Abstract:Superoxide flash represents quantal and bursting production of mitochondrial reactive oxygen species (ROS) instigated by transient opening of the mitochondrial permeability transition pore (mPTP). Given their critical role in metabolism, ischemia-reperfusion injury, and apoptosis, characterization of flash properties would be valuable to further mechanistic and physiological studies of this newly discovered mitochondrial phenomenon. Here we developed the flash detector FlashSniper based on segmentation of two-dimensional feature maps extracted from time-lapse confocal image stacks, and on the theory for correcting optical distortion of flash-amplitude histograms. Through large-scale analysis of superoxide flashes in cardiomyocytes, we demonstrated uniform mitochondrial ROS excitability among subsarcolemmal and intermyofibrillar mitochondria, and exponential distribution of intervals between consecutive flash events. Flash ignition displayed three different patterns: an abrupt rise from quiescence (44%), a rise with an exponential foot (27%), or a rise occurring after a pedestal precursor (29%), closely resembling action-potential initiation in excitable cells. However, the optical blurring-corrected amplitudes of superoxide flashes were highly variable, as were their durations, indicating stochastic automaticity of single-mitochondrion ROS excitation. Simultaneous measurement of mitochondrial membrane potential revealed that graded, rather than all-or-none, depolarization mirrored the precursor and the primary peak of the flash. We propose that superoxide flash production is a regenerative process dominated by stochastic, autonomous recruitment of a limited number of units (e.g., mPTPs) in single mitochondria.

Abstract:In quiescent cells, mitochondria are the primary source of reactive oxygen species (ROS), which are generated by leakiness of the electron transport chain (ETC). High levels of ROS can trigger cell death, whereas lower levels drive diverse and important cellular functions. We show here by employing a newly developed mitochondrial matrix-targeted superoxide indicator, that individual mitochondria undergo spontaneous bursts of superoxide generation, termed "superoxide flashes." Superoxide flashes occur randomly in space and time, exhibit all-or-none properties, and provide a vital source of superoxide production across many different cell types. Individual flashes are triggered by transient openings of the mitochondrial permeability transition pore stimulating superoxide production by the ETC. Furthermore, we observe a flurry of superoxide flash activity during reoxygenation of cardiomyocytes after hypoxia, which is inhibited by the cardioprotective compound adenosine. We propose that superoxide flashes could serve as a valuable biomarker for a wide variety of oxidative stress-related diseases.

Abstract:Mitochondrial research is important to ageing, apoptosis, and mitochondrial diseases. In previous works, mitochondria are usually stimulated indirectly by proapoptotic drugs to study mitochondrial development, which is in lack of controllability, or spatial and temporal resolution. These chemicals or even gene techniques regulating mitochondrial dynamics may also activate other inter- or intra-cellular processes simultaneously. Here we demonstrate a photostimulation method on single-mitochondrion level by tightly-focused femtosecond laser that can precisely activate restorable fragmentation of mitochondria which soon recover their original tubular structure after tens of seconds. In this process, series of mitochondrial reactive oxygen species (mROS) flashes are observed and found very critical to mitochondrial fragmentation. Meanwhile, transient openings of mitochondrial permeability transition pores (mPTP), suggested by oscillations of mitochondrial membrane potential, contribute to the scavenging of redundant mROS and recovery of fragmented mitochondria. Those results demonstrate photostimulation as an active, precise and controllable method for the study of mitochondrial oxidative and morphological dynamics or related fields.

Abstract:Mitochondrial oxidative flashes (mitoflashes) are oxidative burst events in mitochondria. It is crosslinked with numerous mitochondrial molecular processes and related with pivotal cell functions such as apoptosis and respiration. In previous research, mitoflashes were found as spontaneous occasional events. It would be observed more frequently if cells were treated with proapoptotic chemicals. We show that multiple mitoflashes can be initiated by a single femtosecond-laser stimulation that was tightly focused on a diffraction-limited spot in the mitochondrial tubular structure. The mitoflash events triggered by different photostimulations are quantified and analyzed. The width and amplitude of mitoflashes are found very sensitive to stimulation parameters including laser power, exposure duration, and total incident laser energy. This study provides a quantitative investigation on the photostimulated mitoflashes. It may thus demonstrate such optical method to be a promising technique in future mitochondrial research.

