Abstract:Abstract Ultra-deep three-dimensional (3D) optical coherence tomography (OCT) in turbid media is reported. Parallel detection along depth combined with scanning detection across lateral field is exploited in a swept-source OCT system to obtain the depth-resolved reflection matrices. Scanning detection enables the use of a highspeed swept source and a matched detector providing the required depth range for ultra-deep imaging. Disturbances intrinsic to scanning detection are compensated with negligible intra-frame phase variations to ensure satisfactory reconstruction by the inter-frame phase insensitive algorithm. 3D OCT imaging with an ultra-deep imaging depth of 11.5 times the scattering mean-free-path and a triple extension of the imaging depth achieved by a conventional SS-OCT is realized. The measurement time for 3D imaging (125μm×125μm×4mm ) is ~5 min, a significant leap over the time-domain matrix OCT where 6 min is taken for only one specified depth.

Abstract:As a label-free and quantitative imaging technique, optical diffraction tomography has been widely used in biological imaging. However, it is typically limited to weakly-scattering objects. To overcome this limitation, optimization algorithms based on minimizing field differences at the exit/observation plane, including total variation regularization, have been proposed and demonstrated. We propose a novel optimization algorithm to generalize field discrepancies from one plane to multiple planes throughout the scattering area. We numerically demonstrate that minimizing the field discrepancies at multiple planes instead of only one plane improves the robustness and accuracy of reconstructing multiply-scattering objects, without sacrificing the computational efficiency.

Abstract: Conventional approaches for 3D imaging in or through scattering media are usually limited to 2D reconstruction of objects at some discontinuous locations, although the time-consuming iteration, guide-star, or complex system are implemented. How to quickly visualize dynamic 3D objects behind scattering media is still an open issue. Here, by using structured light illumination, we propose a single-shot technique that can quickly acquire continuous 3D surfaces of objects hidden behind the diffuser. The proposed method can realize the 3D imaging of single, multiple, and dynamic targets from the speckled structured light patterns under broad or narrow band light illumination, in which only once calibration of the imaging setup is needed before conducting the imaging. Our approach paves the way to quickly visualize dynamic objects behind scattering media in 3D and multispectral.

Abstract:Multiple scattering of waves in disordered media is a nightmare whether it be for detection or imaging purposes. The best approach so far to get rid of multiple scattering is optical coherence tomography. It basically combines confocal microscopy and coherence time-gating to discriminate ballistic photons from a predominant multiple scattering background. Nevertheless, the imaging depth range remains limited to 1 mm at best in human soft tissues. Here we propose a matrix approach of optical imaging to push back this fundamental limit. By combining a matrix discrimination of ballistic waves and iterative time-reversal, we show both theoretically and experimentally an extension of the imaging-depth limit by at least a factor two compared to optical coherence tomography. In particular, the reported experiment demonstrates imaging through a strongly scattering layer from which only one reflected photon over 1000 billion is ballistic. This approach opens a new route towards ultra-deep tissue imaging.

Abstract:Light scattering imposes a major obstacle for imaging objects seated deeply in turbid media, such as biological tissues and foggy air. Diffuse optical tomography (DOT) tackles scattering by volumetrically recovering the optical absorbance and has shown significance in medical imaging, remote sensing and autonomous driving. A conventional DOT reconstruction paradigm necessitates discretizing the object volume into voxels at a pre-determined resolution for modelling diffuse light propagation and the resulting spatial resolution of the reconstruction is generally limited. We propose NeuDOT, a novel DOT scheme based on neural fields (NF) to continuously encode the optical absorbance within the volume and subsequently bridge the gap between model accuracy and high resolution. Comprehensive experiments demonstrate that NeuDOT achieves submillimetre lateral resolution and resolves complex 3D objects at 14 mm-depth, outperforming the state-of-the-art methods. NeuDOT is a non-invasive, high-resolution and computationally efficient tomographic method, and unlocks further applications of NF involving light scattering.

