Abstract:Diblock and miktoarm star-shaped thermoresponsive copolymers composed of single-stranded DNA (ssDNA) and poly(N-isopropylacrylamide) were successfully synthesized with combination of atom transfer radical polymerization (ATRP) and click chemistry. This approach should be generalizable to other DNA-functionalized copolymers. Such copolymers self-assemble into spherical micelles with ssDNA corona in aqueous solution above the lower critical solution temperature. The micellar size can be tuned from the macromolecular architecture. These DNA-encoded micellar particles are able to encapsulate and release hydrophobic guest molecules upon changing temperature.

Abstract:We report a new type of thermo- and pH-responsive, coacervate-forming highly degradable polymer-hyperbranched poly(β-amino esters) (HPAEs) and its selective cell binding behaviors. The HPAEs were synthesized from 5-amino-1-pentanol (S5) and trimethylolpropane ethoxylate triacrylate (TMPETA) via an A2+B3 type Michael addition. The existence of multiple hydrogen bond pairs as well as tertiary amines makes the S5-TMPETA polymers manifest temperature- and pH-dependent phase transition. By varying the length of the ethylene glycol (EG) spacers in the TMPETA, polymer molecular weight, concentration, and pH value, the phase transition of the S5-TMPETA can be easily tuned in aqueous and buffer solutions, as evidenced by UV-vis spectroscopy and DLS measurements. Especially, the S5-TMPETA prepared from S5 and trimethylolpropane ethoxylate triacrylate 692 (S5-TMPETA692) shows a lower critical solution temperature (LCST) around 33 °C, above which the S5-TMPTEA can form coacervate particles able to encapsulate functional molecules effectively. Importantly, when incubation with HeLa cells, the S5-TMPTETA692 exhibits a temperature- and pH-responsive selective cell binding behaviors. In addition, the S5-TMPETA are highly hydrolyzable and elicit negligible cytotoxicity. This new type of "smart" polymer should find use in a variety of biomedical applications.

Abstract:A type of thermosensitive ionic microgel was successfully prepared via the simultaneous quaternized cross-linking reaction during the surfactant-free emulsion copolymerization of N-isopropylacrylamide (NIPAm) as the main monomer and 1-vinylimidazole or 4-vinylpyridine as the comonomer. 1,4-Dibromobutane and 1,6-dibromohexane were used as the halogenated compounds to quaternize the tertiary amine in the comonomer, leading to the formation of a cross-linking network and thermosensitive ionic microgels. The sizes, morphologies, and properties of the obtained ionic microgels were systematically investigated by using transmission electron microscopy (TEM), dynamic and static light scattering (DLS and SLS), electrophoretic light scattering (ELS), thermogravimetric analyses (TGA), and UV-visible spectroscopy. The obtained ionic microgels were spherical in shape with narrow size distribution. These ionic microgels exhibited thermosensitive behavior and a unique feature of poly(ionic liquid) in aqueous solutions, of which the counteranions of the microgels could be changed by anion exchange reaction with BF4K or lithium trifluoromethyl sulfonate (PFM-Li). After the anion exchange reaction, the ionic microgels were stable in aqueous solution and could be well dispersed in the solvents with different polarities, depending on the type of counteranion. The sizes and thermosensitive behavior of the ionic microgels could be well tuned by controlling the quaternization extent, the type of comonomer, halogenated compounds, and counteranions. The ionic microgels showed superior swelling properties in aqueous solution. Furthermore, these ionic microgels also showed capabilities to encapsulate and release the anionic dyes, like methyl orange, in aqueous solutions.

Abstract:We report on a facile method for fabricating thermosensitive organic/inorganic hybrid hydrogel thin films from a cross-linkable organic/inorganic hydrid copolymer, poly[ N-isopropylacrylamide- co-3-(trimethoxysilyl)propylmethacrylate] [P(NIPAm- co-TMSPMA)]. Fourier transform infrared (FT-IR) spectra confirmed the formation of hybrid hydrogel thin films after hydrolysis of the methoxysilyl groups (Si-O-CH 3) and subsequent condensation of the silanol groups (Si-OH). Atomic force microscopy (AFM) images revealed that the surface morphology of the hydrogel thin films depended on the supporting substrates. Microdomains were observed for the hydrogel thin films on a gold surface, which can be attributed to inhomogeneous network structures. The thermoresponsive swelling-deswelling behavior and the viscoelastic properties of the hydrogel thin films were investigated as a function of temperature (25-45 degrees C) by using a quartz crystal microbalance (QCM) operated in water. The high frequency shear modulus of the P(NIPAm- co-TMPSMA) hydrogel thin films was several hundred kilopascals.

