Abstract:Because of the high reactivity, N-carboxyanhydride (NCA) can be initiated by the thiol group. On the contrary, N-thiocarboxyanhydride (NTA) is more stable and is able to tolerate it. Herein, we apply cysteamine as a regioselective initiator for ring-opening polymerization (ROP) of N-substituted glycine N-thiocarboxyanhydride (NNTA) to synthesize well-defined thiol-capped polypeptoids. ROPs of sarcosine NTA (Sar-NTA) and N-ethylglycine NTA (NEG-NTA) are well controlled when [M]/[I] <= 100 with high yields (>87.5%) producing polypeptoids with designable molecular weights and low polydispersity indices (<1.2). All the polypeptoid chains contain a thiol end group, which is confirmed by NMR analyses, MALDI-ToF MS spectra, and Ellman's assay. Through radical-mediated thiol-ene reaction with styrene, all the thiol chain ends are transferred to oligostyrene, revealing the convenience of further modification. Benefiting from the thiol-ene click chemistry, thiol-capped polysarcosine (PSar) and poly(N-ethylglycine) (PNEG) are promising candidates to replace PEG for their nontoxicity and biocompatibility.

Abstract:ABSTRACTN‐Substituted glycine N‐thiocarboxyanhydrides (NNTAs) are promising cyclic monomers to synthesize polypeptoids with the advantages of easier preparation and higher stability during purification and storage than N‐substituted glycine N‐carboxyanhydrides (NNCAs). NNTAs were commonly considered too stable to polymerize for their low reactivity. In this contribution, we report controlled polymerizations of N‐ethylglycine NTA (NEG‐NTA) and sarcosine NTA (Sar‐NTA) using primary amines as initiator under proper polymerization conditions. The controllability has been fully supported by 1H NMR end group analyses, MALDI‐ToF mass spectra, kinetic data, block copolymerizations by sequential monomer addition, and low polydispersities (1.14–1.17) of polypeptoids. Variation of the [NNTA]/[initiator] ratio allows well control of the molar mass, and degrees of polymerization (DPs) up to 287 can be reached for poly(N‐ethylglycine) or DPs up to 262 for polysarcosine. Understanding Acid-Promoted Polymerization of the N-Substituted Glycine N-Thiocarboxyanhydride in Polar Solvents

Abstract:Polymerization of N-substituted glycine N-thiocarboxyanhydrides (NNTAs) is a promising pathway to prepare functional polypeptoids benefiting from their tolerance to nucleophilic impurities. However, controlled NNTA polymerization is hard to achieve in amide polar solvents, including N,N-dimethylacetamide (DMAc), N,N-dimethylformamide (DMF), and N-methyl pyrrolidone (NMP), the only aprotic solvents for many biomacromolecules and polypeptoids. In the present work, we successfully achieve controlled NNTA polymerization in amide polar solvents by adding acetic acid as a promoter. The promotion is applied to the polymerization of sarcosine NTA, N-ethyl glycine NTA, and N-butyl glycine NTA. DMAc, DMF, and NMP are suitable solvents to prepare polypeptoids with designable molecular weights and low dispersities (1.06-1.21). The polysarcosines with high molecular weights are prepared up to 35.2 kg/mol. A kinetic investigation quantitatively reveals that the presence of acetic acid not only accelerates the polymerization, but also suppresses H2S-catalyzed decomposition of NNTAs by decreasing the concentration of H2S dissolved in polar solvents. Benzoic acid is also able to promote the polymerization, while trifluoroacetic acid, phosphoric acid, and phenol are not appropriate promoters. The moderate acidity of acids is essential. L-Methionine, L-tryptophan, and L-phenylalanine, which are dissolved in DMF, initiate the controlled polymerization of sarcosine-NTA in the presence of acetic acid and introduce functional end groups to polysarcosines quantitatively. In DMAc, hydrophilic vancomycin is grafted by poly(N-butyl glycine). The amphiphilic product dissolves in dichloromethane and stabilizes water-in-oil emulsion.
Direct N-substituted N-thiocarboxyanhydride Polymerization Towards Polypeptoids Bearing Unprotected Carboxyl Groups

