 nanotube arrays (TiNT) by chemical bath deposition method. The resultant CdS/TiNT composite shows largely amplified electrochemiluminescence (ECL) signals due to the vertically aligned nanotubular structure. The ultrasensitive antigen detection is achieved on the composites via an efficient ECL quenching process introduced by sandwiched immunoreaction, and shows a detection limit of 1.0fgmL−1.

Abstract:Near-infrared electrochemiluminescence (NIR ECL) from quantum dots (QDs) has aroused particular attention. However, whether it is possible to achieve NIR ECL sensing has remained an open question. In this article, we reported a NIR ECL immunosensor with amplification techniques for ultrasensitive and selective determination of biomarker. In this sensing platform, NIR-emitting CdTe/CdS core(small)/shell(thick) QDs were first selected as NIR ECL emitters. The NIR ECL nanoprobe (SiO2-QD-Ab2) was designed by covalent assembly of goat antihuman IgG antibody (Ab2) on CdTe/CdS QDs tagged silica nanospheres. Gold nanoparticle-graphene nanosheet (Au-GN) hybrids were prepared by a sonication-induced self-assembly and served as an effective matrix for initial antibodies (Ab1) attachment. After a sandwich immunoreaction, the functionalized silica nanosphere labels were captured onto the glass carbon electrode surface. Integrating the dual amplification from the promoting electron transfer rate of Au-GN hybrids and the increasing QD loading of SiO2-QD-Ab2 labels, the NIR ECL response from CdTe/CdS QDs enhanced 16.8-fold compared to the unamplified protocol and successfully fulfilled the ultrasensitive detection of human IgG (HIgG) with a detection limit of 87 fg mL(-1). Moreover, as a practical application, the proposed immunosensor was used to monitor HIgG level in human serum with satisfactory results obtained.

Abstract:The marriage of energy transfer with electrochemiluminescence has produced a new technology named electrochemiluminescence energy transfer (ECL-ET), which can realize effective and sensitive detection of biomolecules. To obtain optimal ECL-ET efficiency, perfect energy overlapped donor/acceptor pair is of great importance. Herein, we present a sensitive ECL-ET based immunosensor for the detection of tumor markers, using energy tunable CdSeTe/CdS/ZnS double shell quantum dots (QDs) and gold nanorods (GNRs) as the donor and acceptor, respectively. Firstly a facile microwave-assisted strategy for the synthesis of green- to near-infrared-emitting CdSeTe/CdS/ZnS QDs with time- and component-tunable photoluminescence was proposed. And, on the basis of the adjustable optical properties of both CdSeTe/CdS/ZnS QDs and GNRs, excellent overlap between donor emission and acceptor absorption can be obtained to ensure effective ECL-ET quenching, thus improving the sensing sensitivity. This method represents a novel approach for versatile detection of biomolecules at low concentrations.
Ratiometric electrochemiluminescent strategy regulated by electrocatalysis of palladium nanocluster for immunosensing.

Abstract:This work designed a novel ratiometric electrochemiluminescence (ECL) immunosensing approach based on two different ECL emitters: CdS quantum dots (QDs) as cathodic emitter and luminol as anodic emitter. The ECL immunosensor was constructed by a layer-by-layer modification of CdS QDs, Au nanoparticles and capture antibody on a glassy carbon electrode. With hydrogen peroxide as ECL coreactant, the immunosensor showed a cathodic ECL emission of CdS QDs at -1.5 V (vs Ag/AgCl) in air-saturated pH 8.0 buffer. Upon the formation of sandwich immunoassay, the lumiol/palladium nanoclusters (Pd NCs)@graphene oxide probe was introduced to the electrode. Therefore, the cathodic ECL intensity decreased and luminol anodic ECL emission was appeared at +0.3 V (vs Ag/AgCl) owing to the competition of the coreactant of hydrogen peroxide. Using carcino-embryonic antigen as model, this ratiometric ECL strategy could be used for immunoassay with a linear range of 1.0-100 pg mL(-1) and a detection limit of 0.62 pg mL(-1). The enhanced ratiometric ECL signal resulted from the high density and excellent electrocatalysis of the loaded Pd NCs. The immunosensor exhibited good stability and acceptable fabrication reproducibility and accuracy, showing a great promising for clinical application. This electrocatalysis-regulated ratiometric ECL provides a new concept for ECL measurement, and could be conveniently extended for detection of other protein biomarkers. (C) 2015 Elsevier B.V. Efficient Enhancement of Electrochemiluminescence from Tin Disulfide Quantum Dots by Hollow Titanium Dioxide Spherical Shell for Highly Sensitive Detection of Chloramphenicol