Abstract:In diffuse optical tomography (DOT), researchers often face challenges to accurately recover the depth and size of the reconstructed objects. Recent development of the Depth Compensation Algorithm (DCA) solves the depth localization problem, but the reconstructed images commonly exhibit over-smoothed boundaries, leading to fuzzy images with low spatial resolution. While conventional DOT solves a linear inverse model by minimizing least squares errors using L2 norm regularization, L1 regularization promotes sparse solutions. The latter may be used to reduce the over-smoothing effect on reconstructed images. In this study, we combined DCA with L1 regularization, and also with L2 regularization, to examine which combined approach provided us with an improved spatial resolution and depth localization for DOT. Laboratory tissue phantoms were utilized for the measurement with a fiber-based and a camera-based DOT imaging system. The results from both systems showed that L1 regularization clearly outperformed L2 regularization in both spatial resolution and depth localization of DOT. An example of functional brain imaging taken from human in vivo measurements was further obtained to support the conclusion of the study.

Abstract:Optical tomography is a kind of noninvasive technology in CT field using multiple photon diffusion. With the method of Monte Carlo simulation we carried out some numerical computation of the diffusion transmission disciplinarian in two kinds of simple biological texture. A ring distribution of diffuse light intensity is found and is sensitive to the incident light relative position to the absorption sphere at some certain conditions. The resolution and incident depth are also discussed through analysis of the image in numeric simulation.

Abstract:Light scattering by tissue severely limits how deep beneath the surface one can image, and the spatial resolution one can obtain from these images. Diffuse optical tomography (DOT) is one of the most powerful techniques for imaging deep within tissue -- well beyond the conventional $\sim$10-15 mean scattering lengths tolerated by ballistic imaging techniques such as confocal and two-photon microscopy. Unfortunately, existing DOT systems are limited, achieving only centimeter-scale resolution. Furthermore, they suffer from slow acquisition times and slow reconstruction speeds making real-time imaging infeasible. We show that time-of-flight diffuse optical tomography (ToF-DOT) and its confocal variant (CToF-DOT), by exploiting the photon travel time information, allow us to achieve millimeter spatial resolution in the highly scattered diffusion regime ($> 50 $ mean free paths). In addition, we demonstrate two additional innovations: focusing on confocal measurements, and multiplexing the illumination sources allow us to significantly reduce the measurement acquisition time. Finally, we rely on a novel convolutional approximation that allows us to develop a fast reconstruction algorithm, achieving a 100$\times$ speedup in reconstruction time compared to traditional DOT reconstruction techniques. Together, we believe that these technical advances serve as the first step towards real-time, millimeter resolution, deep tissue imaging using DOT.

Abstract:High-efficiency imaging through highly scattering media is urgently desired for various applications. Imaging speed and imaging quality, which determine the imaging efficiency, are two inevitable indices for any optical imaging area. Based on random walk analysis in statistical optics, the elements in a transmission matrix (TM) actually obey Gaussian distribution. Instead of dealing with large amounts of data contained in TM and speckle pattern, imaging can be achieved with only a small number of the data via sparse representation. We make a detailed mathematical analysis of the elements-distribution of the TM of a scattering imaging system and study the imaging method of sparse image reconstruction (SIR). More specifically, we focus on analyzing the optimum sampling rates for the imaging of different structures of targets, which significantly influences both imaging speed and imaging quality. Results show that the optimum sampling rate exists in any noise-level environment if a target can be sparsely represented, and by searching for the optimum sampling rate, it can effectively balance the imaging quality and the imaging speed, which can maximize the imaging efficiency. Abstract:Imaging through a dynamical opaque scattering medium is an almost impossible task, where strong multiple light scattering from moving scatters dynamically prevents imaging formations even with state-of-art techniques like correlation imaging or adaptive optics. Meanwhile, a small number of ballistic photons can still penetrate through but require demanding detection in terms of a ultrashort time gate and high sensitivity. However, visible light is strongly scattered for most of scattering media. Here we experimentally demonstrate a non-invasive coherent imaging scheme based on two-photon absorption capable of imaging through dynamical scattering media with a length equivalent to 28 times mean free paths for single photon transport, where two-photon absorption in a conventional semiconductor photodetector when phase matching is not required works over a wide bandwidth so it can support a fast time gate down to femtosecond level, short enough to distinguish ballistic photons from scattering background, and allows accessing longer wavelengths for deeper penetration. This technique combined with successful optical coherence tomography may pave a new way for imaging through fog, storm, and rain as well as biomedical imaging applications.
Deep imaging inside scattering media through virtual spatiotemporal wavefront shaping