Abstract:Synthesis of poly(α-amino acid)s bearing carboxyl groups is a critical pathway to prepare biomaterials to simulate functional proteins. The traditional approaches call for carboxyl-protected monomers to prevent degradation of monomers or wrong linkage. In this contribution, we synthesize N -carboxypentyl glycine N -thiocarboxyanhydride (CPG-NTA) and iminodiacetic acid N -thiocarboxyanhydride (IDA-NTA) without protection. Initiated by amines, CPG-NTA directly polymerizes into polyCPG bearing unprotected carboxyl groups with controlled molecular weight (2.8–9.3 kg mol −1 ) and low dispersities (1.08–1.12). Block and random copolymerizations of CPG-NTA with N -ethyl glycine N -thiocarboxyanhydride (NEG-NTA) demonstrate its versatile construction of complicated polypeptoids. On the contrary, IDA-NTA transforms amines into cyclic IDA dimer-capped species with carboxyl end group in decent yields (>89%) regio-selectively. Density functional theory calculation elucidates that IDA repeating unit is prone to cyclize to be the six-membered ring product with low ΔG . The polymer is a good adhesive reagent to various materials with adhesive strength of 33–229 kPa.
Controlled Polymerization of N-Substituted Glycine N-Thiocarboxyanhydrides Initiated by Rare Earth Borohydrides Toward Hydrophilic and Hydrophobic Polypeptoids

Abstract:N-substituted glycine N-thiocarboxyanhydrides (NTAs) are alternative monomers to prepare polypeptoids with large-scale producing potential compared to the corresponding N-carboxyanhydrides (NCAs) due to their easily synthetic approach and stability during purification and storage. Novel monomer N-butylglycine NTA (NBG-NTA) has been synthesized and well characterized for the first time. Rare earth borohydrides [RE(BH4)(3)(THF)(3), RE = Sc, Y, La, Nd, Dy, and Lit] have been first applied in the polymerization of sarcosine NTA (Sar-NTA) and NBG-NTA to achieve high molecular weight (MW) hydrophilic and hydrophobic polypeptoids. Polysarcosines (PSars), poly(N-butylglycine)s (PNBGs), and their copolymers with high yields, high MWs, and moderate MW distributions are synthesized at 60 degrees by using RE(BH4)(3)(THF)(3) initiators. MWs of polypeptoids are controlled by feed molar ratios. For instance, PSar with an absolute M-n, of 27.7 kDa (DP = 390) and PDI of 1.14 is produced successfully from Sar-NTA. Thermoresponsive random copolypeptoids poly(sarcosine-r-N-butylglycine)s [P(Sar-r-NBG)s] have reversible phase transitions (cloud point temperature) in aqueous solution and minimal cytotoxicity comparable to PEG and PSar, which is promising in various biomedical and biotechnological applications. Thermal properties of homo- and co-polypeptoids are investigated by TGA and DSC measurements.
Water Tolerated Polymerization of &ITN&IT-Substituted Glycine &ITN&IT-Thiocarboxyanhydride Initiated by Primary Amines

