Abstract:A concise total synthesis of rac-alsmaphorazine D has been described for the first time. The efficient synthetic strategy features four key transformations: 1) a catalytic intramolecular oxidative cyclization for the delta-lactamindole backbone; 2) an oxidative cyclic aminal formation for the hexahydropyrrolo[2,3-b]pyrrole framework; 3) a transannular radical cyclization for the construction of the diazabicyclo-[3.3.1]nonane structure; and 4) a one-pot desilylation/double epimerization reaction that affirms the relative stereochemistry.

Abstract:Novel total syntheses of oxoaporphine alkaloids such as liriodenine, dicentrinone, cassameridine, lysicamine, oxoglaucine and O-methylmoschatoline were developed. The key step of these total syntheses is Cu-catalyzed conversion of 1-benzyl-3,4-dihydro-isoquinolines (1-Bn-DHIQs) to 1-benzoyl-isoquinolines (1-Bz-IQs) via tandem oxidation/aromatization. This novel Cu-catalyzed conversion has been studied in detail, and was successfully used for constructing the 1-Bz-IQ core.

Abstract:We describe the first total synthesis of the unusual cyclopropane-containing indole alkaloid (–)-rauvomine B via a strategy centered upon intramolecular cyclopropanation of a tetracyclic N-sulfonyl triazole. Preparation of this precursor evolved through two generations of synthesis, with the ultimately successful route involving a palladium-catalyzed stereospecific allylic amination, a cis-selective Pictet–Spengler reaction, and ring-closing metathesis as important bond-forming reactions. The key cyclopropanation step was found to be highly dependent on the structure and conformational strain of the indoloquinolizidine N-sulfonyl triazole precursor, the origins of which are explored computationally through DFT studies. Overall, our synthesis proceeds in 11 total steps and 2.4% yield from commercial materials.

Abstract:Isoquinoline alkaloids are attractive natural products due to their diverse chemical structures as well as remarkable bioactivities. Herein, we report the concise total syntheses of two isoquinoline alkaloids, menisporphine and daurioxoisoporphine C, through a mild and efficient photoredox-catalyzed direct C-H arylation of isoquinoline core with aryldiazonium salt. This new strategy is complementary to the conventional isoquinoline synthesis and would provide us a useful means to achieve a more convergent and flexible approach to access diverse isoquinoline structures.

Abstract:A novel synthetic route for the total syntheses of 1-benzoyl-3,4-dihydroisoquinoline alkaloids was developed. Nelumstemine was synthesized for the first time via 6 steps in 50% overall yield starting from 3,4-dimethoxybenzaldehyde, and longifolonine was also synthesized for the first time via 9 steps in 35% overall yield starting from vanillin. The key step of these total syntheses is photo-oxidation of 1-benzyl-3,4-dihydroisoquinolines to 1-benzoyl-3,4-dihydroisoquinolines by air under visible-light irradiation at room temperature. The unique mild photo-oxidation of 1-benzyl-3,4-dihydro-isoquinolines has been studied in detail.

Abstract:We describe the first total synthesis of tabernanthine and ibogaline. Entry to these iboga alkaloid natural products is enabled by a thermal coupling of indoles and aziridines to furnish the requisite nosyl tryptamine starting materials. This route features a Friedel‐Crafts type alkylation to form the key indole‐isoquinuclidine C‐C bond. Finally, a regio‐ and diastereoselective hydroboration‐oxidation enables C‐N bond formation to close the isoquinuclidine ring system and deliver tabernanthine and ibogaline in 10 and 14% yield respectively. Both syntheses were completed in eight steps.

Abstract:Starting from suitably substituted homophthalic acids, total synthesis of titled alkaloids have been demonstrated in very good yields. The obtained natural products laudanosine and romneine were utilized to accomplish synthesis of two isoquinoline-based alkaloids glaucine and dicentrine. Base-induced selective generation of two different types of benzylic carbanions, their coupling reactions with 3,4-dimethoxybenzyl mesylate, and the regioselective iodination followed by intramolecular aryl–aryl coupling reactions to form the fused biaryl systems were the strategic steps.

Abstract:The Rutaceae family is one of the most studied plant families due to the large number of alkaloids isolated from them with outstanding biological properties, among them the quinoline-based alkaloids Graveoline 1 and Dubamine 2. The most common methods for the synthesis of alkaloids 1 and 2 and their derivatives involves cycloaddition reactions or metal-catalyzed coupling processes but with some limitations in scope and functionalization of the quinoline moiety. As a continuation of our current studies on the synthesis and chemical transformation of 2-aminochalcones, we are reporting here an efficient metal-free approach for the total synthesis of alkaloids 1 and 2 along with their analogues with structural diversity, through a two-step sequence involving intramolecular cyclization, oxidation/aromatization, N-methylation and oxidative C-C bond processes, starting from dihydroquinolin-4-ones as common precursors for the construction of the structures of both classes of alkaloids.