Abstract:In this study, modified hollow titanium dioxide and SnS2 quantum dots (SnS2 QDs) were used to build a novel electrochemiluminescence immunoassay (ECLIA) for ultrasensitive detection of chloramphenicol (CAP). The titanium dioxide hollow spheres (THS) modified with polyethyleneimine (PEI) and gold nanoparticles (Au NPs) was synthesized as a probe material for the novel ECLIA, meanwhile it also could be a coreaction accelerator which could greatly improve the interaction efficiency between SnS2 QDs and S2O82-. Then the modified THS was combined with chloramphenicol antibody to form ECLIA probes, and colloidal gold synthesized as a platform for an ECLIA by ascorbic acid (AA) reduction. A competitive immunoassay strategy was used for the detection of CAP, where CAP in the sample would compete with the coating antigen for the limited antibodies. The proposed ECLIA for CAP detection exhibited high sensitivity with a wide linear range from 0.01 ng mL(-1) to 100 ng mL(-1) and a low detection limit at 3.1 pg mL(-1). Furthermore, it could be seen from electrochemical analysis that the effect of THS on the enhancement of ECL signal is about twice as high as that of ordinary titanium dioxide. Importantly, this work not only successfully applied THS to amplify electrochemical-luminescence signal in the ECLIA, but also successfully prepared a stable and highly selective sensor for ultrasensitive detection of CAP.

Abstract:In this work, a novel methodology to design bifunctional ECL-luminophores with self-enhanced and TSA-amplified characteristics was proposed for improving the sensing performance of ECL-immunosensor toward trace cytokine analysis. Thanks to the qualitative- and quantitative- dual signal amplification technique, the as-prepared ECL biosensor demonstrated excellent detection performance. By analyzing the prospective cytokine biomarkers (IL-6), the ECL immunosensor exhibited a broad examination range with quite low detection limit and quite high selectivity, which was far superior to commercial ELISA kits and ever reported works. In particular, the novel ECL nanoprobe developed here could also be applied to monitor other immune toxicities or disease-related cytokines by using the respective antibodies corresponding to these targets. Moreover, the concept and construction strategy of self-amplified ECL-luminophores presented here could be further extended to design a series of Pdots-derived multicolored ECL probes to meet the needs of multipathway detection applications.

Abstract:In this work, a promising electrochemiluminescence (ECL) immunosensor, based on dual-stabilizers capped CdTe quantum dots (QDs) as signal labels and Fe3O4-Au nanocomposites as nanocarriers, was fabricated for the first time. Magnetic Fe3O4-Au nanocomposites served as effective supporters for the primary antibodies (Abi), due to the high surface area to volume ratio, the ease of bioconjugation, and the convenience of magnetic separation. Au nanoparticles (NPs) on the Fe3O4 surface could not only immobilize Ab(1) via coordination and electrostatic interactions, but also supply the good electroconductivity to the immunosensor. The dual-stabilizers-capped CdTe QDs with completely passivated surface exhibited excellent ECL response and stability in the presence of peroxydisulfate (S2O8-), and thus were used as signal labels for the conjugation with secondary antibodies (Ab(2)). The CdTe Os-labeled Ab(2) was characterized by agarose gel electrophoresis. Successful fabrication of the immunosensor has been proved by cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), transmission electron microscopy (TEM), and energy dispersive X-ray spectroscopy (EDS). The developed immunosensor showed a wide linear range from 0.005 ng mL(-1) to 80 ng mL(-1) for the detection of carcinoembryonic antigen (CEA, as a model). And the detection limitation was 1 pg mL(-1) (S/N = 3). Furthermore, the application of the immunosensor in serum sample verified its accuracy and reliability, indicating that it may open a new avenue for clinical bioassay. (C) 2016 Elsevier B.V. General Strategy to Fabricate Electrochemiluminescence Sandwich-Type Nanoimmunosensors Using CdTe@ZnS Quantum Dots As Luminescent Labels and Fe3O4@SiO2 Nanoparticles As Magnetic Separable Scaffolds

