Abstract:Using the phoA-fusion technology, the recombinant metallothionein (MT) from freshwater crab (Sinopotamon yangtsekiense) has been successfully produced in Escherichia coli. MT purified from the bacterial suspension showed one polypeptide with a molecular weight of 7 kDa by tricine-sodium dodecyl sulfate-polyacrylamide gel electrophoresis (Tricine-SDS-PAGE). Western-blotting confirmed the polypeptides had a specific reactivity with mouse polyclonal MT anti-serum. Based on the purified MT and MT anti-serum, the reaction parameters for an enzyme-linked immunosorbent assay (ELISA) were developed. The direct coating ELISA showed a higher linear relationship compared to antibody sandwich coating ELISA. The optimal dilution rates of purified MT anti-serum and coating period were shown to be 1:160,000 and 12 hours at 4°C. At 37°C, the appropriate reaction duration of the first antibody and the second antibody were 2 hours and 1 hour, respectively. According to these optimal parameters, the standard linear equation, y = 0.0032x + 0.1769 (R2 = 0.9779, x, y representing MT concentration and OD450 value), was established for the determination of MT concentration with a valid range of 3.9-500 ng/ml. In verification experiments, the mean coefficients of variation of the intra-assay and inter-assay were 3.260% and 3.736%, respectively. According to the result of MT recovery, ELISA with an approaching 100% MT recovery was more reliable and sensitive than the Cd saturation assay. In conclusion, the newly developed ELISA of this study was precise, stable and repeatable, and could be used as a biomarker tool to monitor pollution by heavy metals.

Isolation, Purification and Identification of Metallothionein in Chinese Soft-Shelled Turtle (pelodiscus Sinensis) Liver

Abstract:Metallothioneins ( MTs ) are a group of low molecular weight, metal-binding and cysteine rich proteins. MTs are widely distributed throughout living organisms and are fairly well identified in mammals, amphibians, fishes, plants, fungi, and cyanobacteria. To compare with the MT's- character, the high degree of conservation of amino acid sequence and the central segments in MTs from many sources will provide significant information for investigation of MT's structure and function or the molecular evolution. Thus far, there are no reports for MT isolated from reptiles. In the present paper, we report the presence of MT from turtle liver. Each extract of Chinese soft-shelled turtle (Pelodiscus sinensis) liver after induction with subcutaneous injection of ZnSO 4, CuSO 4 or CdCl 2 was separated by gel filtration chromatography Sephadex G-50 column and subsequent DEAE Sepharose CL-6B, than Sephadex G-25 for desalination, respectively. According to the results of mass spectrometry and HPLC filtration chromatography detection, the molecular weight of the MT is shown to be about 6 300 dalton. The MT containing 61 amino acid residues has a high cysteine content (17%), aromatic amino acids (tyrosine, phenylalanine), arginine and histidine are considerably low. Zn-MT, Cu-MT or Cd-MT has an ultraviolet absorption shoulder near 220 nm, 270 nm or 250 nm, respectively. In the present work we show that the properties of turtle liver MT are similar to the mammals in molecular weight and cysteine residues of MT. However, the other properties (including the amino acid composition of MT and the capability of binding metal ions) are similar to the MT from the earthworm and yeast. The characteristic of MT in turtle lies between the MT of mammals and lower living organisms. This result presents the distinguishing characteristics of biological evolution of MT in turtle.
[Purification of cadmium ion binding metallothionein-3 by proteinase digestion on affinity chromatographic column].

