Abstract:OBJECTIVE:To observe the effects of ionic dissolution products on nano-sized 58S bioactive glass (nano-58S) on proliferation and specific osteogenic genes expression in MG-63 cells.METHODS:Ionic dissolution products were prepared by incubating nano-58S or sol-gel bioactive glass 58S (58S) particulates in Dulbecco's modified Eagle's medium (DMEM) at 1% w/v for 24 hr and filtrated through 0.22 µm filters to remove the particulates. MG-63 cells were cultured in the nano-58S extraction, 58S extraction, and DMEM, respectively, for different time periods to assay the proliferation, mRNA expression of alkaline phosphatase (ALP), Cbfa1, Collagen type I (Col-I) and osteocalcin (OCN), as well as ALP staining, activity and matrix mineralisation.RESULTS:In the nano-58S group, cell proliferation and mRNA expression of ALP, Cbfa1 and OCN were significantly enhanced in a time-dependent manner compared with the control group. mRNA expression of Cbfa1 on day 4 and OCN on day 7 was significantly higher than that in the 58S group. Moreover, there was significantly more ALP protein expression and mineralisation in the nano-58S group than in the 58S group.CONCLUSION:The nano-58S enhanced proliferation, osteogenic markers expression in MG-63 cells and induced stronger mineralization than 58S.
Mesoporous Bioactive Glass Promoted Osteoblast Proliferation and Differentiation in Vitro

Abstract:This study aimed to evaluate the bioactivity and cytocompatibility of mesoporous bioactive glass. The structure and components of mesoporous bioactive glass 80S MBG were characterized. Mouse osteoblasts were isolated and treated with 80S MBG. The viability and alkaline phosphatase activity of osteoblasts were evaluated. Hydroxyapatite was at the surface of 80S MBG after 4 h, and Ca, P and Si concentration rapidly reached the peak within 1-4 h. pH change was very slight. Mouse osteoblasts seeded in the flask could adhere to the surface and maintain normal morphology. The extract of 80S MBG promoted the proliferation of osteoblasts at lower concentrations, and promoted the differentiation of osteoblasts at higher concentrations. The toxicity gradation of 80S MBG was 0-1. In conclusion, 80S MBG has higher biological activity and good cell compatibility in vitro. The influence for body fluid microenvironment is mild. 80S MBG could be used as a substitute material or scaffold in bone tissue engineering.
Promotive effect of a new type of mesoporous bioactive glass on proliferation and differentiation of human osteoblasts

Abstract:Objective:To investigate the effect of a new type of mesoporous bioactive glass with a hierarchical pore structure on the proliferation and differentiation of osteoblasts,and to clarify its osteogenetic differentiation effect.Methods:The MBG structure was observed by scanning electron microscope (SEM);the MG63 cells at logarithmic growth phase were selected and divided into MBG group (cultured with MBG),NBG group (cultured with NBG)and blank control group (cultured with blank cultrue medium),and they were cultured for 1 , 3 , and 5 d.The proliferation rate of MG63 cells was analyzed by MTT method;enzyme-linked immunoassay method was used to analyze the alkaline phosphates (ALP)activity in MG63 cells;real-time PCR was used to detect the relative expression levels of Sp7 mRNA and Runx2 mRNA in MG63 cells after cultured for 24 h.Results:The SEM results showed that the MBG had porous scaffold structure. Compared with blank control group and NBG group, the proliferation rates of MG63 cells in MBG group at 1, 3, and 5 d were significantly increased (P<0.01 ). Compared with blank control group and NBG group,the activities of ALP in the MG63 cells in MBG group at 1,3, 5 d were significantly increased (P<0.01 ).Compared with blank control group and NBG group, the relative expression level of Sp7 mRNA in the MG63 cells in MBG group was significantly increased 24 h after culture (P<0.01),while the relative expression level of Runx2 mRNA in the MG63 cells in MBG group had no significant differences (P > 0.05 ). Conclusion:Compared with NBG, the MBG presents the promotive effect on the proliferation and differentiation of osteoblasts invitro with low cytotoxicity.

Abstract:Objective Mesoporous bioactive glass (MBG) has good biocompatibility without immune reaction after implanting into tissue as biomaterial which was used in the treatment of bone defect. Genistein (G), a phytoestrogen, could be used in the treatment of osteoporosis in postmenopausal women. Results Here, we report that MBG with large pores (MBG-L) and MBG-L adsorbed with G (MBG-L/G) sustained-release G could enhance osteoblast differentiation and matrix mineralization. Interestingly, we observed that MBG-L enhanced the formation of bone-like deposit and Ca deposition in vitro. In the other side, we also found that MBG-L/G substrate could promote osteoblast differentiation and matrix mineralization through Erk activated Runx2 pathway. Interestingly, the expression of osteoblast-specific marker gene Osteopontin (Opn) was also increased in MC3T3-E1 cells cultured on MBG-L/G substrate. Conclusions We conclude that MBG-L/G is a potential biomaterial for the treatment of bone defect.
Effect of the Nanostructure of Porous Alumina on Growth Behavior of MG63 Osteoblast-Like Cells.

