Abstract:Designed to achieve high two-photon absorptivity, new near-infrared (NIR) emitting squaraine dyes, (E)-2-(1-(2-(2-methoxyethoxy)ethyl)-5-(3,4,5-trimethoxystyryl)-1H-pyrrol-2-yl)-4-(1-(2-(2-methoxyethoxy)ethyl)-5-(3,4,5-trimethoxystyryl)-2H-pyrrolium-2-ylidene)-3-oxocyclobut-1-enolate (1) and (Z)-2-(4-(dibutylamino)-2-hydroxyphenyl)-4-(4-(dibutyliminio)-2-hydroxycyclohexa-2,5-dienylidene)-3-oxocyclobut-1-enolate (2), were synthesized and characterized. Their linear photophysical properties were investigated via UV-visible absorption spectroscopy and fluorescence spectroscopy in various solvents, while their nonlinear photophysical properties were investigated using a combination of two-photon induced fluorescence and open aperture z-scan methods. Squaraine 1 exhibited a high two-photon absorption (2PA) cross-section (δ2PA), ∼20 000 GM at 800 nm, and high photostability with the photochemical decomposition quantum yield one order of magnitude lower than Cy 5, a commercially available pentamethine cyanine NIR dye. The cytotoxicity of the squaraine dyes were evaluated in HCT 116 and COS 7 cell lines to assess the potential of these probes for biomedical imaging. The viability of both cell lines was maintained above 80% at dye concentrations up to 30 μM, indicating good biocompatibility of the probes. Finally, one-photon fluorescence microscopy (1PFM) and two-photon fluorescence microscopy (2PFM) imaging was accomplished after incubation of micelle-encapsulated squaraine probes with HCT 116 and COS 7 cells, demonstrating their potential in 2PFM bioimaging.

Abstract:Extensive efforts have been devoted to the development of near-infrared (NIR) dye-based imaging probes and/or photothermal agents for cancer theranostics in vivo. However, the intrinsic chemical instability and self-aggregation properties of NIR dyes in physiological condition limit their widely applications in the pre-clinic study in living animals. Squaraine dyes are among the most promising NIR fluorophores with high absorption coefficiencies, bright fluorescence and photostability. By introducing dicyanovinyl groups into conventional squaraine (SQ) skeleton. These acceptor-substituted SQ dyes not only show superior NIR fluorescence properties (longer wavelength, higher quantum yield) but also exhibit more chemical robustness. In this work, we demonstrated highly stable and biocompatible supramolecular adducts of SQ and the natural carrier protein, i.e., bovine serum albumin (BSA) (SQ⊂BSA) for tumor targeted imaging and photothermal therapy in vivo. SQ was selectively bound to BSA hydrophobic domain via hydrophobic and hydrogen bonding interactions with up to 80-fold enhanced fluorescence intensity. By covalently conjugating target ligands to BSA, the SQ⊂BSA was capable of targeting tumor sites and allowed for monitoring the time-dependent biodistribution of SQ⊂BSA, which consequently determined the protocol of photothermal therapy in vivo. We envision that this supramolecular strategy for selectively binding functional imaging agents and/or drugs into human serum albumin might potentially utilize in the preclinical and even clinic studies in the future.

Abstract:Many efforts have been made to develop near-infrared (NIR) fluorescent dyes with high efficiency for the NIR laser-induced phototherapy of cancer. However, the low tumor targetability and high nonspecific tissue uptake of NIR dyes in vivo limit their applications in preclinical cancer imaging and therapy. Among the various NIR dyes, squaraine (SQ) dyes are widely used due to their high molar extinction coefficient, intense fluorescence, and excellent photostability. Previously, benzoindole-derived SQ (BSQ) was prepared by incorporating carboxypentyl benzoindolium end groups into a classical SQ backbone, followed by conjugating with cyclic RGD peptides for tumor-targeted imaging. In this study, we demonstrate that the structure-inherent tumor-targeting BSQ not only shows a high fluorescence quantum yield in serum but also exhibits superior reactive oxygen species (ROS) generation capability under the 671 nm laser irradiation for effective photodynamic therapy (PDT) in vitro and in vivo. Without targeting ligands, the BSQ was preferentially accumulated in tumor tissue 24 h post-injection, which was the optimal timing of the laser irradiation to induce increments of ROS production. Therefore, this work provides a promising strategy for the development of photodynamic therapeutic SQ dyes for targeted cancer therapy.