Abstract:The gene encoding human metallothionein-3 (hMT-3) was synthesized and inserted into the poly-cloning sites of fusion expression vector pALEX, and fused downstream to its glutathione S-transferase (GST) fusion partner. Fusion protein GST-Cd2+-hMT-3 was expressed after isopropyl-beta-D-thiogalactopyranoside (IPTG) induction and addition of 0.1 mmol/L CdSO4 into the culture medium, and mainly existed in cellular soluble fraction as revealed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis. Recombinant MT was purified by two purification procedures: "proteinase digestion after purification" method, e. g. by elution of GST-Cd2+-hMT-3 from GST affinity chromatography first, then proteinase digestion and GST affinity chromatographic purification again subsequently; and "proteinase digestion in situ" method, e. g. digestion of GST-Cd2+-hMT-3 directly on the column while its binding to the GST affinity chromatographic resin and collection of Cd2+-MT directly from the flow through fraction after the digestion. It was confirmed that the later procedure exhibited more effective and more convenient by avoiding the conventional elution, dialysis and lyophilization processes and increasing the purity, recovery or yield of the final product. After further purification by a Superdex 75 HR 10/30 column, finally 6-7 mg of Cd2+-hMT-3 was obtained from 3 L of flask culture with the recovery of about 1.8%. SDS-PAGE, amino acid composition and inductively coupled plasma atomic emission spectrometer (ICP-AES) analysis showed that the relative molecular mass of Cd2+-hMT-3 is about 7 000, with a purity above 90%. Its amino acid composition is consistent with the expected value of natural hMT-3, particularly no aromatic amino acid and histidine, and the atomic ratio of 21: (7.5 +/- 0.1) for S: Cd, is also consistent with the theoretical value of 21: 7.

Abstract:The metallothioneins (MT) in Boettcherisca peregrina larvae were induced and produced after their feeding on the dietary containing 800 μg cadmium chlorides per gram diet for 48 hours. Two forms of MTs (MT-Ⅰ and MT-Ⅱ) were isolated and purified from the supernatant of the whole body homogenates from the larvae after exposure to cadmium chlorides, using gel chromatography on Sephadex G-50, ion-exchange chromatography on UNO TM Q1, and desalination on Bio-Gel P-6. Molecular weight for both MT-Ⅰ and MT-Ⅱ was 9 kD. Both MT-Ⅰ and MT-Ⅱ contained 7 cadmium atoms and 20 sulfhydryl groups per molecule, and showed the spectral characteristic of cadmium-thionein, i.e. a broad shoulder at 254 nm. As to amino acid composition, the content of cysteine residues was the highest, accounting for 36.6% and 31.8% of all amino acids for MT-Ⅰ and MT-Ⅱ respectively; however, the contents of aromatic amino acid and histidine were very low, just sharing 1%-2% of all amino acids.

Abstract:After we modified the protocol of purification, monkey metallothionein-1 (mkMT-1) and its mutant at position 33 (C33M mutant) were efficiently expressed and purified by using the glutathione-S-transferase fusion protein system. The protein yield has been considerably improved (8 mg/L culture for mkMT-1 and 10 mg/L culture for C33M mutant). The recombinant MT-1 and C33M mutant were characterized by ESI-MS, UV, and CD spectra. The reactions of MI-1 and C33M mutant with 5,5′-dithiobis(2-nitrobenzoic acid) and EDTA also have been carefully studied. The pH titration of MT-1 and C33M mutant has been studied by UV and CD spectra. The mutation of cysteine-to-methionine at position 33 mostly maintains the α-domain structure similar to that in wild-type mkMT-1, but the C33M mutant has significant loss of stability and cooperative properties of the domain.

Abstract:Research on metallothionein (MT), unlike studies on any other metalloprotein, captures best the difficulties that investigators faced, and continue to face, when relating protein structure to protein function. This article discusses the structural complexity of the family of human MTs with the goal of providing a background for future investigations of their functions in health and disease by molecular and atomic spectroscopies. The analytical (bio)chemist needs to overcome formidable challenges in terms of sample preparation and speciation. MTs are expressed tissue- and isoprotein-specifically; their expression responds to a variety of physiological and environmental stimuli; they are present as closely related products of at least ten different genes, several of which can be expressed simultaneously at considerably different levels in a given tissue; they have additional polymorphic forms, variable metal load, oxidation states, and degrees of polymerization; they react with different metals and thiol-modifying agents; they are localized in the extracellular and intracellular space and re-distribute subcellularly. Results from the application of highly sensitive fluorimetric techniques suggest that the active form of the zinc/thiolate clusters in MT is not the one described by X-ray crystallography or NMR spectroscopy and that the thiol chemistry is the central aspect of MT's mechanism of action. MT is not saturated with zinc ions under normal physiological conditions, and it exists to variable extent in the apoform (thionein) and in partially oxidized forms (thionin). The complexity of human MTs calls for metallomics and metalloproteomics approaches that integrate the knowledge from several scientific disciplines. Any interpretation of MT functions is based on the linkage between the metal ions and their sulfur ligands from the cysteines. Studies on the antioxidant or reactive species-scavenging properties of the cysteine sulfurs of MT need to take the effects on zinc metabolism into account, while any studies with transition metal ions need to consider the effect of metal binding on the reactivity of the cysteine sulfurs.