Abstract:It is well known that cellular responses to materials, in terms of adhesion, migration and proliferation, are highly affected by the surface characteristics of the materials. The investigation of the effect of material surface topography on cell behaviors is of great importance for the development of implanted biomaterials in tissue engineering. Alumina is one of the most popular implant materials used in orthopedics, but few data are available concerning the potential cellular responses of MG63 to nanoporous alumina. The present study investigated the size effect of nanoporous alumina substrates on MG63 cell behaviors in terms of cell viability, expression of integrin β1, alkaline phosphatase (ALP) activity and changes of cell morphology, respectively. Cell viability was measured by means of MTT assay and integrin β1 expression was detected by immunofluorescence staining and real-time PCR. Scanning electron microscopy (SEM) was used to observe cell morphology. Cell function was evaluated by detecting the ALP activity and mineralization. Results showed that cell viability and expression of integrin β1 were decreased with the increasing pore size, however, the increasing pore size of the alumina resulted in elongated cell morphology, enhanced ALP activity and mineralization. This study showed that the surface topography of nanoporous alumina plays an important role in regulating the behaviors of MG63 osteoblast-like cells and porous alumina can be regarded as useful substrate in tissue engineering.
Biocompatibility and Osteogenesis of Micro-and Nano-Bioactive Glasses

Abstract:Because of the special morphology and characterization,micro-and nano-bioactive glass caused much attention by researchers,but the morphology of micro-and nano-bioactive glass on cell performance has been underappreciated and underinvestigated.The special morphology of micro-and nano-bioactive glass was synthesized by the Sol-gel method combined with the template bionic.Human mesenchymal stem cells(hMSCs) were seeded on bioactive glass for 6 days and 10 days in vitro for alkalic phosphatase(ALP) activity and 5days for osteogenic gene expression analysis.Based on the results of ALP activity and osteogenic gene expression analysis,the responses of hMSCs to the SBG exhibited a higher degree of osteogenic differentiation than those on IBG in vitro.This work will provide a reference for design of specific morphology of the bioactive glass in the future.

Abstract:The effects in vitro of a novel multiphase glass-ceramic (with nominal composition of 43.19% CaO, 7.68% MgO, and 49.13% SiO2 in weight percent) on cell adhesion, proliferation, differentiation and ultrastructure of human osteosarcoma cell line MG63, mouse fibroblasts L929, and human lung adenocarcinoma epithelial cell line A549 were investigated in this research. Scanning electron microscopy (SEM) micrographs revealed that the surface morphology of this glass-ceramic was beneficial to cell adhesion. The glass-ceramic extracts at certain concentrations could stimulate the proliferation and differentiation of MG63 and L929 cells, whereas inhibit A549 proliferation, which might be resulted from the released Si ions. In addition, when cultured with 0.1 mg/mL glass-ceramic powder suspension, the cell ultrastructure of MG63 showed abundant organelles and L929 displayed the phenomena of cellular stress response. While more interestingly, A549 exhibited chromatin condensation, mitochondria swell and RER expansion, which was presumed to be early signs of apoptosis. These results suggest that this novel CaO-MgO-SiO2-based multiphase glass-ceramic has potential for bone regeneration and tissue engineering applications. (C) 2013 Elsevier B.V. All rights reserved.

Abstract:The bio-mineralization properties of the melt-derived bioglass 45S5 and the sol-gel derived bioglass 58S and 77S in vitro were investigated. The effect of ionic products from bioactive glass dissolution on osteoblast proliferation and ALP activity was also discussed. The results show that osteoblasts cultured with 77S conditioned DMEM show better cellular viability and ALP activity when compared to 45S5 and 58S. though the bio-mineralization of 77S shows a little worse-only HA can form from 77S when HCA can form from 45S5 and 58S. By comparing the Si concentrations in bioglass conditioned DMEM and control DMEM. it can be concluded that solutions containing high Si concentrations in certain range are mitogenic for bone cells.