Abstract:N-Substituted glycine N-carboxyanhydride (NNCA) polymerization cannot tolerate H2O, because the presence of H2O leads to oligopolymerization or deterioration of NCA. In contrast, its analogue N-substituted glycine N-thiocarboxyanhydride (NNTA) is stable to H2O. In this work, by comparing H-1-NMR spectra signal of NNTA with that of internal standard (trioxane, TOX) before and after heating for 24 h in presence of water, it is shown the tolerance of NNTA to water amount was monomer-equivalent (14000 mu g/g). The polymerization of NNTA initiated by benzylamine was carried out in presence of water. H-1-NMR, MALDI-ToF-MS and SEC were used to confirm the structure of polypeptoid products. The polymerization of N-ethyl glycine NTA (NEG-NTA) initiated by benzylamine showed good tolerance to initiator-equivalent water amount (100 - 650 mu g/g). From this polymerization, high yield (> 70%) was obtained with controlled molecular weights (1600 - 7500) and low molecular weight distribution (PDI, i.e. polydispersity index: 1.22 - 1.25). Increasing amount of water (14000 mu g/g) suppressed the polymerization, resulting in low yields and low molecular weights to some extent, while PDI remained low. All polymers contained benzylamine end group according to MALDI-ToF-MS, which demonstrated that all the polymerization of NNTA followed the normal amine mechanism (NAM) regardless of different water contents. Polymerizations of sarcosine NTA (Sar-NTA) and N-butyl glycine NTA (NBG-NTA) in presence of initiator-equivalent water amount were also investigated. Polymerization of sarcosine NTA resulted in high yield (89%) with controlled molecular weight and low PDI (1.13), which showed that the polymerization of NNTA with higher activity was more tolerant to water. Furthermore, the polymerization kinetics confirmed the controllability of the resulting polymer by demonstrating that the polymerization followed pseudo-first order kinetics with regard to monomer up to high conversion (similar to 90%). In addition, molecular weights of polypeptoids exhibited linear relationship with monomer conversion while PDIs kept low. It is of great interest that well-controlled NEG-NTA polymerization was carried out in commercial THE with water content (about 190 mu g/g) without dehydration. This work demonstrated that the polymerization of NNTA was a robust approach to prepare polypeptoids since both the monomer synthesis and the polymerizations did not require an anhydrous environment, which would benefit the further application of polypeptoids in bioengineering and medical field.
Hydroxyl Group Tolerated Polymerization of N-Substituted Glycine N-Thiocarboxyanhydride Mediated by Aminoalcohols: A Simple Way to Α-Hydroxyl-ω-aminotelechelic Polypeptoids

Abstract:N-Carboxyanhydride (NCA) polymerization cannot tolerate nucleophilic groups that have the ability of initiation, e.g., hydroxyl group. In contrast, N-thiocarboxyanhydride (NTA) is a much more stable monomer to tolerate them. In this contribution, we investigate aminoalcohols including 2-amino-1-ethanol (AE), 3-amino-1-propanol (AP), 4-aminomethylbenzyl alcohol (AMB), 6-amino-1-hexanol (AH), and 12-amino-1-dodecanol (AD) as initiators for ring-opening polymerization of N substituted glycine N-thiocarboxyanhydride (NNTA) to prepare alpha-hydroxyl-omega-aminotelechelic water-soluble polypeptoids. Hydroxyl groups of AE, AP, and AMB are activated by hydrogen bonding with amino groups, which results in a mixture of alpha,omega-diaminotelechelic and alpha-hydroxyl-omega-aminotelechelic polypeptoids confirmed by H-1 NMR, MALDI-ToF, and SEC measurements. Pure alpha-hydroxyl-omega-aminotelechelic polypeptoids are synthesized for the first time initiated by AEI and AD with controlled molecular weights (1.3-12.4 kg/mol) and low polydispersity indices (<1.30). Hydroxyl groups in AH and AD remain inactive to generate hydrogen bonding due to the long distance from amino groups. Water-soluble polypeptoids with special functional end groups are attractive alternatives of PEG for their nontoxicity and biocompatibility having great potential in biomedical applications.
Identifying the Hydrolysis of Carbonyl Sulfide as a Side Reaction Impeding the Polymerization of N-Substituted Glycine N-Thiocarboxyanhydride

Abstract:Polypeptoids are noticeable biological materials due to their versatile properties and various applications in drug delivery, surface modification, self-assembly, etc. N-Substituted glycine N-thiocarboxyanhydrides (NNTAs) are more stable monomers than the corresponding N-carboxyanhydrides (NNCAs) and enable one to prepare polypeptoids via ring-opening polymerization even in the presence of water. However, larger amounts of water (>10,000 ppm) cause inhibition of the polymerization. Herein, we discover that during polymerization hydrogen sulfide evolves from the hydrolysis of carbonyl sulfide, which is the byproduct of ring-opening reaction, and reacts with NNTA to produce cyclic oligopeptoids. The capture of N-ethylethanethioic acid as an intermediate product confirms the reaction mechanism together with density functional theory quantum computational results. By bubbling the polymerization solution with argon, the side reaction can be suppressed to allow the synthesis of polysarcosine with high molar mass ( Mn = 11,200 g/mol, Đ = 1.25) even in the presence of ∼10,000 ppm of water.