Abstract:The purification mechanisms of rare earths metals were briefly explained. The methods for purifying rare earth metal mainly included vacuum melting, electrolytic refining, vacuum distillation/sublimation, fused-salt extraction, zone refining, solid-state electrotransport, electrochemical deoxidation, external getter method, plasma arc melting (Ar or Ar+H-2), etc. The research progress and purification effects were summarized. Multiple means for removing impurities should be combined and the level of equipment should be promoted in order to improve the effects of purification, reduce the cost and shorten production cycle.

Abstract:Metallothionein (MT), a metal-binding protein induced primarily by heavy metals in vertebrates, is considered a biomarker for environmental heavy-metal contamination. To investigate heavy metal pollution in the freshwater environment, MT-I and MT-II were purified from livers of crucian carp (Carassius carassius) by gel exclusion chromatography and ion exchange chromatography. To detect the purified MT-II, a specific monoclonal antibody (mAb) against crucian carp MT-II was produced from the hybridoma strains by cell-cell fusion. By using Enzyme-Linked Immunosorbent Assay (ELISA) with this mAb, the purified crucian carp MT-II was detected with a high specificity and sensitivity. There was a good correlation between the amount of MT-II in carp livers and the concentration of heavy metals in water. ELISA was then used to evaluated the degree of heavy metal pollution in two freshwater systems. The results indicate that the MT-II content in carp liver tissue can be used as an indicator of environmental heavy-metal pollution.

Abstract:Sinopotamon Henanense expresses two metal‒induced metallothioneins (MTs), Cd‒induced MT and Cu‒induced MT (ShCuMT). The Cd‒induced MT has been characterized as a Cd‒thiolate MT. However, it is unknown whether ShCuMT is a Cu‒thiolate MT. In the present study, ShCuMT was expressed heterologously in Escherichia coli and purified by Ni‒NTA column and superdex‒75 column. And its metal‒binding feature was evaluated by DTNB reaction, circular dichroism spectroscopy (CD), isothermal microtitration (ITC), electrospray flight mass spectrometry (ESI‒TOF‒MS), and matrix‒assisted laser desorption ionization flight mass spectrometry (MALDI‒TOF‒MS). Bioinformatics analysis demonstrated that ShCuMT possessed the cysteine‒triplet motif of a Cu‒specific MT. Expression and purification of ShCuMT illustrated that SUMO tag used as the production system for ShCuMT resulted in a high production yield. The stability order of ShCuMT binding metal ions were Cu (Ⅰ) > Cd (Ⅱ) > Zn (Ⅱ). The CD spectrum indicated that ShCuMT binding with Cu (I) exhibited a compact thiol metal clusters structure. Besides, there emerged no a visible nickel‒thiol absorption after Ni‒NTA column affinity chromatography. The ITC results implied that Cu‒ShCuMT possessed the optimal thermodynamic conformation and the highest stoichiometric number of Cu (Ⅰ). Overall, the results suggested that SUMO fusion system is a robust and inexpensive approach for ShCuMT expression and Ni‒NTA column had no influence on metal binding of ShCuMT and Cu(Ⅰ) was considered its cognate metal ion, and ShCuMT possessed canonical Cu‒thiolate characteristics. The metal binding feature of ShCuMT reported here contributes to elucidating the structure‒function relationship of ShCuMT in S. Henanense.
[Rapid Determination of Metallothioneins in Foods by Capillary Zone Electrophoresis].

Abstract:A rapid method for the analysis of metallothioneins (MT) in foods by capillary zone electrophoresis(CZE) was developed. Two isomers of MT (MT1, MT2) in foods were separated and determined. After a series of optimization, the separation and determination of MT1 and MT2 were obtained within 10 min by using the phosphate buffer system consisting of 0.02 mol/L Na2HPO4 and 0.02 mol/L NaH2PO4 (pH 7.0), and UV detection at 200 nm. Under the optimal experimental conditions, the minimum detectable limit was 1 mg/L, and the added standard recoveries of MT1, MT2 in foods were found to be in the range of 82.0%-93.4%. The relative standard deviations (RSD) were found to be lower than 10%. Therefore, with this simple and rapid method, the contents of two isomers in foods can be determined by external standard method after sample pretreatment.