Abstract:Aims Mitochondrial flashes detected with an N- and C-terminal circularly-permuted yellow fluorescent protein (cpYFP) have been thought to represent transient and quantal bursts of superoxide production under physiological, stressful and pathophysiological conditions. However, the superoxide nature of the cpYFP-flash has been challenged, considering the pH-sensitivity of cpYFP and the distinctive regulation of the flash versus the basal production of mitochondrial reactive oxygen species (ROS). Thus, the aim of the study is to further determine the origin of mitochondrial flashes. Main methods We investigated the origin of the flashes using the widely-used pH-insensitive ROS indicators, mitoSOX, an indicator for superoxide, and 2, 7-dichlorodihydrofluorescein diacetate (DCF), an indicator for H2O2 and other oxidants. Key findings Robust, quantal, and stochastic mitochondrial flashes were detected with either mitoSOX or DCF in several cell-types and in mitochondria isolated from the heart. Both mitoSOX-flashes and DCF-flashes showed similar incidence and kinetics to those of cpYFP-flashes, and were equally sensitive to mitochondria-targeted antioxidants. Furthermore, they were markedly decreased by inhibitors or an uncoupler of the mitochondrial electron transport chain, as is the case with cpYFP-flashes. The involvement of the mitochondrial permeability transition pore in DCF-flashes was evidenced by the coincidental loss of mitochondrial membrane potential and matrix-enriched rhod-2, as well as by their sensitivity to cyclosporine A. Significance These data indicate that all the three types of mitochondrial flashes stem from the common physiological process of bursting superoxide and ensuing H2O2 production in the matrix of single mitochondrion.
Synergistic triggering of superoxide flashes by mitochondrial Ca2+ uniport and basal reactive oxygen species elevation

Abstract:Mitochondrial superoxide flashes reflect a quantal, bursting mode of reactive oxygen species (ROS) production that arises from stochastic, transient opening of the mitochondrial permeability transition pore (mPTP) in many types of cells and in living animals. However, the regulatory mechanisms and the exact nature of the flash-coupled mPTP remain poorly understood. Here we demonstrate a profound synergistic effect between mitochondrial Ca(2+) uniport and elevated basal ROS production in triggering superoxide flashes in intact cells. Hyperosmotic stress potently augmented the flash activity while simultaneously elevating mitochondrial Ca(2+) and ROS. Blocking mitochondrial Ca(2+) transport by knockdown of MICU1 or MCU, newly identified components of the mitochondrial Ca(2+) uniporter, or scavenging mitochondrial basal ROS markedly diminished the flash response. More importantly, whereas elevating Ca(2+) or ROS production alone was inefficacious in triggering the flashes, concurrent physiological Ca(2+) and ROS elevation served as the most powerful flash activator, increasing the flash incidence by an order of magnitude. Functionally, superoxide flashes in response to hyperosmotic stress participated in the activation of JNK and p38. Thus, physiological levels of mitochondrial Ca(2+) and ROS synergistically regulate stochastic mPTP opening and quantal ROS production in intact cells, marking the flash as a coincidence detector of mitochondrial Ca(2+) and ROS signals.

Abstract:Mitochondrial research is important to the study of ageing, apoptosis, and metabolic diseases. Over the years, mitochondria have been studied with stimulation by chemical agents in a global manner for basic and applied research. This approach lacks of precision and accuracy in terms of spatial and temporal resolution. Here we demonstrate a direct and well-defined photostimulation targeting on single mitochondrial tubular structure using a tightly-focused femtosecond (fs) laser that could precisely activate mitochondria at single tubule level to show restorable fragmentation and subsequent recovery after tens of seconds. In these two processes, a series of mitochondrial reactive oxygen species (mROS) flashes was observed and found critical to the mitochondrial fragmentation. Meanwhile, transient openings of mitochondrial permeability transition pores (mPTP) were seen with oscillations of mitochondrial membrane potential. These activities were crucial for the recovery through scavenging the mROS. Without the feedback mechanisms, the fragmented mitochondria could not return back to their original tubular structure. These interesting observations show that photostimulation by fs laser is an active, precise, clean and well-defined approach to dissect the role of mitochondria in normal physiology and different kinds of diseases.