Abstract:In this work, we develop new glycine‐derived polymers that exhibit thermoresponsive properties in water. Therefore, a series of monomers containing one, two or three amide functional groups and one terminal cyanomethyl group is synthesized. The resulting homopolymers, obtained by free radical polymerization (FRP) and reversible addition fragmentation chain transfer (RAFT) polymerization, display a sharp and reversible upper critical solution temperature (UCST)‐type phase transition in water. Additionally, we show that the cloud point (TCP) can be adjusted over more than 60 °C by the number of glycyl groups present in the monomer structure and by the polymer's molar mass. These novel thermoresponsive polymers based on cyanomethylglycinamide enrich the range of non‐ionic UCST polymers and are promising to find applications in various fields. Abstract:A series of novel amphiphilic alternating CPEG copolymers were synthesized through an amine–epoxy click reaction comprising aliphatic amine and polyethylene glycol diglycidyl ether (PEGDE). These polymers were characterized in detail via nuclear magnetic resonance (NMR), gel permeation chromatography (GPC), Fourier-transform infrared spectroscopy (FTIR), and thermogravimetric analysis (TGA) to confirm the successful synthesis. Due to their amphiphilic structure, these polymers display thermoresponsiveness, with tunable cloud points (Tcps) that are adjustable from 20.8 °C to 46.8 °C by altering the side-chain length of the aliphatic amine, varying the mixing ratios of copolymers, the solution's pH, and salt additions. This tunable thermoresponsive behavior positions CPEG copolymers as promising candidates for a range of functional material applications.

Abstract:Thermoresponsive water-soluble polymers are of great importance since they typically show a lower critical solution temperature (LCST) in aqueous media. In this research, the LCST change in broad temperature ranges of copolymers composed of natural glycyrrhetinic acid (GA)-based methacrylate and N,N'-dimethylacrylamides (DMAs) was investigated as a function of the concentration and the content of GA pendants. By complexation of GA pendants with β-cyclodextrin (β-CD), a side-chain polypseudorotaxane was obtained, which exhibited a significant increase in the LCST of copolymers. Moreover, the precisely reversible control of the LCST behavior was realized through adding a competing guest molecule, sodium 1-admantylcarboxylate. This work illustrates a simple and effective approach to endow water-soluble polymers with broad temperature tunability and helps us further understand the effect of a biocompatible host-guest complementary β-CD/GA pair on the thermoresponsive process.

Abstract:The fascinating functions of proteins and peptides in biological systems have attracted intense interest to explore their mimics using polymers, including polypeptides synthesized from polymerization. The folding, structures and functions of proteins and polypeptides are largely dependent on their sequence. However, sequence-tunable polymerization for polypeptide synthesis is a long-lasting challenge. The application of polypeptides is also greatly hindered by their susceptibility to enzymatic degradation. Although poly-α/β-peptide has proven to be an effective strategy to address the stability issue, the synthesis of poly-α/β-peptide from polymerization is not available yet. Hereby, we demonstrate a living and controlled copolymerization on α-NCA and β-NTA to prepare sequence-tunable poly-α/β-peptides. This polymerization strategy shows a prominent solvent-driven characteristic, providing random-like copolymers of poly-α/β-peptides in THF and block-like copolymers of poly-α/β-peptides in a mixed solvent of CHCl3/H2O (95/5, v/v), and opens new avenues for sequence-tunable polymerization and enables facile synthesis of proteolysis tunable poly-α/β-peptides for diverse applications.