Abstract:The bioactivity of coatings on alloy implants plays critical roles in the fast osteointegration and maintenance of a long‐term life span of the implants. The aim of this study is to incorporate nanopores and bioactive components into the plasma‐sprayed coatings by constructing nanopores and bioactive elements on the surface of coatings and further improve their in vitro bone‐forming activity. In this study, mesoporous bioactive glasses (MBG) were used for modifying the surface of ZrO2 coatings on Ti‐6Al‐4V by spin‐coating methods. The result shows that MBG nanolayer has been successfully coated on the surface of ZrO2 coatings. The MBG‐modified ZrO2 coatings have improved apatite‐mineralization ability and stable bonding strength. In addition, the attachment, proliferation, alkaline phosphate activity, and bone‐related gene expression of rabbit bone marrow stromal cells on MBG‐ZrO2 coating were also enhanced as compared with conventional bioactive glass‐modified ZrO2 coatings and pure ZrO2 coatings. The results indicated that MBG‐modified ZrO2 coatings on Ti alloy are promising for orthopedic application.
Effect of Process Conditions on Microstructure and Properties of Bioactive Glass

Abstract:The bioactive glass is used widely as bone-repairing material due to its excellent bioactivity, biocompatibility and the function for bone restoration, which depended, to a great extent, on the composition, microstructure of the glass and the process conditions. In this study, effect of process conditions (the melting temperature and time) on compositions, microstructure and the properties of bioactive glass was analyzed using EDS, Density measurement, XRD and FTIR techniques. The results indicated that compositions and microstructure changed because of the selective evaporation of constituent and the change in homogeneity of components.The bioactivities of the glasses prepared under different process conditions were characterized by immersing the glass particles in simulated body fluid (SBF) combined with FTIR technique. The results indicated that hydroxyl-carbonate-apatite (HCA) layer formed on the surface of the samples after 24 hours soaking in SBF. A quantitative method was used to compare the bioactivity of samples. It indicated the different preparation processes had influence on the bio-mineralization properties. Then the optimal preparation condition (1450°C, 30min) was obtained.

Abstract:It is important to understand the interaction mechanisms between nanomaterials and adipose-derived stem cells for biomedical application. Nanoscale bioactive glass has positive effects on guiding osteoblasts differentiation and bone regeneration. However, the effects and molecular mechanism of monodispersed bioactive glass nanoparticles on the osteogenic differentiation of adipose-derived stem cells are still not clear up to now. In this study, the effects and underlying molecular mechanism of monodispersed bioactive glass nanoparticles on the osteogenic differentiation of adipose-derived stem cells are investigated in minute detail. The results show that nanoparticles (100-200 nm) can be absorbed by stem cells and is distributed in cytoplasm and nucleus. In both culture conditions (normal and osteoinductive), nanoparticles (80 mu g mL(-1)) can significantly enhance the osteogenic differentiation of stem cells through upregulating the alkaline phosphatase activity, osteogenic genes and protein expressions, as well as calcium deposition. Further study suggests that the activation of transforming growth factor-beta/Smad3 signaling pathway plays an important role in the osteogenic differentiation of adipose-derived stem cells enhanced by monodispersed nanoparticles. This study may have important implications for better understanding of stem cells fate induced by monodispersed nanoparticles and provide a promising approach toward stem cells-based bone regeneration.
[Effects of a Novel Borate Bioglass on Osteoblast Behavior in Vitro].

Abstract:OBJECTIVE:As a bioactive material, the osteogenic activity of borate bioglass has been proved. To design a novel borate bioglass according to an improved formula and to investigate the effects of the borate bioglass on osteoblasts in vitro for further research and potential clinical application. METHODS:The novel Na2O-K2O-MgO-CaO-P205-B203-SrO borate bioglass was prepared by melting process. The initial and secondary extracts were prepared according to ISO10993-12: 2007 respectively with different extract time of 0-24 hours and 24-48 hours. The osteoblasts (MC3T3-E1) of the 5th-15th passages from mouse were cocultured with the initial (initial extract group) and secondary (secondary extract group) extracts, respectively, to assess the effects of the borate bioglass on the cell proliferation, protein synthesis, alkaline phosphatase (ALP) activity, cell apoptosis, and cell migration; while alpha-MEM medium without addition of extract served as control group. RESULTS:The absorbance values at 450 nm were 0.3560 +/- 0.0187, 0.331 0 +/- 0.025 4, and 0.2040 +/- 0.0138 in initial extract, secondary extract, and control groups, respectively, showing significant differences among 3 groups (P < 0.05). The total protein contents were (382.847 +/- 9.521), (226.071 +/- 5.847), and (220.248 +/- 8.213) U in initial extract, secondary extract, and control groups, respectively; there were significant differences between initial extract group and control group, and between initial extract group and secondary group (P < 0.05), but there was no significant difference between secondary extract group and control group (P > 0.05). However, no significant difference was observed in the ALP activity [(0.01301 +/- 0.00039), (0.012 93 +/- 0.00044), and (0.012 92 +/- 0.000 35) U/mg], apoptosis rate (7.03% +/- 1.95%, 6.46% +/- 2.88%, and 6.18% +/- 2.21%), horizontal migration [(137.50 +/- 11.43), (134.98 +/- 10.50), (135.21 +/- 8.66) microm], and transmembrane cell number [(10.92 +/- 4.99), (10.07 +/- 2.50), and (9.81 +/- 2.64) cells/field] among initial extract, secondary extract, and control groups (P > 0.05). CONCLUSION:This novel borate bioglass has excellent cytocompatibility, which plays regulatory effects on the cell proliferation, secretion, and migration.
The Effect Of Bioactive Glass Ceramics On The Expression Of Bone-Related Genes And Proteins In Vitro