Abstract:Mitochondria play a key role in all cellular physiology, processes, and behaviors. It is very difficult to precisely stimulate single mitochondria noninvasively in traditional biomedical research. In this study, we report that femtosecond laser can stimulate fragmentation or swelling of single mitochondria in human mesenchymal stem cells rather than physical disruption or ablation. In experiments, fragmented mitochondria can recover normal very soon but swelling ones cannot. At the same time, laser-induced generation of mitochondrial reactive oxygen species and opening of mitochondria permeability transition pores are involved in mitochondrial responses to photostimulation. Furthermore, the localized translocation of proapoptotic molecules are found in those stimulated mitochondria. Those results suggest femtosecond-laser photostimulation as a noninvasive and precise method for mitochondrial manipulation and related research.

Abstract:Respiratory mitochondria spontaneously undergo quantal, brief bursts of superoxide production, named "superoxide flashes". A property common to all species and cell types examined, the generation of superoxide flashes is coupled to transient opening of the mitochondrial membrane permeability transition pore and depends on the functional integrity of the electron-transfer chain (ETC). The unitary properties of superoxide flashes (amplitude and duration) appear to be stereotypical, at levels from isolated mitochondria to whole organs (e.g. the beating heart) and even to live animals. Functionally, superoxide flashes act as elemental reactive oxygen species (ROS) signaling events ("signaling ROS") that participate in diverse cellular processes, whereas the constitutive electron leakage of molecular oxygen from the ETC produces the bulk of ROS for the regulation of redox homeostasis ("homeostatic ROS"). In particular, superoxide flash incidence provides a digital readout to gauge glucose- and insulin-stimulated mitochondrial metabolism in live animals, and a novel biomarker of the oxidative stress in hyperosmotic stress, ischemia-reperfusion injury, and neurodegenerative diseases. Such dynamic, locally-high levels of ROS in the form of superoxide flashes constitute one of the earliest signals that initiate the cell-death program without appreciably affecting the global ROS level. Moreover, in contrast to the effects of global ROS, superoxide flashes negatively regulate neuronal progenitor cell self-renewal in the developing cerebral cortex. Thus, recent advances in "flashology" have bolstered the notion that ROS act as second messengers in physiological and pathophysiological contexts, and demonstrate the paramount importance of the spatiotemporal organization of ROS signals in determining their signaling efficiency and modality.

Abstract:The role of mitochondrial reactive oxygen species (mitoROS) in cellular function remains obscure. By synthesizing recent data, we propose here that local dynamic mitoROS in the form of “superoxide flashes” serve as “signaling ROS” rather than “homeostatic ROS”, distinguishable from basal mitoROS due to constitutive leakage of the electron transfer chain (ETC). Individual superoxide flashes are 10-s mitoROS bursts that are compartmentalized to a single mitochondrion or local mitochondrial networks. As a highly-conserved universal mitochondrial activity, it occurs in intact cells, in ex vivo beating hearts, and even in living animals. Unlike basal mitoROS, superoxide flashes are ignited by transient openings of a type of mitochondrial permeability transition pore (mPTP), and their incidence is richly regulated by an array of factors that converge on either the mPTP or ETC. Emerging evidence has shown that superoxide flashes decode dietary and metabolic status or exercise, gauge oxidative stress (e.g., during reoxygenation after hypoxia or anoxia), and constitute early mitochondrial signals that initiate oxidative stress-related apoptosis in a context-dependent manner. That they make only a miniscule contribution to global ROS attests to the high efficiency of local ROS signaling. However, the exact mechanisms underlying superoxide flash formation, regulation and function remain uncertain. Future investigation is warranted to uncover the cellular logic and molecular pathways of local dynamic mitoROS signaling in heart muscle cells and many other cell types.