Abstract:Poly(α-amino acid)s, containing amino acid repeat units, are remarkable biomaterials with similar structures to protein, which are mainly synthesized by solid phase peptide synthesis, ring opening polymerization of N-carboxyanhydrides, polymerization of activated urethane derivatives of α-amino acids and so on. α-Amino acid N-thiocarboxyanhydride (NTA) and N-substituted glycine N-thiocarboxyanhydride (NNTA) have recently emerged as a subject of scientific interest due to their easy synthesis and purification in open air, long shelf-life and well controlled polymerization to produce poly(α-amino acid)s. In this feature article, we review the history and the recent development on the design of NTA monomers, and summarize the recent advances in poly(α-amino acid)s synthesis through ring-opening polymerization of NTA and NNTA by using different initiators and reaction mediums. We also highlight the applications of poly(α-amino acid) materials resulting from NTA polymerization.

Abstract:In this study, a new pH-tunable thermoresponsive hydroxyl-terminated hyperbranched polyether (HTHP 2) was successfully prepared via a one-pot cationic polymerization technique and postmodification. In the first step, hydroxyl-terminated hyperbranched polyether containing double bonds (HTHP 1) were synthesized. Then, through thiol-ene click reaction, pH-responsive carboxyl groups were introduced to the target polymer of HTHP 2. The products were characterized via Fourier-transform infrared spectra (FTIR), nuclear magnetic resonance (NMR), and size-exclusion chromatography-multiangle laser light scattering (SEC-MALLS). Moreover, dynamic light scattering (DLS) and UV-Vis spectroscopy was employed to study the pH- and thermoresponsiveness in detail. Results showed that HTHP 2 possessed typical pH-controllable thermoresponsive behavior. By regulating the solution pH value range 3.0-5.4, LCST of HTHP 2 could be changed from 12.8 to 68.0 degrees C. Meanwhile, the cell viabilities of A549 cells were more than 80% for in vitro cytotoxicity tests of HTHP 2, suggested that HTHP 2 polymers are of good biocompatibility for up to 24 h.

The convenient monomer synthesis without rigorous anhydrous requirements makes the ring‐opening polymerization of NTA a competitive alternative to prepare polypeptoid‐containing materials with potential of large‐scale production. Polysarcosines (PSars) with high yields (>90%) and low polydispersities (<1.2) are synthesized from sarcosine N‐thiocarboxyanhydride (Sar‐NTA) at 60 °C initiated by primary amines including poly(ethylene glycol) amine (PEG–NH2). The lengths of PSar segments are controlled by various feed ratios of Sar‐NTA to initiator. PEG‐b‐PSar products, a class of novel double‐hydrophilic diblock copolymers, are effective in stabilizing oil‐in‐water emulsions at nano‐ and microscale, which demonstrates promising encapsulation applications in food, cosmetics, and drug delivery. Due to the different solubility of PEG and PSar blocks, PEG‐b‐PSar copolymers form micelles in organic solvents with the capability to incorporate metal cations including Cu2+ and Ni2+. image
Block Polypeptoids: Synthesis, Characterization, and Response Toward Irradiation with UV Light and Temperature

Abstract:Stimuli-responsive polymers have received considerable attention due to their ability to change properties in response to environmental changes. Polypeptoids are biocompatible and candidates for biomedical applications such as in controlled drug release and tissue engineering. In this work, we synthesized UV-sensitive diblock polypeptoids poly(sarcosine-r-butylglycine)-b-PNB (P(Sar-r-NBG)-b-PNB) and triblock polypeptoids polysarcosine-b-polybutylglycine-b-PNB (PSar-b-PNBG-b-PNB). Exposure to 254 nm UV light triggers the cleavage of nitrobenzyl groups leading to an increase in hydrophilicity, which is confirmed by UV/vis, H-1 NMR, and dynamic light scattering (DLS) analysis. Consequently, micelles formed by these block polypeptoids can undergo UV-trigged release of encapsulated cargo, herein exemplified using Nile red or pyrene as model payload. Furthermore, the diblock polypeptoids containing random P(Sar-r-NBG) segment exhibit thermoresponsive behavior, which is investigated by optical transmittance and DLS, and their cloud point temperatures (T(cp)s) can be adjusted by the feed molar ratio of Sar-NTA and NBG-NTA monomers. Furthermore, the block polypeptoids are able to self-assemble in water and show morphological changes from spherical nanoparticles to linear aggregates of several millimeters in length when the concentration increases. CCK-8 cell viability assays prove that the block polypeptoids' micellar aqueous solution is noncytotoxic before and after UV irradiation. Such novel biocompatible block polypeptoids with dual-responsive properties and varying morphology are of interest for applications in the field of biomedicine.