Abstract:Using biodegradable bone substitutes in alveolar ridge augmentation avoids second-site surgery for autograft harvesting. Considerable efforts have been undertaken to develop rapidly resorbable bone substitute materials with a higher degree of biodegradability than tricalcium phosphate (TCP). This study examines the effect of novel biodegradable glass ceramics on the expression of bone-related genes and proteins by human bone-derived cells (HBDC) and compares this behavior with that of TCP. Test materials used were alpha-TCP, a surface-treated glass ceramic GB9N with crystalline phase Ca2KNa(PO4)(2) and a small amount of amorphous silica phosphate; AP40 - a glass ceramic based on crystalline phases of apatite and wollastonite; and a glass ceramic Mg5 composed of 20.6% CaO, 58.5% P2O5, 14.4% Na2O, 4.1% MgO and 2.4% CaF2 (wt%). HBDC were grown on the substrata for 3, 5, 7, 14 and 21 days, counted and probed for various bone-related mRNAs and proteins (type I collagen (Col I), osteocalcin (OC), osteopontin (OP), osteonectin (ON), alkaline phosphatase (ALP) and bone sialoprotein (BSP)). The substrata supported continuous cellular growth for 21 days. By day 21, GB9N had the highest number of HBDC. GB9N induced significantly enhanced expression of Col I, ALP, OP, OC and ON mRNA at 3 days; of OP, OC and ON mRNA and protein at 7 and 14 days; and of ALP, OP and OC mRNA and Col I, ALP, BSP, ON and OP protein at 21 days. Since all novel glass ceramics supported cellular proliferation together with expression of bone-related genes and proteins at least as much as TCP, these ceramics can be regarded as potential bone substitutes. GB9N had the most effect on osteoblastic differentiation, thus suggesting that this material may possess a higher potency to enhance osteogenesis than TCP.
Size Control And Biological Properties Of Monodispersed Mesoporous Bioactive Glass Submicron Spheres

Abstract:We report a facile method for fabricating monodispersed mesoporous bioactive glass sub-micron spheres (MBGS) using dodecylamine (DDA) as a catalyst and template agent in a sol-gel process by self-assembly between a neutral organic surfactant and neutral inorganic precursor ((SI0)-I-0). Moreover, we investigate the effect of sub-micron particle size on the physicochemical properties, apatite-forming ability, and biocompatibility of MBGS. Results showed that all samples exhibited regular spherical morphology and favorable mono-dispersibility. The average particle diameters of MBGS (200-800 nm) were controlled by adjusting the concentration of DDA. All samples induced the formation of rod-like apatite precipitates, which closely resembled the natural nanoscale apatite crystal, showing their high apatite-forming ability. Furthermore, MBGS surfaces also supported the attachment and promoted proliferation of alkaline phosphatase (ALP) activity of MG-63 cells, showing the good biocompatibility of MBGS. MBGS-1 had the smallest particle size and it was found that it could significantly enhance MG-63 proliferation and differentiation because of the smaller particle size and higher specific surface area. It is hoped that this study may motivate the development and applications of submicron biomaterials for bone repair applications.
Monodispersed Β‐glycerophosphate‐decorated Bioactive Glass Nanoparticles Reinforce Osteogenic Differentiation of Adipose Stem Cells and Bone Regeneration in Vivo

Abstract:Design and development of highly bioactive nanoscale biomaterials with enhanced osteogenic differentiation on adipose stem cells is rather important for bone regeneration and attracting much attention. Herein, monodispersed glycerophosphate‐decorated bioactive glass nanoparticles (BGN@GP) are designed and their effect is investigated on the osteogenic differentiation of adipose mesenchymal stem cells (ADMSCs) and in vivo bone regeneration. The surface‐modified BGN@GP can be efficiently taken by ADMSCs and shows negligible cytotoxicity. The in vitro results reveal that BGN@GP significantly enhances the alkaline phosphatase activity and calcium biominerialization of ADMSCs either under normal or osteoinductive medium as compared to BGNs. Further studies find that the osteogenic genes and proteins including Runx2 and Bsp in ADMSCs are significantly improved by BGN@GP even under normal culture medium. The in vivo animal experiment confirms that BGN@GP significantly promotes the new bone formation in a rat skull defect model. This study suggests that bioactive small molecule decorating is an efficient strategy to improve the osteogenesis capacity of inorganic ceramics nanomaterials.