Abstract:Temperature-responsive hydrogels, otherwise known as thermogels, are promising biomaterials which undergo phase change from liquid to a solid-like gel state when warmed. Although poly (ethylene glycol) (PEG) is employed extensively as the hydrophilic segment in thermogelling polymer design, recent years has witnessed development of new families of PEG-free thermogelling copolymers which offer new opportunities in expanding material property profiles for new biomedical applications. Herein, we study the supramolecular self-assembly process of PEG-free amphiphilic branched graft copolymer containing polyethylenimine (PEI) and poly (propylene glycol) (PPG), which shows both thermogelling ability and pH-responsive gelation behaviour. Additionally, we demonstrate the ease of post-synthetic modulation of thermogel properties by amine functionalisation on the PEI copolymer segments through convenient nucleophilic substitution and amide formation reactions. The synthetic platform offered by PEI-PPG thermogels allow facile customisation of these versatile materials for targeted biomedical applications.

Abstract:It is a challenging work to design novel responsive materials by controlling the secondary structure of polypeptide, especially β-sheet. We designed and synthesized a novel dual responsive diblock copolymer of methyl poly (ethylene glycol)-b-poly (O-benzyl-l-threonine) (mPEG-b-PBnLT). The copolymer solutions exhibited unusual and reversible gel-to-sol-to-gel transition behavior with temperature, and the transition temperature window could be easily adjusted by changing copolymer concentration or PBnLT block length. It was confirmed that low temperature gel-to-sol transition was due to the disassembly of the preliminary β-sheet layered nano-assemblies that induced the transformation of self-organized morphology from nanosized fibrils to spherical aggregates, while the high temperature sol-to-gel transition was ascribed to the dehydration of PEG segments. The disassembly of β-sheet structures was caused by the change in the polarity of the pendant benzyl ether bond of PBnLT block in water with temperature. This stimuli-responsiveness of secondary self-assembled nanostructures and the corresponding change in macroscopic phase transition inspires a brand-new strategy for structural design and functional control of polypeptide materials.

Abstract:Poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) (PEO-PPO-PEO) triblock copolymer (Pluronic F127) was modified by introducing poly(N-isopropylacrylamide) (PNIPAM) at both the PEO ends, and the pentablock copolymer (PNIPAM(41)-F127-PNIPAM(41), PN41) so prepared was characterized using gel permeation chromatography and H-1 NMR spectroscopy. The degree of polymerization of NIPAM blocks at the two ends was 41. The solution behaviour and microstructure of PN41 aggregates in water were examined using UV-visible spectroscopy, micro-differential scanning calorimetry and small-angle neutron scattering (SANS) and compared with F127. Two lower critical solution temperatures (LCSTs) were observed for the pentablock copolymer, corresponding to PPO and PNIPAM blocks, respectively. The adsorption of PN41 on thiol-grafted hydrophobic gold surfaces at various temperatures was investigated using a quartz crystal microbalance. It was found that the adsorption behaviour and mechanism of PN41 were mainly determined by the interactions of the pentablock copolymers with different chain conformations in dilute aqueous solutions at various temperatures. SANS measurements were used to determine the temperature-dependent structural evolution of polymer micelles in aqueous solution. A NOESY study revealed that above the LSCT of PNIPAM, the interaction of PPO and PNIPAM protons increases and the distance between PPO and PNIPAM decreases. (c) 2019 Society of Chemical Industry

Abstract:Supramolecular polymers are usually thermomechanically unstable, as their mechanical strength decrease drastically upon heating, which is a fatal shortcoming for their applications. Herein, inspired by heat shock proteins (HSPs) which enable living organisms to tolerate lethal high temperatures, we design an HSPs-like response to impart supramolecular elastomer with high thermomechanical stability. The HSPs-like response relies on the reversible hydrolysis of boronic acid and the tunable association strength of boron dative bonds. As the temperature increases, the boronic acid dehydrates and transforms into boroxane. The boroxane, acting as a heat shock chemical, prevents the disintegration of the supramolecular network through formation of multi and stronger dative bonds with imidazole-containing polymers, thereby enabling the material to retain its mechanical strength at high temperatures. Such chemical transformation and network change induced by the HSPs-like response are fully reversible during the heating and cooling processes. Moreover, due to the dynamic nature of the supramolecular network, the elastomer possesses recycling and self-healing abilities.