Abstract:In this work, we rationally design a new type of squaraine dye SQ, modified with the dicyanovinyl motif that can specifically detect DQ (Diquat) by virtue of aggregation-disaggregation mechanism and electrostatic interaction between DQ and SQ. It undergoes H-aggregation in aqueous solution, resulting in fluorescence quenching. After the addition of DQ, the dicyanovinyl group on the probe reacts with DQ to deagglomerate so that the fluorescence is enhanced, tracing DQ in the environment with high sensitivity (LOD=4.9×10-7 M), selectivity and good stability. In addition, the actual sample spike recovery experiment and the confocal fluorescence imaging experiment of DQ in zebrafish also confirmed the detection method. At present, the "turn-on" type fluorescence probe for detecting DQ by using near-infrared squaraine dye has not been reported. The application of SQ provides a new strategy for the quantification of DQ.

Abstract:The study of supramolecular interactions and aggregation behaviour of functional materials is of great importance to tune and extend their spectral sensitivity and, hence, improve the optoelectronic response of related devices. In this study, we resolve spatially and spectrally the absorption and emission features of a squaraine aggregate by means of confocal microscopy and absorption/photoluminescence spectroscopy. We observe that the aggregate affords both a broad absorption spectrum (centred at 670 nm), likely originated by a dyes configuration with allowed J- and H- arrangements, and a strong and relatively narrow emission in the near-infrared (NIR) part of the spectrum (centred at 780 nm), with a remarkable Stokes shift of 110 nm that is among the largest exhibited by squaraine dyes. These peculiarities would be beneficial for extending the spectral sensitivity of bot photovoltaic and light-emitting diodes, and extremely appealing for possible applications of these aggregates as NIR fluorescent probes in biomedical applications.

Abstract:Traditional organic fluorophores generally have hydrophobic conjugated backbones and exhibit an aggregation-caused quenching emission property, which limits greatly their applications in the biological field. Aggregation-induced emission (ME) fluorophores can breakthrough this shortcoming and are more promising in biological imaging. In this paper, we synthesized three novel donor-acceptor-donor-type second near-infrared (NIR-II) fluorophores and studied their geometric and electronic structures and photophysical properties by both theoretical and experimental studies. All the three fluorophores had typical AIE characteristics, and their emission wavelength spanned the traditional near-infrared and NIR-II regions. They exhibited much stronger fluorescence after being encapsulated in polymer nanoparticles (NPs) than in solutions, and the fluorophore-loaded NPs had desirable biosafety and significant tumor accumulation, indicating that they have great application potentials in tumor detection.

Abstract:Organic fluorescent probes are widely used in bioimaging and bioassays, but the notorious photobleaching hampers their applications. Encapsulation of organic dyes into nanoparticles (NPs) is an effective strategy to minimize photobleaching, but classical organic dye molecules tend to have their fluorescence quenched in aggregate states, which is termed aggregation-caused quenching (ACQ). Here we demonstrate our attempt to tackle this problem through the aggregation-induced emission (AIE) strategy. 3,4: 9,10-Tetracarboxylic perylene bisimide (PBI) is a well-known organic dye with a serious ACQ problem. By attaching two tetraphenylethene (TPE) moieties to the 1,7-positions, the ACQ-characteristic PBI-derivative was converted to an AIE-characteristic molecule. The obtained PBI derivative (BTPEPBI) exhibits several advantages over classical PBI derivatives, including pronounced fluorescence enhancement in aggregate state, red to near infrared emission, and facile fabrication into uniform NPs. Studies on the staining of MCF-7 breast cancer cells and in vivo imaging of a tumor-bearing mouse model with BTPEPBI-containing NPs reveal that they are effective fluorescent probes for cancer cell and in vivo tumor diagnosis with high specificity, high photostability and good fluorescence contrast.

Abstract:This article reports three new two-photon absorption (TPA) materials that are quinolinium-carbazole derivates. They are 3-(N-methyl-4-ethylquinolinium iodide)-9-ethylcarbazole (M4), 3-(N-methyl-4-ethylquinolinium iodide)-9-ethylcarbazole (H2), and 3-(N-methyl-4-ethylquinolinium iodide)-9-ethylcarbazole (H4). Their TPA cross-sections are 491, 515, and 512 GM, respectively. Under the excitation of near-infrared light, their fluorescence emission is about 650 nm. The compounds can stain nucleic acid DNA with the same level of nuclear localization as Hoechst 33342. Under continuous irradiation with a near-infrared laser, the three new compounds showed less fluorescence decay than DAPI, and the average fluorescence decay rates were 0.016%/s, 0.020%/s, and 0.023%/s. They are expected to become new two-photon fluorescent probes of nucleic acid DNA because of their excellent performance.