Abstract:The mitochondrion is essential for energy metabolism and production of reactive oxygen species (ROS). In intact cells, respiratory mitochondria exhibit spontaneous “superoxide flashes”, the quantal ROS-producing events consequential to transient mitochondrial permeability transition (tMPT). Here we perform the first in vivo imaging of mitochondrial superoxide flashes and tMPT activity in living mice expressing the superoxide biosensor mt-cpYFP, and demonstrate their coupling to whole-body glucose metabolism. Robust tMPT/superoxide flash activity occurred in skeletal muscle and sciatic nerve of anesthetized transgenic mice. In skeletal muscle, imaging tMPT/superoxide flashes revealed labyrinthine three-dimensional networks of mitochondria that operate synchronously. The tMPT/superoxide flash activity surged in response to systemic glucose challenge or insulin stimulation, in an apparently frequency-modulated manner and involving also a shift in the gating mode of tMPT. Thus, in vivo imaging of tMPT-dependent mitochondrial ROS signals and the discovery of the metabolism-tMPT-superoxide flash coupling mark important technological and conceptual advances for the study of mitochondrial function and ROS signaling in health and disease.

Abstract:Femtosecond lasers have been advancing Biophotonics research in the past two decades with multiphoton microscopy, microsurgery, and photodynamic therapy. Nevertheless, laser irradiation is identified to bring photodamage to cells via reactive oxygen species (ROS) generation with unclear mechanism. Meanwhile, currently in biological researches, there is no effective method to provide controllable ROS production precisely, which originally is leaked from mitochondria during respiration and plays a key role in a lot of important cellular processes and cellular signaling pathways. In this study, we show the process of how the tightly focused femtosecond-laser induces ROS generation solely in mitochondria at the very beginning and then release to cytosol if the stimulus is intense enough. At certain weak power levels, the laser pulses induce merely moderate Ca2+ release but this is necessary for the laser to generate ROS in mitochondria. Cellular original ROS are also involved with a small contribution. When the power is above a threshold, ROS are then released to cytosol, indicating photodamage overwhelming cellular repair ability. The mechanisms in those two cases are quite different. Those results clarify parts of the mechanism in laser-induced ROS generation. Hence, it is possible to further this optical scheme to provide controllable ROS generation for ROS-related biological researches including mitochondrial diseases and aging.
Regulation of Superoxide Flashes by Transient and Steady Mitochondrial Calcium Elevations

Abstract:The mitochondria play essential roles in both intracellular calcium and reactive oxygen species signaling. As a newly discovered universal and fundamental mitochondrial phenomenon, superoxide flashes reflect transient bursts of superoxide production in the matrix of single mitochondria. Whether and how the superoxide flash activity is regulated by mitochondrial calcium remain largely unknown. Here we demonstrate that elevating mitochondrial calcium either by the calcium ionophore ionomycin or by increasing the bathing calcium in permeabilized HeLa cells increases superoxide flash incidence, and inhibition of the mitochondrial calcium uniporter activity abolishes the flash response. Quantitatively, the superoxide flash incidence is correlated to the steady-state mitochondrial calcium elevation with 1.7-fold increase per 1.0 Δ F / F 0 of Rhod-2 signal. In contrast, large mitochondrial calcium transients (e.g., peak Δ F / F 0 ∼2.8, duration ∼2 min) in the absence of steady-state elevations failed to alter the flash activity. These results indicate that physiological levels of sustained, but not transient, mitochondrial calcium elevation acts as a potent regulator of superoxide flashes, but its mechanism of action likely involves a multi-step, slow-onset process.

Abstract:Although the mitochondrial permeability transition pore (mPTP) was first discovered almost 30 years ago [1], it did not attract significant research attention until the 1990's when several studies implicated mPTP in apoptosis [2]. Today, the dogma suggests that opening of mPTP is detrimental to the cell and mPTP activation is widely thought to contribute to disease in cancer, neurodegenerative diseases, stroke, muscular dystrophy, and cardiac reperfusion injury [3]. Multiple factors including Ca(2+), OH(-), P(i), cyclophilin D, reactive oxygen and nitrogen species (ROS and RNS) trigger mPTP opening [4]. However, whether mPTP activation feeds back to alter mitochondrial ROS generation remains unclear. We recently demonstrated that under normal conditions, individual mitochondria undergo spontaneous transient bursts of quantal superoxide generation, termed "superoxide flashes" [5]. Superoxide flashes are observed in all cell types investigated to date and are triggered by a surprising functional coupling between mPTP activation and electron transport chain (ETC) dependent superoxide production. Additionally, reoxgenation following anoxia leads to uncontrolled superoxide flash genesis in cardiomyocytes. This positive feedback mechanism for mPTP/ETC-dependent ROS generation may drive localized redox signaling in individual mitochondria under physiological conditions, and when left unchecked, contribute to global cellular oxidative stress under pathological conditions in cardiac disease. The mPTP activity-dependent cell life and death determination imposes new challenges and opportunities in the pursuit of therapeutic agents for treating diseases in which oxidative stress has been implicated such as cardiac ischemia-reperfusion injury.