Abstract:Biocompatible and biodegradable thermoresponsive polymers are of great interest in "smart" biomedical applications. Herein, triblock copolymers (6a and 6b) comprised of hydrophobic poly(propylene glycol) (PPG) and OEGylated polypeptides bearing Y-shaped or linear pendants are prepared by ring-opening polymerization of reactive N-carboxyanhydride monomers and subsequent side chain modification of polypeptides. OEGylated triblock copolymer bearing Y-shaped (6a) or linear pendants (6b) with similar main chain length and the same amount of oligo(ethylene glycol) (OEG) units are successfully prepared as verified by H-1 NMR, FTIR, and GPC. CD analysis reveals that 6a with Y-shaped OEG pendants shows thermal- or salt-induced destabilization of alpha-helical conformation in water. Both 6a and 6b show reversible lower critical solution temperature (LCST)-type phase transitions in DI H2O or PBS. Incorporation of a PPG segment leads to a significant decrease of the LCST-type cloud point temperature (T-cp). Consequently, 6a and 6b show T-cp to human body temperature at low polymer concentrations (<= 0.5 mg mL(-1) in PBS). Moreover, 6a and 6b show low in vitro cytotoxicity and biodegradability in the presence of proteinase K. The results indicate that the thermoresponsive properties of LCST-type polymers can be readily tuned by constructing Y-shaped pendants as well as incorporation of hydrophobic polymer building blocks.
Biocompatible Thermoresponsive Polymers with Pendent Oligo(Ethylene Glycol) Chains and Cyclic Ortho Ester Groups

Abstract:Two acrylate monomers with six-member cyclic ortho ester groups, i.e., 2-(1,3-dioxan-2-yloxy)ethyl acrylate (DEA) and 2-(5,5-dimethyl-1,3-dioxan-2-yloxy) ethyl acrylate (DMDEA), were synthesized. These two monomers were copolymerized with an oligo(ethylene glycol) acrylate (OEGA) under atom transfer radical polymerization conditions to afford two series of thermoresponsive copolymers, poly(DEA-co-OEGA)s and poly(DMDEA-co-OEGA)s. All the copolymers were soluble in water to form transparent solutions at low temperature, however, some of them exhibited association behaviors below the cloud point (CP) as evidenced by (HNMR)-H-1, dynamic light scattering (DLS) and fluorescence method. The aggregation tendency of the copolymers depends on their composition as well as the structure of the ortho ester units. With a similar composition, poly(DMDEA-co-OEGA) showed a stronger aggregation tendency than poly(DEA-co-OEGA). In addition, increasing molar content of the ortho ester units in a copolymer promoted its aggregation in water. Thermally induced phase transitions of these copolymers were studied by various methods including turbidimetry, temperature-dependent (HNMR)-H-1, DLS, and microscopy. The results indicate that CP of the copolymers increased with increasing the content of OEGA units, but the changing behaviors of CP were rather different for two types of copolymers, which can be ascribed to the difference in hydrophobicity of the ortho ester units. Little hysteresis was observed for the copolymers with more OEGA units while those with more alto ester units showed significant hysteresis probably due to the hydrophobic character of the ortho ester. The formation of coacervate droplets above CP reveals that the copolymers underwent a liquid liquid phase separation upon heating. pH-dependent hydrolyses of the copolymers were studied by turbidimetry and H-1 NMR methods. The hydrolysis rate depends greatly on pH and the hydrophilic/hydrophobic balance of the copolymers: lower pH and more hydrophilic character resulted in a faster hydrolysis rate. Finally, this type of acid-labile thermoresponsive copolymers and the acid-catalyzed hydrolysis products have low cytotoxicity.

Abstract:An intelligent and biodegradable hydrogel film was prepared via polymerization of a well-designed macromer, and in situ encapsulation of model protein drugs in the hydrogel was performed in this paper. Hydrogels were prepared by polymerization of a synthetic macromer, in which the central part is a temperature-sensitive triblock copolymer composed of poly(ethylene oxide)–poly(propylene oxide)–poly(ethylene oxide), the extension along each end is biodegradable oligo (ε-caprolactone) (oligoCL) terminated with an acrylate group. The biodegradability of the resultant hydrogels was confirmed. The swelling/deswelling experiments revealed the negative and reversible temperature-sensitivity of the hydrogels. The volume phase transition temperature is between the refrigerator temperature and human body temperature. Four model protein drugs (insulin, lysozyme, SOD and BSA) were encapsulated into the hydrogel via in situ polymerization of macromer aqueous solutions mixed with proteins. Proteins were found to be controlled released and the release rate was strikingly influenced by temperature. The in vitro release profiles of proteins of different molecular weights (MW) were measured. The half release time is found to be positively related to the molecular weight of examined model proteins. The novel chemically crosslinked hydrogels might be prospective as controlled-release carriers of biomacromolecular drugs.