Abstract:This work aims to develop universal sandwich type electrochemiluminescence (ECL) nanoimmunosensors for quantitative detection of biomarkers. A series of low-toxic CdTe@ZnS QDs with different core sizes have been synthesized via hydrothermal method and characterized by UV-vis spectra, photoluminescence spectra, TEM, EDS, and XRD. Especially, the ECL behaviors of CdTe@ZnS QDs have been investigated carefully. The CdTe@ZnS QDs with the highest ECL quantum yields have been chosen as ECL labels for conjugation with secondary antibodies. The QDs-labeled antibodies have been characterized by agarose gel electrophoresis. Meanwhile, Fe3O4@SiO2 magnetic nanoparticles were utilized as nanocarriers for the immobilization of primary antibodies due to magnetic separation ability, large specific surface area, and ease of amination for biofunctionalization. Successful fabrication of the nanoimmunosensor was confirmed by SEM and electrochemical impedance spectroscopy. Carcinoembryonic antigen was detected as a model to prove the feasibility of the above strategy. Under the optimal conditions, the proposed nanoimmunosensor exhibited a wide linear range of 0.01 to 125 ng mL(-1) for carcinoembryonic antigen determination with a low detection limit of 3.0 pg mL(-1) (S/N = 3). Moreover, the nanoimmunosensor also displayed excellent selectivity, good stability, and acceptable reproducibility, indicating its potential applications in clinical diagnostics and immune research.

Abstract:A highly sensitive competitive immunosensor based on the electrochemiluminescence (ECL) of quantum dots (QDs) was proposed by coupling with an enzymatic amplification. The fabrication process of the immunosensor was traced with atomic force microscopic images and electrochemical impedance spectra. The strong cathodic ECL emission of the immobilized QDs could be detected at a relatively low emission potential. The reduction of dissolved oxygen during the cathodic process provided a self-produced coreactant, H(2)O(2), for the ECL emission. Using human IgG (HIgG) as a model protein, upon the immuno-recognition of the immobilized HIgG to its antibody labeled simply with horseradish peroxidase, the ECL intensity decreased due to the steric hindrance of the proteins to electron transfer. The decrease could be greatly amplified by an enzymatic cycle to consume the self-produced coreactant, leading to a wide calibration range of 0.05 ng mL(-1) approximately 5 microg mL(-1) and a low limit of detection for the competitive immunoassay of HIgG. This immunosensor showed good stability and fabrication reproducibility. The immunoassays of practical samples showed acceptable results. This facile immunosensing strategy opened a new avenue for detection of proteins and application of QDs in ECL biosensing.

Abstract:A novel and sensitive electrochemiluminescence (ECL) immunosensor based on CdS quantum dots (QDs)-carbon nanotubes (CNTs) and gold nanoparticles-chitosan (GNPs-CHIT) was presented. CdS QDs ECL was much enhanced by combing poly(diallyldimethylammonium chloride) functionalized CNTs. GNPs-CHIT nanohybrids was used to construct an effective antibody immobilization matrix with excellent stability and bioactivity. The principle of ECL detection for target human IgG is based on the increment of steric hindrance after immunoreaction, which resulted in the decrease in ECL intensity. The linear response range was between 0.006 and 150ngmL−1, and the detection limit was 0.001ngmL−1. This approach offers obvious advantages of being simpler, faster, and more stable compared with other immunosensors, which possesses great potential for protein detection in clinical laboratory.

Abstract:A novel strategy for the enhancement of electrochemiluminescence (ECL) was developed by combining CdSe nanoctystals(NCs),carbon nanotube-chitosan(CNT-CHIT), and 3-aminopropyl-triethoxysilane (APS). A label-free ECL immunosensor for the sensitive detection of human IgG (HIgG) was fabricated. The colloidal solution containing CdSe NCs/CNT-CHIT composite was first covered on the Au electrode surface to form a robust film, which showed high ECL intensity and good biocompatibility. After APS as a cross-linker was covalently conjugated to the CdSe NCs/CNT-CHIT film, the ECL intensity was greatly enhanced. And, an intensity about 20-fold higher than that of the CdSe NCs/CNT-CHIT. film was observed. After antibody was bound to the functionalized film via glutaric dialdehyde (GLD), the modified electrode could be used as an ECL immunosensor for the detection of HIgG. The specific immunoreaction between HIgG and antibody resulted in the decrease in ECL intensity. The ECL intensity decreased linearly with HIgG concentration in the range of 0.02-200 ng mL(-1), and the detection limit was 0.001 ng mL(-1). The immunosensor has the advantages of high sensitivity, speed, specificity, and stability and could become a promising technique for protein detection.
Enhanced Anodic Electrochemiluminescence of CdTe Quantum Dots Based on Electrocatalytic Oxidation of a Co-Reactant by Dendrimer-Encapsulated Pt Nanoparticles and Its Application for Sandwiched Immunoassays.