Abstract:In orthopedic surgery, bioactive glass film coating is extensively studied to improve the synthetic performance of orthopedic implants. A lot of investigations have confirmed that nano-porous structure in bioactive glasses can remarkably improve their bioactivity. Nevertheless, researches on preparation of nano-porous bioactive glasses in the form of film coating and their cell response activities are scarce. Herein, we report the preparation of nano-porous bioactive glass film on commercial glass slide based on a sol-gel technique, together with the evaluation of its in vitro bioactivity through immersion in simulated body fluid and monitoring the precipitation of apatite-like layer. Cell responses of the samples, including attachment, proliferation and osteogenic differentiation, were also investigated using BMSCS (bone marrow derived mesenchymal stem cells) as a model. The results presented here provide some basic information on structural influence of bioactive glass film on the improvement of bioactivity and cellular effects. (C) 2013 Elsevier B. V. Abstract:BACKGROUND:As the bone substitute materials,the bioactive glasses with excellent bioactivity and biocompatibility are used to repair the bone defects.Especially,porous bioactive glasses are benefiicial to form bone in virtue of its high specific surface area and pore volume.OBJECTIVE:To utilize citric acid as a substitution of inorganic acid so as to synthesize self-assembled mesoporous-macroporous bioactive glass(MMBG).METHODS:Citric acid and P123 were dissolved in ethanol,and the mixture was continuously stirred for 2 hours.Tetraethyl orthosilicate,Ca(NO3)2·4H2O,and triethyl phosphate were then added to the solution and stirred for 24 hours,followed by putting the solution in a beaker for 7 days under static conditions.Finally,calcination was performed for removing the organic composition.Samples were evaluated by immersion studies in simulated body fluid.RESULTS AND CONCLUSION:The synthesized material had mesoporous-macroporous structure with high porosity and BET surface.The samples also showed good bone-forming activity.It is anticipated that the synthesized MMBG can be used in bone defect repairing and bone tissue engineering.

Abstract:Mesoporous bioactive glass (MBG) is performed as a bone tissue engineering material because of its good bioactivity, biocompatibility and osteoinducion characteristics. Here, we propose MBG with larger pores (MBG-L) adsorbed fibroblast growth factor (FGF) to facilitate osteoblast differentiation and matrix mineralization. Specifically, we observed that MBG-L promotes calcium deposit precipitation in vitro. In addition, adhesion, proliferation, differentiation and matrix mineralization were promoted after osteoblast cultured on MBG-L/FGF. Interestingly, we found that the transcriptional activity of the critical transcription factor Runx2 was increased through MAPK pathway after osteoblast cultured on MBG-L/FGF. Support for this result, we found that the expression of osteoblastic marker genes, Osteocalcin (Ocn), Osteopontin (Opn), and Runx2 were increased. Thus, our findings provided that MBG-L/FGF could be a promising new material in bone tissue engineering.

Abstract:In this paper, the boron‐containing mesoporous bioactive glass (MBG) nanospheres have been successfully synthesized by modified sol‐gel method assisted by surfactant, and the effect of boron substitution on structure and bioactivity was evaluated by combining experiments and ab initio molecular dynamics (AIMD) simulations. All of the samples exhibit regularly uniform mesoporous spherical microstructure with an average size about 60 nm, and the boron‐containing MBGs show higher specific surface area with the value up to 416.20 m2/g. The simulated body fluid (SBF) immersion test confirms that the deposited hydroxyapatite (HA) evidently increases with the increasing of boron content, indicating that the biological behavior has been significantly improved resulting from incorporation of boron. Additionally, our results also reveal that B2O3 substitution has positive impact on cell proliferation of human periodontal ligament cells (hPDLCs) at lower extracted concentration. Furthermore, AIMD simulation is employed to understand the relationship between structural changes and in vitro bioactivity in terms of structural information, especially the boron coordination number. The results illustrate that the boron‐containing MBG nanospheres with excellent bioactivity are great potential for biomedical applications.