Abstract:A novel thermosensitive amphiphilic copolymer of PNIPAm grafted polyphosphazene (PNIPAm-g-PPP) has been synthesized through a co-substitution reaction of poly(dichlorophosphazene) with amino terminated NIPAm oligomer and glycine ethyl ester (GlyEt). The polymer obtained had a molar ratio of 1:5.25 PNIPAm to GlyEt and had a lower critical solution temperature (LCST) near 30 degreesC. The formation of polymeric micelles was confirmed by fluorescence, dynamic light scattering and transmission electron microscopy measurements.

Abstract:A series of short intrinsically disordered polypeptide conjugated oligonucleotides (IDPOCs) were rationally developed and assembled into well-defined nanospheres. The nanospheres exhibited excellent reversible thermoresponsive regulation of their contraction and expansion. Furthermore, the nanospheres showed biocompatibility, drug encapsulation and effective cellular uptake.

Abstract:A simple methodology for the preparation of thermosensitive organic-inorganic hybrid hydrogels using silica nanoparticle-mediated polymer networks is described. A thermosensitive copolymer poly[N-isopropylacrylamide-co-(3-methacryloxypropyltrimethoxysilane)], (pNS), with reactive side chains (Si-O-CH3) was first synthesized by free radical polymerization using N-isopropylacrylamide (NIPAAm) and 3-methacryloxypropyltrimethoxysilane (MAPTS). Then, simple mixing of the aqueous solution of this copolymer with silica nanoparticle (SiP) suspensions at room temperature led to the formation of thermosensitive hybrid hydrogels cross-linked with silica nanoparticles (SiP) which did not require any other processing like washing for the removal of unreacted monomers and initiators. The effects of SiP content on gelation abilities, temperature-responsive behaviors, swelling and deswelling kinetics, and mechanical properties of the hydrogels were investigated. The results showed that transparent hybrid hydrogels with adjustable network structures were obtained within a few minutes to a couple of hours depending on the concentration of the copolymers and the silica nanoparticles. The hybrid hydrogels exhibited a lower critical solution temperature (LCST) of around 33 °C with no significant deviation from conventional poly(N-isopropylacrylamide) hydrogels; the LCST was not significantly affected by the concentration of silica nanoparticles (which in these systems serve as the cross-linkers). In addition, the hydrogels showed significantly large equilibrium swelling ratios, improved mechanical strength, and suitable deswelling behavior, which can easily be tuned by varying the composition of the hybrid hydrogels.

Abstract:Constructing noncovalent interactions has been a benign method to tune the stimuli responsivity and assembled structure of polymers in solution; this is essential for controlling the functions and properties of stimuli-responsive materials. Herein, we demonstrate a novel supramolecular strategy to manipulate the cloud point (T-cp) and assembled structure of thermoresponsive polymers in solution by using H-bonding interactions. We use poly(lactide-co-glycolide)-b-poly(ethylene glycol)-b- poly(lactide-co-glycolide) (PLGA-PEG-PLGA) as a model thermoresponsive polymer and functionalize its chain terminals by the self-complementary quadruple H-bonding motif, 2-ureido-4[1H]-pyrimidinone (UPy). UPy end functionalization and increasing PLGA block length decrease the T-cp of copolymer. Both UPy- and nonfunctionalized copolymers form the spherical micelles at low temperature. They undergo the intermicellar aggregation and form large compound micelles during heating; this thermally induced structural transition causes the presence of T-cp. Due to the UPy-UPy H-bonding interactions, UPy end functionalization leads to more copolymer chains to associate in one micelle, thus, enhancing the hydrodynamic, gyration radii, core size, as well as the packing density of PLGA in micelle core and grafting density of PEG on core-shell interface. The decreased T-cp of UPy-functionalized copolymer stemmed from the stronger intermicellar attractions at high temperature. Furthermore, UPyfunctionalized copolymers exhibit higher drug loading content, slower drug release rate, and better separation efficiency in removing the hydrophobic substances from water than PLGA-PEG-PLGA precursors.