Abstract:Herein, we synthesized Pt dendrimer-encapsulated nanoparticles (Pt DENs) using amine-terminated sixth-generation polyamidoamine dendrimers. The enhanced and stable anodic electrochemiluminescence (ECL) of 3-mercaptopropionic acid-capped CdTe quantum dots (QDs) in a tripropylamine solution was achieved owing to Pt DENs. The reason may be that Pt DENs exhibit high catalytic electrochemical oxidation in the presence of tripropylamine and excellent conductive property. Inspired by this, Pt DENs were conjugated with Fe3O4@SiO2 nanoparticles and served as nano-carriers. The capture antibodies were immobilized on the Fe3O4@SiO2-Pt DEN nanocomposites, which possess many attractive advantages such as the ease of bioconjugation, large specific surface area, and convenience of magnetic separation. Fluorescence microscopy images and UV-vis spectra were used to verify the immobilization of capture antibodies on the nanocomposites. The CdTe QDs were applied as signal labels for conjugation of nanocomposites with detection antibodies, which were characterized by agarose gel electrophoresis. Electrochemical impedance spectroscopy and cyclic voltammetry demonstrated the successful preparation of an ECL immunosensor. Under the optimal conditions, the proposed immunosensor provided a wide linear range from 0.005 ng mL-1 to 150 ng mL-1 with a detection limit of 0.2 pg mL-1 (S/N = 3) for the detection of carcinoembryonic antigen. Moreover, the immunosensor showed good performance for the detection of carcinoembryonic antigen in serum samples as well as great potential in clinical bioassay.

Abstract:A new, ultrasensitive photoelectrochemical immunosensing platform was established on the basis of CdSeTe@CdS:Mn core-shell quantum dots-sensitized TiO2 coupled with signal amplification of CuS nanocrystals conjugated signal antibodies. In this proposal, carcinoembryonic antigen (CEA, Ag) was selected as an example of target analyte to show the analytical performances of the platform. Specifically, TiO2-modified electrode was first assembled with CdSeTe alloyed quantum dots (AQDs) via electrostatic adsorption assisted by oppositely charged polyelectrolyte, and then further deposited with CdS:Mn shells on the surface of CdSeTe AQDs via successive ionic layer adsorption and reaction strategy, forming TiO2/CdSeTe@CdS:Mn sensitization structure, which was used as photoelectrochemical matrix to immobilize capture CEA antibodies (Ab1); signal CEA antibodies (Ab2) were labeled with CuS nanocrystals (NCs) to form Ab2-CuS conjugates, which were employed as signal amplification elements when specific immunoreaction occurred. The ultrahigh sensitivity of this immunoassay resulted from the following two aspects. Before detection of target Ag, the TiO2/CdSeTe@CdS:Mn sensitization structure could adequately harvest the exciting light with different bands, evidently expedite the electron transfer, and effectively depress the charge recombination, resulting in noticeably increased photocurrent. When target Ag existed, the Ab2-CuS conjugates could dramatically decrease the photocurrent due to competitive absorption of exciting light and consumption of electron donor for CuS NCs coupled with steric hindrance of Ab2 molecules. The fabricated photoelectrochemical immunosensor showed a low limit of detection of 0.16 pg/mL and a wide linear range from 0.5 pg/mL to 100 ng/mL for CEA detection, and it also exhibited good specificity, reproducibility, and stability.

The mSiO2/CdTe was successfully fabricated by loading CdTe QDs into mesoporous silica (mSiO2) and characterized by ultraviolet-visible spectra, photoluminescence spectra, transmission electron microscopy, fourier transform infrared spectrum and zeta potential. The electrochemiluminescence intensity of mSiO2/CdTe is much higher than that of CdTe QDs because a single mSiO2 can encapsulate hundreds of QDs. Therefore, mSiO2/CdTe has been chosen as ECL labels for conjugation with secondary antibodies. Meanwhile, Fe3O4@SiO2@PS was used as a carrier of broad primary antibodies owing to convenient magnetic separation capability, large specific surface area, and the special force between the PS and antibodies. Carcinoembryonic antigen as a model tumor mark has been detected to prove the feasibility of our immunosensing platform. The immunosensor for carcinoembryonic antigen was successfully confirmed by cyclic voltammograms and electrochemical impedance spectroscopy. Under the optimal conditions, the immunosensor presented a linear range of 0.001 to 80ng/mL for carcinoembryonic antigen detection with a low detection limit of 0.3pgmL−1 (S/N=3). Furthermore, the immunosensor also displayed outstanding selectivity, good stability, and acceptable reproducibility, indicating its potential applications in clinical diagnostics.
High-Efficient Energy Funneling Based on Electrochemiluminescence Resonance Energy Transfer in Graded-Gap Quantum Dots Bilayers for Immunoassay