Abstract:The structural, optical spectroscopic, and density functional theory calculations of 2,4-Bis[4-(N,N-diisobutylamino)-2,6-dihydroxyphenyl] squaraine dye (SQ) are promising investigations that can be useful in photochemical mechanisms explanation and in designing promising chemosensors. The nature of the bonds, the chemical composition of SQ, crystallographic structure, morphology, and quality characteristics were explored by FTIR, XRD, and SEM investigations. The crystallite size, the dislocation density, and the lattice strain were found to be 57.3 nm, 3.045×10-4 nm-2, and 3.065×10-3, respectively. The spectrum behavior of the absorbance and molar absorption coefficient spectra of SQ in DCM with three concentrations were analyzed to get some important optical properties of SQ dye. The determined optical gap transition equals〖 E〗_gap^opt= 1.81 eV. Quantum chemical computations and optimized molecular geometries identified where electrophilic attacks are likely and analyzed the molecule's electrostatic potential. Using DFT calculations at the B3LYP/6-31+G(d,p) level, the electronic structure of the squaraine molecule was confirmed, showing a HOMO-LUMO gap of 1.73 eV. MEP analysis indicated that the oxygen atoms are electron-rich, suggesting potential for hydrogen bonding and electrophilic interactions.

Azulene-Containing Squaraines for Photoacoustic Imaging and Photothermal Therapy

Abstract:Photoacoustic imaging (PAI) guided photothermal therapy (PTT) can realize real-time diagnosis and in situ treatment of cancer at the same time. Absorption in the near-infrared (NIR) region with large molar extinction coefficient (ε) and high value of photothermal conversion efficiency (PCE) are key prerequisites for photothermal agents (PTAs) to realize dual PAI and PTT treatments. Squaraines have stable quinoid structures with strong planarity and rigidity, in favor of the NIR absorption and high ε values. On the other hand, azulene derivatives mostly have very faint fluorescence emission, which is beneficial for photothermal transformation. Herein, two azulene-containing squaraines Az-SQ-1 and Az-SQ-2 are synthesized as high-performance PTAs. In comparison with Az-SQ-1, Az-SQ-2 possesses larger εmax of 3 × 105 M-1 cm-1 at 780 nm in organic solution and higher PCE of 53.2% in the form of nanoparticles under 808 nm laser irradiation. Accordingly, Az-SQ-2 NPs present stronger photoacoustic signals (about 15.1-times the background signal) and more efficient suppression of tumor growth. Our research indicates that the introduction of azulene unit to traditional NIR dyes is a simple but effective approach to obtain outstanding PTAs in the aspect of phototheranostics.

Abstract:We introduce a novel strategy to enhance the fluorescence brightness of organic-molecule-based nanoparticles in the second near-infrared window (NIR-II, 1000-1700 nm) by fabricating J-aggregate nanoparticles SQP-NPs(J). Our prepared J-aggregate nanoparticles SQP-NPs(J) show an emission maximum near 1100 nm, and the emission intensity is 4.8-fold higher than that of H-aggregate SQP-NPs(H). In addition, SQP-NPs(J) can be used for NIR-II imaging guided photothermal therapy on MCF-7 tumor-bearing mice due to the fact that SQP-NPs(J) have highly effective photothermal properties, which are significant for precise tumor diagnostics and treatments.

Abstract:Traditional luminescent materials including fluorescent probes suffer from notorious aggregation‐caused quenching in aqueous solutions. Although several approaches such as the aggregation‐induced emission effect have been developed, it remains a significant challenge to identify an effective and efficient strategy to resolve this issue. Herein, quaternary ammonium salts Q8PBn and Q8PNap as a novel class of bright near infrared window II (NIR‐II, 1000–1700 nm) probes are designed and synthesized, and the twisted intramolecular charge transfer formation at the excited state can be effectively suppressed for the newly designed probes. Furthermore, Q8PNap complexation with fetal bovine serum (Q8PNap/FBS) significantly increases the quantum yield by ≈32‐fold compared with PEGylated tertiary amine Q8P, and Q8PNap/FBS is successfully used to achieve high spatial and temporal resolution imaging of hind limb vasculature, lymphatic system, and small tumor metastasis, as well as precise NIR‐II imaging‐guided tumor and lymph node surgery in small animal models for the first time.