Abstract:Respiratory mitochondria undergo stochastic, intermittent bursts of superoxide production accompanied by transient depolarization of the mitochondrial membrane potential and reversible opening of the membrane permeability transition pore. These discrete events were named 'superoxide flashes' for the reactive oxygen species (ROS) signal involved, and 'mitochondrial flashes' (mitoflashes) for the entirety of the multifaceted and intertwined mitochondrial processes. In contrast to the flashless basal ROS production of 'homeostatic ROS' for redox regulation, bursting ROS production during mitoflashes may provide 'signalling ROS' at the organelle level, fulfilling distinctly different cell functions. Mounting evidence indicates that mitoflash frequency is richly regulated over a broad range, and represents a novel, universal, and 'digital' readout of mitochondrial functional status and of the mitochondrial stress response. An emerging view is that mitoflashes participate in vital processes including metabolism, cell differentiation, the stress response and ageing. These recent advances shed new light on the role of mitochondrial functional dynamics in health and disease.

Abstract:Mitochondrial respiration through electron transport chain (ETC) activity critically regulates energy homeostasis and heart performance. Insufficient energy supply and excessive oxidative stress underlie the mechanism of heart failure. How mitochondrial respiration and reactive oxygen species production are fine-tuned in the healthy heart and dys-regulated in the failing heart remain largely unknown. Recently, we discovered stochastic and transient superoxide production events, named superoxide flashes, in single mitochondria of intact cardiac myocytes. Here, we show that superoxide flash is tightly coupled to mitochondrial respiration and diminished superoxide flash activity is a hallmark of the transition to heart failure. Increase in either metabolic substrate availability (glucose 10 mM, palmitate 0.5 mM, or pyruvate 10 mM) or energy demand (isoproterenol 1 µM or pacing) boosted superoxide flash activity. In permeabilized myocytes, addition of the specific substrates for each ETC complex (Complex I, II or IV) elevated superoxide flash activity 3-4 folds over basal level, indicating that superoxide flash arises from the ETC electron flow. Further, myocytes from a Complex I deficient mice (Ndufs4H-/-) showed significantly lower flash activity, which was stimulated by palmitate but not pyruvate. This indicates that Complex I is likely the primary site for electron entrance under normal conditions. However, when Complex I activity is compromised, electron entrance at other sites (associated with palmitate oxidation) may be enhanced. Interestingly, inhibition of the FoF1 ATP synthase by oligomycin A acutely increased flash activity followed by a decline to below normal level. Thus, ETC activity is transiently accelerated upon blockade of the downstream ATP generation. In the pressure overload induced heart failure model, the decline of superoxide flash precedes the contractile dysfunction, suggesting that impaired mitochondrial respiration occurs at the early stage of heart failure. In summary, our data support the mechanistic coupling of superoxide flash with mitochondrial respiration, and suggest that superoxide flash is a sensitive indication of single mitochondrial dysfunction in heart failure.