Abstract:The content of metallothionein (MT) in fish can serve as a biomarker for heavy metal contamination in an aquatic environment. In this paper, an improved sample separation and quantification method using cadmium saturation combined with anion exchange high performance liquid chromatography–inductively coupled plasma mass spectrometry (HPLC–ICP-MS) was developed to measure the two major isoforms of MT (MT-I and MT-II) in fish. Replacing sulfur with cadmium as the quantitative element by cadmium saturation reduced the instrumental requirement and analysis cost. In the chromatographic separation, NH3–NH4Cl buffer was applied as the mobile phase instead of Tris–HCl buffer to prevent carbon deposition on the ICP cone. The chloride concentration was found to be a key factor to the separation of MT-I and MT-II, and the optimum chromatographic condition was 27 mM of NH3–NH4Cl in isocratic mode, with a pH of 8.6 and a flow rate of 0.8 mL min−1. The limits of detection were 0.7 μg g−1 for MT-I and 1.0 μg g−1 for MT-II, with recoveries ranging from 96.2 to 104.2% for MT-I and 84.7 to 90.4% for MT-II in different fish tissues. The relative standard deviations were less than 8% for both isoforms.Graphical abstract

Abstract:A purification procedure using microporous membrane and oxidation in air for single-walled carbon nanotubes (SWCNTs) prepared by arc-discharge was discussed which eliminated coexisting metal nanoparticles, carbon nanoparticles and other impurities. The characterization of SWCNTs was carried out at different steps of the purification by TGA, HRTEM and Raman spectra. Experiments show that the method we used is effective to purify the SWCNTs produced by arc-discharge, SWCNTs with the purity of more than 90% could be obtained.

Abstract:Tailor-made materials for the purification of proteins with His-tag was designed through synergizing the selectivity of surface sieving and metal ion affinity. By excluding impurity proteins out of the surface polymer network, such materials could purify His-tagged proteins from the crude cell lysis with purity up to 90%, improved by 14% compared to that obtained by the commercial metal chelating affinity materials. This study might promote the His-tagged protein purification to a new level.

Abstract:The principle and application of seven purification methods for metals in solid state are investigated. Ultra high vacuum degassing and external gettering can effectively remove interstitial impurities in refractory metals. Interstitial impurity can be decreased to very low level in fused halide salt at low temperature, especially when current flows through. Solid state electro-transport technology can remove both interstitial impurities and many metallic impurities. The application and limitation of solid state thermo-transport technology are also discussed.

Abstract:The development of highly efficient membranes technology using low-pressure driven filtration process, is one of the principal challenges in the wastewater treatment field, especially those aimed at the removal of trace heavy metals. In this work, a novel positively charged tight ultrafiltration (PCTUF) membrane was developed to remove heavy metal cations (Mn2+, Co2+, Ni2+, Zn2+ and Cd2+) from contaminated waters via electrostatic repulsion mechanism. The PCTUF membrane was fabricated from a new polymer with poly (vinyl chloride co dimethylaminoethyl methacrylate), P (VC-co-DMA) via a nonsolvent induce phase separation (NIPS) process and following facile surface quatemization. The quatemization conditions, the pore structures and chemical properties of the membranes were investigated in detail. The optimally quatemized membrane possessed a positively charged surface and 3.27 nm charged channel with the water permeability of 84 L m(-2) h(-1) bar(-1). The rejections of heavy metal cations surpassed 95% for feed solutions containing 10 ppm heavy metal. Moreover, the influences of feed concentrations and the operating condition with pressure and pH on the membrane performances were also investigated. The results revealed that the prepared PCTUF membrane with its high perm-selectivity performance provides a worthy reference for highly efficient removal of heavy metal cations.
Partial purification, characterization and translation in vitro of rat liver metallothionein messenger ribonucleic acid