Abstract:The multiple scattering of light makes materials opaque and obstructs imaging. Wavefront shaping can reverse the scattering process, but imaging with physical wavefront shaping has limitations such as requiring physical guidestars, being restricted within a small isoplanatic volume, and relying on slow wavefront updates, with some approaches only working for planar targets outside the scattering media. Here, we introduce scattering matrix tomography (SMT): measure the hyperspectral scattering matrix of the sample, use it to digitally scan a synthesized confocal spatiotemporal focus and construct a volumetric image of the sample, and then use the tomograms as virtual guidestars in a nonconvex optimization to find the pulse shape, input wavefront, and output wavefront that can compensate for aberrations and scattering. SMT combines the strengths of wavefront shaping, spatiotemporal gating, and computational adaptive optics, eliminating physical guidestars and enabling digital double-path wavefront corrections tailored for every isoplanatic volume. We demonstrate sub-micron lateral resolution and one-micron axial resolution at one millimeter beneath ex vivo mouse brain tissue and over three transport mean free paths inside an opaque colloid, where existing imaging methods all fail due to the overwhelming multiple scattering. As a noninvasive and label-free method that works both inside and outside the scattering media, SMT may be applied broadly across medical imaging, biological science, device inspection, and colloidal physics.

Full-wave simulations of tomographic optical imaging inside scattering media

Abstract:Label-free tomographic optical imaging inside scattering media is important for medical diagnosis, biological science, colloidal physics, and device inspection. An outstanding challenge is that the ground-truth structure is often unknown, so one cannot rigorously assess and compare different imaging schemes. Here we demonstrate full-wave simulations of tomographic optical imaging deep inside scattering media, which provide not only the ground truth but also the flexibility to tailor the structure and the convenience of comparing different imaging schemes in the same virtual setup with minimal cost. We model reflectance confocal microscopy, optical coherence tomography, optical coherence microscopy, interferometric synthetic aperture microscopy, and the recently proposed scattering matrix tomography for imaging nanoparticle targets embedded in a large scattering medium. The ground truth enables the identification of artifacts that would typically be mistaken as being correct while setting a rigorous and uniform standard across different methods. This work shows how full-wave simulations can fill the gaps of experiments for studying imaging inside scattering media.

Abstract:We discuss image reconstruction algorithms for diffuse optical tomography that allow utilization of extremely large data sets. Image reconstruction is performed with experimental data obtained with the use of a CCD camera-based noncontact imager. We demonstrate that more than 107 measurements can be acquired and utilized. This is two orders of magintude or more larger than the data sets which are typically used in diffuse optical imaging.

Abstract:Human ability to visualize an image is usually hindered by optical scattering. Recent extensive studies have promoted imaging technique through turbid materials to a reality where color image can be restored behind scattering media in real time. The big challenge now is to recover a 3D object in a large field of view with depth resolving ability. Here, we reveal a new physical relationship between speckles generated from objects at different planes. With a single given point spread function, 3D imaging through scattering media is achieved even beyond the depth of field (DOF). Experimental testing of standard scattering media shows that the original DOF can be extended up to 5 times and the physical mechanism is depicted. This extended 3D imaging is expected to have important applications in science, technology, bio-medical, security and defense.

Abstract:Near-infrared diffuse optical tomography imaging (DOT) suffers from a poor depth resolution due to the depth sensitivity decreases markedly in tissues. In this paper, an intelligent method, which is called layered maximum-singular-values adjustment (LMA), is proposed to compensate the decrease of sensitivity in depth dimension, and hence obtain improved depth resolution of DOT imaging. Simulations are performed with a semi-infinite model, and the simulated results for objects located in different depths demonstrate that the LMA technique can improve significantly the depth resolution of reconstructed objects. The positional errors of less than 3 mm can be obtained in the depth dimension for all depths from -1 cm to -3 cm.

Abstract:Optical diffraction tomography (ODT) is a powerful non-invasive 3D imaging technique, but its combination with broadband light sources is difficult. In this study, we introduce ultrabroadband ODT, covering over 150 nm of visible spectral bandwidth with a lateral spatial resolution of 150 nm. Our work addresses a critical experimental gap by enabling the measurement of broadband refractive index changes in 3D samples, a crucial information that is difficult to assess with existing methodologies. We present broadband, spectrally resolved ODT images of HeLa cells, obtained via pulse-shaping based Fourier transform spectroscopy. The spectral observations enabled by ultrabroadband ODT, combined with material-dependent refractive index responses, allow for precise three-dimensional identification of the nanoparticles within cellular structures. Our work represents a crucial step towards time and spectrally-resolved tomography of complex 3D structures with implications for life and materials science applications.