Abstract:ABSTRACTObjective: Although several methods have been used to detect the intracellular reactive oxygen species (ROS) generation, it is still difficult to determine where ROS generate from. This study aimed to demonstrate whether ROS generate from mitochondria during oxidative stress induced mitochondria damage in cardiac H9c2 cells by laser scanning confocal microscopy (LSCM). Methods: Cardiac H9c2 cells were exposed to H2O2 (1200μM) to induce mitochondrial oxidant damage. Mitochondrial membrane potential (ΔΨm) was measured by staining cells with tetramethylrhodamine ethyl ester (TMRE); ROS generation was measured by staining cells with dichlorodihydrofluorescein diacetate (H2DCFDA). Results: A rapid/transient ROS burst from mitochondria was induced in cardiac cells treated with H2O2 compared with the control group, suggesting that mitochondria are the main source of ROS induced by oxidative stress in H9c2 cells. Meanwhile, the TMRE fluorescence intensity of mitochondria which had produced a great deal of ROS decreased significantly, indicating that the burst of ROS induces the loss of ΔΨm. In addition, the structure of mitochondria was damaged seriously after ROS burst. However, we also demonstrated that the TMRE fluorescence intensity might be affected by H2DCFDA. Conclusions: Mitochondria are the main source of ROS induced by oxidative stress in H9c2 cells and these findings provide a new method to observe whether ROS generate from mitochondria by LSCM. However, these observations also suggested that it is inaccurate to test the fluorescence intensities of cells stained with two or more different fluorescent dyes which should be paid more attention to. Abstract:Normal cardiac excitation-contraction (EC) coupling relies on highly coordinated intracellular calcium (Ca2+) handling and adequate mitochondrial energy metabolism. Whether mitochondria can sense the oscillating Ca2+ signals during EC coupling to adjust its respiration status and other functions is largely unknown. Recently, we discovered stochastic and transient mitochondrial superoxide production events, named superoxide flashes, in resting cardiac myocytes. Superoxide flashes are coupled with mitochondrial respiration and triggered by physiological relevant transient permeability transition pore (mPTP) openings. Using superoxide flash as the optical determination of mitochondrial respiration status in living myocytes, we found that EC coupling promotes mitochondrial respiration and reactive oxygen species (ROS) production as evidenced by the significantly increased superoxide flash activity by electrical stimulation (pacing frequency 0.5 - 2 Hz). Pre-incubation with Ru360 (50-100 uM), a mitochondrial Ca2+ uniporter inhibitor, or cyclosporine A (1 uM), an mPTP blocker, prevented this increase. Simultaneous monitoring of mitochondrial Ca2+ (by rhod-2) and superoxide (by mt-cpYFP, a mitochondrial targeted superoxide indicator) revealed step-wise accumulation of mitochondrial Ca2+ during cytosolic Ca2+ transients in individual mitochondria, which is followed by superoxide flash production and matrix Ca2+ release. Importantly, mitochondrial respiration only responds to transient Ca2+ signals (acute addition of ionophore or pacing) but not chronic manipulations of steady state Ca2+ (depleting or elevating resting cytosolic Ca2+). Through spatiotemporal mapping of superoxide flash and local sarcoplasmic reticulum (SR) Ca2+ releasing events, Ca2+ sparks, we found that significant amount of Ca2+ sparks clustered near the flashing mitochondria. Antioxidant pretreatment attenuated the superoxide flash associated Ca2+ sparks. In summary, mitochondrial Ca2+ uptake during cardiac EC coupling leads to stochastic acceleration of mitochondrial respiration, mPTP activity and free radical production, which in turn modulate local Ca2+ signaling through promoting SR Ca2+ release.

Abstract:Irreversible mitochondrial permeability transition and the resultant cytochrome c release signify the commitment of a cell to apoptotic death. However, the role of transient MPT (tMPT) because of flickering opening of the mitochondrial permeability transition pore remains elusive. Here we show that tMPT and the associated superoxide flashes (i.e. tMPT/superoxide flashes) constitute early mitochondrial signals during oxidative stress-induced apoptosis. Selenite (a ROS-dependent insult) but not staurosporine (a ROS-independent insult) stimulated an early and persistent increase in tMPT/superoxide flash activity prior to mitochondrial fragmentation and a global ROS rise, independently of Bax translocation and cytochrome c release. Selectively targeting tMPT/superoxide flash activity by manipulating cyclophilin D expression or scavenging mitochondrial ROS markedly impacted the progression of selenite-induced apoptosis while exerting little effect on the global ROS response. Furthermore, the tMPT/superoxide flash served as a convergence point for pro- and anti-apoptotic regulation mediated by cyclophilin D and Bcl-2 proteins. These results indicate that tMPT/superoxide flashes act as early mitochondrial signals mediating the apoptotic response during oxidative stress, and provide the first demonstration of highly efficacious local mitochondrial ROS signaling in deciding cell fate.