Abstract:Poly(beta-peptoid) is a class of polypeptide mimics that possesses excellent biocompatibility and resistance to proteolysis. However, the synthesis of poly(beta-peptoid) s with functionalities is a long-standing challenge that greatly hinders the functional study and application of poly(beta-peptoid)s. We report a controllable and easy synthesis of poly(beta-peptoid)s bearing diverse functionalities via the ring-opening polymerization on N-substituted beta-alanine N-thiocarboxyanhydrides (beta-NNTAs). The polymerization can be carried out in open vessels under mild conditions using amines as the initiators to provide poly(beta-peptoid)s with targeted molecular weights, narrow dispersities, and diverse functionalities in the side chains and termini. The beta-NNTAs polymerization is even compatible with initiators bearing unprotected hydroxyl groups. The amphiphilic/cationic poly(beta-peptoid)s exhibit a broad spectrum and potent antibacterial activities against multidrug-resistant bacteria. In addition, the highly favored stability of beta-NNTA monomers for purification and storage highlights the advantages of this beta-NNTA polymerization strategy for poly(beta-peptoid)s synthesis, functional study, and application as a synthetic mimic of polypeptides. A New Thermoresponsive Polymer of Poly(n-Acryloylsarcosine Methyl Ester) with a Tunable LCST

Abstract:Poly(N-acryloylsarcosine methyl ester) (PNASME), a thermoresponsive N-ester substituted polyacrylamide that has a well-defined molecular weight and a low Đ, was synthesized by solution RAFT polymerization. The synthesized PNASME exhibited a cloud point temperature (Tcp) or lower critical solution temperature (LCST) of 44.0 °C. It was found that various factors could affect the Tcp of PNASME, e.g., the polymer molecular weight, the polymer concentration, the salt effect on Tcp following the order of the Hofmeister series, urea induced increasing Tcp, and phenol induced decreasing Tcp. The mechanism of urea and phenol inducing the phase transition of PNASME was revealed by 2D NOESY spectroscopy, and how the hydrogen bonding between PNASME and the solutes of urea and phenol affected the phase transition of PNASME was clarified. The contrary effect on the Tcp of PNASME induced by urea and phenol makes it easy to adjust the thermoresponse of PNASME, and the tunable thermoresponse of the diblock copolymer of poly(N-acryloylsarcosine methyl ester)-block-poly(N-isopropylacrylamide) was demonstrated. The present study is believed to offer a new route to tune the solution behavior of thermoresponsive polymers.

Abstract:Biocompatible amphiphilic block copolymers composed of polysarcosine (PSar) and poly(e-caprolactone) (PCL) were synthesized using ring-opening polymerization of sarcosine N-thiocarboxyanhydride initiated by oxyamine-ended PCL and characterized by NMR, SEC, and DSC. Self-assembling of two triblock copolymers PSar(8)-b-PCL28-b-PSar(8) (CS7) and PSar(16)-b-PCL40-b-PSar(16) (CS10) in dilute solution was studied in detail toward polymersome formation using thin-film hydration and nano-precipitation techniques. A few giant vesicles were obtained by thin-film hydration from both copolymers and visualized by confocal laser scanning microscope. Unilamellar sheets and nanofibers (with 8-10 nm thickness or diameter) were obtained by nanoprecipitation at room temperature and observed by Cryo-TEM. These lamellae and fibrous structures were transformed into worm-like cylinders and spheres (D similar to 30-100 nm) after heating to 65 degrees C (>T-m,T-PCL). Heating CS10 suspensions to 90 degrees C led eventually to multilamellar polymersomes (D similar to 100-500 nm). Mechanism II, where micelles expand to vesicles through water diffusion and hydrophilic core forming, was proposed for polymersome formation. A cell viability test confirmed the self-assemblies were not cytotoxic.