Abstract:A surprising electrochemiluminescence (ECL) enhancement effect in graded-gap CdSeTe@ZnS-SiO2 quantum dot (QD) bilayers was observed and used to create an ultrasensitive immunoassay. CdSeTe@ZnS-SiO2 QDs of two different sizes were used as a donor-acceptor pair, owing to their tunable energy and low biotoxicity. The graded-gap CdSeTe@ZnS-SiO2 QD bilayers were fabricated by layer-by-layer assembly of differently sized CdSeTe@ZnS-SiO2 QDs on a glutaraldehyde-activated electrode. Benefiting from a short interlayer distance and perfect spectral overlap in the graded-gap QD bilayers, highly efficient ECL resonance energy transfer (ECLRET)-based energy funneling was observed, wherein excitons from trapped states could be effectively recycled. Consequently, the observed ECL enhancement for the bilayers was more than four times greater than that observed for reference samples. The graded-gap QD bilayers were utilized in an ECL biosensor for the detection of carcinoembryonic antigen (CEA). The proposed method featured a detection limit of 0.4 pg mL(-1) CEA with a linear calibration range from 1 pg mL(-1) to 200 ng mL(-1). This method represents a novel approach for versatile detection of biomolecules in research and clinical applications.
CdZnTeS Quantum Dots Based Electrochemiluminescent Image Immunoanalysis.

Abstract:In this work, quaternary CdZnTeS quantum dots (QDs) with a particularly strong electrochemiluminescence (ECL) were synthesized as ECL signal labels. The strong ECL signals can be obtained at both cathode and anode with the ECL efficiencies of 19.78% and 1.62%, respectively. The sandwich-structured ECL immunosensors for the detection of alpha-fetoprotein (AFP) and cancer antigen 125 (CA125) were accomplished with direct ECL image analysis. Under optimal conditions, the QDs-based ECL image immunoanalysis possessed good linearity from 0.5 ng/mL to 20 ng/mL for AFP and from 20 U/mL to 500 U/mL for CA125 with the detection limit of 0.1 ng/mL and 6 U/mL, respectively (S/N = 3), and the lower detection limit obtained by photomultiplier tube were 0.1 fg/mL for AFP and 0.03 mU/mL for CA125 with the wide dynamic range from 0.5 fg/mL to 20 ng/mL and from 0.1 mU/mL to 500 U/mL, respectively (S/N = 3). Furthermore, the ECL immunoanalysis was evaluated with commercial enzyme-linked immunosorbent assay in human serum samples. The good results indicated that CdZnTeS QDs-based ECL biosensor has great potential for fast biomedical screening and multi-assays in clinical diagnosis.

Abstract:Electrochemiluminescent (ECL) performance and cytotoxicity of CdTe quantum dots (QDs)-based nanocomposites and its possible application for ECL immunoassay were investigated. Two types of CdTe-based nano composites, i.e., SiO2-coated CdTe (CdTe@SiO2) and CdTe-functionalized SiO2 (SiO2@CdTe), were synthesized and comprehensively compared in regarding of the cytotoxicity and ECL performance. The in vitro cytotoxicity of SiO2@CdTe and CdTe@SiO2 nanoparticles was assessed in L02 cells using standard CCK-8 assay, and their ECL performance was investigated by constructing sandwiched immunosensor using SiO2@CdTe and CdTe@ SiO2 as tags for the labelled antibody, respectively. The results showed that CdTe@SiO2 exhibited much lower cytotoxicity and a higher ECL intensity than SiO2@CdTe. Taking the analysis of alpha-fetoprotein (AFP) as an example, the ECL immunosensor using CdTe@SiO2 as an emitter was proved to have a wide linear dynamic range from 1.0 pg mL(-1) to 100 ng mL(-1) with a low detection limit of 0.22 pg mL(-1) (S/N ratio of 3). The ECL immunosensor also demonstrated satisfactory recovery and excellent reproducibility and stability, indicating that this method has prospects in practical application in the clinical diagnosis of AFP.