Abstract:Poly(A)+ (polyadenylated) mRNA coding for metallothioneins was purified 13-fold from rat liver polyribosomes and was identified by its ability to direct the biosynthesis of these proteins in a wheat-germ cell-free system. The carboxymethylated products of the protein-synthesizing system in vitro were analysed with sodium dodecyl sulphate/20% polyacrylamide-gel electrophoresis. The labelled compounds [3H]serine and [35S]cysteine were incorporated at high specific radioactivity into proteins that co-migrated with authentic metallothioneins. No [3H]leucine incorporation was found, in agreement with the amino acid composition of the metallothioneins. Metallothionein mRNA had a sedimentation coefficient of 9 S and carried a maximum of four ribosomes. At 5 h after a subcutaneous injection of ZnCl2 or CdCl2 (10 mumol/kg body wt.), the amount of this mRNA increased approx. 2- and 4-fold respectively, on the basis of translation in vitro. The increase in metallothionein mRNA (defined by translation in the wheat-germ system) was transient and, after CdCl2 treatment, fell back to control values by 17 h. Metallothioneins constituted a maximum of 0.8% of the total protein products synthesized in the wheat-germ system by total mRNA isolated from rat liver after CdCl2 treatment.
Sensitive and Selective Sensing System of Metallothioneins Based on Carbon Quantum Dots and Gold Nanoparticles.

Abstract:In this study, we developed a sensitive and selective sensing system for the detection of metallothioneins (MTs). The system is based on the fluorescence resonance energy transfer (FRET) between carbon quantum dots (CQDs) and gold nanoparticles (AuNPs). In this method, the fluorescence emission of CQDs was quenched by AuNPs due to FRET. When MTs were added to the CQD-AuNP system, the strong combination between thiol group and Au made the CQDs release from AuNPs, and the fluorescence of CQDs was recovered. The CQD-AuNP system can detect the MTs in aqueous solution (pH 3.0, citrate-HCl buffer) selectively and sensitively with a short response time (15 min). Results show that the fluorescence recovery efficiency has a good linear relationship with the MTs concentration in the range of 12-210 nmol L-1, and the limit of detection was 5.25 x 10(-9) mol L-1. Furthermore, the sensing system was utilized to determine MTs in human urine samples with satisfactory results. The proposed system exhibits the advantages of high sensitivity, high selectivity, easy operation and most importantly, low cost and non-cytotoxicity to detect protein MTs. (C) 2020 Elsevier B.V. Electrochromatography, as a new chromatography technique, is one of the promised separation techniques suitable for mu-TAS, because it has not only the high performance efficiency of conventional capillary electrophoresis and high selectivity of high performance liquid chromatography. Latest progress on chip electrochromatography is reviewed.
Genetic modifications of metallothionein enhance the tolerance and bioaccumulation of heavy metals in Escherichia coli

Abstract:Metallothioneins (MTs) are low molecular weight cysteine-rich proteins that bind to metals. Owing to their high cysteine (Cys) content, MTs are effective mediators of heavy metal detoxification. To enhance the heavy metal binding ability of MT from the freshwater crab Sinopotamon henanense (ShMT), sequence-based multiple sequence alignment (MSA) and structure-based molecular docking simulation (MDS) were conducted in order to identify amino acid residues that could be mutated to bolster such metal-binding activity. Site-directed mutagenesis was then used to modify the primary structure of ShMT, and the recombinant proteins were further enhanced using the SUMO fusion expression system to yield SUMO-ShMT1, SUMO-ShMT2, and SUMO-ShMT3 harboring one-, two-, and three- point mutations, respectively. The resultant modified proteins were primarily expressed in a soluble form and exhibited the ability to readily bind to heavy metals. Importantly, these modified proteins exhibited significantly enhanced heavy metal binding capacities, and they improved Cd2+, Cu2+ and Zn2+ tolerance and bioaccumulation in Escherichia coli (E. coli) in a manner dependent upon the number of introduced point mutations (SUMO-ShMT3 > SUMO-ShMT2 > SUMO-ShMT1 > SUMO-ShMT > control). Indeed, E. coli cells harboring the pET28a-SUMO-ShMT3 expression vector exhibited maximal Cd2+, Cu2+, and Zn2+ bioaccumulation that was increased by 1.86 ± 0.02-, 1.71 ± 0.03-, and 2.13 ± 0.02-fold relative to that in E. coli harboring the pET28a-SUMO-ShMT vector. The present study offers a basis for the preparation of genetically engineered bacteria that are better able to bioaccumulate and tolerate heavy metals, thus providing a foundation for biological heavy metal water pollution treatment.