Abstract:A new two-photon fluorescent Cd(2+) probe APQ is developed by introducing a N(1),N(1)-dimethyl-N(2)-(pyridin-2-ylmethyl)ethane-1,2-diamine binding group and a 4-methoxyphenylvinyl conjugation-enhancing group to the 2- and 6-positions of quinoline. This probe shows a large red shift and good emission enhancement under Cd(2+) binding. It also exhibits a high ion selectivity for Cd(2+) (especially over Zn(2+)) and a large two-photon absorption cross section at 710 nm. Two-photon microscopy imaging studies reveal that the new probe is non-toxic and cell-permeable and can be used to detect intracellular Cd(2+) under two-photon excitation.
NIR C-Myc Pu22 G-quadruplex probe as a photosensitizer for bioimaging and antitumor study

Abstract:Despite the fact that C-Myc G-quadruplex in the oncogene promoter regions is one of the crucial targets of antitumor drugs, the selectivities and proliferation inhibitions of its probes towards tumor cells remain a big challenge. Until now, no effective C-Myc G-quadruplex probes have been reported as a photosensitizer to increase their antitumor activities. Here, the first NIR C-Myc G-quadruplex probe PDS-SQ has been designed, compristing a G-quadruplex binder PDS and a squaraine dye SQ as a photosensitizer. Conjugate PDS-SQ could selectively NIR image C-Myc Pu22 G-quadruplex in tumor cells, and show stronger antitumor activity in the irradiation by a chemo-photodynamic method than in the dark. The study provides a new way to develop the novel NIR C-Myc G-quadruplex probes with more potent antitumor activities.

Abstract:This article describes the design and development of squaraine-based semiconducting polymer dots (Pdots) that show large Stokes shifts and narrow-band emissions in the near-infrared (NIR) region. Fluorescent copolymers containing fluorene and squaraine units were synthesized and used as precursors for preparing the Pdots, where exciton diffusion and likely through-bond energy transfer led to highly bright and narrow-band NIR emissions. The resulting Pdots exhibit the emission full width at half-maximum of ∼36 nm, which is ∼2 times narrower than those of inorganic quantum dots in the same wavelength region (∼66 nm for Qdot705). The squaraine-based Pdots show a high fluorescence quantum yield (QY) of 0.30 and a large Stokes shift of ∼340 nm. Single-particle analysis indicates that the average per-particle brightness of the Pdots is ∼6 times higher than that of Qdot705. We demonstrate bioconjugation of the squaraine Pdots and employ the Pdot bioconjugates in flow cytometry and cellular imaging applications. Our results suggest that the narrow bandwidth, high QY, and large Stokes shift are promising for multiplexed biological detections.

Abstract:Fluorescence imaging in the second near-infrared window (NIR-II) allows visualization of deep anatomical features with an unprecedented degree of clarity. NIR-II fluorophores draw from a broad spectrum of materials spanning semiconducting nanomaterials to organic molecular dyes, yet unfortunately all water-soluble organic molecules with >1,000 nm emission suffer from low quantum yields that have limited temporal resolution and penetration depth. Here, we report tailoring the supramolecular assemblies of protein complexes with a sulfonated NIR-II organic dye (CH-4T) to produce a brilliant 110-fold increase in fluorescence, resulting in the highest quantum yield molecular fluorophore thus far. The bright molecular complex allowed for the fastest video-rate imaging in the second NIR window with ∼50-fold reduced exposure times at a fast 50 frames-per-second (FPS) capable of resolving mouse cardiac cycles. In addition, we demonstrate that the NIR-II molecular complexes are superior to clinically approved ICG for lymph node imaging deep within the mouse body.

Abstract:Effective photothermal agents (PTAs) are of particular importance for anticancer phototherapy. Nevertheless, conventional PTAs are frequently plagued by inadequate photostability, short-wavelength absorption, and low photothermal conversion efficiency (PCE). Herein, we designed and synthesized a near-infrared squaraine dye (SQ) with the maximum absorption wavelength at ∼710 nm. The squaraine dye demonstrates much enhanced photostability compared with indocyanine green (ICG), a commercial PTA. To improve the tumor targeting capacity, SQ was encapsulated into polymeric nanoparticles (SQ-NPs), which show a high photothermal conversion of 52.8% with strong photoacoustic signals upon 760 nm NIR laser irradiation. Through intravenous injection into tumor-bearing mice, SQ-NPs accumulate at the tumor site and efficiently inhibit the growth of tumor under photoacoustic image-guided photothermal therapy.