Abstract:The poly(ethylene glycol)/poly(2-(N,N-dimethylamino)ethyl methacrylate) (PEG/PDMAEMA) double hydrophilic block copolymers were synthesized by atom transfer radical polymerization using mPEG-Br or Br-PEG-Br as macroinitiators. The narrow molecular weight distribution of PEG/PDMAEMA block copolymers was identified by gel permeation chromatography results. The thermosensitivity of PEG/PDMAEMA block copolymers in aqueous solution was revealed to depend significantly on pH, ionic strength, chain structure, and concentration of the block copolymers. By optimizing these factors, the cloud point temperature of PEG/PDMAEMA block copolymers can be limited within body temperature range (30-37 degrees C), which suggests that PEG/PDMAEMA block copolymers could be a good candidate for drug delivery systems. Thermoresponsive Polyphosphoester via Polycondensation Reactions: Synthesis, Characterization, and Self-Assembly

Abstract:Using a novel strategy, amphiphilic polyphosphoesters based on poly(oxyethylene H-phosphonate)s (POEHP) with different poly(ethylene glycol) segment lengths and aliphatic alcohols with various alkyl chain lengths were synthesized using polycondensation reactions. They were characterized by 1H NMR, 13C {H} NMR 31P NMR, IR, and size exclusion chromatography (SEC). The effects of the polymer structure on micelle formation and stability, micelle size, and critical micelle temperature were studied via dynamic light scattering (DLS). The hydrophilic/hydrophobic balance of these polymers can be controlled by changing the chain lengths of hydrophilic PEG and hydrophobic alcohols. A solubilizing test, using Sudan III, revealed that hydrophobic substances can be incorporated inside the hydrophobic core of polymer associates. Loading capacity depends on the length of alkyl side chains. The results obtained indicate that these structurally flexible polymers have the potential as drug carriers.

Hydrogen-Bonded Thermostable Liquid Crystalline Complex Formed by Biodegradable Polymer and Amphiphilic Molecules

Abstract:We report a simple method to prepare biodegradable polymer-amphiphile complexes by solution mixing of poly(propylene carbonate) (PPC) with octadecanoic acid (OA). The complexes were characterized by combination of thermogravimetry analysis (TGA), differential scanning calorimetry (DSC), wide-angle X-ray diffraction (WAXD), small-angle X-ray scattering (SAXS), polarizing optical microscopy (POM), transmission electron microscopy (TEM), and Fourier transform infrared spectroscopy (FT-IR). Compared with the amorphous PPC copolymer, the PPC-OA-x complexes show excellent thermal stability and form thermotropic liquid crystalline state without rigid mesogenic units. The corresponding mechanism has been proposed to elucidate the observed phenomena. The stabilization effect induced by hydrogen-bonding interactions between PPC and OA molecules is responsible for the thermostability and formation of liquid crystalline state. The present findings may extend the applications of such biodegradable aliphatic polycarbonate by improving its glass transition temperature and thermoplastic processability.
Preparation of a thermosensitive and biodegradable microgel via polymerization of macromonomers based on diacrylated Pluronic/oligoester copolymers

Abstract:A novel biodegradable and thermosensitive hydrogel microparticle was prepared via suspension polymerization of a kind of block copolymer macromonomers. According to the molecular design, the macromonomer is composed of a thermosensitive triblock copolymer poly(ethylene oxide)–poly(propylene oxide)–poly(ethylene oxide) (PEO–PPO–PEO) and two oligomers of biodegradable polyester such as oligo(lactic acid) or oligo(ε-caprolactone), and end-capped with acryloyl groups. Microgels were obtained by inverse suspension polymerization of the macromonomer aqueous droplets initiated by a redox initiator. Thermosensitivity and in vitro biodegradation of the resultant microgels were confirmed. The gel microparticles in an aqueous solution were swollen at low temperature and shrunken at high temperature (human body temperature). Degradation rate could be adjusted by controlling the composition and the degree of polymerization of oligoester. Thus, the microgels exhibit combinatory and tunable properties.
Preparation and Properties of UCST‐Type Thermoresponsive Polypeptide Bearing Amide Pendants

Abstract:Thermoresponsive polypeptides bearing amide pendants with an upper critical solution temperature (UCST) under physiologically relevant solution conditions are developed. A series of poly(gamma-propanyl-l-glutamate)s bearing amide pendants (PPLG(n)-Am) with different molecular weight (or degree of polymerization, DP) is prepared by copper mediated 1,3-dipolar cycloaddition. While fourier transform infra-red and CD analysis reveal random coil conformation of the PPLG(n)-Am in the solid-state and in aqueous solutions, respectively, PPLG(n)-Am shows reversible UCST-type phase behaviors in deionized water (DI) water and phosphate-buffered saline. Variable temperature UV-vis spectroscopy indicates that the UCST-type cloud point temperature (T-cp) in the range of 25-75 degrees C can be readily tuned by molecular weight (or DP), polymer concentration, and incorporation of hydrophobic (i.e.,n-hexyl) pendants. Moreover, PPLG(n)-Am is readily degraded by proteinase K within 48 h. The results indicate that UCST-type thermoresponsive and biodegradable polypeptides in aqueous solutions can be constructed by conjugation of amide pendants which shall provide guidance for future UCST polymer designs.