Abstract:Dimeric indolosesquiterpenoids drew significant interest in the synthetic community due to their higher potential of biological activity than their monomeric congeners. Biosynthetically it has been shown that xiamycin A (2a) could dimerised under the oxidative condition to form the more potent all possible N-N, C-N, and C-C dimers. The pursuit of effective methods for creating these linkages has driven the advancement of synthetic chemistry, leading to new strategies and tools that enhance the ability to produce these biologically significant molecules. Till date the synthesis and biological evaluation of dimer of the oridamycin A remains unexplored. Herein we have described a synthetic design towards the concise total synthesis of dioridamycin (1a) and dixiamycin (1b) following a strategic use of hypervalent iodine(III) to forge intramolecular cross amination leading to the indolosesquiterpenoid pentacyclic core under mild and atom economical condition. Our study reveals the utility of PhI(OAc)2 for introduction of intramolecular C-N bond for indolosesquiterpenoids is functional group effective, provides a wider variety of products in good to excellent yields and also have conventionally been viewed as challenging.

Abstract:An efficient, green, and novel method for the synthesis of N-heterocycle-fused quinoxalines is reported herein. Dimethyl sulfoxide was used as both a reactant and a solvent in this reaction. A wide range of products in moderate to excellent yields were obtained, including pyrrolo[1,2-a]quinoxalines, indolo[1,2-a]quinoxalines, 1H-pyrrolo[3,2-c]quinolines, and benzo[4,5]imidazo[1,2-c]quinazolines.

Abstract:[structure: see text]. Catalyzed by CuI/N,N-dimethylglycine, oxiranecarboxamides underwent a highly efficient and stereoselective N-vinylation reaction with (Z)-1-aryl-2-bromoethenes to afford the corresponding enamides. The method has been applied to a straightforward synthesis of (-)-(2R,3S)-SB204900, the enantiomer of the natural product. Following a hypothetic biomimetic pathway, both (+)-(5R,6S)-xi-Clausenamide and (-)-(5R,6S)-balasubramide have been efficiently synthesized for the first time through the unprecedented intramolecular 8-endo-epoxy-arene cyclization of (Z)-N-(phenylvinyl)oxiranecarboxamides.

Abstract:Total synthesis of tropane alkaloids (-)-cocaine and (-)-ferruginine were accomplished in nine steps each and in 55% and 46% overall yields, respectively, starting from the known Betti base derivative (+)-(7aR,10R,12S)-10-(1H-benzotriazol-1-yl)-7a,8,9,10-tetrahydro-12-phenyl-12H-naphtho-[1,2-e]pyrrolo[2,1-b][1,3]oxazine. In this novel route, RCM reaction and 1,3-dipolar cycloaddition were employed as key steps for the enantioselective construction of tropane skeleton and the regioselective introduction of 3-bromo-2-isoxazoline ring as masked cis-2,3-disubstituents. To obtain the desired precursor (2S,5R)-2-allyl-5-vinylpyrrolidine for RCM reaction, we developed a general and practical method for the preparation of enantiopure cis-2,5-disubstituted pyrrolidines bearing alkene- and/or alkyne-containing substituents. We also offered two highly efficient pathways for the conversion of the 3-bromo-2-isoxazoline ring into the desired cis-2,3-disubstituted groups in (-)-cocaine and (-)-ferruginine.

Abstract:The synthesis of diversely substituted quinazoline-2,4(1H, 3H)-diones by cyclization of tert-butyl (2-cyanoaryl) carbamates using readily accessible Boc protected o-amino nitriles is reported. The reaction proceeds smoothly at room temperature using just 1 equiv. of H2O2 as an oxidant under basic conditions. This reaction is compatible with a variety of aromatic/heteroaromatic substrates with different functional groups. This strategy can be utilized for simplified synthesis of Goshuyuamide II and an alkaloid isolated from Zanthoxylum arborescens in good yield. This method was also applied to the synthesis of quinazoline-2,4(1H, 3H)-diones that are precursors to medicinally important compounds: Alfuzosin, Terazosin, Prazosin, Terazosin, IAAP, Doxazosin, FK 366 (Zenarestat) and KF31327.

Abstract:Creating an expedient method for synthesizing biologically significant functionally rich octahydroquinoline is a highly sought‐after yet challenging endeavour. In this report, we document an unprecedented approach to the construction of novel spirooctahydroquinoline oxindole architectures. The reaction proceeds through Pd (II) catalyzed cycloisomerisation and quinine catalyzed [4+2]‐addition sequence to afford the desired adduct in moderate to excellent yield (up to 91%) in a single diastereomer with three contiguous stereocenters including one spirocenter. An overwhelming number of libraries were generated (35 examples) reflecting the synthetic versatility and functional groups/substituents tolerance. Further, the adduct is transformed into medicinally important fluoro‐decahydroepoxyethanoquinoline spirooxindole scaffold with five contiguous stereocenters in excellent yield and selectivity (95% yield and >99:1 dr) through a three steps sequence which signifies the synthetic utility of the developed methodology.

Abstract:The total synthesis of (-)-goniomitine has been accomplished in 11 steps starting from commercially available diethyl L-malate. The synthesis features a chiral pool approach to prepare the chiral C-9 unit containing a quaternary carbon center, an Ir-catalyzed C-H borylation to synthesize the 2-indoleboronic acid pinacol ester, and a Suzuki reaction to couple together the two key intermediates. Notably, the high degree of convergence of this strategy makes it particularly amenable to the total synthesis of other aspidosperma family natural products.
