Modification of Polycaprolactone Microspheres-Aggregated Scaffolds with Galactosylated Chitosan for Modulating the Differentiation of Bone Mesenchymal Stem Cells

Abstract:The solid matrices are highly demanded for adhesion and preservation of functions of hepatocytes in a bioartificial liver (BAL) system. Herein, polycaprolactone (PCL) microspheres-aggregated scaffold with defined topography was modified by galatosylated chitosan (GC) for controlling the differentiation of bone mesenchymal stem cells (BMSCs) toward hepatocytes. By being hydrolyzed in 1 M NaOH, carboxyl groups with a density of 2.67 mmol/mg were introduced on the surface of the PCL microspheres-aggregated scaffold, which were used to covalently couple the GC molecules. Most of the immobilized GC was remained on the scaffold after incubation in PBS for 168 h. In vitro culture of BMSCs showed that they formed a more spherical morphology with higher viability and a level of albumin secretion on the GC-modified scaffolds than those on the blank control scaffolds.
